        〈프로듀스 101〉시즌2의 서사연구 경쟁과 육성을 중심으로

        본 연구는 2016년 한국에서 인기가 가장 많은 오디션 리얼리티 프로그램 〈프로듀스 101〉 시즌2를 대상으로 경쟁과 육성에 대한 내용을 중심으로 서사분석을 통해서 분석한다. 구체적인 제작 방식, 제작 방식을 설정하는 이유와 의의, 그리고 이 프로그램의 성공한 경험과 장점을 연구를 통하여 밝혀보고자 한다. 이를 위해서 서사 분석 이야기 층위에서 〈프로듀스 101〉 시즌2 제작 중 경쟁 방식, 육성 방식, 그리고 시청자의 위치를 서사구조로 분석한다. 분석 결과를 보면, 〈프로듀스 101〉 시즌2는 생존과 방출을 중심으로 아이돌그룹을 육성 하기 위한 오디션 프로그램으로 먼저 경쟁과 육성의 중요성을 강조한다. 또한, 시청자의 참여는 이 프로그램에 전체적으로 영향을 준다. 순위발표식에서 나타나는 시청자의 100% 선택권과 다른 결정적인 참여 형식은 〈프로듀스 101〉 시즌2의 시청자가 엄청 큰 권력을 가진다고 강조한다. This study conducted a narrative analysis on the audition program, 〈Produce 101〉 season2 which was the most popular in Korean in 2016, focusing on the competition and training participators. Results indicated that the narrative structure of the program had three factors; 1) the competition, 2) training, and 3) audience participation. And, 〈Produce 101〉 season2 was a talent program employing the process of survival and elimination to develop an idol group with audience participations.

        Fibroblast Growth Factor 21 Attenuates Diabetes- Induced Renal Fibrosis by Negatively Regulating TGF- β-p53-Smad2/3-Mediated Epithelial-to-Mesenchymal Transition via Activation of AKT

        Background: Epithelial-to-mesenchymal transition (EMT) is required for renal fibrosis, which is a characteristic of diabetic nephropathy (DN). Our previous study demonstrated that fibroblast growth factor 21 (FGF21) prevented DN associated with the suppressing renal connective tissue growth factor expression, a key marker of renal fibrosis. Therefore, the effects of FGF21 on renal fibrosis in a DN mouse model and the underlying mechanisms were investigated in this study. Methods: Type 1 diabetes mellitus was induced in C57BL/6J mice by intraperitoneal injections of multiple low doses of streptozotocin. Then, diabetic and non-diabetic mice were treated with or without FGF21 in the presence of pifithrin-α (p53 inhibitor) or 10-[4´-(N,N-Diethylamino)butyl]-2-chlorophenoxazine hydrochloride (10-DEBC) hydrochloride (Akt inhibitor) for 4 months. Results: DN was diagnosed by renal dysfunction, hypertrophy, tubulointerstitial lesions, and glomerulosclerosis associated with severe fibrosis, all of which were prevented by FGF21. FGF21 also suppressed the diabetes-induced renal EMT in DN mice by negatively regulating transforming growth factor beta (TGF-β)-induced nuclear translocation of Smad2/3, which is required for the transcription of multiple fibrotic genes. The mechanistic studies showed that FGF21 attenuated nuclear translocation of Smad2/3 by inhibiting renal activity of its conjugated protein p53, which carries Smad2/3 into the nucleus. Moreover pifithrin-α inhibited the FGF21-induced preventive effects on the renal EMT and subsequent renal fibrosis in DN mice. In addition, 10-DEBC also blocked FGF21-induced inhibition of renal p53 activity by phosphorylation of mouse double minute-2 homolog (MDM2). Conclusion: FGF21 prevents renal fibrosis via negative regulation of the TGF-β/Smad2/3-mediated EMT process by activation of the Akt/MDM2/p53 signaling pathway.

