Recent studies showed that the PD-1/PD-L1 checkpoint blockade is a dramatic therapy for melanoma by enhancing antitumor immune activity. Currently, major strategies for the PD-1/PD-L1 blockade have mainly focused on the use of antibodies and compounds. Seeking an alternative approach, others employ endogenous proteins as blocking agents. The extracellular domain of PD-1 (ePD1) includes the binding site with PD-L1. Accordingly, we constructed a PD-1-based recombinantly tailored fusion protein (dFv-ePD1) that consists of bivalent variable fragments (dFv) of an MMP-2/9-targeted antibody and ePD1. The melanoma-binding intensity and antitumor activity were also investigated. We found the intense and selective binding capability of the protein dFv-ePD1 to human melanoma specimens was confirmed by a tissue microarray. In addition, dFv-ePD1 significantly suppressed the migration and invasion of mouse melanoma B16-F1 cells, and displayed cytotoxicity to cancer cells in vitro. Notably, dFv-ePD1 significantly inhibited the growth of mouse melanoma B16-F1 tumor cells in mice and in vivo fluorescence imaging showed that dFv-ePD was gradually accumulated into the B16-F1 tumor. Also the B16-F1 tumor fluorescence intensity at the tumor site was stronger than that of dFv. This study indicates that the recombinant protein dFv-ePD1 has an intensive melanoma-binding capability and exerts potent therapeutic efficacy against melanoma. Immobilization of ammonium and phosphate by zeolites synthesized from low-calcium (LC-Z) and highcalcium (HC-Z) fly ashes is studied. LC-Z has higher ammonium immobilization capacity while HC-Z has better performance in phosphate uptake. As part of Ca2+ released from HC-Z compete with ammonium the exchangeable sites in LC-Z, LC-Z can affect phosphate immobilization of HC-Z, and HC-Z also lowers ammonium uptake of LC-Z. Separate dosing of LC-Z and HC-Z, i.e., first LC-Z and then HC-Z, is proved better than mixed dosing at simultaneous immobilization of ammonium and phosphate. The effect parameters for phosphate and ammonium immobilization of LTH-Z are investigated.
Studies on Residue Stress and Deformation Behavior of GCr15 Subjected to Whirlwind Milling

Whirlwind milling is gaining extensive application for processing a large screw. Due to its specific forming principle, the profile of residual stresses and the deformation behavior would been given significant emphasis. Under conditions of large cutting depth (0.06–0.12 mm), the effect of cutting parameters on three characteristic parameters of the residual stress, such as surface residual stress, maximum residual stress and residual stress extreme value, was always different from that under small cutting depth (6–10 μm). Comparing to the right arc of the screw, the plastic deformation in the left arc is more obvious and far-reaching. The most extensive deformation region caused by WM cutting force was found for the topmost surface. As cutting speed increased, the nanohardness variation reflected the decreasing plastic deformation. Therefore, the research on physical change by WM is instructive for extending the fatigue life of the screw.

The delta 6-desaturase gene was cloned from Rhizopus stolonifer, which could accumulate up to 49% of gamma-linolenic acid (GLA, C18:3 Δ^(6,9,12)) to the total fatty acids. The cloned DNA contains a 1,380 bp open reading frame encoding a protein of 460 amino acids, which showed high similarity to those of fungal delta 6-desaturases with three conserved histidine-rich motifs and HPGG motif. Notably, this deduced sequence had a shorter C-terminus. Results demonstrated that the cDNA sequence exhibited delta 6-desaturase activity by accumulation of about 22.4% of GLA to the total fatty acids in the recombinant Pichia pastoris strain GS115.
