Human Sapovirus (HuSaV) is one of the major causes of acute gastroenteritis in humans, and it is used as a molecular diagnostic technique based on polymerase chain reaction (PCR) from humans, food, shellfish, and aquatic environments. In this study, the HuSaV diagnosis technique was used in an aquatic environment where a number of PCR inhibitors are included and pathogens, such as viruses, are estimated to exist at low concentration levels. HuSaV-specific primers are improved to detect 38 strains registered in the National Center for Biotechnology Information (NCBI). The established optimal condition and the composition, including the RT-nested PCR primers and SL<SUP>®</SUP> Non-specific reaction inhibitor, were found to have 100 times higher sensitivity based on HuSaV plasmid than the previously reported methods (100 ag based on HuSaV plasmid 1 ng/μL). Through an artificial infection test, the developed method was able to detect at least 1 fg/μL of HuSaV plasmid contaminated with total nucleic acid extracted from groundwater. In addition, RT-nested PCR primer sets for HuSaV detection can react, and a positive control is developed to verify false positives. This study is expected to be used as a HuSaV monitoring method in the future and applied to the safety response to HuSaV from water environments.
Development and Practical Use of RT-PCR for Seed-transmitted Prune dwarf virus in Quarantine

Among imported plants, seeds are the items that have many latent pathogens and are difficult to inspect. Also, they are the import and export items whose market is expected to expand. The biggest problem with seeds is viruses. Prune dwarf virus (PDV) is the virus that is commonly inspected in Prunus cerasifera, P. persica, P. armeniaca, P. mandshurica, P. cerasus, P. avium or P. serotina seeds. In this study, two RT-PCR primer sets, which can promptly and specifically diagnose plant quarantine seed-transmitted PDV, were developed; and nested PCR primers, where products amplify 739 and 673 nucleotides (nt), and an nested PCR-product, 305 nt, can be obtained as these products are amplified again, were developed. Also, a modified-positive control plasmid was developed, where the restriction enzyme XhoI, which can identify the contamination of samples from the control, was inserted. The method developed in this study has detected PDV in 18 cases since 2007, and is expected to continuously contribute to the plant quarantine in Korea.

An advanced tangential X-ray pinhole camera (TXPC) has been developed for KSTAR by utilizing a 2-D duplex multi-wire proportional counter (MWPC) detector. The KSTAR MWPC employs a 2-D parallel type readout system for high temporal resolution and adopts a duplex type for the capability of electron temperature measurement via the multi-color method. This paper presents the performance test result of the developed MWPC system utilizing a Fe-55 X-ray source. As a preliminary experimental result from the 2012 KSTAR campaign, the clear presentation of sawtooth activities and its frequency change, and 2-D plasma images during the vertical disruption event are given.

<P>A novel yellow-pigmented bacterial strain, designated ARSA-19<SUP>T</SUP>, was isolated from a freshwater reservoir in Cheonan, Korea. Cells were Gram-staining-negative, rod-shaped, strictly aerobic and positive for catalase and oxidase. The phylogenetic tree based on 16S rRNA gene sequences showed that strain ARSA-19<SUP>T</SUP> belonged to the genus <I>Flavobacterium</I> and was related to <I>Flavobacterium macrobrachii</I> an-8<SUP>T</SUP> (96.5 % sequence similarity), <I>F. koreense</I> KACC 14969<SUP>T</SUP> (96.2 %), <I>F. chungnamense</I> KACC 14971<SUP>T</SUP> (96.1 %), <I>F. cheonanense</I> KACC 14972<SUP>T</SUP> (96.4 %) and <I>F. aquatile</I> ATCC 11947<SUP>T</SUP> (95.6 %). The isolate contained MK-6 as the predominant menaquinone and iso-C15 : 0 (21.8 %), iso-C16 : 0 3-OH (10.4 %), iso-C15 : 1 G (9.3 %), summed feature 3 (iso-C15 : 0 2-OH and/or C16 : 1ω7<I>c</I>; 8.2 %), iso-C15 : 0 3-OH (7.7 %) and iso-C16 : 0 (7.3 %) as the major fatty acids. The polar lipids of strain ARSA-19<SUP>T</SUP> were phosphatidylethanolamine, four unknown aminolipids and two unidentified polar lipids. The DNA G+C content of strain ARSA-19<SUP>T</SUP> was 33.3 mol%. Based on its phenotypic and genotypic characteristics and on the phylogenetic evidence presented, strain ARSA-19<SUP>T</SUP> is considered to represent a novel species in the genus <I>Flavobacterium</I>, for which the name <I>Flavobacterium dankookense</I> sp. nov. is proposed. The type strain is ARSA-19<SUP>T</SUP> ( = KCTC 23179<SUP>T</SUP> = JCM 17065<SUP>T</SUP> = KACC 14968<SUP>T</SUP>). Emended descriptions of <I>Flavobacterium cheonanense</I>, <I>Flavobacterium chungnamense</I>, <I>Flavobacterium koreense</I> and <I>Flavobacterium aquatile</I> are also proposed.</P>

Human Aichivirus A1 (HuAiV-A1) is a waterborne human pathogenic virus classified as Picornaviridae and Kobuvirus. In this study, we developed a method that can detect about 35 minutes faster with the same detection sensitivity level than the previously reported HuAiV-A1 diagnostic RT-PCR primer. The RT-PCR primer sets developed in this study are capable of detecting HuAiV-A1 at a level of about 100 ag and formed 563 bp amplification product. In addition, the RT-nested PCR method was able to amplify 410 bp using the RT-PCR product as a template. The detection sensitivity of our method was 10 times higher than the method with the highest detection sensitivity to date. Therefore, the detection method of HuAiV-A1 developed in this study is expected to be used in the water environment in which a small amount of virus exists. Also, this detection method is expected to be used as HuAiV-A1 diagnostic technology in both clinical and non-clinical field.
Development of a particle injection system for impurity transport study in KSTAR.

<P>A solid particle injection system is developed for KSTAR. The system has a compact size, compatibility with a strong magnetic field and high vacuum environment, and the capability to inject a small amount of solid particles with a narrow injection angle. The target flight-distance of 10 cm has been achieved with a particle loss rate of less than 10%. Solid impurity particles such as tungsten and carbon will be injected by this system at the midplane in KSTAR. The impurity transport feature will be studied with a soft X-ray array, a vacuum ultra-violet diagnostic, and Stand Alone Non-Corona code.</P>

We developed a loop-mediated isothermal amplification (LAMP) method to rapidly diagnose Wheat streak mosaic virus (WSMV) during quarantine inspections of imported wheat, corn, oats, and millet. The LAMP method was developed as a plant quarantine inspection method for the first time, and its simplicity, quickness, specificity and sensitivity were verified compared to current reverse transcription-polymerase chain reaction (RT-PCR) and nested PCR quarantine methods. We were able to quickly screen for WSMV at quarantine sites with many test samples; thus, this method is expected to contribute to plant quarantine inspections.
