This study was designed to determine biochemical and pharmacological properties of a newly synthesized benzimidazole derivative, 2-amino-4, 5-dihydropyrido [1, 2-a] thiazolo [5, 4-g] benzimidazole (YJA20379-5) in vitro and in vivo. In the leaky membrane vesicles of pig gastric mucosa, YJA20379-5 inhibited the $K^+$-stimulated $H^+$, $K^+$-ATPase activity in a concentration- and time-dependent manner, with $IC_{50}$ values being $43{\mu}\textrm{M}$ and $43{\mu}\textrm{M}$ at pH 6.4 and 7.4, respectively. YJA20379-5, given intraduodenally, had a potent inhibitory effect on the gastric acid secretion in pylorus-ligated rats. The $ED_{50}$ value for acid secretion was 15.4 mg/kg. YJA20379-5, administered orally, also suppressed gastric damages induced by water-immersion stress, indomethacin and ethanol, and duodenal damage induced by mepirizole in rats; the $ED_{50}$ values were 17.6, 4.7, 3.0 and 18.7 mg/kg, respectively. Furthermore, repeated oral administration of YJA20379-5 accelerated the spontaneous healing of acetic acid-induced gastric ulcers in rats. It is concluded that the a-ntisecretory activity of YJA20379-5 appears to be associated with inhibition of $H^+$, $K^+$-ATPase, while its antigastric and antiduodenal lesion activities are primarily related to the antisecretory effect.

<P>We used 2-DE and MALDI-TOF/TOF to identify proteins of vascular smooth muscle cells whose expression was or was not altered by exposure to 500 μM H<SUB>2</SUB>O<SUB>2</SUB> for 30 min. We detected more than 800 proteins on silver-stained gels of whole protein extracts from rat aortic smooth muscle strips. Of these proteins, 135 clearly unaffected and 19 having levels altered by exposure to H<SUB>2</SUB>O<SUB>2</SUB> were identified. Protein characterization revealed that the most prominent vascular smooth muscle proteins were those with antioxidant, cytoskeletal structure, or muscle contraction. In addition, cofilin, an isoform of the actin depolymerizing factor family, shifted to its basic site on the 2-DE gel as a result of H<SUB>2</SUB>O<SUB>2</SUB> treatment. In Western blot analysis of proteins from A7r5 aortic smooth muscle cells, the phosphorylation, but not the expression, of cofilin was decreased by H<SUB>2</SUB>O<SUB>2</SUB> in a dose-dependent manner. The H<SUB>2</SUB>O<SUB>2</SUB>-induced dephosphorylation of cofilin and apoptosis was inhibited by Na<SUB>3</SUB>VO<SUB>4</SUB>, an inhibitor of protein tyrosine phosphatase (PTP). These results suggest that cofilin is one of the proteins regulated by H<SUB>2</SUB>O<SUB>2</SUB> treatment in vascular smooth muscle, and has an important role in the induction of vascular apoptosis through PTP-dependent mechanisms.</P>

<P><B>Abstract</B></P> <P>The pathogenesis of atopic dermatitis (AD) involves T helper 2 (Th2) cells, and effective therapies remain elusive due to the paucity of animal models. We aimed to develop a mouse model of an immune system aberration caused by allergen. Experiments were conducted in two phases. In experiment 1, BALB/c mice were sensitized with one of four chemical allergens – toluene diisocyanate (TDI), hexamethylene diisocyanate (HDI), trimellitic anhydride (TMA), or 2,4-dinitrochlorobenzene (DNCB) – for 3 weeks. Based on results of experiment 1, immunological features were compared between TMA-sensitized BALB/c mice and NC/Nga mice, after exposure to mite extracts, harmful chemicals and detergents in experiment 2. Sensitization by allergen caused a large number of pathological changes in the skin, and an increase in mast cell number. TMA-sensitized BALB/c mice models showed higher sensitivity to an environmental allergen than NC/Nga mice did. Overall, the initial sensitization with TMA leads to disturbances in Th2-mediated immunity.</P> <P><B>Highlights</B></P> <P> <UL> <LI> To develop a mouse model of an immune system aberration caused by a chemical allergen. </LI> <LI> Th2-skewed immune response in TMA-sensitized mice closely mimics human AD. </LI> <LI> TMA-sensitized mice showed higher sensitivity to an environmental allergen than NC/Nga mice. </LI> </UL> </P>

We investigated whether increased contractile responsiveness to epidermal growth factor (EGF) is associated with altered activation of mitogen-activated protein kinase (MAPK) in the aortic smooth muscle of deoxycorticosterone acetate (DOCA)-salt hypertensive rats. EGF induced contraction and MAPK activity in aortic smooth muscle strips, which were significantly increased in tissues from the DOCA-salt hypertensive rats compared with those from sham-operated rats. AG1478, PD98059, and LY294002, inhibitors of EGF receptor (EGFR) tyrosine kinase, MAPK/extracellular signal-regulated kinase (ERK) kinase, and phosphatidylinositol 3-kinase (PI3K), respectively, inhibited the contraction and the activity of ERK1/2 that were elevated by EGF. Y27632 and GF109203X, inhibitors of Rho kinase and protein kinase C, respectively, attenuated EGF-induced contraction, with no diminution of ERK1/2 activity. Although EGF also elevated the activity of EGFR tyrosine kinase in both sham-operated and DOCA-salt hypertensive rats, the expression and the magnitude of activation did not differ between strips. These results strongly suggest that EGF induces contraction by the activation of ERK1/2, which is regulated by the PI3K pathway in the aortic smooth muscle of DOCA-salt hypertensive rats.

