        CTCF as a multifunctional protein in genome regulation and gene expression

        <P>CCCTC-binding factor (CTCF) is a highly conserved zinc finger protein and is best known as a transcription factor. It can function as a transcriptional activator, a repressor or an insulator protein, blocking the communication between enhancers and promoters. CTCF can also recruit other transcription factors while bound to chromatin domain boundaries. The three-dimensional organization of the eukaryotic genome dictates its function, and CTCF serves as one of the core architectural proteins that help establish this organization. The mapping of CTCF-binding sites in diverse species has revealed that the genome is covered with CTCF-binding sites. Here we briefly describe the diverse roles of CTCF that contribute to genome organization and gene expression.</P>

      • Remote Memory and Cortical Synaptic Plasticity Require Neuronal CCCTC-Binding Factor (CTCF)

        <P>The molecular mechanism of long-term memory has been extensively studied in the context of the hippocampus-dependent recent memory examined within several days. However, months-old remote memory maintained in the cortex for long-term has not been investigated much at the molecular level yet. Various epigenetic mechanisms are known to be important for long-term memory, but how the 3D chromatin architecture and its regulator molecules contribute to neuronal plasticity and systems consolidation is still largely unknown. CCCTC-binding factor (CTCF) is an 11-zinc finger protein well known for its role as a genome architecture molecule. Male conditional knock-out mice in which CTCF is lost in excitatory neurons during adulthood showed normal recent memory in the contextual fear conditioning and spatial water maze tasks. However, they showed remarkable impairments in remote memory in both tasks. Underlying the remote memory-specific phenotypes, we observed that female CTCF conditional knock-out mice exhibit disrupted cortical LTP, but not hippocampal LTP. Similarly, we observed that CTCF deletion in inhibitory neurons caused partial impairment of remote memory. Through RNA sequencing, we observed that CTCF knockdown in cortical neuron culture caused altered expression of genes that are highly involved in cell adhesion, synaptic plasticity, and memory. These results suggest that remote memory storage in the cortex requires CTCF-mediated gene regulation in neurons, whereas recent memory formation in the hippocampus does not.</P>

        Comparison of Trends in Blood Pressure and the Prevalence of Obesity Among Korean and American Adolescents: A 12-Years Cross-sectional Study

        Objectives: This study was conducted to explore recent trends in the prevalence of pediatric elevated blood pressure and hypertension (HTN) in Korea and the United States, applying the new HTN reference values for adolescents. Methods: This study analyzed 17 339 (8755 Korean and 8584 American) adolescents aged 10 to 17 who participated in the Korea National Health and Nutrition Examination Survey and the National Health and Nutrition Examination Survey, respectively, between 2005 and 2016. HTN was defined using percentile-based reference values for non-overweight adolescents from 7 nations, and obesity was defined by body mass index (BMI) based on country-specific growth charts. All analyses were stratified by sex and year-over-year changes were evaluated by calculating the p for trend. Results: Systolic blood pressure showed a statistically meaningful upward trend in Korean boys and girls, while diastolic blood pressure did not show any significant changes. Both systolic and diastolic blood pressure levels declined among United States boys and girls. The prevalence of elevated blood pressure and HTN remained similar in Korean adolescents, but declined in both sexes among United States adolescents. BMI increased in both sexes among Korean adolescents, although the overweight and obesity rates stayed the same. No significant trends were found in any obesity indices among United States adolescents. The prevalence of elevated blood pressure and HTN by obesity level was higher among Korean adolescents than among their United States peers in both sexes, and the gap became higher at more severe levels of obesity. Conclusions: Despite the strong correlation between obesity and HTN, recent trends in the prevalence of HTN and obesity among Korean and United States adolescents were strikingly different. Follow-up studies are necessary to determine why the prevalence of HTN was more than twice as high among Korean adolescents than among their United States counterparts.

        현대 사회에서는 결정에 어려움을 겪는 사람들이 증가하는 추세이다. 이에 본 연구는 선택이 필요한 상황을 공유하여 여러 사람의 다양한 의견을 듣고 결정을 내리는 데 도움을 얻기 위한 투표 중점 커뮤니티 시스템을 개발하고자 한다. 해당 시스템은 커뮤니티 기능 외에도 사용자 협업 필터링을 이용한 사용자 맞춤 게시글 추천 기능, 반응형 웹 구현, 투표 종료 후 이메일 알림 기능 등의 사용자 편리성과 접근성을 강화하여, 각자 처한 상황에서 도움이 되는 의사 결정을 효율적으로 도출할 수 있는 시스템을 제안한다. The contemporary era witnesses a growing prevalence of decision-making challenges among individuals. In response, this study endeavors to develop a voting-centric community system aimed at aiding individuals in reaching decisions by sharing decision-requiring scenarios and soliciting diverse perspectives from the community. The proposed system seeks to enhance user convenience and accessibility by integrating features such as community functionalities, personalized post recommendations employing user collaboration filtering, responsive web implementation, and post-voting email notification capabilities. The overarching objective is to offer an efficacious system that facilitates proficient decision-making in varied individual contexts.

