Development of Nanobio Sensors & Diagnosis Chip for Potential Applications based on Insect Models

Microfluidics based on nanobio sensors technologies can provide convenient and accurate diagnosis tools. In this talk, we present recent developments of nanobio sensors & diagnosis chip using microfluidics, with special emphasis on disposable plastic devices format. In detail, we overview of the common methods used in the fabrication of polymer microfluidic systems, including replica and injection mold-ing. Also we explain the different methods by which on-chip operations—such as the pumping and valving of fluid flow, the mixing of different reagents, and the separation and detection of different biochemical species implemented in a microfluidic format. Finally, a few select biotechnological applications of microfluidics are presented to illustrate both the utility of this technology and its potential applications with insect models in the near future.

Objective: This study aimed to compare the surgical outcomes between robotic single-site myomectomy (RSSM) and single-port laparoscopic myomectomy (SPLM) by propensity score (PS) matching analysis. Methods: We included the retrospective data of 276 patients who underwent SPLM and 82 patients who underwent RSSM. Then, we conducted a PS matching analysis to control selection bias using age, gravidity, parity, body mass index, previous history of pelvic surgery, preoperative hemoglobin (Hb) level, preoperative c-reactive protein (CRP) level, and the number, type, and weight of myoma. We selected 76 patients for each group. Results: The matched RSSM group had a shorter hospital stay (OR=0.614, 95% CI=0.4140.913, p=.016) but had a longer operation time (OR=1.022, 95% CI=1.0101.034, p=.000) than the SPLM group. The former group also showed comparable blood loss (OR=1.006, 95% CI=0.9981.014, p=.138), Hb drop (OR=0.910, 95% CI=0.6571.263, p=.574), CRP change (OR=1.174, 95% CI=0.9991.379, p=.051), and complication rate (OR=0.846, 95% CI=0.1076.704, p=.874). Conclusion: The exact difference between SPLM and RSSM is difficult to determine because of the heterogeneity of the disease. The clinical outcomes of RSSM after PS matching analysis revealed the advantages of shorter hospital stays; comparable blood loss, Hb drop, CRP change, and complication rate; and longer operation times compared with those of SPLM.

<P><B>A high‐yield solution‐processed ultrathin (<10 nm) trigonal tellurium (t‐Te) nanowire (NW)</B> is introduced as a new class of piezoelectric nanomaterial with a six‐fold higher piezoelectric constant compared to conventional ZnO NWs for a high‐volume power‐density nanogenerator (NG). While determining the energy‐harvesting principle in a NG consisting of t‐Te NW, it is theoretically and experimentally found that t‐Te NW is piezoelectrically activated only by creating strain in its radial direction, along which it has an asymmetric crystal structure. Based upon this mechanism, a NG with a monolayer consisting of well‐aligned t‐Te NWs and a power density of 9 mW/cm<SUP>3</SUP> is fabricated.</P>

Objective : While balanced crystalloid (BC) could be a relevant fluid regimen with buffer system compared with normal saline (NS), there have been no studies on the optimal fluid for surgery of an unruptured intracranial aneurysm (UIA). This study aimed to compare the effects of fluid regimens between NS and BC on the metabolic and clinical outcomes of patients who underwent surgery for UIA. Methods : This study was designed as a propensity score matched retrospective comparative study and included adult patients who underwent UIA clipping. Patient groups were categorized as NS and BC groups based on the types of pre-operative fluid and the amount of fluid administered during surgery. The primary outcomes were defined as electrolyte imbalance and acidosis immediately after surgery. The secondary outcomes were the length of stay in the intensive care unit (ICU) and duration from the end of the operation to extubation. Results : A total of 586 patients were enrolled in this study, with each of 293 patients assigned to the NS and BC groups, respectively. Immediately after surgery, serum chloride levels were significantly higher in the NS group. Compared to the NS group, the BC group had lower incidence rates of acidemia (6.5% vs. 11.6%, p=0.043) and metabolic acidosis (0.7% vs. 4.4%, p=0.007). As compared to NS group, BC group had significantly shorter duration from the end of the operation to extubation (250±824 vs. 122±372 minutes, p=0.016) and length of stay in ICU (1.37±1.11 vs. 1.12±0.61 days, p=0.001). Throughout multivariable analysis, use of BC was found to be significant factor for favorable post-operative results. Conclusion : This study showed that the patients who received BC during UIA clipping had lower incidence of metabolic acidosis, earlier extubation and shorter ICU stay compared to those who received NS. Therefore, using BC as a peri-operative fluid can be recommended for patients who undergo surgery for UIA.

