Differential regulation of TLE3 expression in the mouse testis during postnatal development

Spermatogonial stem cells (SSCs; also known as Asingle [As] spermatogonia in mice) divide to self-renew or to produce progenitor cells known as Apaired(Apr) spermatogonia in basal compartment of seminiferous tubules of mammalian testis. These characterized cells are the finally differentiated product of a developmental process referred to as “spermatogenesis.” In the development of SSCs it is critical to maintain a balance between self-renewal and differentiation. because an excess of either process will lead to infertility. these two processes are tightly controlled by intrinsic signals of SSCs and extrinsic signals from the microenvironment, known as the SSC niche. The SSC niche is formed by Sertoli cells, the only somatic cells found inside the seminiferous tubules. The WNT/β-catenin pathway is known to regulate Sertoli cell functions critical to their capacity to support spermatogenesis in the postnatal testis, but The mechanisms and factors of the pathway are not well known. We found a factor TLE3 (Transducin Like Enhancer Of Split 3). The transcriptional co-repressor TLE family is known to function as transcription co-repressors within the context of Wnt signaling by interacting with histone deacetylase HDAC2. We examined the expression level of TLE3 in various mouse tissues. As a result of RT-PCR, TLE3 showed significantly higher expression in testis than that in other tissues. Immunofluorescent analysis revealed that TLE3 and HDAC2 expression are differentially regulated in the mouse testis during postnatal development. In adult testis, TLE3 and HDAC2 were co-expressed in Sertoli cells. TLE3 and HDAC2 protein are also located in nucleus in mouse TM4 Sertoli cells. Taken together, TLE3 may play a role in regulating WNT/β-catenin pathway via interaction with HDAC2 in Sertoli cell. Futher studies are needed to look into factors that regulated by siTLE3 in Sertoli cell and interated with TLE3 in WNT/β-catenin pathway.

<P><B>Abstract</B></P> <P>The present study investigates the fresh and hardened properties of controlled low-strength materials (CLSM) utilizing circulating fluidized bed combustion (CFBC) ashes, activated with cement or sodium carbonate. Engineering properties such as flowability, settlement, setting behavior, compressive strength, and hydration characteristics were evaluated. The results provided new insights, demonstrating that CFBC ashes can be effectively utilized in producing CLSM with suitable material design as the CLSM could achieve the required performances specified in the ACI 299R-13. The CLSM mixtures showed a self-destruction characteristic due to the formation of ettringite, while it showed stable strength development in a dry condition.</P> <P><B>Highlights</B></P> <P> <UL> <LI> High volume of CFBC fly ash and bottom ash were utilized in CLSM. </LI> <LI> The CLSM strength was significantly fluctuated by curing condition. </LI> <LI> CFBC ash led expansion of CLSM. </LI> <LI> Cement or sodium carbonate could be used as activator for CLSM. </LI> <LI> CLSM with CFBC fly ash could pass heavy metal leaching criteria. </LI> </UL> </P>
Dynamic Regulation of TLE3 Expression during Spermatogenesis in Mouse Testis

Spermatogenesis is the process of proliferation and differentiation of spermatogonial stem cells into adult spermatozoa within seminiferous tubules. Sperm cells undergo extensive epigenetic modifications during spermatogenesis. One of the epigenetic modifications, histone acetylation induces a loose chromatin structure and active transcription which facilitates binding of transcription factors at sperm chromosome. Therefore, histone acetylation is known to play an important role in spermatogenesis. However, factors involved in histone acetylation during spermatogenesis are not well known. We found a factor TLE3(Transducin Like Enhancer of Split 3). The transcriptional co-repressor TLE family is known to regulate histone acetylation by interacting with histone deacetylase( HDAC). We examined the expression level of TLE3 in various mouse tissues. As a result of RT-PCR, TLE3 showed significantly higher expression in testis than that in other tissues. Immunofluorescent and Immunohistochemical analysis revealed that TLE3 protein was located in sertoli cell and spermatid in seminiferous tubules of adult mouse testis. In addition, the differential regulation in TLE3 protein expression site was observed in the testis during postnatal development. We also analyzed the expression pattern of HDAC2, which is known to cooperate with TLE3. Both HDAC2 and TLE3 protein were detected in spermatogonial stem cells at one-weeks-old mouse, however, Their expression site were transferred to sertoli cell at six-weeks-old mouse. aken together, TLE3 may play a role in regulating histone acetylation via interaction with HDAC2 in the testis. Futher studies are needed to look into a relation between TLE3 and histone acetylation during spermatogenesis and postnatal development in testis.

