In this study, we aimed to evaluate the effect of a-cyperone on S. aureus. We used a hemolysin test to examine the hemolytic activity in supernatants of S. aureus cultured with increasing concentrations of a-cyperone. In addition, we evaluated the production of a-hemolysin (Hla) by Western blotting. Real-time RT-PCR was performed to test the expression of hla (the gene encoding Hla) and agr (accessory gene regulator). Furthermore, we investigated the protective effect of a-cyperone on Hla-induced injury of A549 lung cells by live/dead and cytotoxicity assays. We showed that in the presence of subinhibitory concentrations of a-cyperone, Hla production was markedly inhibited. Moreover, a-cyperone protected lung cells from Hla-induced injury. These findings indicate that a-cyperone is a promising inhibitor of Hla production by S. aureus and protects lung cells from this bacterium. Thus, a-cyperone may provide the basis for a new strategy to combat S. aureus pneumonia.

A disintegrin-like and metalloprotease (reprolysin type) with thrombospondin type 1 motif (ADAMTS1) plays a critical role in follicular rupture and represents a major advance in the proteolytic events that control ovulation. In this study, a 9,026-bp DNA sequence containing the full coding region, all 8 introns and part of the 5'and 3' untranslated region of the porcine ADAMTS1 gene was obtained. Analysis of the ADAMTS1 gene using the porcine radiation hybrid panel indicated that pig ADAMTS1 is closely linkage with microsatellite marker S0215, located on SSC13q49. The open reading frame of its cDNA covered 2,844 bp and encoded 947 amino acids. The coding region of porcine ADAMTS1 as determined by sequence alignments shared 85% and 81% identity with human and mouse cDNAs, respectively. The deduced protein contained 947 amino acids showing 85% sequence similarity both to the human and mouse proteins, respectively. Comparative sequencing of three pig breeds revealed one single nucleotide polymorphism (SNP) within exon 7 of which a G-C substitution at position 6006 changes a codon for arginine into a codon for proline. The substitution was situated within a PvuII recognition site and developed as a PCR-RFLP marker for further use in population variation investigations and association analysis with litter size. Allele frequencies of this SNP were investigated in seven pig breeds/lines. An association analysis in a new Qingping female line suggested that different ADAMTS1 genotypes have significant differences in litter size (p<0.01).
New Evidence of Alleles (V199I and G52S) at the PRKAG3 (RN) Locus Affecting Pork Meat Quality

The porcine PRKAG3 (RN) gene encodes the regulatory gamma subunit of adenosine monophosphate-activated protein kinase (AMPK), which is a good candidate gene affecting meat quality. In this study, the effects of two missense mutations A595G (Ile199Val) and G154A (Gly52Ser) in porcine PRKAG3 gene on meat quality traits were studied in M. Longissimus dorsi (LD), M. Semispinalis capitis (SC) and M. Biceps femoris (BF) from different populations of 326 pigs. The PRKAG3 alleles 199I, 199IV, 52S and 52G were identified with PCR-RFLPs and all genotypes - 199I/199I, 199I/199V, 199V/199V, 52S/52S, 52S/52G and 52G/52G - were found. The frequency of V allele was larger than that of I allele in all populations. I allele frequency was zero in Chinese Meishan pigs (population D) especially. G allele frequency was larger than that of S allele in all populations except Large White (population A). Both variations at the PRKAG3 locus significantly affected these meat quality traits. The pork meat quality has not previously been established in Meishan or crosses thereof. The results suggested that generally pH of LD, SC and BF was higher in Meishan pigs than that in other populations. Moreover, Meishan pigs showed higher water-holding capacity and intramuscular fat (IMF), lower water content and water loss percentage compared to other populations in terms of the two variations. The results present here supply new evidence that alleles V199I and G52S at the PRKAG3 locus affect pork meat quality and provide useful information on pork production.
Some Factors Affecting Freezing of Boar Semen in 5 ml Maxi-straws

