The effects of drone pupa have been recorded worldwide since about seven hundred years ago. However, there is still a lack of scientific evidence. In response, this study evaluates the activity of ADH, one of the liver health-related functional evaluation biomarkers, in drone pupa extracts. ADH activity was assessed for two fractions (Hexane, water) of the drone pupa. These samples were manufactured using two solvents and had a difference in the time of dissolving the extract samples. According to our results, ADH activity of water fraction was higher than hexane fraction. Using DMSO as a solvent, the highest ADH activity of hexane fraction was 120 minutes, and the water fraction was 180 minutes. The difference of ADH activity between hexane fraction and water fraction were 23.58 mU/mL and 35.04 mU/mL, respectively. As a result, the drone pupa extracts showed ADH activity, which is expected to be a diet supplement that can help with alcohol metabolism and drug metabolism.

In recent years, edible insects have caught the attention of researchers, because of their potential as an alternative source of food and nutraceuticals. Based on the record of use for edible and medicinal purpose, drone pupa was approved as a new food ingredient in 2021. To investigate the possibility of antiproliferative effect for malignancy, we observed the viability of HepG2 cells by treating the extract. When the extract was treated in HepG2 cells at 10 mg/mL and 5 mg/mL for 24 hours, the cell viability was 49.33 and 66.98%, respectively. Detection nucleosomes in cytoplasmic fractions showed that drone pupa extract induced apoptosis in HepG2. Western blotting was used to evaluate apoptosis-related protein expression such as Bcl-2, Bcl-xL, Bax, caspase 3. Our result suggest that drone pupa extract have apoptosis-induced activity in HepG2 cancer cell.

This paper discussed the nutrients and sensory analysis of food enriched with selected edible insect species which are labelled as a novel food-drone pupa. Breads of two different compositions with the addition of freezing dried flour and fat extract were evaluated by electronic tongue and analysis of nine nutrients. Through nutritional analysis, it was observed that the addition of drone pupa freeze-dried powder to bread led to an increase in both protein and fat content, along with a decrease in carbohydrate content. Similarly, bread with drone pupa fat extract added also exhibited an increase in protein and fat content, while carbohydrate content decreased. It was observed that the bread with the freeze-dried flour of drone pupa showed a higher salty and sour taste bread with fat extract through electronic tongue analysis. On the other hand, bread added with fat extract showed a higher umami and sweetness. In addition, the salty and bitter taste was the least in bread with 5% of freeze-dried drone pupa flour, and the higher the content of fat extracts the higher the umami and sweet taste. When adding processed drone pupa products, it is considered appropriate to do so at a level of 5~10%. As a result of this study, it is supposed that drone pupa could be applied to baking in various processing forms and can be used as basic data for sensual characteristics.

Tracheal mite ( Acarapis woodi ) is an internal parasite that is parasitic on the bronchus of adult bees and sucks fluid from the trachea. Since its first report by Rennie, it has been spread throughout Europe and in some Asian regions, with adjacent Japan and China reported in 2011 and 2012, respectively. Korea detected specific genes of A. woodi in 2015, but only one of 99 samples has been identified and the being of A. woodi has not been confirmed. In this study, we established a specific nested PCR method to confirm for detecting low-copy number of A. woodi-specific gene in bee samples. As a result, A. woodi-specific COI gene was amplified in 15 of 23 samples, and they were judged positive by melting point analysis and sequencing analysis. Although we could not observe the existence of the mites in bees, our results suggest that tracheal mit might exist in nature.

