박과류 관련 종자전염 검역바이러스 3종의 PCR 진단시스템 개발

박과는 스쿼시, 호박, 애호박, 조롱박, 오이 및 수박 등 100여 속이 넘는 속이 포함된 식물과이며, 종자의 수입량이 매우 많은 작물이다. 이들이 우리나라로 수입될 때, 규제바이러스인 Squash mosaic virus (SqMV), Cucumber green mottle mosaic virus (CGMMV) 및Kyuri green mottle mosaic virus (KGMMV)에 대하여 검역을 수행한다. 본 연구에서는 SqMV, CGMMV 및 KGMMV를 신속·높은 감도로 검출할 수 있는 특이적 RT-PCR 및 nested PCR 프라이머 조합과 유전자변형-양성대조구 플라스미드를 개발하였다. 또한 본 연구에서 개발한 진단시스템을 2010년부터 2014년 상반기까지 현장적용하여, SqMV 47건, CGMMV 67건 및 KGMMV 17건을 검출하였다. The Cucurbitaceae are a plant family that consist of over a hundred genera, the most important of which are squash, pumpkin, zucchini, some gourds, cucumber and watermelon. These are among the top imported seeds in Korea. At the time of their import, the Squash mosaic virus (SqMV), the Cucumber green mottle mosaic virus (CGMMV) and the Kyuri green mottle mosaic virus (KGMMV) are designated as regulated viruses for quarantine in Korea. This study was conducted to develop specific primer sets for easy and rapid detection of SqMV, CGMMV and KGMMV. RT-PCR with the nested PCR primer sets and modified positive control plasmids were capable of highly sensitive detection and verification of such viruses. In addition, developed diagnostic PCR systems applied to quarantine sites detected 47 cases of SqMV, 67 cases of CGMMV and 17 cases of KGMMV between 2010 and the first half of 2014.
Recovery and Adsorption Rate of Murine Norovirus Using NanoCeram^® Filters

This study investigated the recovery and absorption rates of murine norovirus, a surrogate for human norovirus, by using NanoCeram^® filters which served as a tool for recovering viruses. In the study, two types of NanoCeram^® filters were employed: one was a cartridge type and the other was a disc type (Φ 47 mm) whose surface area is 75 times smaller than the cartridge type. The analytical method was the real-time reverse transcription-polymerase chain reaction (RT-PCR). The study found that the average recovery rates of the cartridge type and the disc type were 30.9% and 29.5% respectively. Since these two rates were very close to each other, the adsorption rate of the cartridge type could be predicted with the disc type. Analyzing recovery and absorption rates of the disc type based on different filtered volumes showed that when the volume increased from 0.5 L to 20 L, the average recovery rate rose from 14.78% to 30.41 %, while the average absorption rate dropped from 56.33% to 10.48%. The increase in turbidity from less than 1NTU to less than 3 NTU raised the average recovery rate from 47.23% to 82.84%.

팔당호 주변에서 자생하는 식물체를 이용하여 실험실내에서 배양된 남조류에 투여하여 조류증식 억제 효율을 조사하였다. 조류 성장 억제 효과에 있어 육상식물을 이용하는 경우 조류 밀도가 10^4cells/ml 범위에서의 0.5g/l 투여량은 전혀 효과를 나타내지 못했던 반면, 2.5g/l 투여량에서는 95% 이상의 효과를 보였다. 조류 밀도가 10^6cells/ml범위에서의 육상식물 투여량은 2.5g/l 이상이 필요했다. 조류성장 억제에 육상식물은 잣잎, 수생식물은 애기마름이 큰 효과를 보였다. 애기마름 투여조는 조류 밀도가 10^6cella/ml범위에서도 2.5g/l 투여량으로 50%의 억제 효율이 나타났다. 식물을 이용한 조류 증식 억제 효과는 식물 투여 후 7일째 후 가장 효과가 좋은 것으로 나타났다. 습체, 건체, 추출물 비교시 추출물이 가장 효과가 좋은 것으로 육상식물 투여조의 경우 1.25g/l 투여량에 비해 2.5g/l 투여량에서 현저한 효과를 보였고, 애기마름은 1.25g/l 투여량에서도 99%의 효과를 보였다. 건체식물은 습체식물에 비해 초기에도 높은 효과를 보였다. 조류의 비증식속도로 보았을 때도 식물체 추출물을 투여했을 때 가장 효과가 좋았고 다음으로 건체, 습체식물 투여순으로 조류 증식억제 효과를 나타내었다.

Physicochemical properties of oil in water microemulsions containing soybean oil and egg phosphatide were observed for 3 weeks under the storage condition of 4℃ refrigerator. Changes in major fatty acid content, particle size distribution, rheogram, acid value and pH value were measured by gas chromatograph, laser particle sizer, Coulter counter and rheometer. From above experiments following conclusions were obtained; 1) Mean particle diameter was shifted from 240 to 266㎜. 2) No significant changes were observed in the content of major fatty acids of soybean oil, rheogram, acid value and pH value.

Norovirus (NoV), which belongs to the family Caliciviridae, is one of the major causes of nonbacterial acute gastroenteritis in the world. In this study, we purified proteins from the epitope region of norovirus for development of the rapid diagnosis system using polyclonal antibodies. As antigens, parts of the ORF (open reading frame) 2, ORF2-P domain, ORF2-Epi, and ORF3 regions were selected and their expressions were induced. The antigenicity of the purified proteins was identified by Western blotting. Each of the purified proteins was injected into mice for the production of novel antibodies and after 3 months of immunization, sera from the mice were obtained. The polyclonal antibody titer was tested by enzyme-linked immunosorbent assay (ELISA) and antibody against ORF2-Epi showed the highest titer. Those polyclonal antibodies can be used in further immunoassay for the rapid detection of NoVs from food and clinical specimens.
