<P>Graphene oxide (GO) is a nanomaterial with newly developing biological applications. Autophagy is an intracellular degradation system that has been associated with the progression of neurodegenerative disorders. Although induction of autophagic flux by GO has been reported, the underlying signaling pathway in neurodegenerative disorders and how this is involved in neuroprotection remain obscure. We show that GO itself activates autophagic flux in neuronal cells and confers a neuroprotective effect against prion protein (PrP) (106–126)-mediated neurotoxicity. GO can be detected in SK-N-SH neuronal cells, where it triggers autophagic flux signaling. GO-induced autophagic flux prevented PrP (106–126)-induced neurotoxicity in SK-N-SH cells. Moreover, inactivation of autophagic flux blocked GO-induced neuroprotection against prion-mediated mitochondrial neurotoxicity. This is the first study to demonstrate that GO regulates autophagic flux in neuronal cells, and that activation of autophagic flux signals, induced by GO, plays a neuroprotective role against prion-mediated mitochondrial neurotoxicity. These results suggest that the nanomaterial GO may be used to activate autophagic flux and could be used in neuroprotective strategies for treatment of neurodegenerative disorders, including prion diseases.</P>

근골격계 조직의 병변을 정확하게 진단하기 위해서는 골 조직의 이상뿐 아니라, 다양한 연부조직의 상태를 평가하는 것이 중요하다. 연부조직의 상태를 평가하기 위해서 자기공명영상이 널리 사용되어 왔으나 검사비용이 비싸고, 실시간 검사가 불가능한 단점이 있다. 최근에는 근골격계 조직의 평가를 위해 초음파 검사가 사용되고 있고, 점차 유용성이 증가하고 있다. 초음파 영상은 조직 고유의 음향저항 등에 따라 음영이 구현된다. 초음파가 잘 반사되는 뼈, 석회, 인대, 건 등은 고음영으로, 반사가 덜 일어나는 근육, 신경 등은 저음영으로 표현된다. In order to accurately diagnose lesions of musculoskeletal tissue, evaluation not only of the abnormality of the bone, but the condition of soft tissue is important. Magnetic resonance imaging has been widely used in evaluation of the state of soft tissue, however, it has the disadvantage that testing is expensive and real-time scanning is not possible. In recent years, ultrasonography has been used for evaluation of musculoskeletal tissue and its usefulness has shown a gradual increase. The ultrasound image is determined by the tissue specific acoustic impedance and other factors. Highly reflective tissues such as bone, calcification, ligament, and tendon are expressed as hyperechoic images, and less reflective tissues such as muscle and nerve are expressed as hypoechoic images.
고령 환자의 대퇴 경부 골절에 시행한 양극성 고관절 반치환술

목적: 65세 이상 노년층의 대퇴경부 골절에서 양극성 반치환술의 임상적 결과를 분석하고자 하였다. 대상 및 방법: 대퇴경부 골절로 양극성 반치환술을 시행한 65세 이상의 환자 중 3년 이상 추시가 가능하였던 43명 (46예)을 대상으로 하였다. 임상적 평가는 Harris 고관절 점수를 측정하였고 동통지수, 지지지수 등을 분석하였으며, 방사선학적 평가로는 대퇴스템 해리와 비구컵의 전이 등을 관찰하였다. 결과: Harris 고관절 점수는 평균 88.7점 (62~96점)이었고 임상적 평가는 우수 34예, 양호 7예, 보통 3예, 불량 2예였다. 동통지수는 평균 39.6점 (20~44점)이었고 동통이 없었던 경우가 20예, 경미한 동통 18예, 경한 동통 6예, 중등도 동통 2예였다. 지지지수는 평균 9.6점 (4~11점)이었고, 32예에서 단장이나 목발 등 도구 없이 보행이 가능하였다. 대퇴스템의 무균성 해리 또는 양극성 비구컵의 중심부 전이로 인하여 2예 (4%)에서 재치환술을 시행하였다. 결론: 노년층의 대퇴경부 골절에서 양극성 반치환술은 조기 보행 및 기능회복을 위해 권장할 만한 치료 방법이라 판단된다. Purpose: To evaluate the clinical results of bipolar hemiarthroplasty in elderly patients more than 65 years of age with a femoral neck fracture. Materials and Methods: Forty-six bipolar hemiarthroplasties in 43 patients more than 65 years of age which could be followed more than 3 years were included in this study. The clinical outcomes were evaluated using Harris hip score, pain score and support score. The radiological results were analyzed by femoral stem loosening and bipolar cup migration. Results: The average Harris hip score was 88.7 (62~96) points. An excellent score was recorded in 34 cases, good in 7 cases, fair in 3 cases and poor in 2 cases. The average pain score was 39.3 points and there were no pain in 20 cases, slight pain in 17 cases, mild pain in 6 cases and moderate pain in 2 cases. The average support score was 9.6 points and 32 patients could walk without the use of any assistive devices. Two cases were converted to total hip arthroplasty due to femoral stem loosening with or without bipolar cup migration. Conclusion: For the early ambulation and functional recovery of elderly patients with femoral neck fracture, bipolar hemiarthroplasty was considered as one of recommendable methods.

The purpose of this study was to investigate the alternative splicing in equine cordon-bleu WH2 repeat protein-like 1 (COBLL1) gene that was identified in horse muscle and blood leukocytes, and to predict functional consequences of alternative splicing by bioinformatics analysis. In a previous study, RNA-seq analysis predicted the presence of alternative spliced isoforms of equine COBLL1, namely COBLL1a as a long form and COBLL1b as a short form. In this study, we validated two isoforms of COBLL1 transcripts in horse tissues by the real-time polymerase chain reaction, and cloned them for Sanger sequencing. The sequencing results showed that the alternative splicing occurs at exon 9. Prediction of protein structure of these isoforms revealed three putative phosphorylation sites at the amino acid sequences encoded in exon 9, which is deleted in COBLL1b. In expression analysis, it was found that COBLL1b was expressed ubiquitously and equivalently in all the analyzed tissues, whereas COBLL1a showed strong expression in kidney, spinal cord and lung, moderate expression in heart and skeletal muscle, and low expression in thyroid and colon. In muscle, both COBLL1a and COBLL1b expression decreased after exercise. It is assumed that the regulation of COBLL1 expression may be important for regulating glucose level or switching of energy source, possibly through an insulin signaling pathway, in muscle after exercise. Further study is warranted to reveal the functional importance of COBLL1 on athletic performance in race horses.
회전근 개 질환에서 초음파, 자기공명영상, 관절경 소견의 연관성

초음파 검사는 다양한 견관절 질환을 검사하는 강력하고 유용한 방법이다. 최근 들어 고해상도 변환기의 사용과 술기의 발전으로 회전근 개 질환을 진단하는 정확성이 향상되고 있다. 하지만, 시술자의 숙련도에 따라 다양한 결과를 나타내는 단점이 있어 주의를 요한다. 본 강좌에서는 회전근 개 질환의 초음파 소견을 자기 공명 영상, 관절경 소견과 비교하여 초음파 영상의 해석에 도움을 주고자 하였다. Ultrasonography is a powerful and useful method for the examination of the various shoulder diseases. The use of high-resolution transducer and technical evolution allowed the improvement of the accuracy of detection of the rotator cuff disease. However, its limitation is that there is marked disparity in the interpretation according to the operators' experience. This article describes the ultrasonographic findings of rotator cuff pathology compared to the MR (magnetic resonance) imaging and arthroscopic findings for helping the interpretation of ultrasonographic picture.
