Transgenic rice expressing Bacillus thuringiensis (Bt) Cry1Ac and Cry2Aa toxins have proven resistant to lepidopteran pests in China, including the striped stem borer, Chilo suppressalis. Identifying the molecular mechanisms through which Cry toxins affect insect pests is essential, both for understanding the nature of their toxicity, and for assessing the potential for the development of resistance to such toxins. In this study, both onedimensional gel electrophoresis (1-DE), combined ligand blots and mass spectrometry, were used to identify proteins binding to the Cry1Ac and Cry2Aa toxins in brush border membrane vesicles (BBMVs) of the C. suppressalis larvae midgut. Aminopeptidase-N (APN), alkaline phosphatase (ALP), cadherin-like (CAD), ATP-binding cassette transporter subfamily C1 protein (ABCC1), actin, ATPase, alpha amylase, and heat shock protein cognate (HSPC), were detected in Cry1Ac binding bands, and APN, actin, ATPase and HSPC were identified as Cry2Aa potential binding proteins. The novel potential binding protein, delta-aminolevulinic acid dehydratase, was also detected in both Cry1Ac and Cry2Aa binding bands. This is the first study to comprehensively identify the proteins that bind to Cry1Ac and Cry2Aa toxins in C. suppressalis.

Spoofing attacks, especially replay attacks, pose great security challenges to automatic speaker verification (ASV) systems. Current works on replay attacks detection primarily focused on either developing new features or improving classifier performance, ignoring the effects of feature variability, e.g., the channel variability. In this paper, we first establish a mathematical model for replay speech and introduce a method for eliminating the negative interference of the channel. Then a novel feature is proposed to detect the replay attacks. To further boost the detection performance, four post-processing methods using normalization techniques are investigated. We evaluate our proposed method on the ASVspoof 2017 dataset. The experimental results show that our approach outperforms the competing methods in terms of detection accuracy. More interestingly, we find that the proposed normalization strategy could also improve the performance of the existing algorithms.

Several organophosphorus (OP) insecticides can selectively kill the silkworm maggot, Exorista sorbillans (Es) (Diptera: Tachinidae), while not obviously affecting the host (Bombyx mori) larvae, but the mechanism is not yet clear. In this study, the cDNA encoding an acetylcholinesterase (AChE) from the field Es was isolated. One point mutation (Gly353Ala) was identified. The Es-353G AChE and Es-353A AChE were expressed in baculovirus- insect cell system, respectively. The inhibition results showed that for eserine and Chlorpyrifos, Es-353A AChE was significantly less sensitive than Es-353G AChE. Meanwhile, comparison of the I(50) values of eserine, dichlorvos, Chlorpyrifos and omethoate of recombinant Es AChEs with its host (Bombyx mori) AChEs indicated that, both Es AChEs are more sensitive than B. mori AChEs. The results give an insight of the mechanism that some OP insecticides can selectively kills Es while without distinct effect on its host, B. mori. [BMB reports 2010; 43(8): 573-578]

Evaluation of the implementation and performance of a firm's green supply chain management (GSCM) is an ongoing process. Balanced scorecard is a multi-criteria evaluation concept that highlights implementation and performance measures. The literature on the framework is abundant literature but scarce on how to build a hierarchical framework under uncertainty with dependence relations. Hence, this study proposes a hybrid approach, which includes applied interpretive structural modeling to build a hierarchical structure and uses the analytic network process to analyze the dependence relations. Additionally, this study applies the fuzzy set theory to determine linguistic preferences. Twenty dependence criteria are evaluated for a GSCM implemented firm in Taiwan. The result shows that the financial aspect and life cycle assessment are the most important performance and weighted criteria.

