        Transcriptome-wide characterization of the WRKY family genes in Lonicera macranthoides and the role of LmWRKY16 in plant senescence

        Background: Lonicera macranthoides is an important woody plant with high medicinal values widely cultivated in southern China. WRKY, one of the largest transcription factor families, participates in plant development, senescence, and stress responses. However, a comprehensive study of the WRKY family in L. macranthoides hasn't been reported previously. Objective: To establish an extensive overview of the WRKY family in L. macranthoides and identify senescence-responsive members of LmWRKYs. Methods: RNA-Seq and phylogenetic analysis were employed to identify the LmWRKYs and their evolutionary relationships. Quantitative real-time (qRT-PCR) and transgenic technology was utilized to investigate the roles of LmWRKYs in response to developmental-, cold-, and ethylene-induced senescence. Results: A total of 61 LmWRKY genes with a highly conserved motif WRKYGQK were identified. Phylogenetic analysis of LmWRKYs together with their orthologs from Arabidopsis classified them into three groups, with the number of 15, 39, and 7, respectively. 17 LmWRKYs were identified to be differentially expressed between young and aging leaves by RNA-Seq. Further qRT-PCR analysis showed 15 and 5 LmWRKY genes were significantly induced responding to tissue senescence in leaves and stems, respectively. What's more, five LmWRKYs, including LmWRKY4, LmWRKY5, LmWRKY6, LmWRKY11, and LmWRKY16 were dramatically upregulated under cold and ethylene treatment in both leaves and stems, indicating their involvements commonly in developmental- and stress-induced senescence. In addition, function analysis revealed LmWRKY16, a homolog of AtWRKY75, can accelerate plant senescence, as evidenced by leaf yellowing during reproductive growth in LmWRKY16-overexpressing tobaccos. Conclusion: The results lay the foundation for molecular characterization of LmWRKYs in plant senescence.

        Physalin D inhibits RANKL-induced osteoclastogenesis and bone loss via regulating calcium signaling

        We investigated the effects of physalin A, B, D, and F on osteoclastogenesis induced by receptor activator of nuclear factor кB ligand (RANKL). The biological functions of different physalins were first predicted using an in silico bioinformatic tool (BATMAN-TCM). Afterwards, we tested cell viability and cell apoptosis rate to analyze the cytotoxicity of different physalins. We analyzed the inhibitory effects of physalins on RANKL-induced osteoclastogenesis from mouse bone-marrow macrophages (BMMs) using a tartrate-resistant acid phosphatase (TRAP) stain. We found that physalin D has the best selectivity index (SI) among all analyzed physalins. We then confirmed the inhibitory effects of physalin D on osteoclast maturation and function by immunostaining of F-actin and a pit-formation assay. On the molecular level, physalin D attenuated RANKLevoked intracellular calcium ([Ca(2+)](i)) oscillation by inhibiting phosphorylation of phospholipase Cγ2 (PLCγ2) and thus blocked the downstream activation of Ca2+/calmodulindependent protein kinases (CaMK)IV and cAMP-responsive element-binding protein (CREB). An animal study showed that physalin D treatment rescues bone microarchitecture, prevents bone loss, and restores bone strength in a model of rapid bone loss induced by soluble RANKL. Taken together, these results suggest that physalin D inhibits RANKL-induced osteoclastogenesis and bone loss via suppressing the PLCγ2-CaMK-CREB pathway. [BMB Reports 2020; 53(3): 154-159]

        There are 10 sets of CNC equipment in a numerical controlled workshop, and set up the CAD/CAPP/CAM center, there is a problem that each production unit is an information island, which can not implement informatization management. This paper takes ZNE-100TL module realized the embedded Ethernet and RS-232C data transfer function as the hardware platform, takes VC++ as software platform, applied Ethernet technology to realize communication the CNC equipment with CAD/CAPP/CAM center. This is good for production management and data transmission in numerical controlled workshop, which can improve the production efficiency.

        In this paper, a practical SPICE model for an IGBT and a PiN diode is proposed based on the Finite Differential Method (FDM). Other than the conventional Fourier model and the Hefner model, the excess carrier distribution can be accurately solved by a fast FDM in the SPICE simulation tool. In order to improve the accuracy of the SPICE model, the Taguchi method is adopted to calibrate the extracted parameters. This paper presents a numerical modelling approach of an IGBT and a PIN diode, which are also verified by SPICE simulations and experiments.

