        Zebrafish Dnd protein binds to 3`UTR of geminin mRNA and regulates its expression

        Dnd (dead end) gene encodes an RNA binding protein and is specifically expressed in primordial germ cells (PGCs) as a vertebrate-specific component of the germ plasma throughout embryogenesis. By utilizing a technique of specific nucleic acids associated with proteins (SNAAP), 13 potential target mRNAs of zebrafish Dnd (ZDnd) protein were identified from 8-cell embryo, and 8 target mRNAs have been confirmed using an RT-PCR analysis. Of the target mRNAs, the present study is focused on the regulation of geminin, which is an inhibitor of DNA replication. Using electrophoretic mobility shift assay (EMSA), we demonstrated that ZDND protein bound the 67-nucleotide region from 864 to 931 in the 3`UTR of geminin mRNA, a sequence containing 60.29% of uridine. Results from a dual-luciferase assay in HEK293 cells showed that ZDND increases the translation of geminin. Taken together, the identification of target mRNA for ZDnd will be helpful to further explore the biological function of Dnd in zebrafish germ-line development as well as in cancer cells.         Susceptibility Genes for Multiple Sclerosis Identifed in a Gene-Based Genome-Wide Association Study

        Background and Purpose Multiple sclerosis (MS) is a demyelinating and infammatory disease of the central nervous system. Te aim of this study was to identify more genes associated with MS. Methods Based on the publicly available data of the single-nucleotide polymorphism-based genome-wide association study (GWAS) from the database of Genotypes and Phenotypes, we conducted a powerful gene-based GWAS in an initial sample with 931 family trios, and a replication study sample with 978 cases and 883 controls. For interesting genes, gene expression in MS-related cells between MS cases and controls was examined by using publicly available datasets. Results A total of 58 genes was identifed, including 20 “novel” genes signifcantly associated with MS (p<1.40×10-4). In the replication study, 44 of the 58 identifed genes had been genotyped and 35 replicated the association. In the gene-expression study, 21 of the 58 identifed genes exhibited diferential expressions in MS-related cells. Tus, 15 novel genes were supported by replicated association and/or diferential expression. In particular, four of the novel genes, those encoding myelin oligodendrocyte glycoprotein (MOG), coiled-coil alpha-helical rod protein 1 (CCHCR1), human leukocyte antigen complex group 22 (HCG22), and major histocompatibility complex, class II, DM alpha (HLA-DMA), were supported by the evidence of both. Conclusion zTe results of this study emphasize the high power of gene-based GWAS in detecting the susceptibility genes of MS. Te novel genes identifed herein may provide new insights into the molecular genetic mechanisms underlying MS.

