발효 황기 추출물의 항산화, 항염, 콜라게나아제 및 엘라스타아제 활성 저해 효과

본 연구는 발효 황기 추출물이 발효 전 황기 추출물 보다 항산화, 항염, 콜라게나아제 및 엘라스테아제 억제 효과가 뛰어난지 평가하고자 하였다. FAE의 총 폴리페놀 및 플라보노이드 함량이 AE보다 높은 것으로 나타났으며, DPPH, ABTS 및 환원력 측정에서 FAE에서 AE보다 높은 항산화 활성을 보였다. HaCaT 세포에 SIN-1을 처리하여 효소 활성을 측정하였을 때 FAE 추출물의 처리는 감소된 SOD, catalase, GPx 활성을 AE보다 증가시키는 것을 확인하였다. 또한 LPS로 자극된 RAW264.7 세포에서 증가된NO, PGE2 및 IL-6의 발현을 AE보다 FAE에서 더 억제하는 것을 확인하였다. 또한, collagenase와 elastase의 억제활성이 AE보다 FAE에서 우수하였다. 종합하자면, FAE는 AE보다 항산화, 항염, 콜라게나아제 및 엘라스타아제 억제 효능이 있는 천연기능성 소재 및 건강기능식품 등의 소재로 활용될 수 있을 것이라 사료된다. We aimed to evaluate the antioxidant, anti-inflammatory, anti-collagenase, and anti-elastase effects of fermented and non-fermented Astragalus membranaceus extracts (FAE and AE). The total polyphenol and flavonoid contents of FAE were higher than those of AE. DPPH, ABTS, and reducing power assays confirmed that the radical scavenging efficacy and reducing power of FAE were higher than those of AE. In SIN-1-stimulated HaCaT cells, reduced superoxide dismutase, catalase, and glutathione peroxidase activity levels were rescued in the FAE group but not in the AE group. FAE also suppressed increases in NO, PGE2, and IL-6 levels more potently than AE in LPS-stimulated RAW264.7 cells. FAE also exhibited more potent collagenase and elastase inhibitory activities than AE. In summary, this study suggests that FAE can be used in functional foods and cosmetics with antioxidant, anti-inflammatory, anti-collagenase, and anti-elastase properties.

<B>Purpose</B><P>Although many studies have demonstrated that smoking, alcohol, obesity, and insulin resistance are risk factors for cancer, the role of those factors on cancer survival has been less studied.</P><B>Patients and Methods</B><P>The study participants were 14,578 men with a first cancer derived from a cohort of 901,979 male government employees and teachers who participated in a national health examination program in 1996. We obtained mortality data for those years from the Korean Statistical Office. We used a standard Poisson regression model to estimate the hazard ratio (HR) for survival in relation to smoking, alcohol, obesity, and insulin resistance before diagnosis.</P><B>Results</B><P>Poor survival of all cancer combined (HR, 1.24; 95% CI, 1.16 to 1.33), cancer of the lung (HR, 1.45; 95% CI, 1.15 to 1.82), and cancer of the liver (HR, 1.36; 95% CI, 1.21 to 1.53) were significantly associated with smoking. Compared with the nondrinker, heavy drinkers had worse outcomes for head and neck (HR, 1.85; 95% CI, 1.23 to 2.79) and liver (HR, 1.25; 95% CI, 1.11 to 1.41) cancer, with dose-dependent relationships. Patients with a fasting serum glucose level above 126 mg/dL had a higher mortality rate for stomach (HR, 1.52; 95% CI, 1.25 to 1.84) and lung (HR, 1.48; 95% CI, 1.18 to 1.87) cancer. Higher body mass index was significantly associated with longer survival in head and neck (HR, 0.54; 95% CI, 0.39 to 0.74) and esophagus (HR, 0.44; 95% CI, 0.28 to 0.68) cancer.</P><B>Conclusion</B><P>Prediagnosis risk factors for cancer development (smoking, alcohol consumption, obesity, and insulin resistance) had a statistically significant effect on survival among male cancer patients.</P>

<P> According to the advent of ubiquitous world, the requirement for the mobile information devices with high display quality and compact device size is surprisingly increased. Among the diverse candidates for this mobile display system for mobile devices, Liquid Crystal on Silicon (LCoS) is the most competitive device due to the high aperture ratio and simple fabrication process. The inorganic liquid crystal (LC) alignment layers are widely used for LCoS devices because of the thermal and photochemical stability. In this work, the reactive sputtering was selected for the preparation method of inorganic LC alignment layers. The nitrogen (N2) gas had the effect on the deposition process of SiO2 and the surface morphology of SiO2 thin layers were affected by the N2 mixing ratio of sputtering gas. In addition, the LC alignment properties on SiO2 thin layer were also closely related with the N2 mixing ratio and other sputtering conditions. In the case of high RF power of reactive sputtering, the N2 mixing ratio has little effect on the LC alignment on SiO2 thin layers. However, in the case of low RF power of reactive sputtering, the LC alignment properties were enfeebled by increasing the N2 mixing ratio. This result might be attributed to the change of the surface morphology of inorganic SiO2 thin layers.</P>

