      • Knowledge Acquisition Approach Based on Rough Set in Online Aided Decision System for Food Processing Quality and Safety

        For the problem that the use effect of food processing information system is poor due to absence of knowledge acquisition measures and knowledge self-updating function, a knowledge acquisition approach based on rough set is put forward. First, the approach establishes a set of predicted samples for the relationship between food processing parameters and product quality; then uses the discretization of continuous attribute, attribute reduction and rule extraction algorithm of rough set to acquire automatically predicted knowledge from a large number of predicted sample sets, and then saves the predicted knowledge in the knowledge base of expert system; finally, realizes the extraction of knowledge of food processing process based on the inference engine, which greatly enhances the effectiveness and practicality of the acquired knowledge in online aided decision system of the food processing quality and safety.

        Protective association of Klotho rs495392 gene polymorphism against hepatic steatosis in non-alcoholic fatty liver disease patients

        Background/Aims: Non-alcoholic fatty liver disease (NAFLD) is closely associated with metabolic dysfunction. Among the multiple factors, genetic variation acts as important modifiers. Klotho, an enzyme encoded by the klotho (KL) gene in human, has been implicated in the pathogenesis of metabolic dysfunctions. However, the impact of variants in KL on NAFLD risk remains poorly understood. The aim of this study was to investigate the impact of KL rs495392 C>A polymorphism on the histological severity of NAFLD. Methods: We evaluated the impact of the KL rs495392 polymorphism on liver histology in 531 Chinese with NAFLD and replicated that in the population-based Rotterdam Study cohort. The interactions between the rs495392, vitamin D, and patatin-like phospholipase domain containing 3 (PNPLA3) rs738409 polymorphism were also analyzed. Results: Carriage of the rs495392 A allele had a protective effect on steatosis severity (odds ratio [OR], 0.61; 95% confidence interval [CI], 0.42–0.89; P=0.010) in Chinese patients. After adjustment for potential confounders, the A allele remained significant with a protective effect (OR, 0.66; 95% CI, 0.45–0.98; P=0.040). The effect on hepatic steatosis was confirmed in the Rotterdam Study cohort. Additional analysis showed the association between serum vitamin D levels and NAFLD specifically in rs495392 A allele carriers, but not in non-carriers. Moreover, we found that the rs495392 A allele attenuated the detrimental impact of PNPLA3 rs738409 G allele on the risk of severe hepatic steatosis. Conclusions: The KL rs495392 polymorphism has a protective effect against hepatic steatosis in patients with NAFLD.

        Centromere protein U enhances the progression of bladder cancer by promoting mitochondrial ribosomal protein s28 expression

        Bladder cancer is one of the most common types of cancer. Most gene mutations related to bladder cancer are dominantly acquired gene mutations and are not inherited. Previous comparative transcriptome analysis of urinary bladder cancer and control samples has revealed a set of genes that may play a role in tumor progression. Here we set out to investigate further the expression of two candidate genes, centromere protein U (CENPU) and mitochondrial ribosomal protein s28 (MRPS28) to better understand their role in bladder cancer pathogenesis. Our results confirmed that CENPU is up-regulated in human bladder cancer tissues at mRNA and protein levels. Gain-of-function and loss-of-function studies in T24 human urinary bladder cancer cell line revealed a hierarchical relationship between CENPU and MRPS28 in the regulation of cell viability, migration and invasion activity. CENPU expression was also up-regulated in in vivo nude mice xenograft model of bladder cancer and mice overexpressing CENPU had significantly higher tumor volume. In summary, our findings identify CENPU and MRPS28 in the molecular pathogenesis of bladder cancer and suggest that CENPU enhances the progression of bladder cancer by promoting MRPS28 expression.

        Sintering technology and properties of sponge city permeable bricks prepared from electrical porcelain was

        The generated electric porcelain waste increases with the rapid development of the electric porcelain industry. Unreasonableutilization results in the waste of resources and environmental pollution. However, at present, many coal gangues and fly ashesare wasted and underutilized in my country. In this paper, electric porcelain waste is used as the main raw material, and itssintering temperature, holding time, and heating rate are discussed to study the performance of the permeable brick preparedfrom electric porcelain waste. Results show that with the increase in sintering temperature, the porosity and waterpermeability coefficient of permeable bricks decrease, and their volume shrinkage rate and compressive strength show anincreasing trend. Furthermore, porosity decreases with the increase in holding time. The volume shrinkage rate andcompressive strength increase, and the water permeability coefficient decreases. The volume shrinkage rate of the permeablebrick decreases, and its porosity, water permeability coefficient, and compressive strength increase with the heating rate. Combined with the comprehensive consideration of the porosity, volume shrinkage rate, water permeability coefficient, andcompressive strength of permeable bricks, the best sintering process for preparing permeable bricks from electrical porcelainwaste is presented with a sintering temperature of 1225 °C, holding time of 1 h, and heating rate of 6 °C/min. The porosityof the prepared permeable brick is 79.5%, the volume shrinkage rate is 25.79%, the water permeability coefficient is 2.82×10-2cm/s, and the compressive strength is 5.79 MPa.

