Streptomyces peucetius ATCC 27952 is responsible for producing clinically important chemotherapeutic ugents, daunorubicin (DNR) and doxorubicin (DXR), adraimycin. We focused to develop hybrid doxorubicin by modifving the daunosamine sugar with other sugar moieties. The attachment of sugar moieties is facilitated with same glycosyltransferase of S.prucetius ATCC 27952 or without using its own glycosyltransferase. Proper modification on gene cluster of daunosamine by gene(s) from other biosynthetic gene cluster of deoxysugar leads to various hybrid doxorubicin. We expect that introduction of doxorubicin/daunorubicin resistance gene, drrC into wild type S.peucetius ATCC 27952 by pSET152 vactor assisted with ermE* promoter enhances the production of antibiotics
Does Anastomosis Configuration Influence Long-term Outcomes in Patients With Crohn Disease?

Purpose: In this study, we evaluated the role of various anastomoses in surgical recurrence for patients with Crohn disease (CD). Methods: We analyzed data retrospectively from consecutive laparotomy cases involving complicated CD between 1991 and 2008. Clinical data were compared in terms of reoperation-free survival (RFS) according to the types of anastomoses, the materials used for the anastomoses, and the operating surgeon. Results: Of 233 patients with entero-enteric or entero-colic anastomoses, 199 (85%), 11 (5%), and 23 (10%) experienced side-to-side (SS), side-to-end (SE), and end-to-end (EE) anastomoses, respectively. The SS group had the following characteristics: more extensive bowel involvement, frequent obstruction, and greater stapler use; the SS anastomoses were also frequently made by specialized surgeons (P < 0.001–0.004). EE anastomoses were frequently made by general surgeons using a hand-sewing technique (P < 0.001). No differences in RFS were noted among the 3 groups according to the type of anastomosis and the operating surgeon. However, the hand-sewn group showed better RFS than the stapler group (P = 0.04). Conclusion: The roles of the anastomotic configuration, the material used, and the operating surgeon were not significantly correlated with reoperations or complications in our retrospective CD cohort, irrespective of the higher risk of anastomosis site stricture for EE anastomoses.

<P>Acrylic acid monomer in a viscous supporting nylon solution was polymerized and fabricated simultaneously via an electrospinning process. This novel polymerization method defines the fiber morphology as a network of interconnected mats. This network consists of smaller poly(acrylic acid) (PAA) fibers, approximately 19 nm in diameter, and larger nylon 6 fibers, approximately 75 nm in diameter. These two different fibers are separated by extraction of PAA from the fibrous mat in water and differentiation of field-emission scanning electron microscopy (FESEM) images of the same mat using before and after extraction of PAA. The structure of the extracted PAA was confirmed by (1)H NMR and Fourier transform infrared (FT-IR) analysis. The observed modification to conventional electrospun mats is due to the presence of an extra phase-separated PAA produced by the electrospinning polymerization process. Finally, fiber morphologies and hybrid property were characterized via FT-IR, FESEM, thermogravimetric analysis, and X-ray diffraction. Similarly extracted samples and synthesis PAA were characterized in FT-IR and (1)H NMR spectroscopy.</P>

<P><B>Abstract</B></P><P>We have found that the thermophilic activity of glucose-1-phosphate thymidylyltransferase (stRmlA) is conditional upon the presence certain substances. In particular, it showed its thermal stability only in the presence of 50mM dTTP or dTMP, thermal stability being the highest at 70°C. The purified enzyme was stable up to 90°C within a broad pH range from 2.0 to 13.0, and its maximum activity was measured at a pH of 11.5 at 70°C. Unlike other mesophilic counterparts, it showed catalytic activity in the presence of various metal ions in the following order of reactivity: Mg<SUP>2+</SUP>>Zn<SUP>2+</SUP>>Cu<SUP>2+</SUP>>Co<SUP>2+</SUP>>Fe<SUP>2+</SUP>>Ca<SUP>2+</SUP>>Fe<SUP>3+</SUP>>Ni<SUP>2+</SUP>. Its catalytic activity was not inhibited even by the denaturants 50mM guanidine hydrochloride and 50mM urea.</P><P>To explore the molecular basis for its unusual thermostability, homology structural modeling, codon usage comparisons, and amino acid composition analyses were performed. The CCC, CUC and UCC codons are significantly higher, and distributed uniformly in <I>strmlA</I> as contrasted with other GPTTs. Our analyses revealed that the GC rich coding sequences may not be the reason for thermostability of stRmlA. The β-sheets are also found more likely in stRmlA, which contributes to thermal stability by increasing the number of hydrogen bonds. Further results suggest that a large number of apolar functional groups exposed to solvent accessible surface area, a significant number of residues sensitive to oxidation or deamination, and a higher hydrophobicity, any or all of the which could be reasons for its unique thermophilic behavior. Finally, we postulate that dTTP or dTMP enters the active site and binds tightly to inhibit the defolding of the protein, which, in turn, increases its thermal stability. Such findings will be useful for further investigations on thermophilic behavior of enzymes.</P>