<P>RT-PCR, nucleotide sequencing, and phylogenetic analysis were performed for genotyping and molecular characterization of noroviruses isolated from Korean groundwater. Among 160 samples collected from 80 sites between 2008 and 2010, 14 samples (8.7 %) from 12 sites were positive for noroviruses (NoVs). The percentages of NoV-positive samples in 2008, 2009, and 2010 were 22.2, 3.2, and 0 %, respectively, representing a yearly decrease. GII-positive samples (n = 9, 5.6 %) outnumbered GI-positive samples (n = 5, 3.1 %). The genotypes of the GI NoVs were GI.2, GI.5, and GI.6, and the genotypes of the GII NoVs were all GII.4. One sample, HM623465, was very similar to CUK-3 and CBNU2 and two GII.4 sequences isolated from the stool of Korean gastroenteritis patients. A BLASTN search revealed several nucleotide sequences highly similar to those of NoVs isolated in this study. The original isolation sources for these similar NoVs were mostly stool (n = 731, 80.0 %) and groundwater (n = 135, 14.8 %), and all the countries from which they were isolated were almost in Asia (96.0 %); specifically, China (n = 192, 21.0 %), Japan (n = 383, 41.9 %), Korea (n = 296, 32.4 %), and other Asian countries (n = 6, 0.7 %). These results suggest that Korean groundwater might be contaminated with NoVs from the stool of infected patients and that these NoVs in turn cause new cases of gastroenteritis through a typical fecal-oral route with region-specific circulation. Therefore, it is important to properly treat sewage, which may include waterborne viruses and manage point sources in groundwater for national health and sanitation. In addition, continuous molecular surveillance remains important for understanding circulating NoVs.</P>
Analyzing and mitigating parasitic capacitances in planar transformers for high‑frequency operation

Planar magnetic components are compact and less susceptible to skin effect due to their thin copper layers. However, the increase in parasitic capacitance has been a challenge in high-frequency operation of planar transformers (PTs). Parasiticcapacitances resonate with inductance and may damage switch devices. Thicker printed circuit board (PCB) and reconfigured windings are suggested in this paper to mitigate the parasitic capacitances. A detailed analysis of the parasitic capacitances is performed with a prototype PT. The resonant frequency increased from 1.27 to 1.63 MHz with a 0.4 mm thicker PCB. The reconfigured PT was 1.38 MHz, 0.3 MHz higher than the original PT.

<P><B>Abstract</B></P> <P>Cyclin-dependent kinase 5 (Cdk5) controls neuronal migration in the developing cortex when multipolar newborn neurons transform to become bipolar. However, by which mechanisms Cdk5 controls cell adhesion in migrating neurons are not fully understood. In this study, we examined the functional interaction between Cdk5 and N-cadherin (Ncad) in newborn neurons when they undergo the multipolar to bipolar transition in the intermediate zone (IZ). Detailed expression analysis revealed that both Cdk5 and Ncad were present in GFP-electroporated migrating neurons in the IZ. Misexpression of dominant negative Cdk5 into the embryonic brains stalled neuronal locomotion in the lower IZ in which arrested cells were round or multipolar. When Ncad was co-introduced with Cdk5DN, however, cells continue to migrate into the cortical plate (CP) and migrating neurons acquired typical bipolar morphology with a pia-directed leading process. Similarly, downregulation of CDK5 resulted in lesser aggregation ability, reversed by the expression of Ncad <I>in vitro</I>. Down-regulation of activity or protein level of CDK5 did not alter the total amount of NCAD proteins but lowered its surface expression in cells. Lastly, expression of CDK5 and NCAD overlapped in the IZ of the human fetal cortex, indicating that the role of Cdk5 and Ncad in neuronal migration is evolutionarily conserved.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Reduced Cdk5 activity inhibits the multipolar-bipolar transition of migrating neurons. </LI> <LI> Ncad expression rescues migration defects caused by the reduction of Cdk5 activity. </LI> <LI> Downregulation of Cdk5 impairs the membrane localization of Ncad. </LI> <LI> Cdk5 and Ncad are present in developing mouse and human brains. </LI> </UL> </P>
