        Effects of Arachidonic Acid on the Calcium Channel Current (<I>I</I><SUB>Ba</SUB>) and on the Osmotic Stretch-induced Increase of <I>I</I><SUB>Ba</SUB> in Guinea-Pig Gastric Myocytes

        <P> We employed the whole-cell patch clamp technique to investigate the effects of arachidonic acid (AA) on barium inward current through the L-type calcium channels (<I>I</I><SUB>Ba</SUB>) and on osmotic stretch-induced increase of <I>I</I><SUB>Ba</SUB> in guinea-pig antral gastric myocytes. Under isosmotic condition, AA inhibited <I>I</I><SUB>Ba</SUB> in a dose-dependent manner to 91.1⁑1.4, 72.0⁑3.2, 46.0⁑1.8, and 20.3⁑2.3% at 1, 5, 10, 30 mM, respectively. The inhibitory effect of AA was not affected by 10 ㄍM indomethacin, a cyclooxygenase inhibitor. Other unsaturated fatty acids, linoleic acid (LA) and oleic acid (OA) were also found to suppress <I>I</I><SUB>Ba</SUB> but stearic acid (SA), a saturated fatty acid, had no inhibitory effect on <I>I</I><SUB>Ba</SUB>. The potency sequence of these inhibitory effects was AA (79.7⁑2.3%) > LA (43.1⁑2.7%) > OA (14.2⁑1.1%) at 30 ㄍM. On superfusing the myocyte with hyposmotic solution (214 mOsm) the amplitude of <I>I</I><SUB>Ba</SUB> at 0 mV increased (38.0⁑5.5%); this increase was completely blocked by pretreatment with 30 mM AA, but not significantly inhibited by lower concentrations of AA (1, 5 and 10 ㄍM) (P>0.05). Unsaturated fatty acids shifted the steady-state inactivation curves of <I>I</I><SUB>Ba</SUB> to the left; the extent of shift caused by AA was greater than that caused by LA. The activation curve was not affected by AA or LA. The results suggest that AA and other unsaturated fatty acids directly modulate L-type calcium channels and AA might modulate the hyposmotic stretch- induced increase of L-type calcium channel current in guinea-pig gastric smooth muscle.

        Simultaneous detection of Salmonella spp., Pseudomonas aeruginosa, Bacillus cereus, and Escherichia coli O157:H7 in environmental water using PMA combined with mPCR

        A multiplex polymerase chain reaction (mPCR) with propidium monoazide (PMA) and internal amplification control (IAC) for the simultaneous detection of waterborne pathogens Salmonella spp., Pseudomonas aeruginosa, Bacillus cereus, and Escherichia coli O157:H7, was developed. This PMA-IAC-mPCR assay used four new specific primers based on the genes for invA, ecfX, cesB, and fliC, respectively. A 16S rRNA primer was chosen for IAC to eliminate false negative results. The photosensitive dye, propidium monoazide (PMA) was used to exclude signals from dead bacteria that could lead to false positive results. In pure culture, the limits of detection (LOD) were 101 CFU/ml for P. aeruginosa, 102 CFU/ml for both Salmonella spp. and E. coli O157:H7, and 103 CFU/ml for B. cereus, respectively. In addition, with a 6–8 h enrichment of all four bacteria that were combined in a mixture that was spiked in water sample matrix, the LOD was 3 CFU/ml for Salmonella spp., 7 CFU/ml for E. coli O157:H7, 10 CFU/ml for B. cereus and 2 CFU/ml for P.aeruginosa. This PMA-IAC-mPCR assay holds potential for application in the multiplex assay of waterborne pathogens.

        Determination of Abamectin Residue in Paprika by High-Performance Liquid Chromatography

        Reversed-phase high-performance liquid chromatography (HPLC) techniques were developed to quantify abamectin (ABM) in paprika (Capsicum annum). Separation was achieved on a $C_{18}$ ODS column with a mobile phase of acetonitrile/water (96/4, v/v) mixture in an isocratic elution at the flow tate of 1.2 mL/min for avermectins (AVMs). The retention times were 8.0 and 9.7mins for AVM $B_{lb}$ and AVM $B_{1a}$, respectively. Residual AVMs (sum of AVM $B_{1a}$, AVM $B_{1b}$ and 8,9-Z-AVM $B_{1a}$) in the vegetable were extracted with acetonitrile, and the silica solid-phase extraction cartridges were used to purify the extract. AVMs were derivatized using trifluoroacetic acid and 1-methylimidazole, and the derivatives were determined with a fluorescence detector (excitation at 365 nm and emission at 470 nm). High and consistent recoveries, ranging from 93% to 115%, were obtained for AVM $B_{1a}$ and 8, 9-Z-AVM $B_{1a}$ at fortified levels of $20{\mu}g/kg\;and\;200{\mu}g/kg$ for paprika. The limit of quantitation (LOQ) was $2{\mu}g/kg$. The residual levels of AVMs in paprika in a field experiment from one day to seven days after the last application decreased from 18.40 to $7.59{\mu}g/kg$. The half-life $(T_{1/2})$ of AVMs in paprika was 1.47 days.

