        異功散이 마우스 黑色腫의 肺轉移 抑制에 미치는 抗癌 및 免疫作用에 關한 硏究

        종양은 그 발생원인과 성장기전이 자세히 밝혀져 있지 않는 질병으로 최근 성인사망의 제 1원인으로 급격히 발생빈도가 증가하고 있다. 종양환자의 수는 점점 증가하는 추세이며, 그에 따른 사망자의 수도 늘고 있다. 종양을 치료하는 방법으로 수술요법·방사선요법·화학약물요법·골수이식·면역요법·호르몬요법 등이 사용되어 왔으나 최근에 천연물 특히 전통적인 한약재를 이용한 종양파괴 및 종양에 있어 성장을 억제하는 약재의 연구가 시도되어 왔다. 한의학의 특정 약재 및 처방의 항암효과가 경험적으로 알려져 왔으며, 과학적이고 객관적인 병태를 통하여 이러한 약재의 효능을 검증하려는 실험이 최근에 시도되고 있다. 이에 본 연구는 補眞氣虛弱 및 治 久嗽不己 脾胃虛弱 등의 효능이 있는 異功散을 이용하여 B16세포의 전이억제능, 흑색종의 증식억제 및 면역능에 대하여 연구하고자 하였다. 본 연구에서는 시험관내 세포독성능을 알아보기 위하여 MTT법으로 각 농도별로 처리된 이공산을 B16세포에 적정농도로 처리하여 생존율 및 IC50을 측정하였다. 또한 생쥐를 이용한 동물실험으로써 흑색종을 폐에 전이시켜 이공산 투여후 흑색종의 집략수를 측정하였다. 동시에 항암효과와 면역능과의 관련성을 알아보기 위하여, Interleukin-2생산능과 NK-Activity를 측정하였다. 이공산의 생쥐에 대한 폐전이 흑색종의 억제효능과 면역반응에 미치는 영향을 알아보기 위한 실험에서 시험관내 세포독성 측정에서는 2㎍/well의 농도로부터 2배 희석시켜 MTT법으로 측정한 결과 유의성있는 억제를 보였으며 2,2124㎍/well의 IC50을 얻었다. 흑색종의 억제능을 측정한 결과 7일과 14일째에 유의성있는 억제를 보였고, 21일째는 흑색종이 폐의 전면에 분포되어 측정되지 않았다. 이 공산 투여후 생쥐의 IL-2의 생산능을 측정한 결과 이공산 투여군에 있어 투여 7일, 14일, 21일째에 모두 유의성있는 증가를 보였다. 그리고 NK-Activity를 측정한 결과 이공산 투여군이 대조군과 비교하여 유의한 차이는 보이지 못하였다. 이상의 결과를 종합하여 볼 때 이공산이 B16세포에 대하여 직접 세포독성능을 나타내며 보조 T 세포를 자극하여 IL-2의 생산능을 증가시켜 흑색종의 전이 및 증식을 억제하는 것으로 추정된다. 따라서 전이억제의 기전과 면역반응에 관련된 연구가 더욱 진행되어야 할것으로 사료된다. Herbal medicine as a candidate for effective cancer treatment recently gain positive concerns in field of therapeutic oncology. that is why some oriental medicine has been empirically safer in toxicity than anticancer drugs used in western medicine, and to show excellent therapeutic efficacy in human trial. Thus, these effects by clinically applied-herbs have not yet fully demonstrated in experimental tumor model. This study was initiated to evaluate antitumor effect of Ikongsan as candidate of antitumor-hebal agent against B16 melanoma metastasized into murine lung, and to examine association between immune parameter an anti-metastasis effect by Ikongsan. In experiment to test whether Ikongsan can directly kill cancer cells in vitro or not, Ikongsan showed direct killng action in concentration or higher against B16 melanoma cells using MTT assay. And also, in another experiment to know whether Ikongsan can inhibit growth and metastasis of cancer cell or not, Ikongsan significantly inhibited lung metastasis induced by intravenous injected-melanoma in inbred C57BL/6 mouse. When we quatiated interleukin-2 production, we could obtain results that in Ikongsan-treated group(7th, 14th, 21th), IL-2 production was significantly increased, But, NK activity in Ikongsan-treated group was not in significantly difference as compared to that of Ikongsan-nontreated group. These results show that Ikongsan can inhibit growth of B16 melanoma cells through biological mechanisms.

        만경강 유역내 하천의 특성별 영향 평가

        본 연구는 만경강 유역의 수질 보전을 위한 기초자료를 얻기 위하여 만경강 본류와 이에 유입되는 지천을 선정하여 2001년 5월부터 11월까지 매월 수질 모니터링을 실시하였다. 만경강 유역내 본류 및 지천의 유기물과 영양염류 함량을 조사한 결과, 본류의 T-N은 3.78~12.68 ㎎/L였으며, 지천중 축산폐수가 유입되는 익산천의 농도가 가장 높았다. 본류의 T-P의 BOD는 각각 0.043~0.864 ㎎/L와 2.59~13.29 ㎎/L였으며, 지천중 익산천이 가장 높았다. 본류의 COD는 12.9~119.5 mg/L였으며, 지천중 도시생활하수가 유입되는 추천이 가장 높았다. 만경강 유역 수질저하의 주 원인은 축산폐수였으며, 도시생활하수가 그 다음으로 중요한 원인으로 평가되었다. 만경강 본류의 T-N과 T-P는 하천의 유량이 증가하는 홍수기인 7~8월보다 장마선 갈수기인 5~6월에 높았고, COD는 장마전 갈수기보다 홍수기에 높았으며 장마가 완전히 끝난 영농후 갈수기인 9~11월에도 수질저하가 지속되었다. 만경강에 유입되는 지천중 전반적으로 수질오염이 심각한 익산천과 추천은 영농후 갈수기에 수질이 가장 저하되었고 상대적으로 홍수기에는 양호하였다. A survey on four tributaries along with Mankyeong River was carried out to get the information for the water quality improvement and control. Typical paddy farming were major agricultural practices in Kosancheon and Soyangcheon. Iksancheon was livestock raising watershed. Chucheon was urban watershed. Water quality in six sites of main stream and four sites of tributaries in Mankyeong River were investigated from May to August in 2001. The concentration of nutrients in main stream of Mankyeong River were in the range of 3.78~12.68 mg/L for total nitrogen, 0.043~0.864 mg/L for total phosphorus, 2.59~13.29 mg/L for BOD and 12.9~119.5 mg/L for COD, respectively. Water quality of Mankyeong River mostly exceeded the standard water quality criteria of Korea. Major causes of water pollution were evaluated as sewage of swine and urban area. Among the four tributaries, water quality in agricultural practices, Kosancheon and Soyangcheon was relatively less polluted. While, the highest level of water pollution measured in Iksancheon was due to livestock The water quality of Iksancheon and Chucheon was generally more polluted in the dry period than in rainy period.

