      • Impact of low dose atorvastatin on development of new-onset diabetes mellitus in Asian population: Three-year clinical outcomes

        Park, J.Y.,Rha, S.W.,Choi, B.,Choi, J.W.,Ryu, S.K.,Kim, S.,Noh, Y.K.,Choi, S.Y.,Akkala, R.G.,Li, H.,Ali, J.,Xu, S.,Ngow, H.A.,Lee, J.J.,Lee, G.N.,Kim, J.,Lee, S.,Na, J.O.,Choi, C.U.,Lim, H.E.,Kim, J.W Elsevier/North-Holland Biomedical Press 2015 INTERNATIONAL JOURNAL OF CARDIOLOGY Vol.184 No.-

Background: High dose atorvastatin is known to be associated with new onset diabetes mellitus (NODM) in patients with high risk for developing diabetes mellitus (DM). However, low dose atorvastatin is more commonly used as compared with high dose atorvastatin. The aim of this study is to investigate the impact of low dose atorvastatin (LDA, 10mg or 20mg) on the development of NODM up to three years in Asian patients. Methods: From January 2004 to September 2009, we investigated a total of 3566 patients who did not have DM. To adjust for potential confounders, a propensity score matching (PSM) analysis was performed using the logistic regression model. After PSM (C-statistics: 0.851), a total of 818 patients (LDA group, n=409 patients and control group, n=409 patients) were enrolled for analysis. Results: Before PSM, the cumulative incidence of NODM (5.8% vs. 2.1%, p<0.001), myocardial infarction (0.5% vs. 0.1%, p-value=0.007), and major adverse cardio-cerebral event (MACCE, 1.8% vs. 0.7%, p-value=0.012) at three-years were higher in the LAD group. However, after PSM, there was a trend toward higher incidence of NODM (5.9% vs. 3.2%, p=0.064) in the LDA group, but the incidence of MACCE (1.2% vs. 1.5%, p-value=1.000) was similar between the two groups. In multivariable analysis, the LDA administration was tended to be an independent predictor of NODM (OR: 1.99, 95% CI: 1.00-3.98, p-value 0.050). Conclusions: In this study, the use of LDA tended to be a risk factor for NODM in Asian patients and reduced clinical events similar to the control group. However, large-scale randomized controlled trials will be needed to get the final conclusion.

Dipeptidyl petidase-IV inhibitor (gemigliptin) inhibits tunicamycin-induced endoplasmic reticulum stress, apoptosis and inflammation in H9c2 cardiomyocytes

        Hwang, H.J.,Jung, T.W.,Ryu, J.Y.,Hong, H.C.,Choi, H.Y.,Seo, J.A.,Kim, S.G.,Kim, N.H.,Choi, K.M.,Choi, D.S.,Baik, S.H.,Yoo, H.J. North-Holland 2014 Molecular and cellular endocrinology Vol.392 No.1

The direct effects of dipeptidyl peptidase-IV (DPP-IV) inhibitors on endoplasmic reticulum (ER) stress-induced apoptosis and inflammation in cardiomyocytes have not been elucidated. H9c2 cell viability, which was reduced by tunicamycin, was increased after DPP-IV inhibitor gemigliptin treatment. Gemigliptin significantly decreased the tunicamycin-mediated increase in glucose regulated protein 78 (GRP78) expression and ER stress-mediated signaling molecules such as protein kinase RNA-like endoplasmic reticulum kinase (PERK)/C-EBP homologous protein (CHOP) and inositol-requiring enzyme 1α (IRE1α)/c-Jun N-terminal kinase (JNK)-p38. Furthermore, gemigliptin effectively induced Akt phosphorylation in a dose-dependent manner. Using flow cytometry and Hoechst staining, we showed that treatment with Akt inhibitor significantly blocked the anti-apoptotic effects mediated by gemigliptin. The reduction in tunicamycin-induced GRP78 level and PERK/CHOP pathway activity by gemigliptin was reversed after treatment with Akt inhibitor. In conclusion, gemigliptin effectively inhibited ER stress-induced apoptosis and inflammation in cardiomyocytes via Akt/PERK/CHOP and IRE1α/JNK-p38 pathways, suggesting its direct protective role in cardiovascular diseases.