        중국 소비자의 ‘超餐’신소매(新零售)유통 이용특성에 대한 연구 - 차오지우종(超级物种) vs 허마센셩(盒马鲜生)을 중심으로 -

        본 연구에서는 중국에서 급성장하고 있는 온·오프라인 결합형 신 소매유통 모델과 관련하여 차오지우종(超级物种)과 허마센셩(盒马鲜生)을 대상으로 ‘超餐(마트+푸드코트)’ 신선식품 신 소매유통에 대한 중국 소비자의 선택특성/신 소매유통 특성이 신 소매유통 수용성과 만족도에 미치는 영향에 대해 살펴보았다. 아울러 차오지우종과 허마센셩을 이용하는 소비집단에 대해 다중집단분석을 통한 집단 간 경로분석 결과의 차이를 살펴보았다. 첫째, 차이분석 결과는 다음과 같다. 소비자 선택특성에서는 차오지우종이 허마센셩 보다 모두 높게 나타났으며 소비자의 수용성도 더 높게 나타났다. 아울러 신 소매유통의 특성(체험성, 편리성, 배송서비스)에서도 체험성은 높게 나타난 반면 편리성과 배송서비스는 허마센셩이 차오지종 보다 더 높은 것으로 나타났다. 성별에 따른 차이 분석결과에서는 차우지우종의 경우, 차오지우종의 4개 변수 평균값 모두 여성보다 남성이 높게 나타나고 있다. 한편, 사용횟수에 따른 평균값의 차이는 사용횟수가 많아질수록 평균값도 높아지는 것으로 나타났다. 둘째, 가설검증 결과는 다음과 같다. 신 소매유통 수용성에 미치는 영향과 관련하여 소비자 선택특성(혁신성, 경제성, 맞춤형 서비스)과 신 소매유통 특성(체험성, 편리성, 배송서비스) 변수 모두 신 소매유통 수용성에 정(+)의 영향을 미치는 것으로 나타났다. 집단 간 경로계수의 유의성 및 경로계수에 대한 차이를 검증하기 위한 다중집단 비교분석 결과에서는 차오지우종이 허마센셩 보다 소비자 선택특성/신 소매유통 특성, 신 소매유통 수용성 및 만족도 간 영향관계가 더 큰 것으로 나타났다. 특기할만한 사실은 차오지우종은 ‘배송서비스-소비자 수용성’에서 허마센셩은 ‘맞춤형 서비스-소비자 수용성’에서 더 크게 영향을 미치는 것으로 나타났다. In recent years, new retail has always been the first hot spot in China’s capital market. This paper chooses the fast-expanding ‘Chaojiwuzhong’ and ‘Hema supermarket’ as the analysis object and analyzed the factors of customer selection characteristics of new retailers and new retailer’s use characteristics on new retail acceptance and satisfaction. In addition, the differences in the results of path analysis were examined through multi-group analysis according to the ‘Chaojiwuzhong’ and ‘Hema’ supermarket. Based on the results of this study, our findings can be summarized as follows: First, there are statistically significant differences in research variables between ‘Chaojiwuzhong’ and ‘Hema’ supermarket. Especially, Chaojiwuzhong’s consumer selection characteristics and consumers’ new retail acceptance are higher than ‘Hema’. However, in the case of new retailer’s use characteristics, Hema’s convenience and delivery service except experientiality are higher than Chaojiwuzhong. On the other hand, it was found that the higher the number of uses, the higher the acceptability for new retailers. Therefore, it is of utmost importance to ensure the quality and safety of fresh food in order to make a steady visit. Secondly, it was found that both the customer selection characteristics of new retailers and the new retailer’s use characteristics have a positive effect on the new retail acceptance. First, it was found that both the consumer selection characteristic and new retailer’s use characteristics have a positive effect on the new retail acceptance. Among them, customized service was found to have the greatest impact. Therefore, it seems that technologies innovating customized services must be continuously developed.

        Experimental Study on Chloride Diffusion in Structural Concrete Considering the Effect of Damages Induced by the Cyclic Impact Loading

        An experimental investigation on chloride diffusion in structural concrete under the cyclic impact loading is conducted. Effects of magnitudes and loading times are studied. Relationship between damage coefficients and chloride diffusion coefficients is obtained. The cyclic impact loading causes the initiation of new cracks and the propagation of existing cracks, which brings an obvious promotion for chloride diffusion. There should be a damage threshold. If the magnitude of the external loading is large enough to make the damage exceed the threshold, initiation and propagation of cracks become quite significant and chloride diffusion can be greatly promoted. With the increase of the damage coefficient, the chloride diffusion coefficient increases. The increasing rates decrease with long immersion times. Relationship curves of damage coefficients and chloride diffusion coefficients show a good correlation and can be well described with power functions.

        Identification of novel rheumatoid arthritis-associated MiRNA-204-5p from plasma exosomes

        Rheumatoid arthritis (RA) is an autoimmune disease characterized by infiltration of immune cells in the synovium. However, the crosstalk of immune cells and synovial fibroblasts is still largely unknown. Here, global miRNA screening in plasma exosomes was carried out with a custom microarray (RA patients vs. healthy controls = 9:9). A total of 14 exosomal miRNAs were abnormally expressed in the RA patients. Then, downregulated expression of exosomal miR-204-5p was confirmed in both the replication (RA patients vs. healthy controls = 30:30) and validation groups (RA patients vs. healthy controls = 56:60). Similar to the findings obtained in humans, a decreased abundance of exosomal miR-204-5p was observed in mice with collagen-induced arthritis (CIA). Furthermore, Spearman correlation analysis indicated that plasma exosomal miR-204-5p expression was inversely correlated with disease parameters of RA patients, such as rheumatoid factor, erythrocyte sedimentation rate, and C-reactive protein. In vitro, our data showed that human T lymphocytes released exosomes containing large amounts of miR-204-5p, which can be transferred into synovial fibroblasts, inhibiting cell proliferation. Overexpression of miR-204-5p in synovial fibroblasts suppressed synovial fibroblast activation by targeting genes related to cell proliferation and invasion. In vivo assays found that administration of lentiviruses expressing miR-204-5p markedly alleviated the disease progression of the mice with CIA. Collectively, this study identified a novel RA-associated plasma exosomal miRNA-204-5p that mediates the communication between immune cells and synovial fibroblasts and can be used as a potential biomarker for RA diagnosis and treatment.
