        Effect of Graphene on Growth of Neuroblastoma Cells

        The unique properties of graphene have earned much interest in the fields of materials science and condensedmatter physics in recent years. However, the biological applications of graphene remain largely unexplored. In this study, we investigated the conditions and viability of a cell culture exposed to graphene onto glass and SiO2/Si, using a human nerve cell line, SH-SY5Y. Cell viability was 84% when cultured on glass and SiO2/Si coated with graphene as compared with culturing on polystyrene surface. Fluorescence data showed that the presence of graphene did not influence cell morphology. These findings suggest that graphene may be used for biological applications.

        태양열 건조 시스템에 관한 실험적 연구(Ⅱ) - 벼의 태양열 건조 특성 -

        곡물 bin을 이용한 벼의 건조실험을 수행하여 상온통풍건조 시스템과 태양열 건조 시스템의 건조 특성을 비교하여 분석하였다. 태양열 건조의 시뮬레이션 모형에 대한 컴퓨터 프로그램을 개발하였으며 시뮬레이션 모형에 의한 함수율의 예측치는 실측치와 전체적으로 일치하였다.<br/> 또한, inverter를 사용하여 벼의 건조 상태에 따라 송풍량을 적절하게 제어하는 송풍량 제어 시스템을 개발하였으며, 개발된 송풍량 제어 시스템을 통하여 에너지 절약 효과를 얻을 수 있었다. In-bin grain drying experiments were performed to investigate the drying characteristics between natural air and solar heated-air drying system of rough rice. A computer simulation model for solar drying system of rough rice resulted in a good agreement between the experimental and predicted moisture content.<br/> In order to save the electric energy consumption of fan, airflow rates control system using inverter was developed and resulted in the effect of energy saving.

        유통중인 즉석섭취 새싹채소와 샐러드의 세균오염 분석

        즉석섭취편이 식품중 신선편의식품(fresh-cut-vegetable)인 새싹 채소와 샐러드의 수요는 급증하고 있으나 특히 새싹채소의 미생물 품질평가를 위한 자료가 거의 없는 상황이다. 따라서 국내 대형할인마트에서 유통중인 새싹채소 50제품, 샐러드 30제품을 수집하여 총 호기성균, 대장균군, E. coli, Salmonella spp, S. aureus, B. cereus, L. monocytogenes, Cam. jejuni, C. perfringens의 오염연구를 수행하였다. 총 호기성균은 새싹채소에서는 7.95 log CFU/g 검출되었으며 샐러드에서 평균 6.70 log CFU/g검출되어 샐러드 보다 새싹채소에서 약 1 log CFU/g 더 높게 검출되었다. 대장균군은 새싹채소에서 6.69 log CFU/g 검출되었고 샐러드에서 평균 5.42 log CFU/g 검출되었다. E. coli은 새싹채소 50제품 중 16제품(32%)에서 2.38 log CFU/g으로 검출되었고 샐러드 30제품 중 8제품(27%)에서 2.21 log CFU/g으로 검출되었다. B. cereus는 새싹채소 50제품 중 16제품(32%)이 검출되었고 샐러드 30제품 중 16제품(53%)가 검출되었으나 대부분이 3 log CFU/g 이하의 검출율을 보여 주었다. Salmonella, S. aureus, L. monocytogenes, Cam. jejuni, C. perfringens는 검출되지 않았다. 그러므로 분석된 새싹채소와 샐러드에서 B. cereus이외에 다른 식중독 병원성 미생물의 위해 가능성은 크지 않은 것으로 보인다. 그러나 총 세균수가 높게 검출되었고 위생지표세균인 대장균의 검출율이 높아서 이들 식품의 미생물 안전성을 확보하기 위해서 보다 더 철저한 생산유통관리가 요망되며 특별히 새싹채소에 대한 미생물안전성 확보에 대한 노력이 요구된다. Although bacterial outbreaks from ready-to-eat foods have increased, little information is available on microbial quality of sprouts in markets. Fifty sprouts and 30 salads were collected from wholesale markets. Total aerobic count (TAC), coliform, Escherichia coli, and some pathogens were detected. TAC for sprouts was 7.95 log CFU/g and 6.70 for salads, indicating that sprouts were more contaminated by 1 log CFU/g than that of salads. The numbers of coliform were 6.69 log CFU/g for sprouts and 5.42 for salads. E. coli was detected in 16 of 50 sprout samples at 2.38 log CFU/g and eight of 30 salads at 2.21 log CFU/g. Bacillus cereus was detected in 29 of 50 sprout samples and 16 of 30 salads, and the counts were mostly <3 log CFU/g. Salmonella, Staphylococcus aureus, Listeria monocytogenes, Campylobacter jejuni, and Clostridium perfringens were not detected. Therefore, although pathogens may not be a high risk for these foods, the high TAC and E. coli contamination require improved production and distribution methods, particularly for sprouts.

