Mitochondrial dysfunction plays a major role in the development of intervertebral disc degeneration (IDD). Sirtuin 5 (SIRT5) participates in the maintenance of mitochondrial homeostasis through its desuccinylase activity. However, it is still unclear whether succinylation or SIRT5 is involved in the impairment of mitochondria and development of IDD induced by excessive mechanical stress. Our 4D label-free quantitative proteomic results showed decreased expression of the desuccinylase SIRT5 in rat nucleus pulposus (NP) tissues under mechanical loading. Overexpression of Sirt5 effectively alleviated, whereas knockdown of Sirt5 aggravated, the apoptosis and dysfunction of NP cells under mechanical stress, consistent with the more severe IDD phenotype of Sirt5 KO mice than wild-type mice that underwent lumbar spine instability (LSI) surgery. Moreover, immunoprecipitation-coupled mass spectrometry (IP-MS) results suggested that AIFM1 was a downstream target of SIRT5, which was verified by a Co-IP assay. We further demonstrated that reduced SIRT5 expression resulted in the increased succinylation of AIFM1, which in turn abolished the interaction between AIFM1 and CHCHD4 and thus led to the reduced electron transfer chain (ETC) complex subunits in NP cells. Reduced ETC complex subunits resulted in mitochondrial dysfunction and the subsequent occurrence of IDD under mechanical stress. Finally, we validated the efficacy of treatments targeting disrupted mitochondrial protein importation by upregulating SIRT5 expression or methylene blue (MB) administration in the compression-induced rat IDD model. In conclusion, our study provides new insights into the occurrence and development of IDD and offers promising therapeutic approaches for IDD.

Wound healing is delayed in diabetic patients. Increased apoptosis and endothelial progenitor cell (EPC) dysfunction are implicated in delayed diabetic wound healing. Melatonin, a major secretory product of the pineal gland, promotes diabetic wound healing; however, its mechanism of action remains unclear. Here, EPCs were isolated from the bone marrow of mice. Treatment of EPCs with melatonin alleviated advanced glycation end product (AGE)-induced apoptosis and cellular dysfunction. We further examined autophagy flux after melatonin treatment and found increased light chain 3 (LC3) and p62 protein levels in AGE-treated EPCs. However, lysosome-associated membrane protein 2 expression was decreased, indicating that autophagy flux was impaired in EPCs treated with AGEs. We then evaluated autophagy flux after melatonin treatment and found that melatonin increased the LC3 levels, but attenuated the accumulation of p62, suggesting a stimulatory effect of melatonin on autophagy flux. Blockage of autophagy flux by chloroquine partially abolished the protective effects of melatonin, indicating that autophagy flux is involved in the protective effects of melatonin. Furthermore, we found that the AMPK/mTOR signaling pathway is involved in autophagy flux stimulation by melatonin. An in vivo study also illustrated that melatonin treatment ameliorated impaired wound healing in a streptozotocin-induced diabetic wound healing model. Thus, our study shows that melatonin protects EPCs against apoptosis and dysfunction via autophagy flux stimulation and ameliorates impaired wound healing in vivo, providing insight into its mechanism of action in diabetic wound healing.
Expression of HMGB1 and its Clinical Significance in T-cell Lymphoma

Objectives: To evaluate the clinical significance of HMGB1 expression in T-cell lymphoma. Methods: Immunohistochemical staining for HMGB1 and survivin was performed with specimens from 120 cases of T-cell lymphoma and 40 cases of reactive lymphoid hyperplasia with antibodies against human HMGB1 and survivin. Results: The expression of HMGB1 and survivin was significantly higher in tissues of T-cell lymphoma than in reactive lymphoid hyperplasia. Positive expression of HMGB1 and survivin was observed in 63.7% (65/102) and 61.8% (63/102) of T-cell lymphoma cases, respectively. While was associated with gender, age, and tumor location, significant correlations with malignancy and clinical stage were observed. Spearman rank correlation analysis revealed that the expression of HMGB1 and survivin was positively correlated in T-cell lymphomas (P<0.01). Conclusions: Expression of HMGB1 and survivin in T-cell lymphomas is significantly associated with malignancy and clinical stage, but not with gender, age and tumor location. Elevated expression of HMGB1 may be an important biomarker for the development and progression of T-cell lymphoma.
Cytogenetical Identification in Seven Species of Elytrigia Desv.

The ploid level of 46 germplasms of Elytrigia Desv. were observed and determined with the microscope, which are from 21 countries and areas that United States of America offered. The results showed these accessions were grouped into three distinct ploid levels including diploidy, tetraploidy and hexaploidy in E. intermedia, E. repens, E. elongata, E. pontica, E. caespitosa, E. juncea, E. alatavica. There were 24 diploid accessions, 1 tetraploidy and 1 hexaploidy in E. intermedia; 4 diploid accessions and 3 tetraploidy in E. repens, 6 texaploidy and 1 hexaploidy in E. elongata, 1 diploidy and 2 tetraploidy in E. pontica, all of E. caespitosa, E. juncea and E. alatavica were diploidy.
An Optimized Third Harmonic Injection Method for Three-Phase Cascaded H-Bridge Photovoltaic Inverter

The uneven irradiation and temperature in photovoltaic (PV) modules of three-phase cascaded H-bridge (CHB) inverters will cause the unbalanced output power between solar panels, resulting in unbalanced grid current between three phases. In addition, since each phase can be regarded as a single-phase inverter, the second harmonic voltage pulsation of dc-side capacitors is a problem to be solved. To solve these two problems, a third harmonic injection optimization method on the basis of fundamental frequency zero sequence is proposed in this paper. The third harmonic with the same amplitude of fundamental modulated wave signal is injected into the modulated wave signal when the amplitude of synthesized wave signal is no greater than unity. Otherwise, the amplitude of third harmonic is decreased correspondingly to ensure the amplitude of synthesized wave signal being unity. With this method, the dc-side voltage pulsation is effectively restrained and the power balancing limit of CHB inverter is expanded. The feasibility and effectiveness of this method are verified by an experimental prototype of a three-phase seven-level CHB inverter.

There are 10 sets of CNC equipment in a numerical controlled workshop, and set up the CAD/CAPP/CAM center, there is a problem that each production unit is an information island, which can not implement informatization management. This paper takes ZNE-100TL module realized the embedded Ethernet and RS-232C data transfer function as the hardware platform, takes VC++ as software platform, applied Ethernet technology to realize communication the CNC equipment with CAD/CAPP/CAM center. This is good for production management and data transmission in numerical controlled workshop, which can improve the production efficiency.
Banknote Image Defect Recognition Method Based on Convolution Neural Network

There are shortcomings in the currently used traditional CCD imaging system which can automatically recognize banknote image defect, such as the need to manually extract the defect characteristics and low accuracy rate of detection results. This paper briefly introduced the advantage of convolution Neural Network (CNN) in image classification and designed a image defect identification method based on convolutional neural network (CNN). The experimental results on data sets show that the identification accuracy rate of this method is 95.6%, which is significantly better than traditional identification method.
Patterning of CVD Diamond Films For MEMS Application

To apply diamond films in microelectromechanical systems(MEMS), it is necessary to develop the patterning technologies of diamond films in the micrometer scale. In this paper, three different kinds of technologies for patterning CVD diamond films carried out by us were demonstrated: selective growth by improved diamond nucleation in DC bias-enhanced microwave plasma chemical vapor deposition (MPCVD) system, selective growth of seeding using diamond-particle-mixed photoresist, and selective etching of oxygen ion beam using Al as the mask. It was show that high selectivity and precise patterns had been achieved, and all the processes were compatible with IC process.