      • Expression and Clinical Significance of mTOR in Surgically Resected Non-small Cell Lung Cancer Tissues: a Case Control Study

        Aims: Mammalian target of rapamycin (mTOR) is master regulator of the PI3K/Akt/mTOR pathway and plays an important role in NSCLCs. Here we characterized mRNA and protein expression levels of mTOR and its functional associated molecules including PTEN, IGF-1R and 4EBP1 in surgically resected NSCLCs. Methods: Fifty-four patients with NSCLCs who underwent pulmonary resection were included in current study. mRNA levels of mTOR, PTEN, IGF-1R, and 4EBP1 were evaluated by RT-PCR and protein expression of mTOR, PTEN, and IGF-1R by immunohistochemistry (IHC). Association of expression of the relevant molecules with clinical characteristics, as well as correlations between mTOR and PTEN, 4EBP1 and IGF-1R were also assessed. Results: The results of RT-PCR showed that in NSCLCs, the expression level of mTOR increased, while PTEN, 4EBP1 and IGF-1R decreased. Statistical analysis indicated high IGF-1R expression was correlated with advanced clinical stage (stage III) and PTEN expression was reversely associated with tumor size (P=0.16). The results of IHC showed mTOR positive staining in 51.8% of cases, while IGF-1R positive staining was found in 83.3% and loss of PTEN in 46.3%. Protein expression of mTOR was correlated with its regulators, PTEN and IGF-1R, to some extent. Conclusions: Abnormal activation of mTOR signaling, high expression of IGF-1R, and loss of PTEN were observed in resected NSCLC specimens. The poor expression agreement of mTOR with its regulators, PTEN, and IGF-1R, implied that combination strategy of mTOR inhibitors with other targets hold significant potential for NSCLC treatment.

        Nonlinear stability of the upper chords in half-through truss bridges

        The upper chords in half-through truss bridges are prone to buckling due to a lack of the upper transverse connections. Taking into account geometric and material nonlinearity, nonlinear finite-element analysis of a simple supported truss bridge was carried out to exhibit effects of different types of initial imperfections. A half-wave of initial imperfection was proved to be effective in the nonlinear buckling analysis. And a parameter analysis of initial imperfections was also conducted to reveal that the upper chords have the greatest impact on the buckling, followed by the bottom chords, vertical and diagonal web members. Yet initial imperfections of transverse beams have almost no effect on the buckling. Moreover, using influence surface method, the combinatorial effects of initial imperfections were compared to demonstrate that initial imperfections of the upper chords play a leading role. Furthermore, the equivalent effective length coefficients of the upper chord were derived to be 0.2~ 0.28 by different methods, which implies vertical and diagonal web members still provide effective constraints for the upper chord despite a lack of the upper transverse connections between the two upper chords. Therefore, the geometrical and material nonlinear finite-element method is effective in the buckling analysis due to its higher precision. Based on nonlinear analysis and installation deviations of members, initial imperfection of l/500 is recommended in the nonlinear analysis of half-through truss bridges without initial imperfection investigation.

        <i>Chd7</i> Is Critical for Early T-Cell Development and Thymus Organogenesis in Zebrafish