        성인초기 간호대학생의 긍정심리자본과 스트레스대처반응이 COVID-19 스트레스에 미치는 영향

        본 연구는 간호대학생의 긍정심리자본과 스트레스대처반응이 COVID-19 스트레스에 미치는 영향을 확인하기 위한 서술적 조사연구이다. 대상자는 2021년 7월 23일부터 8월 11일까지 S시에 소재한 일개 대학에 재학 중인 1~4학년의 간호대학생 176명을 편의표집 하였다. 자료수집은 자발적 연구 참여에 동의한 간호대학생에게 자가보고형 설문지를 온라인 구글서베이로 하였고, 자료분석은 SPSS 28.0 프로그램을 이용하여 기술통계, 피어슨 상관계수, 다중회귀분석으로 하였다. 연구 결과, 간호대학생의 연령(β=-0.24, p =.002)이 낮을수록, 스트레스대처반응(β=0.18, p =.024)이 높을 수록 COVID-19 스트레스가 높은 것으로 나타났다. 반면에 긍정심리자본은 COVID-19 스트레스에 유의미한 영향이 확인되지 않았다. 따라서 추후 연구에서 COVID-19 위기 상황에서 성인초기인 간호대학생의 연령 등의 일반적 특성과 스트레스대처반응을 중심으로 COVID-19 스트레스를 통합적으로 접근하고, 간호 교육 및 실무에서 COVID-19 스트레스의 관련 요인을 효과적으로 관리할 수 있는 중재를 제공할 것을 제안한다. This study aimed to identify the effects of positive psychological capital and stress coping response on coronavirus disease 19 (COVID-19) stress of nursing students. A total of 176 nursing students at one university in S city, Korea, were recruited by convenience sampling from July 23 to August 11, 2021. Data were collected via a self-reported questionnaire using an online Google survey and analyzed with descriptive statistics, Pearson"s correlation coefficient and multiple regression analysis to identify the predictors of COVID-19 stress using SPSS 28.0 program. Lower age (β=-0.24, p =.002) and higher stress coping response (β=0.18, p =.024) were observed, corresponding to increased COVID-19 stress. However, positive psychological capital was not an affecting factor of COVID-19 stress. This study suggests an integrated approach in future study and effective interventions in nursing education and practice based on general features, such as age and stress coping response for COVID-19 stress management of nursing students in young adults in COVID-19 pandemic.

      • Endoplasmic Reticulum Stress-Mediated Apoptosis of EBV-Transformed B Cells by Cross-Linking of CD70 Is Dependent upon Generation of Reactive Oxygen Species and Activation of p38 MAPK and JNK Pathway

