      • 서브 - 링의 신드롬 분석을 이용한 하이퍼큐브 진단 알고리즘

        하이퍼큐브의 정규직이며 계층적인 구조적 특성은 효율적인 진단 알고리즘 개발에 유리하게 적용될 수 있다. Feng et al.의 HADA/IHADA와 Choi와 Rhee의 적응적 큐브 분할 방법은 하이퍼큐브의 전체 노드를 하나의 링으로 임베딩하여 링의 진단 특성을 이용하여 위하여 분할 및 정복 방법을 이용하였다. 또한 Kranakis와 Pelc는 결함을 모두 포함하는 최소의 서브링을 하나의 노드로 하는 하이퍼큐브의 형태로 분할하는 HYP-DIAG 알고리즘을 제안하였다. 또한 최악의 경우에, 테스트 수만을 고려하여 2ⁿ+3n/2의 테스트 수를 갖는 FAST-HYP-DIAG 알고리즘과 병렬 시간만을 고려하여 많아야 11테스트 라운드 이내에 진단을 수행하는 EXPRESS-HYP-DIAG 알고리즘을 제안하였다. 본 논문에서는 HYP-DIAG의 첫 번째 단계에서 얻어진 서브링들의 신드롬을 분석하여 테스트의 수와 테스트 라운드를 모두 고려하는 알고리즘을 제안한다.

        한방 닭죽 개발 및 최적 배합비율 확립

        죽의 기능성 향상을 위하여 2:1:1:1의 배합으로 혼합한 한약재료(가시오가피, 숙지황, 산약, 백복령)와 닭가슴살 등의 재료를 사용해 한방 영양죽을 제조하였다. 한방 닭죽의 DPPH radical 소거능, SOD 유사활성, 맛의 선호도, cost는 수학적인 canonical model과 trace plot을 이용하여 분석한 결과, SOD 유사활성 이외에는 각 성분들이 독립적으로 작용하여 상호작용이 없는 선형 모델을 보여주었다. DPPH radical 소거능, SOD 유사활성, 맛의 선호도를 최대로, cost를 최소로 목적 범위를 설정하고, 각각의 canonical 계수를 이용하여 수적 최적화를 통하여 최적 배합비를 구한 결과, 한약 혼합물 101g, 닭가슴살 38.66g, 찹쌀 비율 41.34g의 최적 배합비를 구하였으며, 이 배합비에 따른 맛의 선호도는 6.39, DPPH radical 소거능은 17.25㎎VCE/ℓ, SOD 유사활성은 2.25%, cost는 1,382.39 won으로 나타났으며, 여러 결과들 사이에 최적화된 한 값에 수렴하는 desirability는 0.714로 나타났다. 이는 한약혼합물이 많을수록 기능성 면에서는 높게 나타나지만 맛의 선호도면에서는 낮게 나타냈고, 닭가슴살의 경우에도 그 함량이 많을수록 맛의 선호도는 올라갔지만 그에 반해 cost가 높게 올라간 것을 반영한 결과로 사료된다. The objectives of this study were to develop porridge with medicinal herbs and chicken breast meat and to find out the optimum mixing ratio. Several herb extracts such as Acanthopanacis senticosus, Rehmannia glutinosa, Disocorea japonica, and Poria cocos Wolf were used in the ratio of 2, 1, 1, and 1, respectively. The optimum processing condition for herbal chicken porridge was determined by a design expert program. Seventeen experimental points were selected, and herb extracts (82∼101g), chicken breast meat (30∼50g), and glutinous rice (40∼60g) were chosen as the independent variables. The measured responses were preference of taste, DPPH radical scavenging, SOD-like activity, and cost. The optimum formulation of herbal chicken porridge using the numerical analysis was set at herbal extracts (101g), chicken breast meat (38.66g), and glutinous rice (41.34g) with a 0.714 desirability value. DPPH radical scavenging effect, preference of taste and the cost showed a linear model, whereas SOD-like activity showed a quadratic model indicating a higher interaction among the mixture. As a result of proximate composition of optimized herbal chicken porridge, the contents of moisture, carbohydrate, crude protein, crude lipid, and ash contents were 76.4, 9.0, 1.4, 0.5, and 1.6%, the calories of the porridge was 46.1kcal/100g.

        Metabolic Features of Coffee Beans Depending on Planted Areas

        Coffee is one of the top selling products in the world. There are various coffee bean species around the world. Among them, Coffea Arabica is the most popular species. However, there are few studies on the metabolites of coffee beans so far. This study demonstrates effects of the planted regions on the metabolite concentrations of coffee beans. The metabolites of coffee beans can be affected by growing area even although same species are grown. Accordingly, we studied coffee bean metabolites extracted from the same species in different regions (The brand names, Kona from Hawaii, Mocha Matari from Yemen, and Blue Mountain from Jamaica) by using mixed solvent of methanol: water: chloroform. A comparative analysis by NMR spectroscopy was performed and the statistical techniques were used to figure out the differences. As a result, we found that chlorogenic acid, caffeine, citrate, and sucrose mainly contributed to the separation of the three groups. When compared with Kona and Blue Mountain, concentrations of chlorogenic acid, caffeine, and sucrose in Mocha Matari were observed to be relatively down-regulated. In addition, compared with the two other groups, concentration of citrate in Kona was observed to be up-regulated.