        Environmentally Friendly Waterproof and Breathable Nanofiber Membranes with Thermal Regulation Performance by One-Step Electrospinning

        The waterproof and moisture permeable fabric with thermal regulation not only has the protective function forhuman body, but also meets the comfort of human clothing, which is a challenging and innovative research. Herein, we reportan environmentally friendly, simple, and high-efficiency strategy to fabricate waterproof and breathable nanofibermembranes with outstanding thermal regulation ability by introducing stearic acid (SA) into the hydrophobic siliceouspolyurethane (SIPU) solution by a facile one-step electrospinning. Owing to the addition of SA, the waterproofness of SIPU/SA nanofiber membranes is greatly improved, meanwhile, the addition of SA acted as a phase change material to imparttemperature regulation performance in appropriate temperature range to the SIPU/SA nanofiber membranes. By adjusting theaddition amount of the SA, eco-friendly and multifunctional SIPU/SA nanofiber membranes are systemically optimized withgood waterproofness (79 kPa), moisture breathability (8.0 kg m-2 d-1), and outstanding temperature regulation performance(latent heat up to 40 J/g), satisfied tensile strength (6.2 MPa) and excellent breaking elongation of 182 %. The obtained SIPU/SA nanofiber membranes can be suitable for the design of multifunctional wearable textiles for various applications,especially in outdoor protective clothing, medical clothing, intelligent clothing and military products.

        Therapeutic Effects of Fibroblast Growth Factor-21 on Diabetic Nephropathy and the Possible Mechanism in Type 1 Diabetes Mellitus Mice

        Background: Fibroblast growth factor 21 (FGF21) has been only reported to prevent type 1 diabetic nephropathy (DN) in the streptozotocin-induced type 1 diabetes mellitus (T1DM) mouse model. However, the FVB (Cg)-Tg (Cryaa-Tag, Ins2-CALM1) 26OVE/PneJ (OVE26) transgenic mouse is a widely recommended mouse model to recapture the most important features of T1DM nephropathy that often occurs in diabetic patients. In addition, most previous studies focused on exploring the preventive effect of FGF21 on the development of DN. However, in clinic, development of therapeutic strategy has much more realistic value compared with preventive strategy since the onset time of DN is difficult to be accurately predicted. Therefore, in the present study OVE26 mice were used to investigate the potential therapeutic effects of FGF21 on DN. Methods: Four-month-old female OVE26 mice were intraperitoneally treated with recombinant FGF21 at a dose of 100 μg/kg/day for 3 months. The diabetic and non-diabetic control mice were treated with phosphate-buffered saline at the same volume. Renal functions, pathological changes, inflammation, apoptosis, oxidative stress and fibrosis were examined in mice of all groups. Results: The results showed that severe renal dysfunction, morphological changes, inflammation, apoptosis, and fibrosis were observed in OVE26 mice. However, all the renal abnormalities above in OVE26 mice were significantly attenuated by 3-month FGF21 treatment associated with improvement of renal adenosine 5´-monophosphate (AMP)-activated protein kinase (AMPK) activity and sirtuin 1 (SIRT1) expression. Conclusion: Therefore, this study demonstrated that FGF21 might exert therapeutic effects on DN through AMPK-SIRT1 pathway.

        Photooxidative removal of Hg0 from simulated flue gas using UV/H2O2 advanced oxidation process: Influence of operational parameters

        Element mercury (Hg0) from flue gas is difficult to remove because of its low solubility in water and high volatility. A new technology for photooxidative removal of Hg0 with an ultraviolet (UV)/H2O2 advanced oxidation process is studied in an efficient laboratory-scale bubble column reactor. Influence of several key operational parameters on Hg0 removal efficiency is investigated. The results show that an increase in the UV light power, H2O2 initial concentration or H2O2 solution volume will enhance Hg0 removal. The Hg0 removal is inhibited by an increase of the Hg0initial concentration. The solution initial pH and pH conditioning agent have a remarkable synergistic effect. The highest Hg0 removal efficiencies are achieved at the UV light power of 36W, H2O2 initial concentration of 0.125 mol/L, Hg0initial concentration of 25.3 μg/Nm3, solution initial pH of 5, H2O2 solution volume of 600 ml, respectively. In addition,the O2 percentage has little effect on the Hg0 removal efficiency. This study is beneficial for the potential practical application of Hg0 removal from coal-fired flue gas with UV/H2O2 advanced oxidation process.

