        Effects of Arachidonic Acid on the Calcium Channel Current (<I>I</I><SUB>Ba</SUB>) and on the Osmotic Stretch-induced Increase of <I>I</I><SUB>Ba</SUB> in Guinea-Pig Gastric Myocytes

        <P> We employed the whole-cell patch clamp technique to investigate the effects of arachidonic acid (AA) on barium inward current through the L-type calcium channels (<I>I</I><SUB>Ba</SUB>) and on osmotic stretch-induced increase of <I>I</I><SUB>Ba</SUB> in guinea-pig antral gastric myocytes. Under isosmotic condition, AA inhibited <I>I</I><SUB>Ba</SUB> in a dose-dependent manner to 91.1⁑1.4, 72.0⁑3.2, 46.0⁑1.8, and 20.3⁑2.3% at 1, 5, 10, 30 mM, respectively. The inhibitory effect of AA was not affected by 10 ㄍM indomethacin, a cyclooxygenase inhibitor. Other unsaturated fatty acids, linoleic acid (LA) and oleic acid (OA) were also found to suppress <I>I</I><SUB>Ba</SUB> but stearic acid (SA), a saturated fatty acid, had no inhibitory effect on <I>I</I><SUB>Ba</SUB>. The potency sequence of these inhibitory effects was AA (79.7⁑2.3%) > LA (43.1⁑2.7%) > OA (14.2⁑1.1%) at 30 ㄍM. On superfusing the myocyte with hyposmotic solution (214 mOsm) the amplitude of <I>I</I><SUB>Ba</SUB> at 0 mV increased (38.0⁑5.5%); this increase was completely blocked by pretreatment with 30 mM AA, but not significantly inhibited by lower concentrations of AA (1, 5 and 10 ㄍM) (P>0.05). Unsaturated fatty acids shifted the steady-state inactivation curves of <I>I</I><SUB>Ba</SUB> to the left; the extent of shift caused by AA was greater than that caused by LA. The activation curve was not affected by AA or LA. The results suggest that AA and other unsaturated fatty acids directly modulate L-type calcium channels and AA might modulate the hyposmotic stretch- induced increase of L-type calcium channel current in guinea-pig gastric smooth muscle.

        Sublytic C5b-9 induces glomerular mesangial cell proliferation via ERK1/2-dependent SOX9 phosphorylation and acetylation by enhancing Cyclin D1 in rat Thy-1 nephritis

        Glomerular mesangial cell (GMC) proliferation is a histopathological alteration in human mesangioproliferative glomerulonephritis (MsPGN) or in animal models of MsPGN, e.g., the rat Thy‐1 nephritis (Thy-1N) model. Although sublytic C5b-9 assembly on the GMC membrane can trigger cell proliferation, the mechanisms are still undefined. We found that sublytic C5b-9-induced rat GMC proliferation was driven by extracellular signal‐regulated kinase 1/2 (ERK1/2), sry-related HMG-box 9 (SOX9), and Cyclin D1. Here, ERK1/2 phosphorylation was a result of the calcium influx-PKC-α-Raf-MEK1/2 axis activated by sublytic C5b-9, and Cyclin D1 gene transcription was enhanced by ERK1/2-dependent SOX9 binding to the Cyclin D1 promoter (−582 to −238 nt). In addition, ERK1/2 not only interacted with SOX9 in the cell nucleus to mediate its phosphorylation at serine residues 64 (a new site identified by mass spectrometry) and 181 (a known site), but also indirectly induced SOX9 acetylation by elevating the expression of general control non-repressed protein 5 (GCN5), which together resulted in Cyclin D1 synthesis and GMC proliferation. Moreover, our in vivo experiments confirmed that silencing these genes ameliorated the lesions of Thy‐1N rats and reduced SOX9 phosphorylation, acetylation and Cyclin D1 expression. Furthermore, the renal tissue sections of MsPGN patients also showed higher phosphorylation or expression of ERK1/2, SOX9, and Cyclin D1. In summary, these findings suggest that sublytic C5b-9-induced GMC proliferation in rat Thy-1N requires SOX9 phosphorylation and acetylation via enhanced Cyclin D1 gene transcription, which may provide a new insight into human MsPGN pathogenesis.

