        Prevalence of BRCA1 and BRCA2 Germline Mutations in Breast Cancer Women of Multiple Ethnic Region in Northwest China

        Purpose: The aim of this study is to further understand the status of BRCA1 and BRCA2 mutation among Chinese high-risk breast cancer patients in multiple-ethnic regions of China. Methods: A total of 79 blood samples of high-risk breast cancer patients from Xinjiang Uyghur autonomous region were analyzed by PCR-DHPLC sequencing analysis. Results: Analysis with full length of the two genes identified a total of 6 deleterious mutations (2073delA, 2394C-T [Q759X] and IVS16+1G>A in BRCA1; 1627A-T [K467X], 6873delCTCC and 9481delA in BRCA2) in this cohort. The prevalence of BRCA1/2 germline mutation was about 7.6% (6/79) in the Xinjiang multiple ethnic region of China. Among them, 3 novel deleterious mutations, 2073delA in BRCA1 (Han ethnic Chinese) and BRCA2 variants 6873delCTCC and 9481delA (both are Kazakh ethnic Chinese), were identified and they had never been reported in breast cancer information core (BIC) database before. 2394C-T (Q759X) and IVS16+1G>A, in BRCA1 and BRCA2 variants 1627A-T were previously reported in other populations but not Chinese. Among 6 of the BRCA-related tumors, three BRCA1- and one BRCA2-associated tumors were in triple negative (estrogen receptor, progesterone receptor, and HER2 negative expressed) status and exhibited a high tumor grade. So far none of these 6 deleterious mutations were reported in ethnic Han Chinese. Conclusion: BRCA germline mutation in Chinese multiple ethnicity region may exhibit different genotypes compared to ethnic Han Chinese in other regions. These differences may arise from interaction of genetic background and environmental factors.

        A Two-Point Modulation Spread-Spectrum Clock Generator With FIR-Embedded Binary Phase Detection and 1-Bit High-Order ΔΣ Modulation

        This paper describes a spread-spectrum clock generation method by utilizing a ${\Delta}{\Sigma}$ digital PLL (DPLL) which is solely based on binary phase detection and does not require a linear time-to-digital converter (TDC) or other linear digital-to-time converter (DTC) circuitry. A 1-bit high-order ${\Delta}{\Sigma}$ modulator and a hybrid finite-impulse response (FIR) filter are employed to mitigate the phase-folding problem caused by the nonlinearity of the bang-bang phase detector (BBPD). The ${\Delta}{\Sigma}$ DPLL employs a two-point modulation technique to further enhance linearity at the turning point of a triangular modulation profile. We also show that the two-point modulation is useful for the BBPLL to improve the spread-spectrum performance by suppressing the frequency deviation at the input of the BBPD, thus reducing the peak phase deviation. Based on the proposed architecture, a 3.2 GHz spread-spectrum clock generator (SSCG) is implemented in 65 nm CMOS. Experimental results show that the proposed SSCG achieves peak power reductions of 18.5 dB and 11 dB with 10 kHz and 100 kHz resolution bandwidths respectively, consuming 6.34 mW from a 1 V supply.

      • Prognostic Value of ALDH1A3 Promoter Methylation in Gliob;astoma: A Single Center Experience in Western China

        Background: Aberrations in gene methylation patterns play important roles in gliomagenesis. However, whether the ALDH1A3 promoter methylation is related to prognoses of primary glioblastomas (GBMs) in Western China remains unclear. Materials and Methods: Methylation levels of ALDH1A3 CpG island in 36 GBMs were identified by pyrophosphate sequencing, while ALDH1A3 expression was assessed with matched paraffin section immunohistochemistry. Survival curves were analysed by Kaplan-Meier. Results: The hypermethylation status of ALDH1A3 promoter predicted a better prognosis accompanied with low expression of ALDH1A3 protein. Conclusions: Our results indicate ALDH1A3 promoter methylation correlates with prognosis in primary GBMs.

      • Improved Model Predictive Control for H-bridge Cascaded STATCOM

        H-bridge cascaded static synchronous compensator (STATCOM) is often used to compensate the harmonic and reactive current derived from non-linear load. In order to improve the performance of STATCOM, an improved model predictive control (MPC) method is put forward in this paper. In the proposed MPC, the difference equations which are derived from the transfer functions of controlling variables and controlled variables in STATCOM are used as the predictive control model, the error between the output of STATCOM and predictive model is used to design feedback correction controller and rolling optimization controller. Meanwhile, the introduced repetitive controller is used to eliminate the inherent steady error. The actual H-bridge cascaded STATCOM is constructed and a series of verification tests are executed. The experimental results prove that the output voltage and current of H-bridge cascaded STATCOM with the proposed MPC have smaller distortion and better sinusoidal shape than those of the traditional Proportion Integral (PI) control. Moreover, the performance of H-bridge cascaded STATCOM is great in high power experiment.

