Assessment of In Vitro Assay System for Thyroid HormoneDisruptors Using Rat Pituitary GH3 Cells

The development of in vitro assays has been recommended to screening and test-ing the potential endocrine disruptors (EDs). These assay systems focus only on identifying thethe thyroid hormone (TH) disruptors. The aim of this study was to evaluate a test system to detectTH disruptors using rat pituitary tumor GH3 cells. The test system is based on the TH-dependentincrease in growth rate. As expected, L-3,5,3-triiodothyronine (T3) markedly induced a morphologicalchange in GH 3 cells from flattened fibroblastic types to rounded or spindle-shaped types. T3 stimu-lated GH3 cell growth in a dose-dependent manner with the maximum growth-stimulating effect9 M. In addition, T3 increased the release of growth hor-mone and prolactin into the medium of the GH3 cells culture. Using this assay system, the TH-dis-rupting activities of bisphenol A (BPA) and its related compounds were examined. BPA,dimethylbisphenol A (DMBPA), and TCI-EP significantly enhanced the growth of GH3 cells in therange of 1 × 10-5M to 1 × 10-6M concentrations. In conclusion, this in vitro assay system might bestandardization before it can be used as a broad-based screening tool.

Qigong is on the developmental stage and is expanding to the entire world, while it acknowsedges to the West as a universial method of human healthy-longevity. The conception of health known to thr west is based on the providence of modem civilization and science focusing on the physical practices, however, which usually limit the projected effective execution itself. Recently the eastern method of more effective physical-mental training was explored and by which is replaced for the human-healthy longevity in human life. The western standard form and its criterion was established through researches done by the scheme of biological-medical science, effectively it is revolutionizing forward the the eastern medical science as a formal integration of psychology, physiology and the social environmental elements. According to the eastern thought human life is a unified form of mental and physical integration. In China, the government actually supports in official for the development of qigong so that it is activated, practically suggested and progressed lively throughout the nation. Qigong in Korea is activated today. It will be expanded further from now on. Qigong is considered as a method of natural healing, physical theraphy and healthy longevity while giving attention to a lot of people in our country. As they demand qigong in daily lives, there increased the amount of service institute In our community. However, they are not qualified enough both of the knowledge and practical technique. In a future it may cause social problems and there need countermeasure. Therefore, the government must consider about certain problems such as Qigong clinics and its practicioners, which need to be examined before approval, and should enact the national policy and system so that Qigong in korea can be identifie as physical activity for all, as well as systematically established for a way of national health.
초음속 터빈 익렬 앞전 형상 및 노즐-익렬 간격에 따른 유동 특성에 대한 실험적 연구

본 연구에서는 초음속 충동형 터빈의 유통특성을 알아보기 위해 소형 초음속 풍동을 설계 하였으며 Single pass Schlieren system을 이용하여 유통을 가시화하였다. 실험은 2차원 초음속 노즐과 익렬을 조합하여 블레이드 앞전 형상과 노즐-익렬 간극에 따라 실시하였다. 실험을 통해 충격파를 포함한 복잡한 유동 형태와 노즐-익렬, 충격파-경계층 상호작용 등을 관찰할 수 있었다. In this paper, a small supersonic wind tunnel is designed and built to study the flow characteristics of a supersonic impulse turbine cascade. The flow is visualized by means of a single pass Schlieren system. The supersonic cascade with 2-dimensional supersonic nozzle was tested for various blade leading edge shapes and gaps between the nozzle and cascade. Highly complicated flow patterns including shocks, nozzle-cascade interaction and shock boundary layer interactions are observed.

<P>Phospholipase D (PLD) is an enzyme that catalyzes the hydrolysis of phosphatidyl choline (PC) to generate phosphatidic acid (PA) and choline. PLD is believed to play an important role in cell proliferation, survival signaling, cell transformation, and tumor progression. However, it remains to be determined whether enhanced expression of PLD in liver is sufficient to induce hepatotoxicity. The aim of this study was to investigate the possible role of PLD in di(2-ethylhexyl) phthalate (DEHP)-induced hepatotoxicity in Sprague-Dawley rats. The phthalate, DEHP (500 mg/kg/d), was administered orally, daily to prepubertal rats (4 wk of age, weighing approximately 70-90 g) for 1, 7, or 28 d. In this study, protein expression levels of PLD1/2, peroxisome proliferator-activated receptor (PPAR), and cytochrome P-450 (CYP) were determined by Western blot analysis using specific antibodies. Liver weight was significantly increased in the DEHP treatment groups. Immunohistochemical analysis demonstrated that DEHP produced strong staining of proliferating cell nuclear antigen (PCNA) at 28 d of exposure, suggestive of hepatocyte proliferation. A significant rise in PLD1/2 expression was observed in liver of DEHP-exposed rats after 7 d. Further, PPAR관, constitutive androstane receptor (CAR), pregnane X receptor (PXR), and CYP2B1 protein expression levels were markedly elevated in DEHP-treated groups. Our results suggest that DEHP significantly enhanced the expression of PLD, which may be correlated with PPAR관-induced hepatotoxicity through a complex interaction with nuclear receptors including CAR and PXR.</P>

The aim of this study was to evaluate a test system to detect TH disruptors using rat pituitary tumor $GH_3$ cells. The test system is based on the TH-dependent increase in growth rate. As expected, L-3,5,3-triiodothyronine ($(T_3)$ markedly induced a morphological change in $GH_3$ cells from flattened fibroblastic types to rounded or spindle-shaped types. $T_3$ stimulated $GH_3$ cell growth in a dose-dependent manner with the maximum growth-stimulating effect being observed at a concentration $1{\times}10^9M$. In addition, $T_3$ increased the release of growth hormone and prolactin into the medium of the $GH_3$ cells culture. Using this assay system, the TH-disrupting activities of bisphenol A (BPA) and its related compounds were examined. BPA, dimethy/bisphenol A (DMBPA), and TCI-EP significantly enhanced the growth of $GH_3$ cells in the range of $1{\times}10^{-5}M\;to\;1{\times}10^{-6}M$ concentrations. In conclusion, this in vitro assay system might be useful for identifying potential TH disruptors. However, this method will require further evaluation and standardization before it can be used as a broad-based screening tool.
A Preliminary Design of Flight Test Conditions for a Sub-scale RBCC Engine using a Sounding Rocket

Various R&D programs for rocket-based combined cycle (RBCC) engines have progressed worldwide for the space development and the defense applications. As a way toward indigenous domestic RBCC program, a preliminary design of flight test conditions was studied in this study for a sub-scale RBCC engine using a sounding rocket. Launch and flight profiles were calculated for several booster options and compared with that of HyShot II program. The result shows that the Korea Sounding Rocket-II (KSR-II) is a proper candidate to perform the flight test available in Korea. The recommend flight test conditions with KSR-II are Mach 6.0 with a test vehicle of 230 kg and Mach 7.4 with 50 kg. Present study will soon be followed by a design of sub-scale RBCC for a flight test using a sounding rocket.