        <P>Coloboma, heart defect, atresia choanae, retarded growth and development, genital hypoplasia, ear anomalies/deafness (CHARGE) syndrome is a congenital disorder affecting multiple organs and mainly caused by mutations in CHD7, a gene encoding a chromatin-remodeling protein. Immunodeficiency and reduced T cells have been noted in CHARGE syndrome. However, the mechanisms underlying T lymphopenia are largely unexplored. Herein, we observed dramatic decrease of T cells in both chd7knockdown and knockout zebrafish embryos. Unexpectedly, hematopoietic stem and progenitor cells and, particularly, lymphoid progenitor cells were increased peripherally in nonthymic areas in chd7-deficient embryos, unlikely to contribute to the T-cell decrease. Further analysis demonstrated that both the organogenesis and homing function of the thymus were seriously impaired. Chd7 might regulate thymus organogenesis through modulating the development of both neural crest cell-derived mesenchyme and pharyngeal endoderm-derived thymic epithelial cells. The expression of faxn1, a central regulator of thymic epithelium, was remarkably down-regulated in the pharyngeal region in chd7-deficient embryos. Moreover, the T-cell reduction in chd7-deficient embryos was partially rescued by overexpressingfoxnl, suggesting that restoring thymic epithelium may be a potential therapeutic strategy for treating immunodeficiency in CHARGE syndrome. Collectively, the results indicated that chd7 was critical for thymic development and T-lymphopenia in CHARGE syndrome may be mainly attributed to the defects of thymic organogenesis. The current finding may benefit the diagnosis and therapy of T lymphopenia and immunodeficiency in CHARGE syndrome.</P>

        Comparative Genomics Profiling of Clinical Isolates of Helicobacter pylori in Chinese Populations Using DNA Microarray

        In order to search for specific genotypes related to this unique phenotype, we used whole genomic DNA microarray to characterize the genomic diversity of Helicobacter pylori (H. pylori) strains isolated from clinical patients in China.The open reading frame (ORF) fragments on our microarray were generated by PCR using gene-specific primers. Genomic DNA of H. pylori 26695 and J99 were used as templates. Thirty-four H. pylori isolates were obtained from patients in Shanghai. Results were judged based on ln(x) transformed and normalized Cy3/Cy5 ratios. Our microarray included 1882 DNA fragments corresponding to 1636 ORFs of both sequenced H. pylori strains. Cluster analysis, revealed two diverse regions in the H. pylori genome that were not present in other isolates. Among the 1636 genes, 1091 (66.7%) were common to all H. pylori strains, representing the functional core of the genome. Most of the genes found in the H. pylori functional core were responsible for metabolism, cellular processes, transcription and biosynthesis of amino acids, functions that are essential to H. pylori’s growth and colonization in its host. In contrast, 522(31.9%) genes were strain-specific genes that were missing from at least one strain of H. pylori. Strainspecific genes primarily included restriction modification system components, transposase genes, hypothetical proteins and outer membrane proteins. These strain-specific genes may aid the bacteria under specific circumstances during their long-term infection in genetically diverse hosts. Our results suggest 34 H. pylori clinical strains have extensive genomic diversity. Core genes and strain-specific genes both play essential roles in H. pylori propagation and pathogenesis. Our microarray experiment may help select relatively significant genes for further research on the pathogenicity of H. pylori and development of a vaccine for H. pylori.

        Simultaneous Determination of Hydroquinone and Catechol by N-doped Porous Biochar-modified Electrode

        N-doped porous biochar (NPB) with large conjugated systems could not only be used as enrichment carriers but also be in favor of electron transport in the electrochemical detection. The NPB-modified electrode was fabricated for the simultaneous detection of catechol (CA) and hydroquinone (HQ) to enhance the signal-to-noise ratio and further improve the sensitivity. A detection limit as low as 37 and 47?nM was achieved for CA and HQ, respectively. The proposed strategy with the merits of high sensitivity, selectivity, and reproducibility exhibited a great potential for the detection.

        Gene Expression Profile of Helicobacter pylori in Response to Growth Temperature Variation

        A Helicobacter pylori whole-genome DNA microarray was constructed to study expression profiles of H. pylori in response to a sudden temperature transfer from 37°C to 20°C. The expression level of the genome at each of four time points (15, 30, 60, and 120 min) after temperature downshift was compared with that just before cold treatment. Globally, 10.2% (n=167) of the total predicted H. pylori genes (n=1636) represented on the microarray were significantly differentially expressed (p<0.05) over a 120 min period after shift to low temperature. The expression profiles of the differentially expressed genes were grouped, and their expression patterns were validated by quantitative real-time PCR. Up-regulated genes mainly included genes involved in energy metabolism and substance metabolism, cellular processes, protein fate, ribosomal protein genes, and hypothetical protein genes, which indicate the compensational responses of H. pylori to temperature downshift. Those genes play important roles in adaption to temperature downshift of H. pylori. Down-regulation of DNA metabolism genes and cell envelope genes and cellular processes genes may reflect damaged functions under low temperature, which is unfavorable to bacterial infection and propagation. Overall, this time-course study provides new insights into the primary response of H. pylori to a sudden temperature downshift, which allow the bacteria to survive and adapt to the new host environment.
