        Disperser-induced bacterial disintegration of partially digested anaerobic sludge for efficient biomethane recovery

        <P><B>Abstract</B></P> <P>The present study highlights the options to enhance the methane potential of partially digested anaerobic sludge using disperser-induced bacterial disintegration. Floc dispersion with no biomass disintegration was achieved at a disperser-specific energy input of 9.5 kJ/kg TS. The outcomes of both sole bacterial disintegration (S-BD) and disperser-induced bacterial disintegration (D-BD) were assessed in terms of changes in lysate, biopolymer release and increase in biodegradability and methane generation. A higher lysate solubilization of about 22.4% was achieved in D-BD compared to S-BD (11.3%), indicating the efficiency of floc dispersion prior to bacterial disintegration. The biochemical methane result implies that D-BD shows higher methane potential of 0. 279 gCOD/gCOD.         Gallbladder perforation

        Backgrounds/Aims: Gallbladder perforation is a rare but potentially fatal disease. We herein present our clinical experience in diagnosis and management of 32 cases of gallbladder perforation. Methods: This retrospective study was conducted with inclusion of all cases of gallbladder perforation that presented to our hospital from January 2012 to November 2014. Cases of traumatic gallbladder perforation and patients younger than 12 years of age were excluded from this study. Results: This study included 32 patients (13 males and 19 females). The mean age of patients was 55.9 years. Gallbladder perforation was most common in the 5th and 6th decade of life. The mean age of patients with type I, II, and III gallbladder perforation was 57.0 years, 57.6 years, and 49.8 years, respectively. The most common site of perforation was the fundus, followed by the body and Hartmann’s pouch (24 : 5 : 2). Most of the type I gallbladder perforations were diagnosed intraoperatively, type II gallbladder perforations were diagnosed by enhanced abdominal computed tomography, and type III gallbladder perforations were diagnosed during laparoscopic cholecystectomy converted to open cholecystectomy for cholelithiasis. Mortality was highest in patients with type I gallbladder perforation. The mean hospital stay was 10.1 days, 6.4 days, and 9.2 days in patients with type I, II, and III gallbladder perforation, respectively. The histopathologic analysis in 28 patients who were operated on showed acute cholecystitis in 19 cases, acute-on-chronic cholecystitis in 4 cases, chronic cholecystitis in 4 cases, and mucinous adenocarcinoma of the gallbladder in a single case. Conclusions: Gallbladder perforation represents a special diagnostic and surgical challenge. Appropriate classification and management are essential.

        A Study on Behavioral Traits of Library and Information Science Students in South India

        Human behaviour normally depends on the environment of the incident and the time of its occurrence. The behaviour of people depends on many factors and these behaviour traits are an important aspect in the Library and Information Science (LIS) field. Hence in this paper an attempt has been made to examine the behaviour traits of LIS students in South India. Out of 400 questionnaires distributed 367 have responded and the response rate is 91.75%. In this survey three aspects comprising student behaviour have been analysed such as Work Environment, Natural Environment, and Social Environment. In the case of Work Environment the respondents were grouped as Workaholic, Impatience, Achievement oriented, Rash nature, and Punctuality. Further, in respect to Natural environment, the respondents are grouped as Complacent, Patience, Easygoing, and Relaxed. Last, the respondents were grouped in the Social Environment as Balancing nature, Magnanimity, Naturalistic, Assertive nature, Dependency, Lucrative, Lonely nature, and Time Based personality. Finally the authors conclude that LIS students need to possess these qualities and behaviours to work in different environments.

        Explicit incremental matrices for the postbuckling analysis of thin plates with small initial curvature

        The postbuckling behaviour of thin plates is an important phenomenon in the design of thin plated structures. In reality plates possess small imperfections and the behaviour of such imperfect plates is of great interest. To numerically study the postbuckling behaviour of imperfect plates explicit incremental or secant matrices have been presented in this paper. These matrices can be used in combination with any thin plate element. The secant matrices are shown to be very accurate in tracing the postbuckling behaviour of thin plates.