        CD70 is expressed in normal activated immune cells as well as in several types of tumors. It has been established that anti-CD70 mAb induces complement-dependent death of CD70^+ tumor cells, but how anti-CD70 mAb affects the intrinsic signaling is poorly defined. In this report, we show that ligation of CD70 expressed on EBV-transformed B cells using anti-CD70 mAb induced production of reactive oxygen species (ROS) and subsequent apoptosis. We observed an early expression of endoplasmic reticulum (ER) stress response genes that preceded the release of apoptotic molecules from the mitochondria and the cleavage of caspases. CD70-induced apoptosis was inhibited by pretreatment with the ER stress inhibitor salubrinal, ROS quencher N-acetylcysteine, and Ca^(2+) chelator BAPTA. We supposed that ROS generation might be the first event of CD70-induced apoptosis because N- acetylcysteine blocked increases of ROS and Ca^(2+), but BAPTA did not block ROS generation. We also round that CD70 stimu¬lation activated JNK and p38 MAPK. JNK inhibitor SP600125 and p38 inhibitor SB203580 effectively blocked upregulation of ER stress-related genes and cleavage of caspases. Inhibition of ROS generation completely blocked phosphorylation of JNK and p38 MAPK and induction of ER stress-related genes. Taken together, we concluded that cross-linking of CD70 on EBV-transformed B cells triggered ER stress-mediated apoptosis via ROS generation and JNK and p38 MAPK pathway activation. Our report reveals alternate mechanisms of direct apoptosis through CD70 signaling and provides data supporting CD70 as a viable target for an Ab-based therapy against EBV-related tumors.

        Anti-inflammatory effect of the extracts from the branch of <i>Taxillus yadoriki</i> being parasitic in <i>Neolitsea sericea</i> in LPS-stimulated RAW264.7 cells

        <P><B>Abstract</B></P> <P>Mistletoe has been used as the herbal medicine to treat hypertension, diabetes mellitus, inflammation, arthritis and viral infection. In this study, we evaluated the anti-inflammatory effect of extracts of branch from <I>Taxillus yadoriki</I> being parasitic in <I>Neolitsea sericea</I> (TY-NS-B) using <I>in vitro</I> model. TY-NS-B significantly inhibited LPS-induced secretion of NO and PGE<SUB>2</SUB> in RAW264.7 cells. TY-NS-B was also observed to inhibit LPS-mediated iNOS COX-2 expression. In addition, TY-NS-B attenuated production of inflammatory cytokines such as TNF-α and IL-1β induced by LPS. TY-NS-B blocked LPS-mediated inhibitor of IκB-α, and inhibited p65 translocation to the nucleus and NF-κB activation. Furthermore, TY-NS-B reduced the phosphorylation of MAPKs such as p38 and JNK, but not ERK1/2. In addition, TY-NS-B increased ATF3 expression and ATF3 knockdown by ATF3 siRNA attenuated TY-NS-B-mediated inhibition of pro-inflammatory mediator expression. Collectively, our results suggest that TY-NS-B exerts potential anti-inflammatory effects by suppressing NF-κB and MAPK signaling activation, and increasing ATF3 expression.         Short Term Load Forecasting Algorithm for Lunar New Year's Day

        Short term load forecasts complexly affected by socioeconomic factors and weather variables have non-linear characteristics. Thus far, researchers have improved load forecast technologies through diverse techniques such as artificial neural networks, fuzzy theories, and statistical methods in order to enhance the accuracy of load forecasts. Short term load forecast errors for special days are relatively much higher than that of weekdays. The errors are mainly caused by the irregularity of social activities and insufficient similar past data required for constructing load forecast models. In this study, the load characteristics of Lunar New Year's Day holidays well known for the highest error occurrence holiday period are analyzed to propose a load forecast technique for Lunar New Year's Day holidays. To solve the insufficient input data problem, the similarity of the load patterns of past Lunar New Year's Day holidays having similar patterns was judged by Euclid distance. Lunar New Year's Day holidays periods for 2011-2012 were forecasted by the proposed method which shows that the proposed algorithm yields better results than the comprehensive analysis method or the knowledge-based method.

