      • Susceptibility Loci Associations with Prostate Cancer Risk in Northern Chinese Men

        Background: KLK3 gene products, like human prostate-specific antigen (PSA), are important biomarkers in the clinical diagnosis of prostate cancer (PCa). G protein-coupled receptor RFX6, C2orf43 and FOXP4 signaling plays important roles in the development of PCa. However, associations of these genes with PCa in northern Chinese men remain to be detailed. This study aimed to investigate their impact on occurrence and level of malignancy. Methods: All subjects were from Beijing and Tianjin, including 266 cases with prostate cancer and 288 normal individuals as controls. We evaluated associations between clinical covariates (age at diagnosis, prostate specific antigen, Gleason score, tumor stage and aggressive) and 6 candidate PCa risk loci, genotyped by PCR- high resolution melting curve and sequencing methods. Results: Case-control analysis of allelic frequency of PCa associated with PCa showed that one of the 6 candidate risk loci, rs339331 in the RFX6 gene, was associated with reduced risk of prostate cancer (odds ratio (OR) = 0.73, 95% confidence interval (CI) =0.57-0.94, P = 0.013) in northern Chinese men. In addition, subjects with CX (CC+TC) genotypes had a decreased risk for prostrate cancer compared to those carrying the TT homozygote (OR =0.64, 95% CI = 0.45- 0.90, P = 0.008). The TT genotype of 13q22 (rs9600079, T) was associated with tumor stage (P=0.044, OR=2.34, 95% CI=0.94-5.87). Other SNPs were not significantly associated with clinical covariates in prostate cancer (P > 0.05). Conclusions. rs339331 in the RFX6 gene may be associated with prostate cancer as a susceptibility locus in northern Chinese men.

        Design of 39-GHz Up- and Down-Conversion Mixers for 5G mmWave TDD Applications

        This article presents fully integrated 39-GHz bidirectional up- and down-conversion mixers for 5G millimeter-wave (mmWave) applications. Fabricated in a 65-nm CMOS process with a 1.2-V supply voltage, the up- and down-conversion mixers consume 39 and 43 mW, respectively. For 5G time-division duplexing (TDD) operation, intermediate-frequency (IF)/local-oscillator (LO)/radio-frequency (RF) T/R switches are introduced. For better isolation and low insertion loss between the up- and down-conversion mixer, a series-shunt singlepole double-throw (SPDT) structure and an equivalent lumped λ/4 transmission line are proposed for IF and RF T/R switches, respectively. To realize compact area and wide bandwidth, a transformer-based matching network is adopted in this design. Targeting multi-channel phased array applications, the measurement result shows that the up-conversion mixer achieves a 2.5-dB peak conversion gain with 6.5 GHz 3-dB bandwidth. Including the insertion loss of the switch and IF signal routing, at the maximum gain of 36.5 GHz, the up-conversion mixer achieves an output 1-dB gain compression point (OP1dB) of 2.5 dBm. Furthermore, the down-conversion mixer achieves a 5-dB peak conversion gain with a 9.7-GHz 3-dB bandwidth.

        Simple and green fabrication of a biomass-derived N and O self-doped hierarchical porous carbon via a self-activation route for supercapacitor application

        To meet the increased performance and cost requirements of commercial supercapacitor, a N and O self-doped hierarchical porous carbon is fabricated via a green and simple self-activation route utilizing leaves of wild hollyhock as raw materials. Comparing to commercial activated carbon, the reported material exhibits some marked merits, such as simple and green fabrication process, low cost, and superior capacitance performance. The specifc surface area of the obtained N and O co�doped hierarchical porous carbon arrives 954 m2 g−1, and the content of the self-doped nitrogen and oxygen reaches 2.64 at.% and 7.38 at.%, respectively. The specifc capacitance of the obtained material reaches 226 F g−1 while the specifc capacitance of the symmetric supercapacitor arrives 47.3 F g−1. Meanwhile, more than 90.3% of initial specifc capacitance is kept under a current density of 20 A g−1, and no arresting degradation is observed for capacitance after 5000 times cycle, perfectly demonstrating the excellent cycle and rate capability of the obtained material. The obtained N and O co-doped hierarchical porous carbon are expected to be an ideal substitution for commercial activated carbon.

        The effect of the PLIN1 gene on the metabolism and mitochondria of porcine skeletal muscle satellite cells

        Background: Perilipin 1 (PLIN1) is a lipid droplet scaffolding protein that plays a regulatory role in fat decomposition and mitochondrial function. Objective: In this study, the effects of PLIN1 gene knockout (PLIN1-KO) and PLIN1 gene overexpression (PLIN1-EX) on cell metabolism and mitochondrial function in porcine skeletal muscle satellite cells were assessed. Methods: Porcine skeletal muscle satellite cells were used as the control group (NC). The expression of mitochondrial function-related proteins was detected by western blot. Apoptosis, cell cycle, mitochondrial function-related indices, mitochondrial structure, and morphology were measured by flow cytometry. Results: Our results demonstrated that stable expression of the PLIN1 gene in skeletal muscle satellite cells is critical to maintaining cell metabolism and mitochondrial function. After knockout and overexpression of the PLIN1 gene, the anti-apoptotic ability of cells was enhanced, and the metabolic activity of the cells was accelerated, but at the cost of mitochondrial structural damage, reduction in the number of mitochondria, and decreased mitochondrial function. Conclusion: This study explored the effect of the PLIN1 gene on the mitochondria and metabolism of porcine skeletal muscle satellite cells and provided a theoretical basis for the subsequent study of the effects of PLIN1 on muscle tissue development and meat quality.