Background: Idiopathic pulmonary fibrosis (IPF) is a progressive and lethal lung disease characterized by the accumulation of excessive fibroblasts and myofibroblasts in the extracellular matrix. The transforming growth factor β1 (TGF-β1).induced epithelial-to-mesenchymal transition (EMT) is thought to be a possible source of fibroblasts/ myofibroblasts in IPF lungs. We have previously reported that apolipoprotein A1 (ApoA1) has anti-fibrotic activity in experimental lung fibrosis. In this study, we determine whether ApoA1 modulates TGF-β1.induced EMT in experimental lung fibrosis and clarify its mechanism of action. Methods: The A549 alveolar epithelial cell line was treated with TGF-β1 with or without ApoA1. Morphological changes and expression of EMT-related markers, including E-cadherin, N-cadherin, and α-smooth muscle actin were evaluated. Expressions of Smad and non-Smad mediators and TGF-β1 receptor type 1 (TβRI) and type 2 (TβRII) were measured. The silica-induced lung fibrosis model was established using ApoA1 overexpressing transgenic mice. Results: TGF-β1.treated A549 cells were changed to the mesenchymal morphology with less E-cadherin and more N-cadherin expression. The addition of ApoA1 inhibited the TGF-β1.induced change of the EMT phenotype. ApoA1 inhibited the TGF-β1.induced increase in the phosphorylation of Smad2 and 3 as well as that of ERK and p38 mitogenactivated protein kinase mediators. In addition, ApoA1 reduced the TGF-β1.induced increase in TβRI and TβRII expression. In a mouse model of silica-induced lung fibrosis, ApoA1 overexpression reduced the silica-mediated effects, which were increased N-cadherin and decreased E-cadherin expression in the alveolar epithelium. Conclusion: Our data demonstrate that ApoA1 inhibits TGF-β1.induced EMT in experimental lung fibrosis.

Idiopathic interstitial pneumonia (IIP) is a histologically identifiable pulmonary disease without a known cause that usually infiltrates the lung interstitium. IIP is largely classified into idiopathic pulmonary fibrosis, idiopathic non-specific interstitial pneumonia, respiratory bronchiolitis-interstitial lung disease (ILD), cryptogenic organizing pneumonia, desquamative interstitial pneumonia, and acute interstitial pneumonia. Each of these diseases has a different prognosis and requires specific treatment, and a multidisciplinary approach that combines chest high-resolution computed tomography (HRCT), histological findings, and clinical findings is necessary for their diagnosis. Diagnosis of IIP is made based on clinical presentation, chest HRCT findings, results of pulmonary function tests, and histological findings. For histological diagnosis, video-assisted thoracoscopic biopsy and transbronchial lung biopsy are used. In order to identify ILD associated with connective tissue disease, autoimmune antibody tests may also be necessary. Many biomarkers associated with disease prognosis have been recently discovered, and future research on their clinical significance is necessary. The diagnosis of ILD is difficult because patterns of ILD are both complicated and variable. Therefore, as with other diseases, accurate history taking and meticulous physical examination are crucial.

Background OAT is a key enzyme that converts ornithine into proline, an essential precursor of collagen. We previously reported that OAT expression is increased in IPF lung and inhibition of OAT attenuate bleomycin-induced lung fibrosis. EMT is the main process for fibroblast production during lung fibrosis. We investigate the role of OAT in lung fibrosis especially focused on the EMT pathway and its clinical implication in IPF. Methods OAT knockdown A549 alveolar epithelial cell line was treated with TGF-β1. Morphological changes and expression of EMT-related markers, including E-cadherin, N-cadherin, and α-smooth muscle actin were evaluated. Expressions of Smad and non- Smad mediators and EMT- related transcription factors were measured. We measured the OAT level from bronchoalveolar lavage fluid (BALF) in IPF and controls. Results OAT knockdown inhibited the TGF-β1 induced change of the EMT phenotype. OAT knockdown attenuates the TGF-β1 induced increase in the phosphorylation of Smad 2 and 3 as well as that of ERK and p38 mitogen-activated protein kinase mediators. In addition, OAT knockdown reduced TGF-β1 induced expression of EMT transcription factors; Twist, Slug, and Snail 1. OAT levels in IPF BALF are reversely correlated with FVC (% pred.) and DLCO (% pred.). Conclusions Inhibition of OAT attenuates BLM induced lung fibrosis via suppressing of TGF-β1 induced EMT pathways. OAT lung level is associated with lung function decline in IPF. Taken together, regulation of OAT could be a therapeutic target and surrogate marker in fibrotic lung disease. [Grant :NRF-2019R1A2C100635]

Hydrotalcites are anionic clays that are quite prevalent in nature and their importance is growing more and more because of their very wide range of potential applications and uses. Understanding the structural and compositional changes that occur on the molecular scale during the thermal decomposition of hydrotalcite compounds is essential for the basic prediction and comprehensive understanding of the behavior and technical application of these materials. In this study, several hydrotalcite compounds calcined at different temperatures for applications in a chlorine resistant textile were prepared and 27-Aluminm solid-state nuclear magnetic resonance (NMR) spectroscopy was used as a tool to study their local structure and behavior. The changes in the Al coordination of the hydrotalcite compounds were investigated with one dimensional (1D) solid-state magic angle spinning (MAS) NMR spectroscopy. The two broad resonances arising from the structurally different Al coordinations of these compounds were clearly resolved by two dimensional (2D) triple quantum magic angle spinning (3QMAS) NMR spectroscopy.
