High sugar concentration culturing was commonly used in modern fermentation industry. However, it leads to the reduction of the foam stability in beer brewing due to the excess secretion of proteinase A. To better understand the characterization of proteinase A excretion from Saccharomyces cerevisiae in high sugar stress conditions, the cultures were grown by using YNB medium with a high concentration of glucose. Pro-PrA isolated from the medium was purified by gel exclusion chromatography, and the PrA activity was detected using fluorescent substrate analysis. The relative molecular weight of the purified PrA and pro-PrA was estimated at 42 and 54 kDa, respectively, by SDS-PAGE. It indicated that the metabolic behavior of PrA in the high glucose culturing was quite different from the normal conditions, and glucose concentration may have a big influence on its secreted process. Further study showed that PrA was released at the logarithmic growth phase of the culturing, and the amount of PrA was 11 times higher compared with the normal culturing. PrA was considered to be activated by itself under acidic conditions. And it was also confirmed in this work that the step-wise pathway for the autoactivation known as a pseudo-PrA has a major contribution to the autoactivation process of PrA zymogen outside the cell.
Biphasic effects of TGFβ1 on BMP9-induced osteogenic differentiation of mesenchymal stem cells

We have found that the previously uncharacterized bone morphogenetic protein-9 (BMP9) is one of the most osteogenic factors. However, it is unclear if BMP9 cross-talks with TGFβ1 during osteogenic differentiation. Using the recombinant BMP9 adenovirus, we find that low concentration of rhTGFβ1 synergistically induces alkaline phosphatase activity in BMP9-transduced C3H10T1/2 cells and produces more pronounced matrix mineralization. However, higher concentrations of TGFβ1 inhibit BMP9-induced osteogenic activity. Real-time PCR and Western blotting indicate that BMP9 in combination with low dose of TGFβ1 potentiates the expression of later osteogenic markers osteopontin, osteocalcin and collagen type 1 (COL1a2), while higher concentrations of TGFβ1 decrease the expression of osteopontin and osteocalcin but not COL1a2. Cell cycle analysis reveals that TGFβ1 inhibits C3H10T1/2 proliferation in BMP9-induced osteogenesis and restricts the cells in G0/G1 phase. Our findings strongly suggest that TGFβ1 may exert a biphasic effect on BMP9-induced osteogenic differentiation of mesenchymal stem cells. [BMB Reports 2012; 45(9): 509-514]
Inhibition of Nemo-like Kinase Increases Taxol Sensitivity in Laryngeal Cancer

Several studies have shown that nemo-like kinase (NLK) plays a vital role in apoptosis of cancer cells. The present research concerned effects and mechanisms of Taxol on NLK knockdown human laryngeal cancerHep-2 cell lines in vitro. Using RNAi, methyl-thiazoltetrazolium (MTT) assays, real-time RT-PCR, Western blotting and flow cytometry analysis, growth and the cell cycle progression of NLK knockdown Hep-2 cells and expression of downstream molecules were observed. Cell growth was obviously suppressed in the Taxol treated group (P<0.001, 48 hours). Cell numbers of combined Taxol-based chemotherapy with lentivirus mediated RNAi treatment group (Lv-shNLK+Taxol goup) were significantly different from NLK-specific siRNA lentivirus infected group (Lv-shNLK group) (p<0.001). Flow cytometry analysis revealed that Lv-shNLK+Taxol caused the G0/G1-phase DNA content to decrease from 44.1 to 3.33% (p<0.001) and the S-phase DNA content to increase from 38.4 to 82.0% (p<0.001), in comparison with the Lv-shNLK+Taxol group. Immunoblot analysis showed that knockdown of NLK led to significant reduction in the levels of cyclin D1, PCNA and PARP, whereas cyclin B1 was elevated in. Cell growth was also obviously suppressed in the Hep-2 cell line, knockdown of NLK making them more sensitive to Taxol treatment. NLK is expected to become a target of new laryngeal cancer gene therapies.
A Study on Multi-Layered EM Wave Absorber Using Natural Lacquer as a Binder