Rapid detection of Tropilaelaps, an external parasite of honeybees that lead to malformation of honeybee or colony collapse disorder, is becoming important. But it is very difficult to find with the naked eye of Tropilaelaps. In this study, we have developed a method to detect the specific gene of Tropilaelaps from the hive debris and to know the number of Tropilaelaps in the hive through Tropilaelaps-specific quantitative detection. Tropilaelaps-specific gene amplified in DNA extracted from hive debris by consecutive PCR (1st detection, 2nd nested PCR). It could detect 10<SUP>1</SUP> molecules level of Tropilaelapsspecific gene and confirm the amplification of the Tropilaelaps-specific gene. It was possible to accurately quantify the number of Tropilaelaps from the hive debris sample, which is difficult to discriminate the presence of Tropilaelaps visually, through Tropilaelaps-specific detection. Under the microscope, Tropilaelaps was collected and quantitative detection of Tropilaelaps-specific genes was performed. It was possible to quantify the number of Tropilaelaps present in the hive through the molecules of the quantified Tropilaelaps-specific genes. We suggest that hive debris can represent as a micro-environment to hive and show that it can be a simpler and more accurate sample than using a parasitic host honeybee. We expect that hive debris should facilitate the monitoring of Tropilaelaps in hive.

BQCV multi-point PCR was developed as a rapid multiplex detection method for BQCV, one of the viral pathogens of honeybees. It could detect BQCV specific genes qualitative as well as quantitative detection based on ultra-rapid PCR. Three primer pairs (RNA dependent RNA polymerase, capsid protein, 3C like protease) were specifically designed for accurate the detection and were optimized for minimizing the detection time and increasing the sensitivity. Our advanced diagnostic system have the accuracy by lowering the concern about the variation in the BQCV detection site. In addition, it should be an opportunity to identify mutations that are mixed with other viruses.

Objective: In this study, we evaluated the prevalence of allergic disease in offsprings delivered via the delivery modes of vaginal delivery vs. planned Cesarean section vs. Cesarean section with labor. Methods: This study included 175 mother-neonate pairs from Severance Hospital who were enrolled in the Cohort for Childhood Origin of Asthma and allergic diseases study. Information regarding prenatal environmental factors, delivery, and diagnosis of allergic diseases was obtained from a questionnaire and medical record review. Patients with at least 3 years of follow-up data were included in this study. Results were adjusted for sex, birthweight, gestational age at birth, season of birth, neonatal intensive care unit admission, parity, breastfeeding, and maternal factors. Results: A total of 175 offsprings were eligible for analysis. Among the subjects, 52.0% were delivered by vaginal delivery, 34.3% by planned Cesarean section, and 16.6% by Cesarean section with labor. Fifty-nine offsprings (33.7%) were diagnosed with allergic disease at a median age of 1 year (range 0.5-3 years). The prevalence of allergic disease was not associated with delivery mode after adjusting for confounding variables. Time period from membrane rupture to delivery, duration of the active phase, and the beginning of the pelvic division prior to Cesarean section were not associated with allergic disease development in offsprings. Conclusion: Cesarean section, irrespective of the occurrence of labor before surgery, did not increase the prevalence of allergic disease in infants up to 3 years of age.

<P> Israeli acute paralysis virus (IAPV) is one of the main pathogens involved in the collapse of honey bee colonies. However, because the virus exhibits a high level of genetic variation and some IAPV strains exhibit high degrees of homology with related viruses, the detection of IAPV in infected honey bees is relatively challenging. To address this obstacle, the aim of the present study was to develop a new detection method that relies on multiple detection sites within the IAPV genome and on Ultra-rapid real-time polymerase chain reaction (UR-qPCR). The new system simultaneously targeted a RNA-dependent RNA polymerase gene <I>(RdRp)</I> and two capsid genes (<I>VP3</I> and <I>VP1</I>). This multi-point PCR approach was highly efficient, with the ability to detect 100% of IAPV infections, and outperformed single-point PCR, which was only able to detect 86.96-95.6% of IAPV infections. Sequence analysis indicated that <I>RdRp</I> was more variable than the two capsid genes, and the specificity of the proposed method was demonstrated by the detection of IAPV from samples co-infected by IAPV and KBV. Importantly, both freezing-thawing RNA isolation and UR-qPCR could be performed in 27min and 40s. Therefore, the present provides an useful tool for the rapid identification of IAPV in apiaries. </P>