        Protein Kinase CK2 Modulates the Calcium Sensitivity of Type 3 Small-conductance Calcium-activated Potassium Channels in Colonic Platelet-derived Growth Factor Receptor Alpha-positive Cells From Streptozotocin-induced Diabetic Mice

        Background/AimsThe gastrointestinal symptom of diabetes mellitus, chronic constipation, seriously affects patients’ life. Whereas, the mechanism of chronic constipation is still ambiguous, resulting in a lack of effective therapies for this symptom. As a part of the smooth muscle cells, interstitial cells of Cajal, and platelet-derived growth factor receptor alpha-positive (PDGFRα+) cells syncytium (SIP syncytium), PDGFRα+ cells play an important role in regulating colonic motility. According to our previous study, in PDGFRα+ cells in colons of diabetic mice, the function of the P2Y1 purinergic receptor/type 3 small-conductance calcium-activated potassium (SK3) channel signaling pathway is strengthened, which may lead to colonic dysmotility. The purpose of this study is to investigate the changes in SK3 channel properties of PDGFRα+ cells in diabetic mice. MethodsWhole-cell patch clamp, Western blotting, superoxide dismutase activity measurement, and malondialdehyde measurement were main methods in the present study. ResultsThe present study revealed that when dialysed with low calcium ion (Ca2+) solution, the SK3 current density was significantly decreased in PDGFRα+ cells from diabetic mice. However, the SK3 current density in PDGFRα+ cells was enhanced from diabetic mice when dialysed with high Ca2+ solution. Moreover, hydrogen peroxide-treatment mimicked this phenomenon in SK3 transgenic HEK293 cells. The subunit of SK3 channels, protein kinase CK2, was up-regulated in colonic muscle layers and hydrogen peroxide-treated HEK293 cells. Additionally, protein phosphatase 2A, the subunit of SK3 channels, was not changed in streptozotocin-treated mouse colons or hydrogen peroxide-treated HEK293 cells. ConclusionThe diabetic oxidative stress-induced upregulation of CK2 contributed to modulating SK3 channel sensitivity to Ca2+ in colonic PDGFRα+ cells, which may result in colonic dysmotility in diabetic mice.

      • A RIF Based Mapping of RDB2RDF

        Mapping RDB to RDF (i.e., RDB2RDF) is the key to constructing the Semantic Web, hence has been an active research field during the last decade. Many technically heterogeneous RDB2RDF tools resulted in non-interchangeable and unreusable RDB2RDF mapping descriptions. In 2009, the W3C RDB2RDF Incubator Group Report once strongly suggested that the RDB2RDF mapping language be expressed in rules as defined by the W3C Rule Interchange Format (RIF) Working Group, because rules are an effective way to express mappings between information models, and RIF, as part of the infrastructure for the Semantic Web, is now a standard for exchanging rules among Web rule systems. This paper addresses the issue of RIF-based RDB2RDF mapping and proposes a database semantics-driven, RIF Production Rule Dialect (RIF-PRD) based mapping description approach. The work includes defining a set of generic RIF-PRD mapping rules for RDB2RDF, developing a prototype mapping engine called RIFD2RME (stands for RIF-based RDB2RDF Mapping Engine), and conducting case study experiments with the prototype. The experimental results indicate that the proposed mapping approach is achievable and effective.

      • siRNA Interference with a Proliferation-Inducing Ligand Gene in the Sgr-7901 Gastric Carcinoma Cell Line

        Objective: The present study aimed to investigate the influence of siRNA interference with a proliferation-inducing ligand (APRIL) gene on gastric carcinoma sgr-7901 cell apoptosis. Correlations between APRIL silencing and tyrosine kinase (trka) expression were also explored. Methods: Two APRIL-silencing siRNA vectors were constructed, and transfected into human gastric carcinoma sgr-7901 cells, expression before and after transfection being detected using RT-PCR and western blot analyses. The expression of 15 trka genes was detected using RT-PCR and apoptotic rates of sgr-7901 were assessed by flow cytometry. Results: The expression levels of receptor trka genes were significantly decreased, and the apoptotic rate of sgr-7901 was significantly increased after transfection (P < 0.05). Conclusion: APRIL gene silencing can increase the apoptotic rate of gastric carcinoma cells, and inhibit the expression of receptor trka genes. There is a correlation between the signaling pathways of APRIL and trka.

        Adaptive Cooperative Spectrum Sensing Based on SNR Estimation in Cognitive Radio Networks

        Single-user spectrum sensing is susceptible to multipath effects, shadow effects, hidden terminals and other unfavorable factors, leading to misjudgment of perceived results. In order to increase the detection accuracy and reduce spectrum sensing cost, we propose an adaptive cooperative sensing strategy based on an estimated signal-to-noise ratio (SNR). Which can adaptive select different sensing strategy during the local sensing phase. When the estimated SNR is higher than the selection threshold, adaptive double threshold energy detector (ED) is implemented, otherwise cyclostationary feature detector is performed. Due to the fact that only a better sensing strategy is implemented in a period, the detection accuracy is improved under the condition of low SNR with low complexity. The local sensing node transmits the perceived results through the control channel to the fusion center (FC), and uses voting rule to make the hard decision. Thus the transmission bandwidth is effectively saved. Simulation results show that the proposed scheme can effectively improve the system detection probability, shorten the average sensing time, and has better robustness without largely increasing the costs of sensing system.

        Efficient green emission by up-conversion inYb3+/Er3+ co-doped monoclinic AgGdW2O8

        Yb3+/Er3+ co-doped monoclinic AgGdW2O8 phosphors with intense green emission via up-conversion process were prepared by the high-temperature solid-state method. The samples were structurally characterized using X-ray powder diffraction, after which their luminescent properties were characterized via up-conversion spectroscopy. Green and red emissions centered at 525, 550, and 656 nm were observed when the phosphors were excited at 980 nm, these emissions are ascribed to 2 H11/2, 4 S3/2 → 4 I15/2 (green emission) and 4 F9/2 → 4 I15/2 (red emission) transitions of Er3+, respectively. It was also found that the green and red emissions originated from the two-photon process. By increasing sintering temperature and doping of Mo6+, the structure of monoclinic AgGdW2O8 transformed to a tetragonal one, which resulted a dramatic increase in UC intensity.