        Profitable ultrasonic assisted microwave disintegration of sludge biomass: Modelling of biomethanation and energy parameter analysis

        <P><B>Abstract</B></P> <P>In this study, microwave irradiation has been employed to disintegrate the sludge biomass profitably by deagglomerating the sludge using a mechanical device, ultrasonicator. The outcomes of the study revealed that a specific energy input of 3.5 kJ/kg TS was found to be optimum for deagglomeration with limited cell lysis. A higher suspended solids (SS) reduction and biomass lysis efficiency of about 22.5% and 33.2% was achieved through ultrasonic assisted microwave disintegration (UMWD) when compared to microwave disintegration – MWD (15% and 20.9%). The results of biochemical methane potential (BMP) test were used to estimate biodegradability of samples. Among the samples subjected to BMP, UMWD showed better amenability towards anaerobic digestion with higher methane production potential of 0.3 L/g COD representing enhanced liquefaction potential of disaggregated sludge biomass. Economic analysis of the proposed method of sludge biomass pretreatment showed a net profit of 2.67 USD/Ton respectively.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Floc deagglomeration happens significantly at 3.5 kJ/kg TS of ultrasonic energy. </LI> <LI> Biomass lysis efficiency of 33.2% was achieved in phase separated disintegration. </LI> <LI> Phase separation shows better amenability towards anaerobic biodegradation. </LI> <LI> Net profit of about 2.67 USD/Ton was achieved in phase separated disintegration. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        α-Gal Nanoparticles in CNS Trauma: II. Immunomodulation Following Spinal Cord Injury (SCI) Improves Functional Outcomes

        BACKGROUND: Previous investigations have shown that local application of nanoparticles presenting the carbohydrate moiety galactose-α-1,3-galactose (α-gal epitopes) enhance wound healing by activating the complement system and recruiting pro-healing macrophages to the injury site. Our companion in vitro paper suggest α-gal epitopes can similarly recruit and polarize human microglia toward a pro-healing phenotype. In this continuation study, we investigate the in vivo implications of α-gal nanoparticle administration directly to the injured spinal cord. METHODS: α-Gal knock-out (KO) mice subjected to spinal cord crush were injected either with saline (control) or with α-gal nanoparticles immediately following injury. Animals were assessed longitudinally with neurobehavioral and histological endpoints. RESULTS: Mice injected with α-gal nanoparticles showed increased recruitment of anti-inflammatory macrophages to the injection site in conjunction with increased production of anti-inflammatory markers and a reduction in apoptosis. Further, the treated group showed increased axonal infiltration into the lesion, a reduction in reactive astrocyte populations and increased angiogenesis. These results translated into improved sensorimotor metrics versus the control group. CONCLUSIONS: Application of α-gal nanoparticles after spinal cord injury (SCI) induces a pro-healing inflammatory response resulting in neuroprotection, improved axonal ingrowth into the lesion and enhanced sensorimotor recovery. The data shows α-gal nanoparticles may be a promising avenue for further study in CNS trauma.         Modulation of Activator Protein-1 (AP-1) and MAPK Pathway by Flavonoids in Human Prostate Cancer PC3 Cells

        In last couple of decades the use of natural compounds like flavonoids as chemopreventive agents has gained much attention. Our current study focuses on identifying chemopreventive flavonoids and their mechanism of action on human prostate cancer cells. Human prostate cancer cells (PC3), stably transfected with activator protein 1 (AP-1) luciferase reporter gene were treated with four main classes of flavonoids namely flavonols, flavones, flavonones, and isoflavones. The maximum AP-1 luciferase induction of about 3 fold over control was observed with $20\;{\mu}M$ concentrations of quercetin, chrysin and genistein and $50\;{\mu}M$ concentration of kaempferol. At higher concentrations, most of the flavonoids demonstrated inhibition of AP-1 activity. The MTS assay for cell viability at 24 h showed that even at a very high concentration $(500\;{\mu}M)$, cell death was minimal for most of the flavonoids. To determine the role of MAPK pathway in the induction of AP-1 by flavonoids, Western blot of phospho MAPK proteins was performed. Four out of the eight flavonoids namely kaempferol, apigenin, genistein and naringenin were used for the Western Blot analysis. Induction of phospho-JNK and phospho-ERK activity was observed after two hour incubation of PC3-AP1 cells with flavonoids. However no induction of phospho-p38 activity was observed. Furthermore, pretreating the cells with specific inhibitors of JNK reduced the AP-1 luciferase activity that was induced by genistein while pretreatment with MEK inhibitor reduced the AP-1 luciferase activity induced by kaempferol. The pharmacological inhibitors did not affect the AP-1 luciferase activity induced by apigenin and naringenin. These results suggest the possible involvement of JNK pathway in genistein induced AP-1 activity while the ERK pathway seems to play an important role in kaempferol induced AP-1 activity.