      • Suitability of Fourier Transform Infrared Spectroscopy as a screening method for the production of useful mutant lines in Panax ginseng

        Panax ginseng C.A Meyer is commonly used in Asian traditional medicine to treat a variety of diseases. Ginsenosides are glycosylated triterpenes, referred to saponins, have been especially noted as active compounds contributing to the various efficacy of ginseng. In this study, we are trying to select high saponin content of ginseng lines from the gamma irradiated adventitious roots. Recently, we have generated several mutant ginseng lines improving ginseniside content by gamma radiation. The mutant lines were selected by phenotypes and ginsenoside content (HPLC analysis) of the irradiated adventitious root lines. However, the ginsenoside content of the mutant lines was not sufficient for commercial use and the selection method was not suitable for large scale of mutant line selection. In this study, we are testing Fourier transfer infrared spectroscopy (FT-IR) as a new selection method of mutant lines in Panax ginseng. About 5,000 pieces of Panax ginseng adventitious roots were exposed to gamma radiation (60Co). Irradiation dosages were 0, 25, 50 and 70Gy. Survival rate of the irradiated samples was evaluated by counting the number of survival main roots after 5 weeks culture in the solid MS medium with NAA, IAA and 5% sucrose. In present, we are collecting the survived adventitious root lines (about 900 lines) from the gamma irradiated ginseng roots for FT-IR and HPLC analysis. After analysis of FT-IR and HPLC, we will assess the suitability of the FT-IR as a screening method for the preparation of mutant lines in ginseng.

        Antioxidant and antiproliferative activities of solvent fractions of broccoli (Brassica oleracea L.) sprout

        Crude methanol extract (ME) of broccoli (Brassica oleracea L.) sprout was fractioned by hexane, chloroform, ethyl acetate, butanol, and water. The contents of total polyphenols (19.89 mg GAE/g) and flavonoids (10.06 mg RE/g) were significantly higher in the butanol fraction (BF) than in the other fractions. The BF showed the highest DPPH ( EC50 = 0.524 mg/mL) and ABTS ( EC50 = 0.180 mg/mL) radical scavenging activities. High-performance liquid chromatography (HPLC) of crude ME showed that the most abundant phenolic compounds were rutin, quercetin, chlorogenic acid, catechin, and p-coumaric acid. The contents of quercetin, chlorogenic acid and p-coumaric acid were higher in the ethyl acetate fraction (EF) and BF than in the other fractions. Antioxidant activity and phenolic compound contents were correlated, suggesting that phenolics were responsible for the antioxidant activity. The hexane fraction (HF) and chloroform fraction (CF) decreased the viability of breast cancer stem cells (BCSCs), and the CF had the highest antiproliferative activity ( IC50 = 69.47 mg/mL). The CF also suppressed the stemness characteristics of BCSCs and induced apoptotic cell death. The most abundant characteristic peak in CF was identified as oleic acid (area = 35.05%) by gas chromatography-mass spectrometry (GC–MS). Therefore, the broccoli sprout BF contained high levels of phenolic compounds that contributed to its antioxidant activity, and CF had a marked anti-proliferative effect on BCSCs.

      • Effects of Drying Method on Anti-Oxidant, Anti-Proliferation Activity and Phenolic Compound Content in Different Parts of Cabbage

        Cabbage (Brassica oleracea L. var. capitata) is one of the vegetables grown worldwide. Cabbage has been reported to contain sulforaphane, which has anticancer properties, and antioxidant phytochemical. In this study, the effect of drying method on the antioxidant, anti-proliferation activity and phenolic compound content of cabbage was evaluated. Cabbage was divided into 6 parts including the outer leaf and core, and dried using hot air drying and freeze drying methods. The outer leaf of cabbage contained more phenolic compounds than other parts and showed high antioxidant activity in the evaluation of antioxidant activity through free radical scavenging analysis. In addition, cabbage samples showed an antiproliferative effect on breast cancer cells and pancreatic cancer cells, and especially the outer leaves showed a significant effect. In most experiments, the effect of cabbage according to the drying method was slightly enhanced when hot air dried. The results of this study suggest that the development of food processing methods is a factor that can be utilized in the development of functional foods.

      • Anti-oxidant and anti-proliferative activities of broccoli sprouts extracted with various solvents

        The antioxidant activity of six solvent fractions obtained from the sprouts of broccoli (Brassica oleracea L.), a cruciferous vegetable containing several bioactive compounds, was investigated. Our results showed that the butanol fraction exhibited the highest total polyphenol content (TPC) and total flavonoid content (TFC) with large radical scavenging activity at lower concentrations compared to other fractions. A significant difference in antioxidant activity was observed between the six fractions of broccoli sprouts, and the results of correlation analysis between the antioxidant activity and polyphenol compounds present in each solvent fraction were presented through HPLC analysis. However, in the antiproliferative activity of breast cancer stem cells (BCSCs) measured by MTT assay, the chloroform fraction showed the highest cytotoxicity. We found that the chloroform fraction increased apoptosis-induced cell death. The chloroform fraction of broccoli sprouts showed anticancer activity potential in BCSCs. Thus, these findings suggest a beneficial effect of broccoli sprouts extracted from six solvents through differences in anti-oxidant and anti-proliferative activities.
