Senescence, a hallmark of aging, is a factor in age-related diseases (ARDs). Therefore, targeting senescence is widely regarded as a practicable method for modulating the effects of aging and ARDs. Here, we report the identification of regorafenib, an inhibitor of multiple receptor tyrosine kinases, as a senescence-attenuating drug. We identified regorafenib by screening an FDA-approved drug library. Treatment with regorafenib at a sublethal dose resulted in effective attenuation of the phenotypes of βPIX knockdown- and doxorubicin-induced senescence and replicative senescence in IMR-90 cells; cell cycle arrest, and increased SA-β-Gal staining and senescence-associated secretory phenotypes, particularly increasing the secretion of interleukin 6 (IL-6) and IL-8. Consistent with this result, slower progression of βPIX depletion-induced senescence was observed in the lungs of mice after treatment with regorafenib. Mechanistically, the results of proteomics analysis in diverse types of senescence indicated that growth differentiation factor 15 and plasminogen activator inhibitor-1 are shared targets of regorafenib. Analysis of arrays for phospho-receptors and kinases identified several receptor tyrosine kinases, including platelet-derived growth factor receptor α and discoidin domain receptor 2, as additional targets of regorafenib and revealed AKT/mTOR, ERK/RSK, and JAK/STAT3 signaling as the major effector pathways. Finally, treatment with regorafenib resulted in attenuation of senescence and amelioration of porcine pancreatic elastase-induced emphysema in mice. Based on these results, regorafenib can be defined as a novel senomorphic drug, suggesting its therapeutic potential in pulmonary emphysema.
On-demand acoustic droplet splitting and steering in a disposable microfluidic chip

<P>On-chip droplet splitting is one of the fundamental droplet-based microfluidic unit operations to control droplet volume after production and increase operational capability, flexibility, and throughput. Various droplet splitting methods have been proposed, and among them the acoustic droplet splitting method is promising because of its label-free operation without any physical or thermal damage to droplets. Previous acoustic droplet splitting methods faced several limitations: first, they employed a cross-type acoustofluidic device that precluded multichannel droplet splitting; second, they required irreversible bonding between a piezoelectric substrate and a microfluidic chip, such that the fluidic chip was not replaceable. Here, we present a parallel-type acoustofluidic device with a disposable microfluidic chip to address the limitations of previous acoustic droplet splitting devices. In the proposed device, an acoustic field is applied in the direction opposite to the flow direction to achieve multichannel droplet splitting and steering. A disposable polydimethylsiloxane microfluidic chip is employed in the developed device, thereby removing the need for permanent bonding and improving the flexibility of the droplet microfluidic device. We experimentally demonstrated on-demand acoustic droplet bi-splitting and steering with precise control over the droplet splitting ratio, and we investigated the underlying physical mechanisms of droplet splitting and steering based on Laplace pressure and ray acoustics analyses, respectively. We also demonstrated droplet tri-splitting to prove the feasibility of multichannel droplet splitting. The proposed on-demand acoustic droplet splitting device enables on-chip droplet volume control in various droplet-based microfluidic applications.</P>

연구 목적: 이 연구는 관계를 단절시키는 외부통제 대신 선택이론을 삶에 적용하도록 권장하는 현실치료가 힘을 추구하고 관계를 해쳐 사회적인 문제를 일으킬 뿐만 아니라 남성들의 삶을 불행하게 만드는 남성 성역할 갈등을 완화하는지 확인해 보는 것이다. 연구 방법: 현실치료 프로그램을 중년 남성 대상으로 주 1회 3시간씩 8주간 실시하였다. 연구 내용: 남성 성역할 갈등 척도 총점이 유의미하게 감소하였다. 그리고 남성 성역할 갈등 척도의 하위요인 중 성공·힘·경쟁 문제 요인(SPC) 및 일과 가족 관계에서의 갈등 요인(CBWF)의 감소는 유의미하였으나, 억압된 정서성 요인(RE)의 감소는 유의미하지 않았다. 결론 및 제언: 이러한 결과는 현실치료가 남성 성역할 갈등 완화에 효과적이어서, 남성이 남성 성역할 갈등이라는 부정적 심리상태에서 벗어나 삶의 질을 높일 수 있음을 의미한다. 그러나 남성들이 정서적인 삶의 질을 높이기 위해서는 정서 인식 및 표현 훈련이 필요함을 시사한다. The purpose of this study is to examine whether Reality Therapy, which encourages the application of Choice Theory instead of external control that disconnects relationships, alleviates male gender role conflict, which causes social problems and makes men’s lives unhappy because it seeks strength and harms relationships. Therefore the Reality Therapy program was conducted for 8 weeks, 3 hours once a week for middle-aged men. As a result, the total score of male gender role conflict was significantly reduced. Among the sub-factors of male gender role conflict, the decrease in Success/Power/Competition factor (SPC) and in Conflict Between Work and Family factor (CBWF) was significant, but the decrease in Repressed Emotion fatcor (RE) was not significant. These results imply that Reality Therapy is effective in alleviating male gender role conflicts, so that men can improve their quality of life away from the negative psychological state of male gender role conflict. However, it suggests that men need emotional recognition and expression training to improve their emotional quality of life.