        Graphene Veils and Sandwiches

        Yuk, Jong Min,Kim, Kwanpyo,Alemá,n, Benjamí,n,Regan, William,Ryu, Ji Hoon,Park, Jungwon,Ercius, Peter,Lee, Hyuck Mo,Alivisatos, A. Paul,Crommie, Michael F.,Lee, Jeong Yong,Zettl, Alex American Chemical Society 2011 NANO LETTERS Vol.11 No.8

<P>We report a new and highly versatile approach to artificial layered materials synthesis which borrows concepts of molecular beam epitaxy, self-assembly, and graphite intercalation compounds. It readily yields stacks of graphene (or other two-dimensional sheets) separated by virtually any kind of “guest” species. The new material can be “sandwich like”, for which the guest species are relatively closely spaced and form a near-continuous inner layer of the sandwich, or “veil like”, where the guest species are widely separated, with each guest individually draped within a close-fitting, protective yet atomically thin graphene net or veil. The veils and sandwiches can be intermixed and used as a two-dimensional platform to control the movements and chemical interactions of guest species.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/nalefd/2011/nalefd.2011.11.issue-8/nl201647p/production/images/medium/nl-2011-01647p_0006.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/nl201647p'>ACS Electronic Supporting Info</A></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/nl201647p'>ACS Electronic Supporting Info</A></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/nl201647p'>ACS Electronic Supporting Info</A></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/nl201647p'>ACS Electronic Supporting Info</A></P>

Multiplex bioimaging of piRNA molecular pathway-regulated theragnostic effects in a single breast cancer cell using a piRNA molecular beacon

        Lee, Y.J.,Moon, S.U.,Park, M.G.,Jung, W.Y.,Park, Y.K.,Song, S.K.,Ryu, J.G.,Lee, Y.S.,Heo, H.J.,Gu, H.N.,Cho, S.J.,Ali, B.A.,Al-Khedhairy, A.A.,Lee, I.,Kim, S. IPC Science and Technology Press 2016 Biomaterials Vol.101 No.-

<P>Recently, PIWI-interacting small non-coding RNAs (piRNAs) have emerged as novel cancer biomarkers candidate because of their high expression level in various cancer types and role in the control of tumor suppressor genes. In this study, a novel breast cancer theragnostics probe based on a single system targeting the piRNA-36026 (piR-36026) molecular pathway was developed using a piR-36026 molecular beacon (MB). The piR-36026 MB successfully visualized endogenous piR-36026 biogenesis, which is highly expressed in MCF7 cells (a human breast cancer cell line), and simultaneously inhibited piR-36026-mediated cancer progression in vitro and in vivo. We discovered two tumor suppressor proteins, SERPINA1 and LRAT, that were directly regulated as endogenous piR-36026 target genes in MCF7 cells. Furthermore, multiplex bioimaging of a single MCF7 cell following treatment with piR-36026 MB clearly visualized the direct molecular interaction of piRNA-36026 with SERPINAI or LRAT and subsequent molecular therapeutic responses including caspase-3 and PI in the nucleus. Variation of lithium isotope geochemistry during basalt weathering and secondary mineral transformations in Hawaii

        Ryu, J.S.,Vigier, N.,Lee, S.W.,Lee, K.S.,Chadwick, O.A. Pergamon Press ; Elsevier Science Ltd 2014 Geochimica et cosmochimica acta Vol.145 No.-

Lithium isotopes are a potential tracer of silicate weathering but the relationship between lithium isotope compositions and weathering state still need to be established with precision. Here, we report Li concentrations and Li isotope compositions of soils developed along a 4 million year humid-environment chronosequence in the Hawaiian Islands. Li concentrations are variable with depth and age, ranging from 0.24 to 21.3ppm, and significant Li depletions (up to 92%) relative to parent basalts are systematically enhanced towards the surface. Our calculations show that the relative contribution from atmospheric deposits to the Li soil budget remains small, with a maximum contribution from dust Li of 20% at the oldest site. This is explained by the capacity of the weathering products to retain, within the profiles, the Li coming from basalt alteration, and allows us to explore more specifically the role of alteration processes on soil Li isotope signatures. The δ7Li values display a large range between -2.5%% and +13.9%%. The youngest soils (0.3ka) display the same δ7Li value as fresh basalt, regardless of depth, despite ~30% Li loss by leaching, indicating that there is little Li isotope fractionation during the incipient stage of weathering. δ7Li values for the older soils (≥20ka) vary non-linearly as a function of time and can be explained by progressive mineral transformations starting with the synthesis of metastable short-range order (nano-crystalline) minerals and followed by their transformation into relatively inert secondary minerals. Results highlight significant Li isotope fractionation during secondary mineral formation and in particular during Li uptake by kaolinite. Finally, we suggest that the non-monotonous evolution of the regolith δ7Li value over the last 4Ma is consistent with climatic variations, where congruent release of Li isotopes occurs during warmer periods.