        The horizontal stability of an FLNG with different turret locations

        The horizontal stability of a Single-Point Moored (SPM) Floating Liquefied Natural Gas (FLNG) facility is investigated. Both numerical and experimental studies have been conducted for this SPM FLNG. The numerical simulations feature well the experimental data. The effects of the turret locations are studied based on the validated numerical model. Statistic results of the vessel's motions with different turret locations are conducted and compared. The results show that the longitudinal location of the turret has a significant influence on the horizontal stability, which has a strong relationship with the yaw and roll motions. The calculated top tensions on the hawsers also develop a regular change as changing the turret's location. The investigation will provide a brief of principles with more details for the design of the ongoing project.

        Lattice Ti Structures with Low Rigidity But Compatible Mechanical Strength: Design of Implant Materials for Trabecular Bone

        The development of porous metals to alleviate the effects of stress shielding in bone will help improve the function of metallic biomaterials in orthopaedic applications. A critical step in advancing this technology is to design metallic structures with low rigidity that is comparable with bone tissue, but with good mechanical strength. In this study, porous titanium (Ti) structures with periodic cell topologies were designed to achieve tunable mechanical properties. The versatility of the design scheme was demonstrated by examining lattice designs with different stiffness properties achieved by using the Selective Laser Melting (SLM) technology. The fabricated porous Ti exhibited a low modulus of 1.05 GPa but a high compressive strength of 55 MPa. Large deformation analysis using digital image correlation (DIC) technique indicated uniform strain patterns at micro-trusses, suggesting the overall high quality of the structure with absence of local flaws. A functionally-graded stiffness design was further investigated by varying the diameters of micro-trusses within the structure. A stiffness graded material may be favourable for anatomical site that has strong depthdependent variations, such as in trabecular bone microstructures.

        A New Algorithm for Non-Rigid Shape Matching with Anisotropic-Scaling Transformation Parameters

        Three-dimensional (3D) shape matching finds a wide application in manufacturing industry, such as surface inspection, workpiece localization and reverse engineering. In this paper, a new optimization method is proposed for non-rigid shape matching modeling and solving. The point-tangent distance function is used to construct a nonlinear least-squares model for anisotropic non-rigid shape matching, from which 9 variables with respect to non-rigid matching parameters are computed simultaneously by solving a linear system. In order to strengthen the non-rigid matching robustness to potential outliers, the optimization model is improved by an iteratively reweighted method. The typical characteristic is to weaken the influence of outliers during iterations. Finally, experiments are carried out to evaluate and analyze the proposed method, including matching accuracy, efficiency and robustness. Shape matching is also an important task in computer vision (such as face registration and object recognition), and the proposed method can find its application in this field.

        Effects of Pyridoxine on Growth Performance and Plasma Aminotransferases and Homocysteine of White Pekin Ducks

        A dose-response experiment with seven supplemental pyridoxine levels (0, 0.66, 1.32, 1.98, 2.64, 3.30, and 3.96 mg/kg) was conducted to investigate the effects of pyridoxine on growth performance and plasma aminotransferases and homocysteine of White Pekin ducks and to estimate pyridoxine requirement for these birds. A total of 336 one-day-old male White Pekin ducks were divided to 7 experimental treatments and each treatment contained 8 replicate pens with 6 birds per pen. Ducks were reared in raised wire-floor pens from hatch to 28 d of age. At 28 d of age, the weight gain, feed intake, feed/gain, and the aspartate aminotransferase, alanine aminotransferase, and homocysteine in plasma of ducks from each pen were all measured. In our study, the pyridoxine deficiency of ducks was characterized by growth depression, decreasing plasma aspartate aminotransferase activity and increasing plasma homocysteine. The ducks fed vitamin $B_6$-deficient basal diets had the worst weight gain and feed/gain among all birds and this growth depression was alleviated (p<0.05) when pyridoxine was supplemented to basal diets. On the other hand, plasma aspartate aminotransferase and homocysteine may be the sensitive indicators for vitamin $B_6$ status of ducks. The ducks fed basal diets had much lower aspartate aminotransferase activity and higher homocysteine level in plasma compared with other birds fed pyridoxine-supplemented diets (p<0.05). According to quadratic regression, the supplemental pyridoxine requirements of Pekin ducks from hatch to 28 days of age was 2.44 mg/kg for feed/gain and 2.08 mg/kg for plasma aspartate aminotransferase and the corresponding total requirements of this vitamin for these two criteria were 4.37 and 4.01 mg/kg when the pyridoxine concentration of basal diets was included, respectively. All data suggested that pyridoxine deficiency could cause growth retardation in ducks and the deficiency of this vitamin could be indicated by decreasing plasma aspartate aminotransferase activity and increasing plasma homocysteine.