        Cloning of porcine chemerin, ChemR23 and GPR1 and their involvement in regulation of Lipogenesis

        Chemerin is a novel adipokine which is abundant in adipose tissue to promote adipocyte differentiation and with significant relativity to BMI and insulin sensitivity. We report here the molecular characterization of porcine chemerin and its receptors ChemR23 and GPR1, as well as their transcriptional regulation during lipogenesis. Chemerin was mainly expressed in liver, intestine, kidney and adipose tissue, consistent with the expression pattern of GPR1, but not ChemR23, which was predominantly present in spleen and temperately in adipose tissue. We further investigated the lipogenesis-related transcriptional activation of PPARγ and KLF15 on chemerin and its receptors. The data showed that KLF15, but not PPARγ, can up-regulate the mRNA level of chemerin, ChemR23 and GPR1, which was consistent with the results of luciferase assay that confirmed the effect of KLF15 on ChemR23 promoter. Taken together, our data provide basic molecular information for the further investigation on the function of chemerin in lipogenesis. [BMB reports 2010; 43(7): 491-498]

        Purification and Characterization of an Antioxidant Protein from Fertilized Eggs

        Free radicals may attack cells or tissue, leading to chronic diseases, and antioxidant consumption is potentially useful for removing free radicals. Egg proteins may be used as potential sources of antioxidant considering their ability of scavenging free radicals to apply for food or cosmetics industry. In this study, we obtained a natural antioxidant protein from fertilized eggs, which was a dietary supplement in some Asian countries. Meanwhile, antioxidant activities of these proteins were evaluated using different oxidation systems. With increasing incubation time, the antioxidant activity of these proteins increased during 15 d of incubation. The samples on day 15 were performed for isolation of antioxidant protein. The protein, named P4-1 (MW, 45 kDa), was isolated and purified by consecutive chromatographic methods. P4-1 contained 17 amino acids, which was determined by liquid chromatography-mass spectrometry and Amino Acid Analyzer. Moreover, the amino acid sequence was highly similar to that of ovalbumin. Differential scanning calorimetry showed that the denaturation temperature of P4-1 was 57.16ºC. Furthermore, P4-1 suggested high oxygen radical-absorbance activity in ·OH assays, and its antioxidant activity was stable at 30-50ºC in acidic and neutral pH. Thus, these results revealed that P4-1 may be a potential resource as a natural antioxidant.

        Facile Synthesis of Flower-Like AgI/BiOBr Z-Scheme Nanocomposite with Enhanced Photocatalytic Activity for Degradation of 17α-Estradiol (EE2)

        In the present work, a direct Z-scheme AgI/BiOBr heterojunction displaying improved photocatalytic activity was designed by a chemical bath deposition method. The obtained photocatalysts were systematically characterized through XRD, SEM, XPS, BET and UV-Vis diffuse reflectance spectroscopy. The synthesized AgI/BiOBr photocatalyst displayed superior photocatalytic performance in the case of 17α-ethinylestradiol (EE2), where 97.0% compound has been removed within 9 min of irradiation. The rate constant using AgI/BiOBr photo-catalyst was 16.0 and 138.7 times more than that of pure AgI and BiOBr, respectively. Furthermore, the photocatalytic mechanism was investigated. The results of this study indicated that AgI/BiOBr was a stable and efficient photocatalyst, with promising practical applications.

        Stabilization of a Coupled ODE-wave System with Disturbances

        This paper is devoted to stabilizing an ODE-PDE coupled system with boundary disturbance, where the PDE is chosen to be a wave equation with viscous damping. Applying the active disturbance rejection control (ADRC) technic, the unknown disturbance is estimated by the extended state observer (ESO). Assuming the absence of disturbance, the dynamic boundary connection is designed. The methods of spectral analysis and separation of operator are applied, which ensures the Riesz basis property as well as the spectrum determined growth condition. The exponential stability of the coupled system without disturbance is then concluded. Finally, combining the disturbance estimation and the dynamic boundary connection, a feedback controller designed to stabilize the coupled system, and the closed-loop system is proved to be asymptotically stable.
