        Consequences of Post-grazing Residues Control and Birth Season on the Body Traits, Reproductive Performance and Offspring's Growth of Suckling Goats and Ewes Reared at Pasture in Guadeloupe (FWI)

        In Guadeloupe small ruminants (SR) are reared for meat production under pasture conditions. Intensive rotational grazing systems (irrigated, fertilised and high stocked) allow reasonable levels of production but generate high post-grazing residues. Experiments were designed to control them. A system in which residuals were mown (RM) was tested in comparison to the control system (Residuals Remained, RR). The same design was carried out for two years with Creole goat (G) and Martinik sheep (S). An accelerated reproductive rate (3 parturitions over 2 years) was carried out. Systems were compared at three parturition seasons per year(dry, intermediate and rainy seasons). Each group was composed of 20 goats ($36.0{\pm}2.5kg$) or 20 ewes ($46.8{\pm}2.4kg$). The female body traits did not vary according to pasture management and seasons. The stocking rate averaged 1,400 kg LW/ha. The mean fertility rate for does varied significantly (p<0.05) within the kidding season, from $80.4{\pm}0.5%$ to $93.7{\pm}2.9%$ while the mean litter size was $2.30{\pm}0.07$ total kids born. No effect of pasture system was observed. Corresponding values for ewes were $83.2{\pm}12.8%$ vs. $75.6{\pm}12.5%$ (p<0.05) and $2.43{\pm}0.24$ vs. $2.03{\pm}0.29$ (p<0.01) total lambs born for SRM and SRR ewes, respectively. A seasonal effect was observed upon ewe performances. The preweaning mortality of kids and lambs averaged 16.3% and 14.4%, respectively. It was 7 and 9 percentage points more (p<0.01) for RR than for RM kids and lambs, respectively. For both species, weaning took place at an average age of $81.4{\pm}3.6days$. In Creole kids, live weight at birth and at weaning were $1.9{\pm}0.2kg$ and $8.9{\pm}0.8kg$, respectively. In the Martinik sheep, the traits averaged $2.9{\pm}0.2kg$ and $18.9{\pm}0.9kg$. For both traits in both species, significant (p<0.05) group${\times}$season interactions were recorded. The consequences of elimination of post-grazing residues varied according to the SR species, the environmental conditions and the animal physiological status. The forage characteristics were not limiting factors since forage availability in the RM systems (2,300 and 2,600 kg DM/ha, respectively) and chemical composition were at satisfactory levels (CP content averaged 12 and 10%). It is recommended to develop new grazing system which would allow the use of post-grazing residues instead of mowing the refusals.

        Pathogenic potential of non-typhoidal Salmonella serovars isolated from aquatic environments in Mexico

        Background River water has been implicated as a source of non-typhoidal Salmonella (NTS) serovars in Mexico. Objective To dissect the molecular pathogenesis and defense strategies of seven NTS strains isolated from river water in Mexico. Methods The genome of Salmonella serovars Give, Pomona, Kedougou, Stanley, Oranienburg, Sandiego, and Muenchen were sequenced using the whole-genome shotgun methodology in the Illumina Miseq platform. The genoma annotation and evolutionary analyses were conducted in the RAST and FigTree servers, respectively. The MLST was performed using the SRST2 tool and the comparisons between strains were clustered and visualized using the Gview server. Experimental virulence assay was included to evaluate the pathogenic potential of strains. Results We report seven high-quality draft genomes, ranging from ~ 4.61 to ~ 5.12 Mb, with a median G + C value, coding DNA sequence, and protein values of 52.1%, 4697 bp, and 4,589 bp, respectively. The NTS serovars presented with an open pan-genome, offering novel genetic content. Each NTS serovar had an indistinguishable virulotype with a core genome (352 virulence genes) closely associated with Salmonella pathogenicity; 13 genes were characterized as serotype specific, which could explain differences in pathogenicity. All strains maintained highly conserved genetic content regarding the Salmonella pathogenicity islands (1–5) (86.9–100%), fimbriae (84.6%), and hypermutation (100%) genes. Adherence and invasion capacity were confirmed among NTS strains in Caco-2 cells. Conclusion Our results demonstrated the arsenal of virulence and defense molecular factors harbored on NTS serovars and highlight that environmental NTS strains are waterborne pathogens worthy of attention.

        Molecular diversity of the VP2 of Carnivore protoparvovirus 1 (CPV-2) of fecal samples from Bogotá

        Background: Carnivore protoparvovirus 1, also known as canine parvovirus type 2 (CPV-2), is the main pathogen in hemorrhagic gastroenteritis in dogs, with a high mortality rate. Three subtypes (a, b, c) have been described based on VP2 residue 426, where 2a, 2b, and 2c have asparagine, aspartic acid, and glutamic acid, respectively. Objectives: This study examined the presence of CPV-2 variants in the fecal samples of dogs diagnosed with canine parvovirus in Bogotá. Methods: Fecal samples were collected from 54 puppies and young dogs (< 1 year) that tested positive for the CPV through rapid antigen test detection between 2014–2018. Molecular screening was developed for VP1 because primers 555 for VP2 do not amplify, it was necessary to design a primer set for VP2 amplification of 982 nt. All samples that were amplified were sequenced by Sanger. Phylogenetics and structural analysis was carried out, focusing on residue 426. Results: As a result 47 out of 54 samples tested positive for VP1 screening, and 34/47 samples tested positive for VP2 980 primers as subtype 2a (n = 30) or 2b (n = 4); subtype 2c was not detected. All VP2 sequences had the amino acid, T, at 440, and most Colombian sequences showed an S514A substitution, which in the structural modeling is located in an antigenic region, together with the 426 residue. Conclusions: The 2c variant was not detected, and these findings suggest that Colombian strains of CPV-2 might be under an antigenic drift.