        Tumor Necrosis Factor-producing T-regulatory Cells Are Associated With Severe Liver Injury in Patients With Acute Hepatitis A

        <P><B>Background and Aims</B></P> <P>CD4<SUP>+</SUP>CD25<SUP>+</SUP>Foxp3<SUP>+</SUP> T-regulatory (Treg) cells control immune responses and maintain immune homeostasis. However, under inflammatory conditions, Treg cells produce cytokines that promote inflammation. We investigated production of tumor necrosis factor (TNF) by Treg cells in patients with acute hepatitis A (AHA), and examined the characteristics of these cells and association with clinical factors.</P> <P><B>Methods</B></P> <P>We analyzed blood samples collected from 63 patients with AHA at the time of hospitalization (and some at later time points) and 19 healthy donors in South Korea. Liver tissues were collected from patients with fulminant AHA during liver transplantation. Peripheral blood mononuclear cells were isolated from whole blood and lymphocytes were isolated from liver tissues and analyzed by flow cytometry. Cytokine production from Treg cells (CD4<SUP>+</SUP>CD25<SUP>+</SUP>Foxp3<SUP>+</SUP>) was measured by immunofluorescence levels following stimulation with anti-CD3 and anti-CD28. Epigenetic stability of Treg cells was determined based on DNA methylation patterns. Phenotypes of Treg cells were analyzed by flow cytometry and an RORγt inhibitor, ML-209, was used to inhibit TNF production. Treg cell suppression assay was performed by co-culture of Treg-depleted peripheral blood mononuclear cells s and isolated Treg cells.</P> <P><B>Results</B></P> <P>A higher proportion of CD4<SUP>+</SUP>CD25<SUP>+</SUP>Foxp3<SUP>+</SUP> Treg cells from patients with AHA compared with controls produced TNF upon stimulation with anti-CD3 and anti-CD28 (11.2% vs 2.8%). DNA methylation analysis confirmed the identity of the Treg cells. TNF-producing Treg cells had features of T-helper 17 cells, including up-regulation of RORγt, which was required for TNF production. The Treg cells had reduced suppressive functions compared with Treg cells from controls. The frequency of TNF-producing Treg cells in AHA patients’ blood correlated with their serum level of alanine aminotransferase.</P> <P><B>Conclusions</B></P> <P>Treg cells from patients with AHA have altered functions compared with Treg cells from healthy individuals. Treg cells from patients with AHA produce higher levels of TNF, gain features of T-helper 17 cells, and have reduced suppressive activity.         <P>Cell transplantation using hepatocytes derived from stem cells has been regarded as a possible alternative treatment for various hepatic disorders. Recently, mesenchymal stem cells (MSCs) from the bone marrow have shown the potential to differentiate into hepatocytes in in vitro and in vivo conditions. Noninvasive imaging techniques allowing in vivo assessment of the location of cells are of great value for experimental studies in which these cells are transplanted. We labeled human mesenchymal stem cells (hMSCs) with green fluorescence protein (GFP) and superparamagnetic iron oxide (SPIO) using a transfection agent (GenePORTER). Cellular labeling was evaluated with magnetic resonance (MR) imaging of labeled suspensions, and Prussian blue staining for iron assessment. hMSCs labeled with SPIO and GFP were injected into the portal veins of immunosuppressed, hepatic-damaged rats. MR imaging findings were compared histologically. To identify the differentiation of hMSCs into hepatocytes and to trace the hepatocytes with molecular imaging, we observed the potential of SPIO and GFP double-labeled hMSCs to differentiate into hepatocyte-like cells in the regenerating rat liver. Serial MR imaging showed the possible detection of transplanted cells in the early period of transplantation. Our results indicate that magnetic labeling of hMSCs with SPIO may enable cellular MR imaging and tracking in experimental in vivo settings.</P>

        Characterization of Indium Hydroxide Powders Synthesized by a Precipitation Method

        Indium hydroxide powders were synthesized using indium nitride, and the characteristics of the indium hydroxide powder were investigated according to the precipitate aging time and the pH and concentration of the liquid synthesis medium. Indium hydroxide (In(OH)3) and indium oxide hydroxide (InOOH) phases co-existed in all conditions studied here. The crystallite size of In(OH)3 was more dependent on the concentration and pH of the reaction medium compared to that of InOOH. Hence, the relative crystallite scale of In(OH)3 increased with increasing concentration and pH of the reaction liquid more than that of in InOOH. Although the crystallite size did not change much with an increase of precipitate aging time, the specific surface area decreased greatly. Such results can aid in the synthesis of indium oxide particles that are used for producing ITO targets.

        Surgical Management of Acute Cerebellar Infarction

        Objective : The aim of this study is to determine which patients with progressively deteriorating acute cerebellar infarction would benefit from surgical treatment and which surgical procedure would best benefit them. Methods : Seventy six patients were treated at our hospital for cerebellar infarction over the past 3 years. Sixty nine patients received conservative management in the neurological department of our hospital. Among them, 7 patients [5 males and 2 females; average age, 49 yrs] were referred to neurosurgical department because of mental deterioration and underwent emergency surgery. Five patients underwent external ventricular drainage with suboccipital craniectomy and two patients underwent suboccipital craniectomy alone. Results : Of the 7 surgically treated patients, 4 patients experienced good recovery and 2 patients experienced moderate disability [disabled but independent] and 1 patient experienced severe disability [conscious but disabled]. There was no death. Conclusion : In patients conservatively treated for cerebellar infarction and showing mental deterioration and radiologically evident brainstem compression and ventricular enlargement, we strongly recommend suboccipital craniectomy [plus optional external ventricular drainage in case of showing hydrocephalus] as a first treatment option.
