Purification and Characterization of an Extracellular Protease from Bacillus pumilus CN8

The protease produced by a Bacillus pumilus CN8 strain was purified by DEAE-Cellulose-52 ion exchange. It has a molecular weight of approximately 96,920 Dalton. In the present study, this protease showed strong activity over a broad range of pH (6.5-9.5) and temperature from 40℃ to 60℃, and the protease performed the maximal activity at pH 7.3 at 42℃. The effect of metal ions on protease activity showed that K+ could slightly increase the protease activity, and other ions such as Zn²+, Fe²+, Na+, Ca²+, Mg²+ had no significant activation or inhibition to the protease (P > 0.05), and the more important is that Cu²+, Mn²+, Sn²+, Cd²+ had a strong inhibitory effect on the protease activity.

Immunoglobulin A nephropathy (IgAN) has been recognized as the most prevalent form ofglomerulonephritis with various histologic and clinical phenotypes in the world. The Gal-de¯cientIgA1 with terminally exposed GalNAc residue(s) plays a key role in the pathogenesis of thedisease. In this study, a novel colorimetric biosensor based on Helix aspersa agglutinin (HAA)absorbed on polydiacetylene (PCDA) nanovesicles for N-acetylgalactosamine (GalNAC) mo-lecular as core framework of Gal-de¯cient IgA1 recognition was investigated using the higha±nity between lectin HAA and GalNAC. The PCDA nanovesicles were prepared via UVcrosslinking. The size, morphology and elastic property of the nanovesicles were tested. Theoptimal concentration of HAA for the recognition of GalNAC via naked eyes observation was3 mM and the critical concentration of GalNAC for the sensitive color transition was 2 ? g/mL. This novel method has many advantages such as low prices, intuitive, real-time and easy to carryout and great potential in Gal-de¯cient IgA1 detection.

The thermal stability and elevated temperature mechanical properties of $SiC_P$/Al-11.7Fe-1.3V-1.7Si (Al-11.7Fe-1.3V-1.7Si reinforced with SiC particulates) composites sheets prepared by spray deposition (SD) $\rightarrow$ hot pressing $\rightarrow$ rolling process were investigated. The experimental results showed that the composite possessed high ${\sigma}_b$ (elevated temperature tensile strength), for instance, ${\sigma}_b$ was 315.8 MPa, which was tested at $315^{\circ}C$, meanwhile the figure was 232.6 MPa tested at $400^{\circ}C$, and the elongations were 2.5% and 1.4%, respectively. Furthermore, the composite sheets exhibited excellent thermal stability: the hardness showed no significant decline after annealing at $550^{\circ}C$ for 200 h or at $600^{\circ}C$ for 10 h. The good elevated temperature mechanical properties and excellent thermal stability should mainly be attributed to the formation of spherical ${\alpha}-Al_{12}(Fe,\;V)_3Si$ dispersed phase particulates in the aluminum matrix. Furthermore, the addition of SiC particles into the alloy is another important factor, which the following properties are responsible for. The resultant Si of the reaction between Al matrix and SiC particles diffused into Al matrix can stabilize ${\alpha}-Al_{12}(Fe,\;V)_3Si$ dispersed phase; in addition, the interface (Si layer) improved the wettability of Al/$SiC_P$, hence, elevated the bonding between them. Furthermore, the fine $Al_4C_3$ phase also strengthened the matrix as a dispersion-strengthened phase. Meanwhile, load is transferred from Al matrix to SiC particles, which increased the cooling rate of the melt droplets and improved the solution strengthening and dispersion strengthening.

Background: Dried and red ginseng are well-known types of processed ginseng and are widely used as healthy food. The dried and red ginseng quality may vary with the storage period of raw ginseng. Therefore, herein, the effect of the storage period of fresh ginseng on processed ginseng quality was evaluated through multicomponent quantification with statistical analysis. Methods: A method based on ultrahigh performance liquid chromatography coupled to triple quadrupole mass spectrometry in multiple-reaction monitoring mode (UPLC-MRM-MS) was developed for quantitation of ginsenosides and oligosaccharides in dried and red ginseng. Principal component analysis and partial least squares discriminant analysis were conducted to evaluate the dynamic distributions of ginsenosides and oligosaccharides after different storage periods. Results: Eighteen PPD, PPT and OLE ginsenosides and nine reducing and nonreducing oligosaccharides were identified and quantified. With storage period extension, the ginsenoside content in the processed ginseng increased slightly in the first 2 weeks and decreased gradually in the following 9 weeks. The content of reducing oligosaccharides decreased continuously as storage time extending, while that of the nonreducing oligosaccharides increased. Chemical conversions occurred during storage, based on which potential chemical markers for the storage period evaluation of fresh ginseng were screened. Conclusion: According to ginsenoside and oligosaccharide distributions, it was found that the optimal storage period was 2 weeks and that the storage period of fresh ginseng should not exceed 4 weeks at 0 °C. This study provides deep insights into the quality control of processed ginseng and comprehensive factors for storage of raw ginseng.
Parallel task scheduling under multi-Clouds

In the Cloud, for the scheduling of parallel jobs, there are many tasks in a job and those tasks are executed concurrently on different VMs (Visual machines), where each task of the job will be executed synchronously. The goal of scheduling is to reduce the execution time and to keep the fairness between jobs to prevent some jobs from waiting more time than others. We propose a Cloud model which has multiple Clouds, and under this model, jobs are in different lists according to the waiting time of the jobs and every job has different parallelism. At the same time, a new method-ZOMT (the scheduling parallel tasks based on ZERO-ONE scheduling with multiple targets) is proposed to solve the problem of scheduling parallel jobs in the Cloud. Simulations of ZOMT, AFCFS (Adapted First Come First Served), LJFS (Largest Job First Served) and Fair are executed to test the performance of those methods. Metrics about the waiting time, and response time are used to test the performance of ZOMT. The simulation results have shown that ZOMT not only reduces waiting time and response time, but also provides fairness to jobs.

Background To explore the relationship between neonatal cord blood resistin, adiponectin, CD4 and CD8 and their influence on early neonatal outcomes. Objective A total of 150 newborns delivered at term were selected as subjects to detect the levels of cord blood resistin, adiponectin, CD4 and CD8. The cases of neonatal asphyxia, respiratory distress, macrosomia and amniotic fluid inhalation syndrome were recorded. Results There were 6 cases of high CD4 and 7 cases of low CD8 among the 150 newborns. There were 10 cases of macrosomia and 5 cases of respiratory distress. According to Pearson correlation analysis, there was no statistically significant difference in cord blood resistin between CD4 and CD8 in newborns delivered at term ( r = 0.132, r = 0.098, P > 0.05). There was no significant difference between adiponectin, CD4 and CD8 ( r = 0.104, r = 0.107, P > 0.05). There were 10 cases of macrosomia and 5 cases of respiratory distress in newborns, and there were no newborns with amniotic fluid inhalation syndrome and asphyxia. According to Pearson correlation analysis, cord blood resistin was negatively correlated with the body weight of large newborns and the level of neonatal respiratory distress ( r = − 0.542, r = − 0.467, P < 0.05). Adiponectin levels were negatively correlated with the body weight of large newborns and neonatal respiratory distress ( r = − 0.564, r = − 0.609, P < 0.05).The results of correlation analysis showed that the result of cord blood resistin was positively correlated with adiponectin ( r = 0.612, P < 0.05). Conclusion The level of cord blood resistin and adiponectin can measure fetal growth and development, and are closely related to neonatal early outcomes.
