<P>Salt-tolerance was studied in transgenic potato. It was conferred by overexpression of ascorbate pathway enzyme (D-galacturonic acid reductase, GalUR). As genetic engineering of the GalUR gene in potato enhances its ascorbic acid content (L-AsA), and subsequently plants suffered minimal oxidative stress-induced damage, we now report on the comprehensive aptness of this engineering approach for enhanced salt tolerance in transgenic potato (Solanum tuberosum L. cv. Taedong Valley). Potatoes overexpressing GalUR grew and tuberized in continuous presence of 200 mM of NaCl. The transgenic plants maintained a higher reduced to oxidized glutathione (GSH:GSSG) ratio together with enhanced activity of glutathione dependent antioxidative and glyoxalase enzymes under salinity stress. The transgenics resisted an increase in methylglyoxal that increased radically in untransformed control plants under salinity stress. This is the first report of genetic engineering of ascorbate pathway gene in maintaining higher level of GSH homeostasis along with higher glyoxalase activity inhibiting the accumulation in methylglyoxal (a potent cytotoxic compound) under salt stress. These results suggested the engineering of ascorbate pathway enzymes as a major step towards developing salinity tolerant crop plants.</P>
Cell Cycle Regulation and Induction of Apoptosis by β-carotene in U937 and HL-60 Leukemia Cells

In this communication, we report the efficacy of $\beta$-carotene towards differentiation and apoptosis of leukemia cells. Dose ($20{\mu}M$) and time dependence (12 h) tests of $\beta$-carotene showed a higher magnitude of decrease (significance p < 0.05) in cell numbers and cell viability in HL-60 cells than U937 cells but not normal cell like Peripheral blood mononuclear cell (PBMC). Microscopical observation of $\beta$-carotene treated cells showed a distinct pattern of morphological abnormalities with inclusion of apoptotic bodies in both leukemia cell lines. When cells were treated with $20{\mu}M$ of $\beta$-carotene, total genomic DNA showed a fragmentation pattern and this pattern was clear in HL-60 than U937 cells. Both the cell lines, on treatment with $\beta$-carotene, showed a clear shift in $G_1$ phase of the cell cycle. In addition the study also revealed anti-oxidant properties of $\beta$-carotene since there was reduction in relative fluorescent when treated than the control at lower concentration. Collectively this study shows the dual phenomenon of apoptosis and differentiation of leukemia cells on treatment with $\beta$-carotene.
Level of Self-Efficacy of Science Teachers Towards Engaging Students

The study aimed at investigating the level of self-efficacy of science teachers towards student engagement. Although the general self-efficacy of teachers has been explored a lot but the efficacy of science teacher has not been explored more and student engagement is crucial towards understanding of science concepts. Therefore, the main purpose of the study was to find the self-efficacy of science teachers mainly towards three dimensions of student engagement (Behavioral, Cognitive and Emotional engagement). The study was carried with 150 secondary science teachers as Kathmandu. Secondary science teachers were found to be moderately high efficacious in all aspects of behavioral engagement of students except four aspects in which they showed moderate efficacy. Regarding the self-efficacy on cognitive engagement they were moderately efficacious in two aspects and were found to be moderately high efficacious in other all aspects. However, science teachers' self-efficacy was found to be moderately high. Teachers were found to be efficacious in making clasroom constructive, developing collaborative skill and high order thinking among the students. However in some aspects they were found less efficacious.

Background: Minor saponins or human intestinal bacterial metabolites, such as ginsenosides Rg3, F2, Rh2, and compound K, are more pharmacologically active than major saponins, such as ginsenosides Rb1, Rb2, and Rc. In this work, enzymatic hydrolysis of ginsenoside Rb1 was studied using enzyme preparations from cultured mycelia of mushrooms. Methods: Mycelia of Armillaria mellea, Ganoderma lucidum, Phellinus linteus, Elfvingia applanata, and Pleurotus ostreatus were cultivated in liquid media at $25^{\circ}C$ for 2 wk. Enzyme preparations from cultured mycelia of five mushrooms were obtained by mycelia separation from cultured broth, enzyme extraction, ammonium sulfate (30-80%) precipitation, dialysis, and freeze drying, respectively. The enzyme preparations were used for enzymatic hydrolysis of ginsenoside Rb1. Results: Among the mushrooms used in this study, the enzyme preparation from cultured mycelia of A. mellea (AMMEP) was found to convert ginsenoside Rb1 into compound K with a high yield, while those from G. lucidum, P. linteus, E. applanata, and P. ostreatus produced remarkable amounts of ginsenoside Rd from ginsenoside Rb1. The enzymatic hydrolysis pathway of ginsenoside Rb1 by AMMEP was $Rb1{\rightarrow}Rd{\rightarrow}F2{\rightarrow}$ compound K. The optimum reaction conditions for compound K formation from ginsenoside Rb1 were as follows: reaction time 72-96 h, pH 4.0-4.5, and temperature $45-55^{\circ}C$. Conclusion: AMMEP can be used to produce the human intestinal bacterial metabolite, compound K, from ginsenoside Rb1 with a high yield and without food safety issues.

This research work aims at exploring factors like servicecape, trust and customers’ preference and the relationship with customer satisfaction and loyalty relationship with customer customer satisfaction and loyalty in the restaurant industry. The Indian restaurants are employed as target samples due to easy access in collecting the data and also due to the fact that these restaurants are a single function activity unlike bit hotels and shopping malls. The servicecape and trust has a position influence on the outcome variable, customers’ preference of restaurants which relies on customers’ own knowledge (CPRRK), as is evident from the fact that majority of customers select restaurants on the basis of their past experience. The questionnaire for the research survey was divided into two parts: a. questions relating to servicecape were designed on the basis of Bitner’s, model and b.for question relating to trust, seven aspects designed by Hess were used. Customers were asked to answer, questions regarding restaurant’s reputation or image caused by the servicecape issues and other aspects relating to trust.
A Review: Relating Low Level Features to High Level Semantics in CBIR

Content based image retrieval is the technique of effective retrieval of digital pictures from an oversized store of variety of pictures. In CBIR, the focus is mostly on extracting features from the queried image and from the images stored in the database for finding the similarity between these features to retrieve images which are similar visually. CBIR becomes tougher once focus goes to reducing the semantic gap or the linguistics gap between low level features and high level semantics. This survey provides a short summary regarding low level features and high level linguistics that are thought of in CBIR for economical and correct retrieval.
