        Screening of Mutant Strain Streptomyces mediolani sp. AC37 for (-)-8-O-Methyltetrangomycin Production Enhancement

        Streptomyces mediolani sp. AC37 was isolated from the root system of higher plant Taxus baccata and produced metabolite identified as (-)-8-O-methyltetrangomycin according to LC/MS/MS analysis. In our screening program for improvements of bioactive secondary metabolites from plant associate streptomycetes, mutation was used as a tool for the induction of genetic variations for selection of higher (-)-8-O-methyltetrangomycin producers of isolates. S. mediolani sp. AC37 was treated with UV irradiation and chemical mutagenic treatment (N-nitroso-N-methyl-urea). The radical scavenging and antioxidant capacity of (-)-8-O-methyltetrangomycin and extracts isolated from mutants were tested using EPR spin trapping technique and ABTS˙+ assay. Comparison of electron microscopic images of Streptomyces sp. AC37 and mutant strains of Streptomyces sp. AC37 revealed substantial differences in morphology and ultrastructure.

        A Progressive Physical Therapy Plan of Care for a Patient with Bilateral Knee Pain due to Degenerative Joint Disease following a Cerebral Vascular Accident: A Case Report

        Purpose The purpose of this case report was to describe the effects of a 6-week physical therapy program consisting of strengthening exercises, balance training, stretching and self- managing education of an adult with bilateral knee osteoarthritis (OA) and cerebral vascular accident (CVA) during an outpatient episode of care. Methods The individual was a 62-year-old female with a history of bilateral knee osteoarthritis and recent history of mild CVA. The subject participated in a 6-week physical therapy program consisting of strengthening exercises, balance training, stretching and self- managing education. The subject was seen 3 times a week, with a day of recovery between each therapy session. The Lower Extremity Functional Scale (LEFS), Berg Balance Scale (BBS), Manual Muscle Test (MMT), Time Up and Go (TUG), Numeric Pain Rating Scale (NPRS), 10-Meter Walk Test (10MWT) and Five Time Sit to Stand Test (FTSST) were used to assess progress in this subject. Results Outcome measures were used at evaluation and were administered every two weeks. The Timed Up and Go improved from 22 seconds to 12 seconds. The Five Time Sit to Stand Test improved from 39 seconds to 16 seconds, while the Berg Balance Scale improved from 43 to 52 points. Lower Extremity Functional Scale improved by 9 points and gait speed improved by 0.27m/s. The Numeric Pain Rating Scale was monitored throughout the course of treatment, which served as a guide that pain was not exacerbated and that the exercise program was well tolerated by the subject. Conclusion This case report describes an exercise program that significantly decreasing pain, while improving balance, functional strength, and gait kinematics with no adverse effect in a subject with bilateral knee osteoarthritis with recent mild CVA. We believe that a significant reduction of pain induces a better functional performance in the subject with OA and CVA.

        A review of articles in the Journal of Global Scholars of Marketing Science (JGSMS) special issue on Marketing Management in International Contexts

        This is a review of four articles published in the JGSMS 2019 special issue on Marketing Management in International Contexts. All papers published in this special issue were peer-reviewed, with oversight from the authors of this document in the role of Guest Editors. Three articles examine aspects of digital communication influences on behavior and attitudes, while one studies consumer preferences in the context of differing cultural and technological backgrounds. Generational cohorts and cultural clusters are examined through these studies. Overall, the articles in this special issue contribute to our knowledge of how younger persons use and are shaped by the digital world they have been born into, but also to the persistence of underlying cultures.

        Effect of Solidification Path and Contraction on the Cracking Susceptibility of Carbon Peritectic Steels

        The solidification path and contraction generated during the solidification of three carbon steels were studied. Two of the carbon steels were of hypo-peritectic chemical composition, with 0.11 % C and 0.15 % C, while one of the carbon steels were hyper-peritectic with 0.16 % C. The steels with 0.11 % C and 0.16 % C solidified as expected due to their chemical composition. In contrast, the chemically hypo-peritectic steel with 0.15 % C solidified as hyper-peritectic steel, which was associated with the microsegregation of Mn. For the steel exhibiting a hypo-peritectic solidification path, peritectic transformation occurred at solid fraction values higher than 0.9, where it was assumed that the contraction generated in the mushy shell cannot be fed by the liquid. However, for steels exhibiting a hyper-peritectic solidification path, peritectic transformation began at solid fraction values lower than 0.9, where the contraction generated by the peritectic transformation was partly fed by the liquid. Hence, the highest cracking susceptibility was associated with the hypo-peritectic solidification mode.

        pep27 and LytA in Vancomycin-Tolerant Pneumococci

        Vancomycin therapy failure due to the emergence of tolerance in pneumococci is increasing. The molecular mechanism of tolerance is not clear, but lytA and pep27 are known to be involved. Our aim was to evaluate the expression of both genes in vancomycin-tolerant Streptococcus pneumoniae (VTSP) strains. Eleven VTSP strains from a total of 309 clinical isolates of S. pneumoniae from 1997 to 2006 were classified according to the criteria of Liu and Tomasz. All VTSP strains were evaluated for susceptibility according to CLSI criteria, serotype by the Quellung test, and clonality by PFGE. The expressions of lytA and pep27 were analyzed in different growth phases by RT-PCR with and without vancomycin. Eighty-two percent of VTSP strains showed resistance to penicillin, and 100% were sensitive to vancomycin and cefotaxime. The most frequent serotypes of VTSP strains were 23F (4/11) and 6B (3/11). Clonal relationship was observed in only two strains. No significant changes were observed in pep27 expression in the three phases of growth in VTSP strains with and without vancomycin. Interestingly, pep27 expression in the stationary phase in the non-tolerant reference strain R6 was significantly higher. However, no significant differences in lytA expression were observed between VTSP and R6 strains during the phases of growth analyzed. The absence of changes in pep27 expression in VTSP strains in the stationary phase may be related to their ability to tolerate high antibiotic concentrations, and thus, they survive and remain in the host under the antibiotic selective pressure reflected in therapeutic failure.