        Low Molecular Weight Organic Acids in Brassica pekinensis Rupr. and Growing soil Influenced by Simulated Nitrate Deposition

        We investigated whether carboxylate exudation of Brassica pekinensis Rupr. was affected by nitrate deposition from simulated acid rain. A gas chromatographic (GC) analysis was employed for the determination of low molecular weight organic acids (LOA) in rhizosphere soils, bulk soil, roots and leaves of Brassica pekinensis Rupr.. Rhizosphere soils were collected after 8 weeks of plant growth by first removing the bulk soil from the root system and then by mechanical move off the rhizosphere soil that adhered to the root surface with soft brush. Soil and plant materials were simultaneously extracted with the mixture of methanol and sulfuric acid (100:7, v/v). Seven organic acids, oxalic, malonic, fumaric, succinic, maleic, L-malic and citric acid were identified and quantified by GC equipped with FID. Oxalic, L-malic, and citric acids were found in both the bulk and rhizosphere soils, while most LOAs were not detected in the control treatment. On the contrary, except maleic acid, all other organic acids were detected in the leaves and roots of cabbages treated with nitrate deposition.

      • Trends in the Incidence of 15 Common Cancers in Hong Kong, 1983-2008

        Background: The objective of this study WAS to describe cancer incidence rates and trends among THE Hong Kong population for the period 1983-2008. Methods: Incident cases and population data from 1983 to 2008 were obtained from the Hong Kong Cancer Registry and the Census and Statistics Department, respectively. Agestandardized incidence rates (ASIR) were estimated and joinpoint regression was applied to detect significant changes in cancer morbidity. Results: For all cancers combined, the ASIR showed declining trends (1.37% in men, 0.94% in women), this also being the case for cancers of lung, liver, nasopharynx, stomach, bladder, oesophagus for both genders and cervix cancer for women. With cancer of thyroid, prostate, male colorectal, corpus uteri, ovary and female breast cancer an increase was evident throughout the period. The incidence for leukemia showed a stable trend since early 1990s, following an earlier decrease. Conclusion: Although overall cancer incidence rates and certain cancers showed declining trends, incidence trends for colorectal, thyroid and sex-related cancers continue to rise. These trends in cancer morbidity can be used as an important resource to plan and develop effective programs aimed at the control and prevention of the spread of cancer amongst the Hong Kong population. It is particularly useful in allowing projection of future burdens on the society with the increase in certain cancer incidences.

      • ADPRT Val762Ala and XRCC1 Arg194Trp Polymorphisms and Risk of Gastric Cancer in Sichuan of China

        Objective: Gastric cancer remains a major health problem in China. We hypothesized that XRCC1 Arg194Trp and ADPRT Val762Ala may be associated with risk. Methods: We designed a multicenter 1:1 matched case-control study of 307 pairs of gastric cancers and controls between October 2010 and August 2011. XRCC1 Arg194Trp and ADPRT Val762Ala were sequenced, and demographic data as well as lifestyle factors were collected using a self-designed questionnaire. Results: Individuals carrying XRCC1 Trp/Trp or Arg/Trp variant genotype had a significantly increased risk of gastric cancer (OR, 1.718; 95% CI, 1.190-2.479), while the OR for ADPRT Val762Ala variant genotype (Ala/Ala or Val/Ala) was 1.175 (95% CI, 0.796-1.737). No gene-gene or gene-environment interactions were found. In addition, family history of cancer and drinkers proportion were higher among cases than among controls (P<0.05). Conclusions: XRCC1 194 Arg/Trp or Trp/Trp genotype, family history of cancer, and drinking are suspected risk factors of gastric cancer from our study. Our findings may offer insight into further similar large gene-environment and gene-gene studies in this region.