In this study, we studied whether hydrogen sulfide ($H_2S$) has an effect on the pacemaker activity of interstitial cells of Cajal (ICC), in the small intestine of mice. The actions of $H_2S$ on pacemaker activity were investigated using whole-cell patch-clamp technique, intracellular $Ca^{2+}$ analysis at $30^{\circ}C$ and RT-PCR in cultured mouse intestinal ICC. Exogenously applied sodium hydrogen sulfide (NaHS), a donor of hydrogen sulfide, caused a slight tonic inward current on pacemaker activity in ICC at low concentrations (50 and $100{\mu}m$), but at high concentration ($500{\mu}m$ and 1 mM) it seemed to cause light tonic inward currents and then inhibited pacemaker amplitude and pacemaker frequency, and also an increase in the resting currents in the outward direction. Glibenclamide or other potassium channel blockers (TEA, $BaCl_2$, apamin or 4-aminopydirine) did not have an effect on NaHS-induced action in ICC. The exogenous application of carbonilcyanide p-triflouromethoxyphenylhydrazone (FCCP) and thapsigargin also inhibited the pacemaker activity of ICC as NaHS. Also, we found NaHS inhibited the spontaneous intracellular $Ca^{2+}$ ($[Ca^{2+}]_i$) oscillations in cultured ICC. In doing an RT-PCR experiment, we found that ICC enriched population lacked mRNA for both CSE and CBS, but was prominently detected in unsorted muscle. In conclusion, $H_2S$ inhibited the pacemaker activity of ICC by modulating intracellular $Ca^{2+}$. These results can serve as evidence of the physiological action of $H_2S$ as acting on the ICC in gastrointestinal (GI) motility.

Aim: To evaluate the differential diagnostic potential of lesion stiffness assessed by the sonoelastographic strain index ratio (SR) and elastographic color scoring system (UE) for breast lesions. Materials and Methods: Three hundred and forty two breast masses (158 benign and 184 malignant) from 325 consecutive patients (mean age 44.2 years; range 16-81)who had been scheduled for a sonographically guided core biopsy were examined proposed by Itoh et al, with scoring 1-3=benign and 4-5=malignant. Strain and area ratios of each lesion were calculated within the same machine. Histological diagnosis was used as the reference standard. The area under the curve (AUC) and cut-off point were obtained by receiver operating curve and the cross table Fischer Test was carried out for assessing diagnostic value. Sensitivity, specificity, PPV, NPV, accuracy and false-discovery rates were compared. Results: The mean strain ratios for benign and malignant lesions were 1.87 and 7.9 respectively. (P<0.0001). When a cutoff point of 3.54 was used, SR had a sensitivity of 94.6%, a specificity 94.3%, a PPV of 95.1%, an NPV of 93.7% and an accuracy of 94.4%. The AUC values were 0.90 for the 5 point scoring system (UE) and 0.96 for the strain index ratio. The overall diagnostic performance was SR method was better (P<0.05). Conclusions: Strain ratio measurement could be another effective predictor in elastography imaging besides 5 the point scoring system for differential diagnosis of breast lesions.
Diversity of Cytochrome P450 in Streptomyces peucetius

We have determined the genome sequences of 8.7 Mb chromosome of Streptomyces peucetiusATCC 27952, which produces clinically important anthracyline chemotherapeutic agents of the polyketide class of antibiotics, daunorubicin and doxorubicin. The cytochrome P450 gene superfamily is represented by 19 sequences in the S. peucetius. Amongthose, 15 code for apparently functional genes whereas four are apparently pseudo genes. Four cytochrome P450s are associated with modular PKS of avermectin, and two with doxorubicin biosynthetic gene cluster. CYP252A1 is the new family found in S. peucetius, which shares 38% identity to CYP51 from Streptomyces coelicolor A3 (2).
