MiR-454 Prompts Cell Proliferation of Human Colorectal Cancer Cells by Repressing CYLD Expression

Previous studies have shown that miR-454 plays an important role in a variety of biological processes in various human cancer cells. However, the underlying mechanisms of this microRNA in colorectal cancer (CRC) cells remain largely unknown. In the present study, we investigated the miR-454 role in CRC cell proliferation. We found that miR-454 expression is markedly upregulated in CRC tissues and CRC cells compared with the matched tumor adjacent tissues and the FHC normal colonic cell line. Ectopic expression of miR-454 promoted the proliferation and anchorage-independent growth of CRC cells, whereas inhibition of miR-454 reduced this effect. Bioinformatics analysis further revealed cylindromatosis (CYLD), a putative tumor suppressor as a potential target of miR-454. Data from luciferase reporter assays showed that miR-454 directly binds to the 3'-untranslated region (3'-UTR) of CYLD mRNA and repressed expression at both transcriptional and translational levels. In functional assays, CYLD-silenced in miR-454-in-transfected SW480 cells have positive effect to promote cell proliferation, suggesting that direct CYLD downregulation is required for miR-454-induced CRC cell proliferation. In sum, our data provide compelling evidence that miR-454 functions as an onco-miRNA, playing a crucial role in the promoting cell proliferation in CRC, and its oncogenic effect is mediated chiefly through direct suppression of CYLD expression.

Background: Multidrug resistance (MDR) to chemotherapy drugs remains a major challenge in clinicalcancer treatment. Here we investigated whether and how ginsenoside Rg5 overcomes the MDR mediatedby ABCB1 transporter in vitro and in vivo. Methods: Cytotoxicity and colon formation as well as the intracellular accumulation of ABCB1 substrateswere carried out in MDR cancer cells A2780/T and A549/T for evaluating the reversal effects of Rg5. Theexpressions of ABCB1 and Nrf2/AKT pathway were determined by Western blotting. An A549/T cellxenograft model was established to investigate the MDR reversal activity of Rg5 in vivo. Results: Rg5 significantly reversed ABCB1-mediated MDR by increasing the intracellular accumulation ofABCB1 substrates without altering protein expression of ABCB1. Moreover, Rg5 activated ABCB1 ATPaseand reduced verapamil-stimulated ATPase activity, suggesting a high affinity of Rg5 to ABCB1 bindingsite which was further demonstrated by molecular docking analysis. In addition, co-treatment of Rg5 anddocetaxel (TXT) suppressed the expression of Nrf2 and phosphorylation of AKT, indicating that sensitizingeffect of Rg5 associated with AKT/Nrf2 pathway. In nude mice bearing A549/T tumor, Rg5 and TXTtreatment significantly suppressed the growth of drug-resistant tumors without increase in toxicitywhen compared to TXT given alone at same dose. Conclusion: Therefore, combination therapy of Rg5 and chemotherapy drugs is a strategy for the adjuvantchemotherapy, which encourages further pharmacokinetic and clinical studies.

Giardia lamblia is a common enteric pathogen associated with diarrheal diseases. There are some reports of G. lamblia infection among different breeds of cattle in recent years worldwide. However, it is yet to know whether cattle in Jiangxi province, southeastern China is infected with G. lamblia. The objectives of the present study were to investigate the prevalence and examine the multilocus genotypes of G. lamblia in cattle in Jiangxi province. A total of 556 fecal samples were collected from 3 cattle breeds (dairy cattle, beef cattle, and buffalo) in Jiangxi province, and the prevalence and genotypes of G. lamblia were determined by the nested PCR amplification of the beta-giardin (bg) gene. A total of 52 samples (9.2%) were positive for G. lamblia. The highest prevalence of G. lamblia was detected in dairy cattle (20.0%), followed by that in beef cattle (6.4%), and meat buffalo (0.9%). Multilocus sequence typing of G. lamblia was performed based on sequences of the bg, triose phosphate isomerase and glutamate dehydrogenase loci, and 22, 42, and 52 samples were amplifiable, respectively, forming 15 MLGs. Moreover, one mixed G. lamblia infection (assemblages A and E) was found in the present study. Altogether, 6 novel assemblage E subtypes (E41*-E46*) were identified for the first time. These results not only provided baseline data for the control of G. lamblia infection in cattle in this southeastern province of China, but also enriched the molecular epidemiological data and genetic diversity of G. lamblia in cattle.