<P>The histamine H4 receptor (H4R), a member of the G-protein coupled receptor family, has been considered as a potential therapeutic target for treating atopic dermatitis (AD). A large number of H4R antagonists have been disclosed, but no efficient agents controlling both pruritus and inflammation in AD have been developed yet. Here, we have discovered a novel class of orally available H4R antagonists showing strong anti-itching and anti-inflammation activity as well as excellent selectivity against off-targets. A pharmacophore-based virtual screening system constructed in-house successfully identified initial hit compound <B>9</B>, and the subsequent homology model-guided optimization efficiently led us to discover pyrido[2,3-<I>e</I>]tetrazolo[1,5-<I>a</I>]pyrazine analogue <B>48</B> as a novel chemotype of a potent and highly selective H4R antagonist. Importantly, orally administered compound <B>48</B> exhibits remarkable efficacy on antipruritus and anti-inflammation with a favorable pharmacokinetic (PK) profile in several mouse models of AD. Thus, these data strongly suggest that our compound <B>48</B> is a promising clinical candidate for treatment of AD.</P> [FIG OMISSION]</BR>

In droplet-based microfluidic systems, in-droplet preconcentration of a sample is one of the important prerequisites for biochemical or medical analysis. There have been a few studies on preconcentration in a moving droplet, but they are limited to practical applications since 1) their method are time-consuming or 2) they require specific properties such as electric and magnetic properties. In this study, we demonstrated the position control of polystyrene particles of 5 and 10 μm in diameter inside a moving water-in-oil droplet using traveling surface acoustic waves. Since the frequencies for effective control of each diameter were found, microparticles with no labels could be utilized. In addition, the proposed method enabled on-demand preconcentration inside a polydimethylsiloxane microchannel. In-droplet preconcentration of microparticles was realized by splitting a mother droplet with manipulated particles at a downstream bifurcation zone. Given these advantages, the proposed system is a promising acoustofluidic lab-on-a-chip platform for preconcentration inside a droplet.
여성 성역할 갈등 완화를 위한 긍정심리 프로그램의 효과

여성 성역할 갈등은 내면화된 성역할 사회화로 인해 자신과 타인에게 부정적인 영향을 주는 심리 상태이다. 본 연구의 목적은 여성 성역할 갈등을 완화하기 위해 긍정심리 프로그램을 실시하여 그 효과를 검증하는 것이다. 이것을 위해 파주시 A 어린이집 교사 13명에게 3시간씩 8주 프로그램을 실시하였다. 또한 경기도 지역 여성 모임 10명에게 6시간씩 4일 과정을 2주에 걸쳐 실시하였다. 파주시 B 어린이집 교사 13명과 서울시 여성 모임 10명에게는 프로그램을 실시하지 않았다. 자료 분석을 위해 SPSS 18.0을 사용하여 프로그램 실시 후의 실험집단과 통제집단 간의 성역할 갈등 점수를 비교한 후, 차이가 있는지를 확인하였다. 공분산분석 결과, 실험집단은 통제집단에 비해 여성 성역할 갈등 점수가 유의미하게 감소하였다. 따라서 긍정심리 프로그램은 여성 성역할 갈등을 완화하여 여성의 정신건강에 기여할 수 있을 것으로 예상된다. 이어 본 연구가 상담에서 갖는 의미, 제한점, 그리고 후속 연구에 대하여 제언하였다. Female gender role conflict is a state of mind that negatively affects oneself and others due to internalized gender role socialization. The purpose of this study is to verify its effectiveness by conducting a positive psychology program to mitigate female gender role conflicts. An eight-week program was conducted for 13 teachers of A Daycare Center in Paju city for three hours, and a four-day course was conducted for six hours for 10 women’s groups in Gyeonggi province for two weeks. The program was not implemented for 13 teachers of B daycare center in Paju and 10 women’s groups in Seoul. For data analysis, SPSS 18.0 is used to compare gender role conflict score between the experimental and control groups after program to see if there are any differences. As a result of the covariance analysis, the experimental group significantly reduced the female gender role conflict score compared to the control group. Therefore, positive psychology programs are expected to contribute to women's mental health by easing gender role conflicts. The implications, limitations, and subsequent studies of this study were then suggested.

Currently, the interests on safety of heavy machinery operation is highly increasing due to growth of construction accidents caused by inexperienced operator, worker’s carelessness, and so on. Therefore, the demand of the automation technology on construction machinery comes to the fore. This paper presents an environment monitoring technology of working site for construction machinery based on laser scanner. The proposed monitoring systems consists two parts. The first part is environmental perception based on one-layer laser scanner processing which estimates object states and classifies. The second part is working area estimation and prediction by analyzing position and velocity kinematics of excavator. It is shown that the proposed working area monitoring technology enhances environment perception performance and safety of construction site.
