Streptococcus crista displays polar tufts of fimbriae through which it binds to Fusobacterium nucleatum to form morphological units termed "corncobs". In a previous report (Oral Microbiol. Immunol. 10: 220-226, 1995) Tn916 was used to make binding-deficient mutants of S. crista. In this study the transposon insertion site in one of these mutants, CC5A-B15, was characterized. Electron microscopy showed that the morphology of the fimbrial tufts of the binding-deficient mutant was altered. Genetic linkage between the Tn916 insertion in CC5A-B15 and the binding deficient phenotype was confirmed by back-crossing the mutated locus into the parental strain. The mutated locus was cloned and sequenced using an inverse polymerase chain reaction protocol. The resulting product was then used to obtain the wild type locus from a bacteriophage λEMBL3-CC5A and DNA library. The exact site of the Tn916 insertion was determined by comparison of the mutated and wild-type DNA sequences. Based on comparisons to Streptococcus pyogenes M1 and Escherichia coli K12 genome sequences, the transposable element disrupted an apparent ATP-binding cassette (ABC). This ABC transport locus consists of four open reading frames potentially encoding an ATP-binding protein, a lipoprotein, and two transport-related proteins. The results suggest that the reduction in binding of the S. crista mutant to F. nucleatum may be due to the altered transport and assembly of components of the fimbrial tufts.
Impact of Trust-based Security Association and Mobility on the Delay Metric in MANET

Trust models in the literature of MANETs commonly assume that packets have different security requirements. Before a node forwards a packet, if the recipient's trust level does not meet the packet's requirement level, then the recipient must perform certain security association procedures, such as re-authentication. We present in this paper an analysis of the epidemic broadcast delay in such context. The network, mobility and trust models presented in this paper are quite generic and allow us to obtain the delay component induced only by the security associations along a path. Numerical results obtained by simulations also confirm the accuracy of the analysis. In particular, we can observe from both simulation's and analysis results that, for large and sparsely connected networks, the delay caused by security associations is very small compared to the total delay of a packet. This also means that parameters like network density and nodes' velocity, rather than any trust model parameter, have more impact on the overall delay.

<P>Although mutations in more than 90 genes are known to cause CMT, the underlying genetic cause of CMT remains unknown in more than 50% of affected individuals. The discovery of additional genes that harbor CMT2-causing mutations increasingly depends on sharing sequence data on a global level. In this way—by combining data from seven countries on four continents—we were able to define mutations in <I>ATP1A1</I>, which encodes the alpha1 subunit of the Na<SUP>+</SUP>,K<SUP>+</SUP>-ATPase, as a cause of autosomal-dominant CMT2. Seven missense changes were identified that segregated within individual pedigrees: c.143T>G (p.Leu48Arg), c.1775T>C (p.Ile592Thr), c.1789G>A (p.Ala597Thr), c.1801_1802delinsTT (p.Asp601Phe), c.1798C>G (p.Pro600Ala), c.1798C>A (p.Pro600Thr), and c.2432A>C (p.Asp811Ala). Immunostaining peripheral nerve axons localized ATP1A1 to the axolemma of myelinated sensory and motor axons and to Schmidt-Lanterman incisures of myelin sheaths. Two-electrode voltage clamp measurements on <I>Xenopus</I> oocytes demonstrated significant reduction in Na<SUP>+</SUP> current activity in some, but not all, ouabain-insensitive ATP1A1 mutants, suggesting a loss-of-function defect of the Na<SUP>+</SUP>,K<SUP>+</SUP> pump. Five mutants fall into a remarkably narrow motif within the helical linker region that couples the nucleotide-binding and phosphorylation domains. These findings identify a CMT pathway and a potential target for therapy development in degenerative diseases of peripheral nerve axons.</P>
Review : Clostridium difficile Infection: A Worldwide Disease

Clostridium difficile, an anaerobic toxigenic bacterium, causes a severe infectious colitis that leads to significant morbidity and mortality worldwide. Both enhanced bacterial toxins and diminished host immune response contribute to symptomatic disease. C. difficile has been a well-established pathogen in North America and Europe for decades, but is just emerging in Asia. This article reviews the epidemiology, microbiology, pathophysiology, and clinical management of C. difficile. Prompt recognition of C. difficile is necessary to implement appropriate infection control practices. (Gut Liver 2014;8:1-6)

Acuminolide (1), a cytotoxic labdane diterpene with a new type of carbon skeleton, was isolated from the stem irk of Neouvaria acuminatissima, together with its congeners 17-O-acetylacuminolide (2), and an inactive derivative, spiroacuminolide (3), Their structures were determined on the basis of spectroscopic and chemical methods, as well as X-ray cystallography. Isolation, structure elucidation and bioassay results are described.