        3-Deoxysappanchalcone Inhibits Cell Growth of Gefitinib-Resistant Lung Cancer Cells by Simultaneous Targeting of EGFR and MET Kinases

        The mechanistic functions of 3-deoxysappanchalcone (3-DSC), a chalcone compound known to have many pharmacological effects on lung cancer, have not yet been elucidated. In this study, we identified the comprehensive anti-cancer mechanism of 3-DSC, which targets EGFR and MET kinase in drug-resistant lung cancer cells. 3-DSC directly targets both EGFR and MET, thereby inhibiting the growth of drug-resistant lung cancer cells. Mechanistically, 3-DSC induced cell cycle arrest by modulating cell cycle regulatory proteins, including cyclin B1, cdc2, and p27. In addition, concomitant EGFR downstream signaling proteins such as MET, AKT, and ERK were affected by 3-DSC and contributed to the inhibition of cancer cell growth. Furthermore, our results show that 3-DSC increased redox homeostasis disruption, ER stress, mitochondrial depolarization, and caspase activation in gefitinib-resistant lung cancer cells, thereby abrogating cancer cell growth. 3-DSC induced apoptotic cell death which is regulated by Mcl-1, Bax, Apaf-1, and PARP in gefitinib-resistant lung cancer cells. 3-DSC also initiated the activation of caspases, and the pan-caspase inhibitor, Z-VAD-FMK, abrogated 3-DSC induced-apoptosis in lung cancer cells. These data imply that 3-DSC mainly increased mitochondria-associated intrinsic apoptosis in lung cancer cells to reduce lung cancer cell growth. Overall, 3-DSC inhibited the growth of drug-resistant lung cancer cells by simultaneously targeting EGFR and MET, which exerted anti-cancer effects through cell cycle arrest, mitochondrial homeostasis collapse, and increased ROS generation, eventually triggering anticancer mechanisms. 3-DSC could potentially be used as an effective anti-cancer strategy to overcome EGFR and MET target drug-resistant lung cancer.

        <P>OBJECTIVE: Lipid accumulation in vascular endothelial cells may play an important role in the pathogenesis of atherosclerosis in obese subjects. We showed previously that alpha-lipoic acid (ALA) activates AMP-activated protein kinase (AMPK) and reduces lipid accumulation in skeletal muscle of obese rats. Here, we investigated whether ALA improves endothelial dysfunction in obese rats by activating AMPK in endothelial cells. METHODS AND RESULTS: Endothelium-dependent vascular relaxation was impaired, and the number of apoptotic endothelial cells was higher in the aorta of obese rats compared with control rats. In addition, triglyceride and lipid peroxide levels were higher, and NO synthesis was lower. Administration of ALA improved all of these abnormalities. AMPK activity was lower in aortic endothelium of obese rats, and ALA normalized it. Incubation of human aortic endothelial cells with ALA activated AMPK and protected cells from linoleic acid-induced apoptosis. Dominant-negative AMPK inhibited the antiapoptotic effects of ALA. CONCLUSIONS: Reduced AMPK activation may play an important role in the genesis of endothelial dysfunction in obese rats. ALA improves vascular dysfunction by normalizing lipid metabolism and activating AMPK in endothelial cells.</P>

        Psychometric Properties of the Korean Version of the Clinical Language Disorder Rating Scale (CLANG)

        Objective: Our study aimed to measure inter-rater and test-retest reliability, concurrent and convergent validity, and factor solutions of the Korean version of the Clinical Language Disorder Rating Scale (CLANG). Methods: The Korean version of the CLANG for assessing thought, language, and communication, the Brief Psychiatric Rating Scale, Young Mania Rating Scale, and Calgary Depression Scale for Schizophrenia were used to evaluate language disorder, formal thought disorder, positive and negative symptoms, manic symptoms, and depressive symptoms, respectively, in 167 hospi-talized patients with schizophrenia. The factor solution was obtained by the direct oblimin method. A receiver operating character-istic curve was used to find the optimal cut-off score for discriminating schizophrenia patients with and without disorganized speech. Results: Inter-rater reliability was considered moderate (intraclass coefficient=0.67, F =3.30, p =0.04), and test-retest reliability was considered high ( r =0.94, p <0.001). Five factors, namely, pragmatics, disclosure, production, prosody, and association, were identified. An optimal cut-off score of 7 points with 84.5% sensitivity and 81.7% specificity was proposed for distinguishing schizo-phrenia patients with and without disorganized speech. Conclusion: Our findings suggest that the Korean version of the CLANG is a promising tool for evaluating language disorder in patients with schizophrenia.