Cardiac, respiratory, electrodennal and frontal (F3,F4) EEG responses were analyzed and compared during exposure to slides of International Affective Picture System (lAPS) in the study on 42 students. Physiological responses during 20 s of exposure to slides intended to elicit happiness (nurturant and erotic), sadness, disgust, surprise, fear or anger emotions were Quite similar and were expressed in heart rate (HR) deceleration, decreased HR variability (HRV), specific SCR, increased non-specific SCR frequency (N-SCR), and EEG changes exhibited in theta increase, alpha-blocking and increased beta activity, and frontal asymmetry. However, some emotions demonstrated variations of the response magnitudes, enabling to differentiate some pairs of emotions by several physiological parameters. The profiles showed higher magnitudes of HRV and EEG responses in exciting (i.e., erotic) and higher cardiac and respiratory responses in surprise. The most different pairs were exciting-surprise (by HR, HRV, theta, and alpha asymmetry), exciting-sadness (by theta, alpha, and alpha asymmetry), and exciting-fear (by HRV, theta, F3 alpha, and alpha asymmetry). Nurturant happiness yielded the least differentiation. Differences were found as well within negative emotions, e.g., anger-sadness were differentiated by HRV and theta asymmetry, while disgust-fear by N-SCR and beta asymmetry. Obtained results suggest that magnitudes of profiles of physiological variables differentiate emotions evoked by affective pictures, despite that the patterns of most responses were featured by Qualitative similarity in given passive viewing context.

Fracture combined with traumatic brain injury (TBI) is one of the most common and serious types of compound trauma in the clinic and is characterized by dysfunction of cellular communication in injured organs. Our prior studies found that TBI was capable of enhancing fracture healing in a paracrine manner. Exosomes (Exos), as small extracellular vesicles, are important paracrine vehicles for noncell therapy. However, whether circulating Exos derived from TBI patients (TBI-Exos) regulate the prohealing effects of fractures remains unclear. Thus, the present study aimed to explore the biological effects of TBI-Exos on fracture healing and reveal the potential molecular mechanism. TBI-Exos were isolated by ultracentrifugation, and the enriched miR-21-5 p was identified by qRT‒PCR analysis. The beneficial effects of TBI-Exos on osteoblastic differentiation and bone remodeling were determined by a series of in vitro assays. Bioinformatics analyses were conducted to identify the potential downstream mechanisms of the regulatory effect of TBI-Exos on osteoblasts. Furthermore, the role of the potential signaling pathway of TBI-Exos in mediating the osteoblastic activity of osteoblasts was assessed. Subsequently, a murine fracture model was established, and the effect of TBI-Exos on bone modeling was demonstrated in vivo. TBI-Exos can be internalized by osteoblasts, and in vitro, suppression of SMAD7 promoted osteogenic differentiation, whereas knockdown of miR-21-5 p in TBI-Exos strongly inhibited this bone-beneficial effect. Similarly, our results confirmed that preinjection of TBI-Exos led to enhanced bone formation, whereas knockdown of exosomal miR-21-5 p substantially impaired this bone-beneficial effect in vivo.

A luminescent Tb3+-pyromellitic acid (H4L = pyromellitic acid) coordination polymer (Tb-H4L CP, TbL0.5(H2L)0.5(H2O)DMF) was prepared for multiresponsive fluorescence sensor. The Tb-H4L CP exhibits uniform nanospheres with average diameter of 220 20 nm, good crystalline structure, and good dispersion in water. The Tb-H4L CP emitted the characteristic green fluorescence through antenna effect from H4L which was an excellent antenna molecule when excited at 270 nm and could be used to selectively determine Fe3+ and polychlorinated benzenes based on fluorescence quenching effect. The coordination between Fe3+ and carboxylic oxygen of Tb-H4L to damage the antenna effect and polychlorinated benzenes absorb the excitation light to result the inner-filter effect of fluorescence, which quenched the fluorescence of Tb-H4L CP. The linear range for Fe3+ was 2.0–100mM with a detection limit of 0.7mM and for 1,3,5-trichlorobenzene was 0–0.44 103 ppm with a detection limit of 0.087 ppm. The Tb-H4L CP demonstrates excellent fluorescence sensing performances, indicating that it can be used as a multi-response sensor to effectively measure toxic and harmful substances.

Interacted force between a robot, human and anatomical object in orthopedic surgery provides crucial information for safe and accurate bone cutting. When the robot plays the role of assisting the surgeon in the whole surgical procedure, the robot is trying to comply with the human intention according to various cutting conditions. The reaction force to the robot from the human is for indicating human intention, whereas the force from the anatomical object reflects the cutting condition. In this paper two impedance indices are defined by these two interacted forces and a surgical robot then decides whether assistive or resistant action is activated so that accuracy and safety of bone cutting can be achieved.