        Effect of condensed tannins from Leucaena leucocephala on rumen fermentation, methane production and population of rumen protozoa in heifers fed low-quality forage

        Objective: The aim of the experiment was to assess the effect of increasing amounts of Leucaena leucocephala forage on dry matter intake (DMI), organic matter intake (OMI), enteric methane production, rumen fermentation pattern and protozoa population in cattle fed Pennisetum purpureum and housed in respiration chambers. Methods: Five crossbred heifers (Bos taurus×Bos indicus) (BW: 295±6 kg) were fed chopped P. purpureum grass and increasing levels of L. leucocephala (0%, 20%, 40%, 60%, and 80% of dry matter [DM]) in a 5×5 Latin square design. Results: The voluntary intake and methane production were measured for 23 h per day in respiration chambers; molar proportions of volatile fatty acids (VFAs) were determined at 6 h postprandial period. Molar concentration of VFAs in rumen liquor were similar (p>0.05) between treatments. However, methane production decreased linearly (p<0.005), recording a maximum reduction of up to ~61% with 80% of DM incorporation of L. leucocephala in the ration and no changes (p>0.05) in rumen protozoa population were found. Conclusion: Inclusion of 80% of L. leucocephala in the diet of heifers fed low-quality tropical forages has the capacity to reduce up to 61.3% enteric methane emission without affecting DMI, OMI, and protozoa population in rumen liquor.

        Heat Treated Twin Wire Arc Spray AISI 420 Coatings Under Dry and Wet Abrasive Wear

        The influence of applying two different heat treatments such as: deep cryogenic and tempering on dry/wetabrasive wear resistance of twin wire arc spray martensitic AISI 420 coatings was evaluated by using a modifiedrubber wheel type test apparatus. A load dependency was observed on the abrasive wear rate behavior ofboth; dry and wet tests. Three body (rolling) and two body (sliding) wear mechanisms were identified in dryconditions, prevailing rolling at lower and higher loads. However, at higher loads, more presence of groovingand pits formation was observed. Coatings tempered at 205 °C/1 h displayed better wear resistance thancryogenic treated ones. A change in wear mechanism between dry and wet conditions was observed; two bodywear mechanism predominated respect to three body. In both; dry and wet conditions the microstructure(several inter-splat oxides) as well as strain and residual stress promotes brittle material removal which wasmore evident in cryogenic and as-sprayed samples during dry test and at higher loads in wet conditions.

        Study of the methane flow influence in the micro‑tribology behavior of DLC coatings deposited by PECVD: a Raman analysis

        This paper presents a Raman spectroscopy study of the influence of methane flow on the micro-tribological behavior of diamond-like carbon coatings deposited with an industrial plasma-enhanced chemical vapor deposition system. Results have shown a direct relationship between the methane flow and thickness of the coatings. The analysis of the Raman spectra and deposition parameters allowed establishing the influence of H content with the methane flow, the disorder level and estimation of the sp3 fraction on the carbon coatings. The micro-tribology tests showed a strong dependence of the wear resistance and hardness with Raman parameters. The coating deposited at 72-sccm methane flow presented a thickness of 1.7 μm and a sp3 fraction of 0.33. This sp3 fraction gave rise to a hardness of 24 GPa and an excellent wear resistance of 3.3 × 10–6 mm3 N?1 mm?1 for this DLC coating. Wear tests showed a swelling in the wear profiles on this coating, which was associated with the occurrence of a re-hybridization process.

      • Screening and isolation of antibacterial proteinaceous compounds from flower tissues: Alternatives for treatment of healthcare-associated infections

        Healthcare-associated infection represents a frequent cause of mortality that increases hospital costs. Due to increasing microbial resistance to antibiotics, it is necessary to search for alternative therapies. Consequently, novel alternatives for the control of resistant microorganisms have been studied. Among them, plant antimicrobial protein presents enormous potential, with flowers being a new source of antimicrobial molecules. In this work, the antimicrobial activity of protein-rich fractions from flower tissues from 18 different species was evaluated against several human pathogenic bacteria. The results showed that protein-rich fractions of 12 species were able to control bacterial development. Due its broad inhibition spectrum and high antibacterial activity, the protein-rich fraction of Hibiscus rosa-sinensis was subjected to DEAE-Sepharose chromatography, yielding a retained fraction and a non-retained fraction. The retained fraction inhibits 29.5% of Klebsiella pneumoniae growth, and the non-retained fraction showed 31.5% of growth inhibition against the same bacteria. The protein profile of the chromatography fractions was analyzed by using SDS-PAGE, revealing the presence of two major protein bands in the retained fraction, of 20 and 15 kDa. The results indicate that medicinal plants have the biotechnological potential to increase knowledge about antimicrobial protein structure and action mechanisms, assisting in the rational design of antimicrobial compounds for the development of new antibiotic drugs.