Purpose: This study aims to investigate the feasibility of Zenith Fenestrated AAA Endovascular Graft (Z-FEN, Cook Medical) from a single Korean institution database by evaluating the vascular anatomy of Korean abdominal aortic aneurysm (AAA) patients with hostile aortic neck. Methods: This is a retrospective study on patients with AAA who underwent endovascular aortic repair (EVAR) and open surgery repair between January 2012 and December 2021 (n = 211). The anatomic characteristics of the aortic neck were evaluated using 3-dimensional reconstructed computed tomographic scans. For the juxtarenal AAA patients (n = 39), feasibility of fenestrated stent graft was evaluated under the protocol of fenestrated EVAR. For those who were not suitable for the application of Z-FEN, the reasons for unsuitability were analyzed. Results: Among 211 AAA patients, 108 patients (51.2%) had complex aortic neck, and 39 (18.5%) had insufficient aortic neck length (<15 mm) for conventional EVAR. Of the 39 patients with juxtarenal AAAs, 13 (33.3%) were determined feasible for Z-FEN. Twenty-six patients (66.7%) were noncandidate for Z-FEN due to severe neck angulation, short aortic neck length, inadequate iliac artery anatomy, large aortic neck diameter, and severe calcification and thrombosis. Proximal aortic neck length of the non-feasible group was significantly shorter than that of the feasible group (P = 0.002). Conclusion: Z-FEN was applicable to 33.3% of the juxtarenal AAA patients. As recent studies confirm, the effectiveness and safety of fenestrated EVAR, Z-FEN can be an option for AAA patients with short aortic neck.
Characteristics of an Optical Waveguide with Two Identical Elliptical Structures

본 논문에서는 알코올 측정용 센서의 광학적 특성 향상을 위한 고유한 광학적 구조를 제안하였으며, 모의해석을 실시하였다. 광 도파관은 한쪽에 공통초점을 갖는 두 개의 타원형 광 도파관으로 구성되었으며, 각 광 도파관의 서로 다른 초점에 적외선 광원과 적외선 센서를 갖는다. 두 타원형 광 도파관의 각도가 30도에서 90도로 증가시켰을 때, 단위면적당 최대 입사광량은 2.23×10<SUP>6</SUP> W/m<SUP>2</SUP> 에서 5.74×10<SUP>5</SUP> W/m² 로 감소하였다. 그러나 조사 빔의 반지름은 1.86 mm로 최소값을 나타내었고, 전체 입사 에너지는 90도 각도를 갖는 구조와 비교하여 약 10 % 작은 값을 보였다. 모의해석 결과로부터 본 논문에 제안한 구조는 긴 흡수파장을 갖는 광학적 가스센서의 감도향상에 기여할 것으로 판단된다. A unique optical waveguide structure is proposed to enhance the optical characteristics of alcohol screening sensors. This structure is then simulated. The structure consists of two elliptical waveguides that have a common focus to one side and has an IR source and detector at each of the other focal points of the ellipses. When the angle between the two elliptical waveguides is increased from 30 degrees to 90 degrees, the maximum level of irradiance is greatly decreased, falling from 2.23×10<SUP>6</SUP> W/m<SUP>2</SUP> to 5.74×10<SUP>5</SUP> W/m<SUP>2</SUP>. However, the diameter of the incident rays is at a minimum of 1.86mm and the total incident flux is less than 10% lower when compared to the structure at 90°. It can be seen from the simulation results that this structure might enhance the sensitivity of an optical gas sensor which has a large absorption wavelength.
A comparative survey on SAR image segmentation using deep learning

Synthetic Aperture Radar (SAR) image is a radar system that observes topographic maps using microwaves as an active sensor. Due to the backscattering characteristics of SAR, speckle is distributed in the image, making it difficult to analyze. This paper investigates the classically used unsupervised method of SAR image segmentation that can easily recognize and analyze SAR images and the recently used deep learning algorithm, and compare the accuracy using performance metrics. Although the method using deep learning has the problem of insufficient dataset, it improves performance by 10-20% compared to unsupervised. Also, among deep learning algorithms, how the algorithms used in Electro Optical / Infrared (EO / IR) are used in SAR images and problems are investigated. In a recent study, the SAR image considered as a visible light image and applied it to a deep learning algorithm using eo to obtain results. In the future, more benchmark datasets for SAR images should be built, and research on deep learning algorithms using SAR data information will be conducted.

Recent manufacturing system requires development on new production technology to enable prompt manufacturing of diverse products. Most of the researchers have been working on micro-factory. Especially, focus on manufacturing of micro parts. Present manufacturing system consumes excessive resources in the form of energy and space to manufacture the micro parts. In this study, the micro lens module assembly system was modeled, analyzed with MST (Micro System Technology) Application Module and simulated through UML Language(Unified Modeling Language) with object-oriented logical model analysis method. Digital model of micro-factory was modeled, to execute the new paradigm of digitalization on products, resources and processes of micro-factory.