        An iterative approach for time-domain flutter analysis of bridges based on restart technique

        This paper presents a restart iterative approach for time-domain flutter analysis of long-span bridges using the commercial FE package ANSYS. This approach utilizes the recursive formats of impulse-response-function expressions for bridge's aeroelastic forces. Nonlinear dynamic equilibrium equations are iteratively solved by using the restart technique in ANSYS, which enable the equilibrium state of system to get back to last moment absolutely during iterations. The condition for the onset of flutter instability becomes that, at a certain wind velocity, the amplitude of vibration is invariant with time. A long-span suspension bridge was taken as a numerical example to verify the applicability and accuracy of the proposed method by comparing calculated results with wind tunnel tests. The proposed method enables the bridge designers and engineering practitioners to carry out time-domain flutter analysis of bridges in commercial FE package ANSYS.

        Determination of Abamectin Residue in Paprika by High-Performance Liquid Chromatography

        Reversed-phase high-performance liquid chromatography (HPLC) techniques were developed to quantify abamectin (ABM) in paprika (Capsicum annum). Separation was achieved on a $C_{18}$ ODS column with a mobile phase of acetonitrile/water (96/4, v/v) mixture in an isocratic elution at the flow tate of 1.2 mL/min for avermectins (AVMs). The retention times were 8.0 and 9.7mins for AVM $B_{lb}$ and AVM $B_{1a}$, respectively. Residual AVMs (sum of AVM $B_{1a}$, AVM $B_{1b}$ and 8,9-Z-AVM $B_{1a}$) in the vegetable were extracted with acetonitrile, and the silica solid-phase extraction cartridges were used to purify the extract. AVMs were derivatized using trifluoroacetic acid and 1-methylimidazole, and the derivatives were determined with a fluorescence detector (excitation at 365 nm and emission at 470 nm). High and consistent recoveries, ranging from 93% to 115%, were obtained for AVM $B_{1a}$ and 8, 9-Z-AVM $B_{1a}$ at fortified levels of $20{\mu}g/kg\;and\;200{\mu}g/kg$ for paprika. The limit of quantitation (LOQ) was $2{\mu}g/kg$. The residual levels of AVMs in paprika in a field experiment from one day to seven days after the last application decreased from 18.40 to $7.59{\mu}g/kg$. The half-life $(T_{1/2})$ of AVMs in paprika was 1.47 days.

        Observer-based Controller Design for A T-S Fuzzy System with Unknown Premise Variables

        For the stabilization problem of T-S fuzzy system, a new observer-based controller design approachis proposed when premise variables are not accessible. With a fuzzy observer, the estimated states error system isdescribed as two parts: unknown premise variable caused terms and observer error terms. Consider the property thatthe norm of the unknown premise variable caused terms are under a Lipschitz condition constraint of observer error,an observer and controller errors augmented system is obtained. Then based on the Lyapunov function method, aseries of linear matrix inequality conditions are proposed to asymptotically stabilize the system, the observer gainmatrices are used to overcome the uncertainties caused by UPVs. Finally a simulation example is used to illustratethe effectiveness of the proposed method, comparisons with traditional method shows the conservatism reductioneffects.

        Steel Bridge Construction of Hong Kong–Zhuhai–Macao Bridge

        The 55-km-long Hong Kong–Zhuhai–Macao Bridge (HZMB) is the world’s longest sea-crossing bridge, connecting Hong Kong with Zhuhai and Macao at the mouth of the Pearl River Estuary in China, comprising 22.9-km-long steel bridges. HZMB is the leading steel bridge in China, with top-level manufacturing and installation technology. This paper outlines the steel bridge construction experiences of HZMB to provide comparisons for the construction of other long sea-crossing steel bridges at home or abroad. The main considerations of construction constraints, scheme selection, structural and aesthetic design of HZMB are presented, and the following points related to new strategies in the steel bridge construction of HZMB are elaborated: (1) construction quality assurance, (2) automatic manufacturing technology, (3) large segment off shore installation, (4) eco-friendly paint (content limitation of volatile organic compounds) and new multifunctional inspection gantry, and (5) Guss Mastic Asphalt steel deck pavement system. The successful implementation of those strategies shows that the steel bridge construction of HZMB promotes improvement in the overall construction and management level of the Chinese bridge industry. The advanced experience of HZMB has opened up broad prospects for the design and construction of off shore bridge engineering in China.