Combined intramuscular administration of ketamine 8mg/kg. xylazine 2mg/kg were done to evaluate effect of anesthesia in Siberian tiger White tiger and Bengal tiger. Mean induction time(MIT), mean arousal time-(MAT). mean walking time(MWT) and clinical sign were evaluated. The results were as follows. MIT were taken 16.1$\pm$3.5 minutes for Siberian tiger. 15.5$\pm$2.4 minutes for White tiger and 12.3$\pm$2.5 minutes for Bengal tiger. MAT were taken 44.2$\pm$9.5 minutes for Siberian tiger, 48.3$\pm$8.6 minutes for White tiger and 58.7$\pm$5.8 minutes for Bengal tiger. MWT were taken 110.6$\pm$11.6 minutes for Siberian tiger, 106.7$\pm$13.1 minutes for White tiger and 99.6$\pm$10.2 minutes for Bengal tiger. Nausea. vomiting. salivation. severe convulsion. sudden decreased respiration and dyspnea were observed in Siberian tiger during sedation and anesthesia. Also, nausea, vomiting, salivation and convulsion were observed in White tiger and Bengal tiger but the clinical signs were more mild than Siberian tiger. The Bengal tiger which used combined ketamine 5mg/kg , xylazine 1mg/kg were shown reduced induction time compare with combined administration ketamine 8mg/kg, xylazine 2mg/kg in Bengal tiger as 10.8$\pm$32 minutes for MIT. 32.3$\pm$4.3 minutes for MAT and 78.5$\pm$7.3 minutes for MWT Vomiting and convulsion were observed during induction time but there were no nausea and salivation. The present results suggested that preventive methods against severe convulsion and dyspnea should be required in Siberian tiger when combined anesthesia of ketamine 8mg/kg, xylazine 2mg/kg used. Combined anesthesia of ketamine 5mg/kg, xylazine 1mg/kg in Bengal tiger might be very effective for simple surgical procedure and diagnosis.

In this case, Grant's Zebra which had been bred and exhibited in Farmland Zoological Garden for 4 years, died after treated dystocia. Result in autopsy, she had large hepatic abscess, and blood chemistry were increased G07, GPT and total billirubin. The microbiological culture with the hepatic abscess idetified the Streptococcus equi. It is diagnosed of hepatic abscess which was caused by Streptococcus equi infection.

To study effective dossage and administration route for scaling, ketamine HCl/propionyl promazine HCl(ketamine) combination and tiletamine HCl/zolazepam HCl(zoletil) were administered in one hundred six dogs. The dogs were toy poodle, Yorkshire Terrier, Pekingese and Chihuahua. Scaling and polishing time, possible treatment time after the first injection of anesthetics, the number of anethesia added, presence of tongue movement during anesthesia, the presence of swaying sign during recovery and respiration were evaluated. The possible treatment time after the first Injection of anesthetic in toy poodle were 26.3${\pm}$3.0 minutes with intravenous(IM) treatment of ketamine 10mg/kg, and 21.4${\pm}$6.6 minutes with intramuscula(IM) treatment of zoletil 8mg/kg, In Yorkshire Terrier were 19.51: 1.7 minutes with IV treatment of ketamine 10mg/kg. 19.0${\pm}$5.2 minutes IM and 20.8${\pm}$6.1 minutes with IM treatment of zoletil 5mg/kg,24.8${\pm}$3,5 minutes with IM treatment of zoletil 8mg/kg. In pekingese were 27.5${\pm}$2.1 minutes with IM treatment of ketamine 10mg/kg,28.0${\pm}$4.2 minutes with IM treatment of zoletil 8mg/kg. In Chihuahua were 19.5${\pm}$1.9 minutes with IV treatment of ketamine 7mg/kg, 17.5${\pm}$1.7 minutes with IM treatment of ketamine 10mg/kg and 20.3${\pm}$3.8 minutes with IM treatment of zoletil 5mg/kg, 21.2${\pm}$5.5 minutes with IM treatment of zoletil 8mg/kg. Swaying sign was observed in all group during recovery time, espically, in toy poodle and Yorkshire Terrier which administered zoletil 8mg/kg IM showed more severe swaying sign. The present results suggested that injection of zoletil 8mg/kg IM might be relatively effective for scaling in Chihuahua Within 20 minutes treatment for scaling in Yorkshire Terrier and Chihuahua, IM treatment of ketamine 7 to 10mg/kg is recommended.
