        Perspective on Oncogenic Processes at the End of the Beginning of Cancer Genomics

        <P><B>Summary</B></P> <P>The Cancer Genome Atlas (TCGA) has catalyzed systematic characterization of diverse genomic alterations underlying human cancers. At this historic junction marking the completion of genomic characterization of over 11,000 tumors from 33 cancer types, we present our current understanding of the molecular processes governing oncogenesis. We illustrate our insights into cancer through synthesis of the findings of the TCGA PanCancer Atlas project on three facets of oncogenesis: (1) somatic driver mutations, germline pathogenic variants, and their interactions in the tumor; (2) the influence of the tumor genome and epigenome on transcriptome and proteome; and (3) the relationship between tumor and the microenvironment, including implications for drugs targeting driver events and immunotherapies.         Low-Cost Camera Based Laser Power Monitoring and Stabilizing for Micro-Hole Drilling

        In this study, a laser power monitoring and stabilizing system for micro-hole drilling based on optical outputs of a CMOS sensor is presented. The correlation between the laser power and the average brightness of the beam spots on the images was investigated. The estimated laser power was used to build an on-line closed loop control of micro-hole drilling. Experimental results showed that the shortest response time of the laser monitoring system was only 30 ms, which was much faster than a thermopile power meter. The average measuring error was 3%, compared with thermopile power meter. In the PCB drilling experiments of 30 kHz pulse repetition frequency, the PID controller could compensate the power disturbances from 38.4% to 1.8%, and the aspect ratio from 20% to 5%. This study demonstrates the feasibility to develop a low-cost laser power monitoring and stabilizing system in laser micromachining processes.

        Photooxidative removal of Hg0 from simulated flue gas using UV/H2O2 advanced oxidation process: Influence of operational parameters

        Element mercury (Hg0) from flue gas is difficult to remove because of its low solubility in water and high volatility. A new technology for photooxidative removal of Hg0 with an ultraviolet (UV)/H2O2 advanced oxidation process is studied in an efficient laboratory-scale bubble column reactor. Influence of several key operational parameters on Hg0 removal efficiency is investigated. The results show that an increase in the UV light power, H2O2 initial concentration or H2O2 solution volume will enhance Hg0 removal. The Hg0 removal is inhibited by an increase of the Hg0initial concentration. The solution initial pH and pH conditioning agent have a remarkable synergistic effect. The highest Hg0 removal efficiencies are achieved at the UV light power of 36W, H2O2 initial concentration of 0.125 mol/L, Hg0initial concentration of 25.3 μg/Nm3, solution initial pH of 5, H2O2 solution volume of 600 ml, respectively. In addition,the O2 percentage has little effect on the Hg0 removal efficiency. This study is beneficial for the potential practical application of Hg0 removal from coal-fired flue gas with UV/H2O2 advanced oxidation process.

        Genetic Diversity and Structure of Cordyceps sinensis Populations from Extensive Geographical Regions in China as Revealed by Inter-Simple Sequence Repeat Markers

        Cordyceps sinensis is one of the most valuable medicinal caterpillar fungi native to China. However, its productivity is extremely limited and the species is becoming endangered. The genetic diversity of eighteen C. sinensis populations across its major distributing regions in China was evaluated by inter-simple sequence repeat (ISSR) markers. A total of 141 markers were produced in 180 individuals from the 18 populations, of which 99.3% were polymorphic. The low average of Shannon (0.104) and Nei index (0.07) of the 18 populations indicates that there are little genetic variations within populations. For all 18 populations, estimates of total gene diversity (HT), gene diversity within populations (HS), coefficient of genetic differentiation (GST), and gene flow (Nm) were 0.170, 0.071, 0.583, and 0.357, respectively. This pattern suggests that the genetic diversity of C. sinensis is low and most of the ISSR variations are found among populations with little gene exchange. The 18 populations are divided into five groups based on the genetic distance and the grouping pattern matches with the geographic distribution along the latitudinal gradient. The five groups show obvious difference in the GST and Nm values. Therefore, the genetic diversification of C. sinensis populations may be determined by geographic isolation and the combined effects of life history characters and the interaction with host insect species. The information illustrated by this study is useful for selecting in situ conservation sites of C. sinensis.