        Online monitoring of IGBT junction temperature based on V<sub>ce</sub> measurement

        In this paper, an online junction temperature monitoring method based on the on-state voltage under high collector current density measurements for IGBT power modules is proposed. Unlike the conventional junction temperature monitoring method, the presented method can extract the junction temperature during operation without altering the modulation strategy or topology of the converter. The proposed method is verified by simulations and the JEDEC-51 standard recommended approach. To accurately extract the on-state voltage during operation, a measurement circuit that combines the advantages of both the active MOSFET clamp and the diode clamp is designed and tested. It has been shown to have good accuracy and a rapid response time. After the calibration results are obtained, the presented method is applied to a three-phase voltage source converter controlled by a closed-loop SVPWM modulation strategy.

        A Controllability Synthesis Problem for Dynamic Multi-Agent Systems with Linear High-Order Protocol

        In this paper, a problem on controllability synthesis for dynamic multi-agent systems with high-order linear protocol is addressed. A particular scenario is considered, in which a given system should be adjusted to be controllable by the input information upon the created leaders, with optional communication links from the leaders to each follower vertex in the graph. It is shown that a single leader is always sufficient to satisfy the requirement for controllability of any given graph topology, so long as there are proper links between the leader and the followers.

        Effects of biochar and Trichoderma on bacterial community diversity in continuous cropping soil

        Soil acidifi cation has become a serious issue in protected production in northern China as a consequence of many years ofcontinuous cropping. Besides aff ecting the growth and quality of vegetables, soil acidifi cation also restricts the sustainabledevelopment of vegetables grown in protected environments. A single application of biochar or Trichoderma can repair thesoil environment, optimize the microbial community structure, reduce plant diseases, and promote plant growth. However,it is not known whether biochar combined with Trichoderma has a synergistic eff ect on these parameters. In this study,long-term continuous cropping of cucumbers in a greenhouse was used to study the eff ect of biochar and Trichoderma onmicro-ecological environmental changes in acidifi ed soil using Illumina sequencing. The results showed that the combinedapplication of biochar and Trichoderma promoted cucumber growth; signifi cantly enhanced root vitality and superoxidedismutase, peroxidase, and catalase activities in leaves; and reduced malondialdehyde content. In addition, soil physicaland chemical properties improved; cation exchange capacity, the activity of soil polyphenol oxidase and invertase, and thediversity of soil bacterial communities increased; and the structure and relative abundance of soil bacteria changed after thecombined application of biochar and Trichoderma . Correlation analysis of environmental factors showed that total nitrogenand soil pH had the greatest impact on bacterial communities. The metabolic function of soil bacteria was improved, andthe circulation of soil nutrients was increased as a consequence of the combined application of biochar and Trichoderma .

      • A Genetic Algorithm Based Motor Controller System Automatic Layout Method

        In this paper, a genetic algorithm based automatic motor controller system layout design and optimization method is proposed for EV application. The modeling procedure including 3-D position and physical characteristic interpretation is described. The simplified purpose of optimization is to find a trade-off between volume and electrical performance of motor controller. Compared with the Pareto Front generated by enumeration method, the presented method can find the optimum solution with a higher speed. The proposed method can be easily extended to a complex system and can provide new approaches for motor controller system design.

        Stability-based On-demand Multi-path Distance Vector Protocol for Edge Internet of Things

        In edge computing scenarios, IoT end devices play a crucial role in relaying and forwarding data to significantly improve IoT network performance. However, traditional routing mechanisms are not applicable to this scenario due to differences in network size and environment. Therefore, it becomes crucial to establish an effective and reliable data transmission path to ensure secure communication between devices. In this paper, we propose a trusted path selection strategy that comprehensively considers multiple attributes, such as link stability and edge cooperation, and selects a stable and secure data transmission path based on the link life cycle, energy level, trust level, and authentication status. In addition, we propose the Stability-based On-demand Multipath Distance Vector (STAOMDV) protocol based on the Ad hoc AOMDV protocol. The STAOMDV protocol implements the collection and updating of link stability attributes during the route discovery and maintenance process. By integrating the STAOMDV protocol with the proposed path selection strategy, a dependable and efficient routing mechanism is established for IoT networks in edge computing scenarios. Simulation results validate that the proposed STAOMDV model achieves a balance in network energy consumption and extends the overall network lifespan.
