Distribution of T and B Lymphocytes in Peripheral Blood and Lymphoid Tissues of Bovine

소의 순환혈액 및 림프조직내의 T 및 B 림프구의 분포를 파악하고자 소흉선세포를 토끼에 과면역시켜 얻은 항혈청을 소의 적혈구, 간장분만 및 골수세포에 순차적으로 흡착시킨 항융선세포혈철(RABTS), 성숙 T 림프구에만 특이성을 가진 단클론항체 BLT_2, B림프구와 소수의 대식세포에 특이성을 가진 단클론항체 _6E_12 등을 1차항체로 이용하여 ABPC법으로 T 및 B 림프구를 동정한 바 다음과 같은 결과를 얻었다. RABTS 및 BLT_1으로 동정한 순환혈액내 단핵세포 (PB-MNC)중 T림프구의 비율은 각각 70.9±5.5% 및 59.0±8.7%이었으며, PB-MNC중 nylon wool 비부착세포를 BLT_1 및 _6E_12로 동정한 T 및 B 림프구의 비율은 각각 91.6±1.0%, 9.6±0.8%이였다. 또한 BLT_1 및 _6E_12로 동정한 서혜림프질, 장간막림프절, 비장 및 흉선의 T/B림프구의 비율은 각각 56.4±6.2/45.3±7.4%, 55.6±7.7/42.3±5.8%, 48.6±5.1/48.5±6.2% 및 23.0±4.8/5.6±2.1%이었으며, RABTS로 동정한 이들 림프조직내의 T림프구의 비율은 각각 76.4±3.4%, 74.2±8.2%, 73.9±5.5% 및 95.6±2.8%이였다. 이상의 결과로 미루어 PB-MNC내의 T림프구의 분리는 nylon wool column법이 보다 효과적임을 알 수 있었다.

<P>Background: Psoriasis is one of the most common chronic inflammatory diseases of the skin. Recently, IL-17-producing T cells have been shown to play a critical role in psoriatic inflammation. Programmed cell death 1 (PD-1) is a coinhibitory receptor expressed on T cells in various chronic inflammatory diseases; however, the expression and function of PD-1 during psoriatic inflammation have not previously been characterized. Objective: We examined PD-1 expression on IL-17A-producing T cells from imiquimod-treated mice and patients with psoriasis. Additionally, we investigated the therapeutic effect of recombinant programmed cell death ligand 1 (PD-L1) protein on imiquimod-induced psoriatic inflammation. Methods: PD-1 expression on IL-17A-producing gamma delta T cells from imiquimod-treated mice was examined by means of multicolor flow cytometric analysis. In the psoriatic skin of patients, PD-1 and IL-17A expression was analyzed by using immunofluorescence. The therapeutic effect of PD-L1-Fc fusion protein (PD-L1-Fc) was assessed in imiquimod-treated mice ex vivo and in vivo. Results: During imiquimod-induced psoriatic inflammation, PD-1 is overexpressed on CD27(-)V gamma 1(-) gamma delta T cells. Furthermore, PD-1 expression on IL-17A(+) T cells was confirmed in psoriatic skin tissues from patients and imiquimod-treated mice. In the CD27(-)V gamma 1(-) gamma delta T-cell population, V gamma 4(-) gamma delta T cells with V gamma 6 mRNA expression showed a high level of PD-1 expression. Furthermore, these PD-1(hi)V gamma 4(-)(V gamma 6(+)) gamma delta Tcells were specialized for anti-CD3-induced IL-17A production, which was inhibited by PD-L1-Fc treatment. In imiquimod-treated mice PD-L1-Fc reduced psoriatic inflammation when given alone and enhanced the therapeutic effect of anti-p40 when given in combination. Conclusion: PD-1 is overexpressed in IL-17A-producing T cells in both imiquimod-treated mice and patients with psoriasis. Moreover, recombinant PD-L1-Fc alleviates psoriatic inflammation in imiquimod-treated mice.</P>

The present study conducted a comparison on tree growth and productivity of high-density apple orchards by several orchard management systems: making scaffolds by heading-back and thinning out pruning (T-1), maintaining scaffolds upward and bearing shoots downward (T-2), managing branches slightly upward without heading-back (T-3), keeping leaders downward and shoots pending (T-4), maintaining leaders high and branches horizontal with severe pinching (T-5), making leaders with lower branches vigorous and upper shoots pending (T-6), and controlling very high planting density with bending branches (T-7). In conclusion, the orchards of (T-5) and (T-6) management systems showed a superior performance in controlling tree growth, productivity, and quality of fruits. Also, superior management efficiency was obtained in the orchards of (T-5) and (T-6).

The irreversible properties of a rare-earth high-T<SUB>c</SUB> superconductor are investigated using grain-aligned Sm<SUB>1-x</SUB>Eu<SUB>x</SUB>Ba<SUB>2</SUB>Cu<SUB>3</SUB>O<SUB>7-δ</SUB>; (x = 0.0, 0.3, 0.5, 0.7, and 0.9) samples. The samples were synthesized using the usual solid-state reaction method. It is found that the transition temperature T<SUB>c</SUB> and the critical current density J<SUB>c</SUB> calculated by the Bean model are significantly improved in the optimally doped samples (x = 0.5 and 0.7). Furthermore, the double logarithmic graph of the irreversibility line B<SUB>irr</SUB> in the grain-aligned series samples obeys approximately a power law dependence (1-T/T<SUB>c</SUB>)<SUP>n</SUP>. Results show that in the case of optimally doped samples with x = 0.5 and 0.7 the graph of B<SUB>irr</SUB> is fitted well with the relation (1-T/T<SUB>c</SUB>)<SUP>n</SUP> with the model-dependent parameter n ∼1.5 without any crossover in the entire temperature range, while in samples with x = 0.0, 0.3, and 0.9 B<SUB>irr</SUB> does show a prominent crossover and can be characterized with n ∼1.5 only near T<SUB>c</SUB>, above which B<SUB>irr</SUB> varies more rapidly. Finally, it is suggested that B<SUB>irr</SUB> is strongly dependent on the extrinsic properties of our grain-aligned samples.