        Cross-linking of CD80 and CD86 Diminishes Expression of CD54 on EBV-transformed B Cells through Inactivation of RhoA and Ras

        Background: Epstein Barr virus (EBV) infected B cells are transformed into lymphoblastoid cell lines. Some researchers suggested some a few similarities between this process and carcinogenesis. We observed the expression of CD80 and CD86, co-stimulatory molecules on EBV-transformed B cells and changes of CD54 expression after stimulation of CD80 and CD86. Methods: CD80 and CD86 were stimulated using anti-CD80 and anti-CD86 monoclonal antibodies. To assess apoptosis and surface protein expression, flow cytometric analysis was performed. Intracellular signal molecules were evaluated by RT-PCR and immunoblot. Morphology and localization of proteins were examined using inverted or confocal microscope. Results: Cross-linking of CD80 and CD86 induced apoptosis and interfered with proliferation of EBV-transformed B cells, and dispersion of clumped cells. We also examined that their stimulation induced ROS accumulation and reduced CD54 expression. Interestingly, we observed that CD80 and CD86 diminished the expression of CD54 in different methods. Both CD80 and CD86 downregulated activation of focal adhesion kinase. CD80 stimulus inhibited CD54 expression through mainly RhoA inactivation, while CD86 down-regulated Ras and JNK phosphorylation. Conclusion: These results suggest that co-stimulatory CD80 and CD86 molecules, expressed EBV-transformed B cells, may play a role in apoptosis and cell adhesion.

        Changes in Anxiety Level and Personal Protective Equipment Use Among Healthcare Workers Exposed to COVID-19

        Background: The relationship between changes in anxiety levels and personal protective equipment (PPE) use is yet to be evaluated. The present study assessed this relationship among healthcare workers (HCWs) involved in the care of patients with coronavirus disease 2019 (COVID-19). Methods: An online survey was conducted in a municipal hospital with 195 nationally designated negative pressure isolation units in Korea. Anxiety level was measured using the self-rating anxiety scale (SAS), and changes in anxiety levels were assessed based on the time when COVID-19 vaccine was introduced in March 2021 in Korea. Monthly PPE usage between June 2020 and May 2021 was investigated. Results: The mean SAS score (33.25 ± 5.97) was within normal range and was lower than those reported in previous studies conducted before COVID-19 vaccination became available. Among the 93 HCWs who participated, 64 (68.8%) answered that their fear of contracting COVID-19 decreased after vaccination. The number of coveralls used per patient decreased from 33.6 to 0. However, a demand for more PPE than necessary was observed in situations where HCWs were exposed to body fluids and secretions (n = 38, 40.9%). Excessive demand for PPE was not related to age, working experience, or SAS score. Conclusion: Anxiety in HCWs exposed to COVID-19 was lower than it was during the early period of the pandemic, and the period before vaccination was introduced. The number of coveralls used per patient also decreased although an excessive demand for PPE was observed.

        Wood-cultivated ginseng exerts anti-inflammatory effect in LPS-stimulated RAW264.7 cells

        <P><B>Abstract</B></P> <P>Ginseng (<I>Panax ginseng</I>) has been reported to exert an anti-inflammatory activity in a variety of inflammatory condition. However, inflammation-regulatory activity of wood-cultivated ginseng has not been thoroughly evaluated. In this study, we evaluated the anti-inflammatory effect of wood-cultivated ginseng (WCG) and elucidated the potential mechanisms in LPS-stimulated RAW264.7 cells. WCG-O dose-dependently suppressed NO and PGE<SUB>2</SUB> production in LPS-stimulated RAW264.7 cells. In addition, WCG-O attenuated LPS-mediated overexpression of iNOS and COX-2. In addition, WCG-O blocked the expression of TNF-α and IL-1β. WCG-O inhibited the activation of IκK-α/β, the phosphorylation of IκB-α, and degradation of IκB-α, which results in the inhibition of p65 nuclear accumulation and NF-κB activation. In addition, WCG-O suppressed the activation of ERK1/2, p38 and JNK, which results in the inhibition of ATF2 nuclear accumulation. These results indicate that WCG-O may exert anti-inflammatory activity by inhibiting NF-κB and MAPK signaling.