        Remote sensing based forest canopy opening and their spatial representation

        The use of remote sensing in natural resource management is an easily accessible input for obtaining detailed information on the ground and landscape. There is a wide range of procedures to analyze the forest canopy through satellite images. The purpose of this work is to obtain a map of forest opening with remote sensing by relating several vegetation indices, Kauth-Thomas transformation and texture filters, to a Landsat 8OLI image. A factor analysis was made to evaluate the contribution of these variable to identify the opening of the forest cover, yielding a r2 ¼ 76%. The results show that the Modified Soil Adjusted Vegetation Index (MSAVI), Soil Adjusted Vegetation Index (SAVI), and brightness factor have the best correlation (0.225–0.216 component coefficient). The resulting model was reclassified into five categories of forest opening and associated with land use data from the National Institute of Statistics and Geography (INEGI-M exico). Thus, 95% of human settlements have a canopy opening between medium and very high, the crops areas 72%, and the low deciduous forest with secondary shrub vegetation 100% of the opening. Coniferous and mixed forests have a low to very low canopy opening 46% and 55%, respectively of their surface. The forests with secondary vegetation, both shrub and arboreal, present greater openness than the same forests in the primary state. Verification of the spatial representation data of canopy opening was made by comparing 94 hemispheric photographs with 94 sites located in open areas obtaining an r ¼ 0.57. This work offers a simple and straightforward methodology, easily replicable in different types of vegetation using free satellite imagery. Hence, it is a helpful tool for decision-makers when considering the general status of conservation of forest systems and their spatial distribution.

        Parathyroid Crisis as Presentation of Atypical Parathyroid Adenoma: Two Diagnostically Challenging Cases

        Atypical parathyroid adenoma (APA) is a rare cause of primary hyperparathyroidism (PHPT) and represents a diagnostic challenge since it is an intermediate form of parathyroid neoplasm of uncertain malignant potential with atypical histological features that require differential diagnosis of parathyroid carcinoma (PC). We present 2 cases of parathyroid crisis as a presentation of APA. The first case was that of a 56-year-old man with parathyroid crisis, constitutional syndrome, and anemia, with evidence of APA after en bloc resection, evolving with hungry bone syndrome after surgery and curation criteria at 6 months after parathyroidectomy (PTX). The second case was a 64-year-old woman with acute chronic kidney disease and parathyroid crisis, with evidence of APA after selective PTX and >50% reduction in parathyroid hormone levels after surgery; however, persistent PHPT at 6 months post-surgery was observed. These cases represented a diagnostic challenge due to their rare clinical presentation (parathyroid crisis), with a heterogeneous spectrum of target organ damage and infrequent symptoms (constitutional syndrome and acute chronic renal disease), in turn caused by a rare pathology (APA). The presentation of these patients may be indicative of PC; however, histopathological diagnosis is a key to the diagnosis of APA. The differential diagnosis of APA vs. PC in clinical practice is indispensable.

        Agave tequilana Counteracts Chronic Hypertension and Associated Vascular Damage

        Systemic arterial hypertension (SAH) is a health problem of great importance worldwide, and endothelial dysfunction underlies SAH development. This condition's main characteristics include vasoconstriction, inflammation, oxidative stress, and procoagulant and proliferative states. This study's objective was to evaluate the antihypertensive, anti-inflammatory, and antioxidant effects of the whole extract and fractions of Agave tequilana in a murine model of SAH. SAH was induced in male ICR or CD-1 (Strain obtained from animals from Charles River Laboratories, Massachusetts) mice by intraperitoneal administration of angiotensin II (AGII) (0.1 μg/kg) for 4 weeks, and then A. tequilana treatments were co-administered with AGII. At the end of the experiment, systolic and diastolic blood pressure were measured and the kidneys were dissected to quantify interleukin (IL)-1β, IL-6, tumor necrosis factor-alpha, IL-10, and malondialdehyde (MDA). The whole extract and the fractions of A. tequilana were chemically characterized using gas chromatography-mass spectrometry. The results indicate that the whole extract (At-W) and At-AcOEt fraction treatment are the most efficient in lowering blood pressure, although all the treatments had an immunomodulatory effect on the cytokines evaluated and an antioxidant effect on lipid peroxidation. Finally, the chromatographic profile shows that the integral extract and fractions of A. tequilana contained phytol (M)3,7,11,15-Tetramethyl-2-hexadecen-1-ol; 9,12-octadecadienoic acid; hentriacontane; 9,19-cyclolanost-24-en-3-ol,(3b); t-sitosterol; and stigmasta-3,5-dien-7-one.