A series of experiments were conducted to determine the suitable freezing and thawing temperatures for the freezing of boar semen in 5 ml maxi-straws. The ultrastructure, in vitro fertilization (IVF) and artificial insemination (AI) of frozen-thawed semen were also be evaluated. The 5 cm freezing height gave the best results not only in post-thaw motility rate (54.00%), but also in normal acrosome morphology rate (NAR) (80.23%). There was no significant difference in the post-thaw motility between different thawing temperatures and corresponding thawing times (p>0.05); the group of $52^{\circ}C$ and 25 s gave the highest motility rate (45.00%). As a whole, not only from the motility but also the NAR, thawing at $42^{\circ}C$ was better than the other two treatments. In the freezing packages, 5 ml maxi-straw gave a little lower mobility (40%), viability rate (49.58%), plasma membrane integrity rate (53.91%) and NAR (52.65%) than the 0.25 ml straw, but there was no significant difference between the two straw volumes (p>0.05). The IVF capacity of frozen-thawed semen in this experiment was similar to fresh semen. From ultrastructure observation, the main damage to boar spermatozoa after freezing was seen in the acrosome, such as swelling and formation of vesicles. After AI in recipient Shanghai White sows, frozen-thawed semen from 5 ml maxi-straws and pellets produced 72.2% and 80% conception rate and 7.8 and 8 litter sizes, respectively, and there was no significant difference between the 5 ml maxi-straw and the pellet (p>0.05).

<P><B>Abstract</B></P> <P>In this paper, a spiral-shaped SU-8 cantilever with biocompatibility was proposed to monitor the changes of cardiac contraction in response to injection of drug in real time. The cantilever comprises a thick quadrate body and a filmy spiral cantilever with micro-grooves and gold patterns on the top of its surface. The longitudinally patterned micro-grooves facilitate the aligned growth of cardiomyocytes in order to increase the contraction force. In addition, the spiral shape increases the cantilever’s effective length and decreases its spring constant, which means its bending displacement can be increased for the same force as compared to that observed in a conventional rectangular cantilever. The bending displacement of the spiral SU-8 cantilever induced by the contraction of cardiomyocytes can be precisely measured at the nanoscale using a homemade laser-based measurement system. The displacement in the spiral SU-8 cantilever without micro-grooves was much greater than that in the case of traditional rectangular SU-8 cantilevers. The displacement produced by the spiral cantilever with grooves is 240% greater than that without grooves. Following preliminary experiments, cardiac drugs including Verapamil, Quinidine, Lidocaine, E-4031, and Bay K 8644 were used to evaluate their side effects on cardiomyocytes. Quantitative analysis of measurements shows that not only Verapamil and Quinidine but also Lidocaine and E-4031 treatment can decrease the cell beating frequency, and both the Verapamil and Lidocaine treatment can decrease the cell contraction force. Furthermore, Bay K 8644 assay results indicate that it can decrease the contraction force and increase the beating frequency of cardiomyocytes.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Spiral cantilever integrated with micro-grooves is proposed for a real time monitoring of cardiac contractibility. </LI> <LI> Home-made measurement system is developed to precisely measure the contractibility of cardiomyocytes at the nanoscale. </LI> <LI> Five different kind of drugs have been employed for the preliminary study of drug-induced cardiac toxicity. </LI> <LI> Spiral cantilever arrays are expected to be applied as a new method to support for the drawbacks of conventional methods. </LI> </UL> </P>

In this paper, the problem of object caging and transporting is considered for multiple mobile robots. With the consideration of minimizing the number of robots and decreasing the rotation of the object, the proper points are calculated and assigned to the multiple mobile robots to allow them to form a symmetric caging formation. The caging formation guarantees that all of the Euclidean distances between any two adjacent robots are smaller than the minimal width of the polygonal object so that the object cannot escape. In order to avoid collision among robots, the parameter of the robots radius is utilized to design the caging formation, and the A@? algorithm is used so that mobile robots can move to the proper points. In order to avoid obstacles, the robots and the object are regarded as a rigid body to apply artificial potential field method. The fuzzy sliding mode control method is applied for tracking control of the nonholonomic mobile robots. Finally, the simulation and experimental results show that multiple mobile robots are able to cage and transport the polygonal object to the goal position, avoiding obstacles.
Hydrate nucleation in quiescent and dynamic conditions

Gas hydrate nucleation is the spontaneous formation of an ordered crystalline lattice from a disordered phase. This inherently random process often involves long induction times particularly in quiescent conditions. An experimental study was conducted to explore the kinetics of hydrate formation in the presence of mineral grains and when subjected to mechanical agitation. Results show that tetrahydrofuran THF hydrate nucleation is facilitated in the presence of most minerals, and induction times are a function of mineralogy and surface characteristics. While mechanical vibration does not suppress the inherent stochastic nature of nucleation, mechanical agitation triggers nucleation when the imposed acceleration exceeds ~10m/s<SUP>2</SUP>.