      • Small-Molecule inhibition of Siderophore biosynthesis in Mycobacterium tuberculosis and Yersinia pestis

        Julian A Ferreras,Ryu, Jae-Sang,Federico Di Lello,Derek S Tan,Luis E N Quadri 이화여자대학교 약학연구소 2005 藥學硏究論文集 Vol.- No.16

Mycobacterium tuverculosis and Yersinia pestis, the causative agents of tuberculosis and plague respectively are pathogens with serious ongoing impact on global public health^(1,2) and potential use as agents of bioterrorism^(3). Both pathogens have iron acquisition systems based on siderphores secreted iron-chelating compounds with extremey high Fe^(3+)affinity(4,5) Several lines of evidence suggest that siderophores have a critical role in bacterial iron acquisition inside the human host^(6-9) where the free iron concentration is well bellow that required for bacterial growth and virulence ^(10) Thus, siderophore biosynthesis is an attractive target in the development of new antibiotics to treat tuberculosis and plague^(2,5,8,11). In particular, such drugs alone or as part of combination therapies could provide a valuable new line of defense against intractable muliple-drug-resistant infections Here we report the design synthesis and domain salicylation enzymes required for siderophore biosynthesis siderophore biosynthesis and growth of M. tuberculosis and Y, pestis under iron-limiting conditions.

      • Realization of high performance flexible wire supercapacitors based on 3-dimensional NiCo2O4/Ni fibers

        Ramadoss, A.,Kang, K. N.,Ahn, H. J.,Kim, S. I.,Ryu, S. T.,Jang, J. H. Royal Society of Chemistry 2016 Journal of Materials Chemistry A Vol.4 No.13

<P>The rapidly developing electronics industry is producing miniaturized electronic devices with flexible, portable and wearable characteristics, requiring high-performance miniature energy storage devices with flexible and light weight properties. Herein, we have successfully fabricated highly porous, binder free three-dimensional flower-like NiCo2O4/Ni nanostructures on Ni-wire as a fiber electrode for high-performance flexible fiber supercapacitors. Such a unique structure exhibited remarkable electrochemical performance with high capacitance (29.7 F cm(-3) at 2.5 mA), excellent rate capability (97.5% retention at 20 mA), and super cycling stability (80% retention, even after 5000 cycles). The remarkable electrochemical performance is attributed to the large active area in the 3D porous architecture and direct contact between the active materials and 3D-Ni current collectors, which facilitate easy ionic/electronic transport. The symmetric fiber super-capacitor showed a gravimetric energy density of 2.18 W h kg(-1) (0.21 mW h cm(-3)) and a power density of 21.6 W kg(-1) (2.1 mW cm(-3)) with good flexibility and cycling performance, signifying potential applications in high-performance flexible energy storage devices. Further, performance in a self-powered system was demonstrated by charging these wire type NiCo2O4/Ni supercapacitors by serially wound DSSCs to drive commercial LEDs. These results suggest that the fabricated device has excellent potential as a power source for flexible, portable and wearable applications as well as self-powered systems.</P>

      • Generation of geodesic acoustic mode by nonlinear coupling of magnetic island and island-driven beta-induced Alfvén eigenmode

        Marchenko, V S,Panwar, A,Reznik, S N,Ryu, C M IOP 2017 Plasma physics and controlled fusion Vol.59 No.9

<P>In a recent work, we have shown that the plasma flow around the magnetic island can excite the beta-induced Alfvén eigenmode (BAE) (Marchenko <I>et al</I> 2016 <I>Nucl. Fusion</I> <B>56</B> 106021). In the present communication, it is shown that coupling of this primary BAE and magnetic island generates secondary geodesic acoustic mode (GAM), which has the frequency and mode structure identical to those of the primary BAE. The fixed GAM/BAE amplitude ratio, determined by the plasma neutrality, is comparable with the plasma/magnetic pressure ratio. This nonlinear coupling can be responsible for axis-symmetric modes, which accompany island-driven Alfvénic modes observed on HL-2A tokamak (Chen <I>et al</I> 2013 <I>Nucl. Fusion</I> <B>53</B> 113010).</P>