A survey was conducted to determine the distribution and prevalence of Toxoplasma gondii and Babesia microti infections in small mammals captured in Gyeonggi and Gangwon Provinces, Republic of Korea (ROK). The serological prevalence of T. gondii (ELISA) and B. microti (IFAT) was 2.3% (15/667) and 2.1% (14/667), respectively. DNA extracts from small mammal heart tissues were screened by PCR for T. gondii and B. microti targeting regions of the GRA5 gene and the 18S rRNA and β-tubulin genes, respectively. Only 0.17% (1/578) of Apodemus agrarius was positive of T. gondii by PCR, while 0.52% (3/578) was positive of B. microti. All other small mammal species [Micromys minutus (16), Mus musculus (3), Myodes regulus (22), Microtus fortis (6), and Crocidura lasiura (42)] were negative for both T. gondii and B. microti. Based on sequence polymorphism and phylogenetic analysis, T. gondii closely aligned with Type I, a highly virulent strain, while B. microti positive samples closely aligned with US-type B. microti and others observed in the ROK, Russia, and Japan. These results indicate that A. agrarius is a reservoir for both T. gondii and B. microti in the ROK.

This study was conducted to investigate assay of high fiber diet in vitro digestibility according to digestive enzyme and time. Treatment of this study included: 1) CON (basic method) 2) T1 (sample amount: 2 times; enzyme level: 2 times) 3) T2 (sample amount: 2 times, enzyme level: 2 times, assay time: 1.5 times) 4) T3 (sample amount: 2 times, enzyme level: 2 times, assay time: 2 times). With regard to dry matter (DM), organic matter (OM) digestibility, T3 and CON were higher than T1 and T2 (P<0.05). Also, with regard to DM and OM digestibility by time of digestion, base was similar T2. In conclusion, in vitro digestibility was closely related to digestion time.

High Velocity Oxygen Fuel (HVOF) thermal spray coating of nano size WC-Co powder (nWC-Co) has been studied as one of the most promising candidate for the possible replacement of the traditional hard plating in some area which causes environmental and health problems. nWC-Co powder was coated on Inconel 718 substrates by HVOF technique. The optimal coating process obtained from the best surface properties such as hardness and porosity is the process of oxygen flow rate (FR) 38 FMR, hydrogen FR 57 FMR and feed rate 35 g/min at spray distance 6 inch for both surface temperature 25℃ and 500℃. In coating process a small portion of hard WC decomposes to less hard W₂C, Wand C at the temperature higher than its decomposition temperature 1,250°C resulting in hardness decrease and porosity increase. Friction coefficient increases with increasing coating surface temperature from 0.55-0.64 at 25℃ to 0.65-0.76 at 500℃ due to the increase of adhesion between coating and counter sliding surface. Hardness of nWC-Co is higher or comparable to those of other hard coatings, such as A1₂O₃, Cr, Cr₂O₃ and HVOF Tribaloy 400 (T400). This shows that nWC-Co is recommendable for durability improvement coating on machine components such as high speed spindle.

Hard chrome plating has been used in surface hard coating over 50 years both for applying hard coating and re-building of worn components. Hard chrome plating solution and mist pollute environment with very toxic Cr??(hex-Cr) known as carcinogen which causes lung cancer. High velocity oxy-fuel (HVOF) thermal spray coatings of WC base cermet and Co-alloy powders are the most promising candidates for the replacement of the traditional hard chrome plating. Surface properties, wear, and friction behaviors of micron size Co-alloy (T800) and micron size WC-12Co (WC-Co) have been studied for the application as hard coatings. The temperature dependence of wear and friction behaviors of T800 and WC-Co have been investigated at the temperature of 25℃ and 538℃ for the application to high speed spindle.

As a part of ongoing studies in developing novel anticancer agents, a series of modified 2,4-diaryl-5H-indeno[1,2-b]pyridines were designed, and synthesized by introducing hydroxyl and chlorine moieties. They were evaluated for topoisomerase inhibitory activity and cytotoxicity against HCT15, T47D, and HeLa cancer cell lines. This modification allowed us to demonstrate structure-activity relationship (SAR) study with respect to the non-substituted 2,4-diaryl-5H-indeno[1,2-b]pyridines. Compounds (2, 3, 4, 5, 8, and 9) with meta or para hydroxyl group on 2 or 4-phenyl ring have enhanced topo I and II inhibitory activity and cytotoxicity. However, additional substitution of chlorine group on furyl or thienyl ring (11, 12, 14, 16-18) generally reduced topo I and II inhibitory activity but improved cytotoxicity. The observation of cytotoxic properties and SAR study according to the position of hydroxyl and chlorine group will provide valuable insight for further study of development of novel anticancer agents with related scaffolds.