        Influence of Ga(Al)As substrates on surface morphology and critical thickness of InGaAs quantum dots

        Influence of Ga(Al)As substrates on surface morphology of InGaAs quantum dots and critical thickness of In0.5Ga0.5As film grown by molecular beam epitaxy is investigated. The In0.5Ga0.5As quantum dots are grown on (001) surfaces of GaAs and Al0.25Ga0.75 A at 450 °C, scanning tunneling microscope images show that the size of quantum dots varied slightly for 10 ML of In0.5Ga0.5As grown on GaAs and Al0.25Ga0.75As surfaces. Reflection high energy electron diffraction (RHEED) is used to monitor the growth of 4 monolayers (ML) In0.5Ga0.5As on Al0.25Ga0.75As and GaAs surfaces during deposition. The critical thickness is theoretically calculated by adding energy caused by surface roughness and heat from substrate. The calculations show that the critical thickness of In0.5Ga0.5As grown on GaAs and Al0.25Ga0.75As are 3.2 ML and 3.8 ML, respectively. The theoretical calculation agrees with the experimental results.

        Safety and antifatigue effect of Korean Red Ginseng capsule: A randomized, double-blind and placebo-controlled clinical trial

        Background: In physical activity or labor, the human body is in a state of high intensity stress, and allparts or physiological functions of the body respond positively to maintain or balance the need formovement. The human body has many physiological changes in the process of movement, and fatigue isthe external manifestation of various complex changes inside the human body. Fatigue is also a physiologicalmechanism of self-protection after the body reaches a certain level of activity, which can preventthe occurrence of life-threatening excessive functional failure. The generation of fatigue is a very complexprocess, and its mechanism has not been concluded yet. Therefore, it is an important work to search andscreen the effective components of natural plants that have anti-fatigue effect and to explore theirmechanism. Methods: This was a 8-week, randomized, double-blind, placebo-controlled clinical trial. A total of 110subjects who passed physical examination were included according to the scheme design, and randomlydivided into a test group which was given KRG and a placebo control group. The calculation is carried outaccording to the standard of sub-high-intensity exercise test. Results: There was no adverse effect on safety index of subjects after taking red ginseng capsule. AfterKRG treatment, subjective strength grade is significant lower than placebo treatment. Blood lactic acidcontent is significantly get lower after trial in KRG group, and significant lower than placebo group. Creatine phosphokinase(CK) content is significantly get lower after trial in KRG group, and significantlower than placebo group. Conclusion: According to the criterion in the test scheme, the result shows that KRG is helpful onrelieving physical fatigue.

        Metabonomics study on the hot syndrome of traditional Chinese medicine by rapid resolution liquid chromatography combined with quadrupole time-of-flight tandem mass spectrometry

        The hot syndrome refers to any feverish conditionsduring a pathological development, a sub-healthphenomenon, and is a potential risk for human health. Themetabonomics study on the hot syndrome may provideinsight into understanding of its pathology and play a rolein the prevention and treatment of its related diseases. Inthis paper, the rats were dosed with the hot syndromeprescription, ginseng and water. The corresponding urinesamples were identified by rapid resolution liquid chromatographycombined with quadrupole time-of-flight tandemmass spectrometry. More than 1,000 metaboliccompounds from different urine samples could be furtherdifferentiated by principal component analysis. As a result,the rat body temperature and weight were recognized as thehot syndrome related factors. Some specific metaboliteshave been discovered as a pattern of the potential biomarkers for the hot syndrome. The results showed thatginseng cannot cause the hot syndrome in a reasonabledose, but the hot syndrome prescription can. It is suggestedthat ginseng cannot be used only as a tradition Chinesemedicine but also as a nutrient. The work showed metabonomicsmethod is a valuable tool in studying mechanismof the hot syndrome.

        The Influence of Reconstruction Algorithm and Heart Rate on Coronary Artery Image Quality and Stenosis Detection at 64-Detector Cardiac CT

        Objective: We wanted to evaluate the impact of two reconstruction algorithms (halfscan and multisector) on the image quality and the accuracy of measuring the severity of coronary stenoses by using a pulsating cardiac phantom with different heart rates (HRs). Materials and Methods: Simulated coronary arteries with different stenotic severities (25, 50, 75%) and different luminal diameters (3, 4, 5 mm) were scanned with a fixed pitch of 0.16 and a 0.35 second gantry rotation time on a 64-slice multidetector CT. Both reconstruction algorithms (halfscan and multisector) were applied to HRs of 40-120 beats per minute (bpm) at 10 bpm intervals. Three experienced radiologists visually assessed the image quality and they manually measured the stenotic severity. Results: Fewer measurement errors occurred with multisector reconstruction (p = 0.05), a slower HR (p < 0.001) and a larger luminal diameter (p = 0.014); measurement errors were not related with the observers or the stenotic severity. There was no significant difference in measurements as for the reconstruction algorithms below an HR of 70 bpm. More nonassessable segments were visualized with halfscan reconstruction (p = 0.004) and higher HRs (p < 0.001). Halfscan reconstruction had better quality scores when the HR was below 60 bpm, while multisector reconstruction had better quality scores when the HR was above 90 bpm. For the HRs between 60 and 90 bpm, both reconstruction modes had similar quality scores. With excluding the nonassessable segments, both reconstruction algorithms achieved a similar mean measured stenotic severity and similar standard deviations. Conclusion: At a higher HR (above 90 bpm), multisector reconstruction had better temporal resolution, fewer nonassessable segments, better quality scores and better accuracy of measuring the stenotic severity in this phantom study.