        Verticillium Wilt of Okra Caused by Verticillium dahliae Kleb. in China

        Okra (Abelmoschus esculentus (L.) Moench) has gained more popularity as an economically significant plant for its nutritional and medicinal value, especially in China. During 2014-2016, the root disease of okra was discovered in four okra commercial fields surveyed in China. A fungul was isolated from the infected tissues, and was identified by Verticillium dahliae based on morphological characteristics. Pathogenicity test demonstrated that the fungus was pathogenic on okra, and fulfilled Koch’s postulates. The analysis of three sequences revealed 99-100% identity with the reported V. dahliae strain in GenBank. Neighbor-joining analysis of the gene sequences revealed that the representative isolates were clustered with V. dahliae. To the best of our knowledge, this is the first report of Verticillium wilt of okra in China.

        Effects of Dietary Methionine Levels on Choline Requirements of Starter White Pekin Ducks

        A $2{\times}5$ factorial experiment, using 2 dietary methionine levels (0.28% and 0.48%) and 5 dietary choline levels (0, 394, 823, 1,239, and 1,743 mg/kg), was conducted to study the effects of dietary methionine status on choline requirements of starter white Pekin ducks from 7 to 28 days of age. Four hundred eighty 7-d-old male White Pekin ducks were randomly allotted to ten dietary treatments, each containing 6 replicate pens with 8 birds per pen. At 28 d of age, weight gain, feed intake, and feed/gain were measured and the legs of all ducks from each pen were examined for incidence of perosis. Perosis and growth depression were observed in choline-deficient ducks and supplementation of choline reduced perosis and significantly increased weight gain and feed intake regardless of dietary methionine levels (p<0.05). In addition, significant positive effects of dietary methionine supplementation on weight gain, feed intake, and feed/gain were observed at any choline level (p<0.05). Supplementation of 1,743 mg/kg choline in diets alleviated the depression of weight gain and feed intake caused by methionine deficiency at 0.28% methionine level. The interaction between choline and methionine influenced weight gain and feed intake of ducks (p<0.05). At 0.28% methionine level, 1,743 mg/kg choline group caused 4.92% and 3.23% amount of improvement in weight gain and feed intake compared with 1,239 mg/kg choline group, respectively. According to the broken-line regression, the choline requirements of starter Pekin ducks for weight gain and feed intake were 1,472 and 1,424 mg/kg at 0.28% methionine level and 946 and 907 mg/kg at 0.48% methionine level, respectively. It suggested the choline recommendations of starter Pekin ducks on a semi-purified diet were 1448 mg/kg at 0.28% methionine level and 927 mg/kg at 0.48% methionine level, respectively. Compared with the adequate methionine level, menthionine deficiency markedly increased the choline requirements of ducks.

        Observer-based Controller Design for A T-S Fuzzy System with Unknown Premise Variables

        For the stabilization problem of T-S fuzzy system, a new observer-based controller design approachis proposed when premise variables are not accessible. With a fuzzy observer, the estimated states error system isdescribed as two parts: unknown premise variable caused terms and observer error terms. Consider the property thatthe norm of the unknown premise variable caused terms are under a Lipschitz condition constraint of observer error,an observer and controller errors augmented system is obtained. Then based on the Lyapunov function method, aseries of linear matrix inequality conditions are proposed to asymptotically stabilize the system, the observer gainmatrices are used to overcome the uncertainties caused by UPVs. Finally a simulation example is used to illustratethe effectiveness of the proposed method, comparisons with traditional method shows the conservatism reductioneffects.