In this work, as an inoculant, in-situ BaB6/Al composites was fabricated and used to refine the α-Al dendrites and modify theeutectic Si of A356.2(Al–7Si–0.3Mg) alloy. BaB6/Al ingot and ribbon composites mainly contain two phases, α-Al and BaB6. In BaB6/Al ribbons, nano-size BaB6ceramic particles exist and disperse uniformly which help to better exert the inoculanteffect in A356.2 alloy. YS, UTS, EL and HV of B2 alloy are 36.3%, 18.3%, 46.4% and 20.8% higher than the base alloy (B0),respectively. The ductile fracture characteristics are more obvious. Moreover, B2 alloy shows the best corrosion resistance. Aland BaB6form a clear, stable and well matched interface, so BaB6particles can act as the heterogeneous nucleation of α-Aland refine the A356.2 alloy. Furthermore, the combined of free micro-alloying elements Ba and B in BaB6/Al compositespromote the twins formation, which change the eutectic Si to fine fibrous and granular shape.

Purpose Epidermal growth factor receptor (EGFR) T790M mutation serves as an important predictor of osimertinib efficacy. However, little is known about how it works among patients with various timings of T790M emergence and treatment. Materials and Methods Advanced EGFR-mutant lung adenocarcinoma patients with positive T790M mutation in tumor were retrospectively enrolled and observed to determine the outcomes of osimertinib treatment. We evaluated the association between patients’ characteristics and the efficacy of osimertinib treatment, particularly with respect to the timing of T790M emergence and osimertinib prescription. Results A total of 91 patients were enrolled, including 14 (15.4%) with primary and 77 (84.6%) with acquired T790M mutation. The objective response rate and disease control rate were 60.9% and 85.1%, respectively. The median progression-free survival (PFS) and overall survival were 11.5 months (95% confidence interval [CI], 9.0 to 14.0) and 30.4 months (95% CI, 11.3 to 49.5), respectively. There was no significant difference in response rate and PFS between primary and acquired T790M populations. In the acquired T790M subgroup, patients who received osimertinib after T790M had been confirmed by rebiopsy had a longer PFS than those with intercalated treatments between rebiopsy and osimertinib prescription (14.0 months [95% CI, 9.0 to 18.9] vs. 7.2 months [95% CI, 3.7 to 10.8]; adjusted hazard ratio, 0.48 [95% CI, 0.24 to 0.98; p=0.043]). Rebiopsy timing did not influence the outcome. Conclusion Osimertinib prescription with intercalated treatment following rebiopsy but not the timing of T790M emergence influenced the treatment outcome. We suggest that it is better to start osimertinib treatment once T790M mutation has been confirmed by biopsy.
Development of miniature bar-type structural fuses with cold formed bolted connections

A novel all-steel miniature bar-type structural fuse (MBSF) with cold formed bolted connections is developed in this study, which consists of a central energy dissipation core cut from a smooth round bar, an external confining tube and nuts. Three types of cross sections for the central energy dissipation core, i.e., triple-cut, double-cut and single-cut cross sections, were studied. Totally 18 specimens were axially tested under either symmetric or asymmetric cyclic loading histories, where the parameters such as cut cross sectional area ratio, length of the yielding portion and cross sectional type were investigated. Numerical simulation of 2 representative specimens were also conducted. An analytical model to evaluate the bending failure at the elastic portion was proposed, and a design method to avoid this failure mode was also presented. The experimental results show that the proposed MBSFs exhibit satisfactory hysteretic performance under both the two cyclic loading histories. Average strain values of 8% and 4% are found to be respectively suitable for designing the new MBSFs as the ultimate strain under the symmetric and asymmetric cyclic loadings.

Robust Guaranteed Cost Control for a Class of Nonlinear 2-D Systems with Input Saturation

In this paper, a robust guaranteed cost controller is designed for a class of two-dimensional(2-D) discrete time systems described by Roesser model, where the system simultaneously contains input saturation, parametric uncertainty and sector nonlinearity. Firstly, a convex hull representation is used to describe saturated input of the system. Secondly, a sufficient condition for ensuring the asymptotic stability of the closed-loop system and the existence of the robust guaranteed cost controller is given according to the Lyapunov stability theory. Then, this sufficient condition is transformed into a linear matrix inequality form by using Schur complement lemma and the design of robust guaranteed cost controller is realized by the solution of LMI. Finally, a numerical example is given to illustrate the effectiveness of the proposed method.