Generally, a silicone rubber and a chlorinated polyethylene/ CPE) have been used as a binder for the development of high-performance composite EM(Electro Magnetic) wave absorbers. In this paper, the EM wave absorption performance of natural lacquer, which is newly proposed as a binder was investigated. The prepared MnZn ferrite EM wave absorbers are mixed with natural lacquer showed excellent EM wave absorption characteristics compared with MnZn ferrite EM wave absorbers which are mixed with the conventional binders. MnZn ferrite EM wave absorbers mixed with natural lacquer were prepared and their absorption ability was also investigated The EM wave absorbers are fabricated in different proportions of MnZn, or NiZn ferrite and natural lacquer, and their reflection coefficients are measured The permittivity and permeability are calculated by using the measured reflection coefficients. The EM wave absorption abilities are calculated according to different thicknesses of the EM wave absorbers.
Effect of Muscle Soft Tissue on Biomechanics of Lumbar Spine under Whole Body Vibration

The object of this study was to simulate effect of muscle soft tissue on resonant frequencies and configuration (deformation and displacement) of lumbar spine in vivo under whole-body vibration (WBV). The three-dimensional (3D) finite element (FE) models of human body including skin, muscle soft tissue, viscera, ligaments, lumbar intervertebral disc (LID) and skeleton were created and validated. Modal analysis was performed by FE method. Results were that muscle soft tissue increased the resonant frequencies of lumbar spine. The first vertical resonant frequency (FVRF) changed greatly with the variation of Young’s modulus of muscle soft tissue in range of 0.01 MPa-0.15 MPa. Muscle soft tissue reduced the modal displacement of the lumbar spine. Especially the modal displacement in the anteroposterior direction was reduced by at least 40%. The risk injury area of lumbar was at low lumbar region under vertical WBV. The conclusions were that muscle soft tissue played a very important role in biomechanics of lumbar spine, and we could contract muscles of whole body for protection of lumbar spine under WBV with high vibration amplitude. The findings could be helpful in understanding the biomechanics of lumbar spine under WBV and offer potential references for spinal disease protection and orthopedics.
Mechanical evaluation of polymer microneedles for transdermal drug delivery: In vitro and in vivo

In this study, we reported two types of PMNs based on polylactic acid (PLA) and polyvinyl alcohol (PVA),respectively. Parafilm M film, porcine skin, and rats’ models were operated to evaluate the mechanicalproperties in vitro and in vivo to find optimal parameters for efficient insertion. Insertion depth was measuredusing Digital Force Gauge by changing insertion force and speed, respectively. Results showed thatincreasing the insertion force and speed used for PMNs application led to a significant increase in thedepth of insertion. A force of 18 N under a speed of 330 mm/min was the optimal condition for insertingPMNs array into ParafilmM film and porcine skin. In addition, PLA-MNs exhibited higher robustness andenhanced homogeneity in insertion depth compared with PVA-MNs, but PVA-MNs were able to reachmuch deeper insertion depth. Moreover, Sprague Dawley (SD) rat experiments confirmed the effectivenessof optimal insertion parameters for transdermal drug delivery. This study illustrated not only thedevelopment of novel PMNs but also the mechanical evaluation for the design of PMNs.
Marine (sea) Familiarity Index and Its Influence to National Image

Purposes - This study discusses various aspects of familiarity with the sea and how familiarity with the sea is perceived. This study also summarizes the literature on the factors of previous researches that affect the formation or decision of national image. Research design and methodology - Sea familiarity index was derived from the results of recognition of free association with the sea, friendliness with the sea, association of the sea with life, interest to the sea, Korea's representative marine regions, and sea related emotionalism. The familiarity with the sea (ocean) perceived by foreign visitors or foreigners in the future will be helpful in deriving a comparative research. Results - This study has identified the marine tourism resources, marine ecology, and the cognitive and emotional situations consumers feel that should be developed practically. If a continuous time series research is conducted in the future through these, it will be a chance to provide a basis for consumers' dynamic awareness. Conclusions - Through this study, it is pointed out that the research utilizing the sea and ocean to form national image highly suggested an opportunity to add and utilize new variables academically. This study on the perspective of the formation of national image suggests a new approach to add and utilize new variables to future research using the sea and ocean.
