<P>Inflammatory cytokines, including tumor necrosis factor-α (TNFα), were elevated in patients with cardiovascular diseases and are also considered as crucial factors in the pathogenesis of preeclampsia; however, the underlying pathogenic mechanism has not been clearly elucidated. This study provides novel evidence that TNFα leads to endothelial dysfunction associated with hypertension and vascular remodeling in preeclampsia through down-regulation of endothelial nitric-oxide synthase (eNOS) by NF-κB–dependent biogenesis of microRNA (miR)-31-5p, which targets eNOS mRNA. In this study, we found that miR-31-5p was up-regulated in sera from patients with preeclampsia and in human endothelial cells treated with TNFα. TNFα-mediated induction of miR-31-5p was blocked by an NF-κB inhibitor and NF-κB p65 knockdown but not by mitogen-activated protein kinase (MAPK) and phosphatidylinositol 3-kinase inhibitors, indicating that NF-κB is essential for biogenesis of miR-31-5p. The treatment of human endothelial cells with TNFα or miR-31-5p mimics decreased endothelial nitric-oxide synthase (eNOS) mRNA stability without affecting <I>eNOS</I> promoter activity, resulting in inhibition of eNOS expression and NO/cGMP production through blocking of the functional activity of the eNOS mRNA 3′-UTR. Moreover, TNFα and miR-31-5p mimic evoked endothelial dysfunction associated with defects in angiogenesis, trophoblastic invasion, and vasorelaxation in an <I>ex vivo</I> cultured model of human placental arterial vessels, which are typical features of preeclampsia. These results suggest that NF-κB–responsive miR-31-5p elicits endothelial dysfunction, hypertension, and vascular remodeling via post-transcriptional down-regulation of eNOS and is a molecular risk factor in the pathogenesis and development of preeclampsia.</P>

<P>Thermal therapy is one of the most popular physiotherapies and it is particularly useful for treating joint injuries. Conventional devices adapted for thermal therapy including heat packs and wraps have often caused discomfort to their wearers because of their rigidity and heavy weight. In our study, we developed a soft, thin, and stretchable heater by using a nanocomposite of silver nanowires and a thermoplastic elastomer. A ligand exchange reaction enabled the formation of a highly conductive and homogeneous nanocomposite. By patterning the nanocomposite with serpentine-mesh structures, conformal lamination of devices on curvilinear joints and effective heat transfer even during motion were achieved. <P><B>Abstract</B></P> <P>Hybridization with conductive 2D nanosheets (NSs) attracts plenty of research activities because of its effectiveness for improving the photocatalyst performance of diverse semiconductors. Here, versatile roles of conductive NSs in hybrid-type photocatalysts are systematically investigated with three representative conductive 2D NSs to synthesize highly efficient visible light-active photocatalysts. Among several conductive NS-based nanohybrids, the RuO<SUB>2</SUB> NS-based nanohybrid exhibits the highest photocatalytic activities. Based on systematic spectroscopic analyses, polar RuO<SUB>2</SUB> NS appears to be more effective as electron reservoir, photosensitizer, cocatalyst and charge carrier pathway in hybrid-type photocatalyst than MoS<SUB>2</SUB> and graphene NSs. The high efficiency of RuO<SUB>2</SUB> NS as hybridization matrix is attributable to the high surface hydrophilicity, high surface bond polarity, and enhanced interfacial electronic coupling of this hydrophilic NS with semiconductor. Objectives:Collagen decrease of Skin appears through various path ways. One of causes may be the Advanced glycation endproducts (AGEs) that combine formation of glucose and protein. The aim of this study was to explore the prevent wrinkle formation of Paeoniae radix (PR) and heated Paeoniae radix (HPR) via AGEs path way. Methods:AGEs formation inhibitory activities of PR and HPR measured using bovine serum albumin, glucose, and fructose. To evaluate the protective effects of PR and HPR in diabetic rats induced with streptozotocin (STZ) and methyl glyoxal (MGO), SD rat were distributed into four groups. Normal rats (Nor), AGEs-induced rats (Con), AGEs-induced rats treated with 100 ㎎/㎏ PR(PR), AGEs-induced rats treated with 100 ㎎/㎏ HPR (HPR). To induce AGEs, streptozotocin (50 ㎎/㎏) was administered intraperitoneally and after 3 days administrated 100mM methyl glyoxal for 3 weeks. Results:The oral administration of HPR inhibited AGEs in skin tissues compared with PR. The increased reactive oxygen species (ROS) levels in the serum were diminished by HPR treatment. The analyses of kidney and skin tissues proteins indicated that HPR treatment effectively reduced AGEs related protein levels as compared to that by PR treatment. Also, HPR decreased anti-oxidant related protein levels in skin tissues such as catalase, glutathione peroxidase. Moreover, it inhibited the reduction of COL1A2 by decreasing MMP-1. Conclusion:Based on these results, it was suggested that PR and HPR could have Improving effects on wrinkle formation. These evidences provide useful information for the development wrinkle formation treated agent.

대부분의 여성은 폐경 이후 신체적 변화를 겪게 된다. 폐경의 증상으로는 홍조, 발한, 몸무게의 증가와 질 건조증 등이 있다. 본 연구에서는 여성의 폐경기 증상 완화에 효능이 있는 열 가지 종류의 생약을 조합하여 만든 혼합 추출물이 동물의 폐경 모델인 난소절제(ovariectomized, OVX) 레트에 어떠한 영향을 미치는지 알아보고자 하였다. 경구투여를 통해 각각 500 mg/kg/d, 1,000 mg/kg/d 농도의 열수 추출물(자음미단)을 8주간 섭취시켰으며, 양성대조군으로 에스트라디올(50 μg/mL)을 사용하였다. 실험을 통해 자음미단이 난소절제 레트에서 몸무게를 유의적으로 감소시켰으며, 간에서의 지방 함량 증가를 억제함을 확인하였다. 자음미단의 섭취는 페경 후 증가하는 혈액의 중성지방과 콜레스테롤 수치도 감소시켰다. 또한, 자음미단은 질의 무게를 증가시켰으며, 특히 질 내벽의 상피세포의 두께에 영향을 주었다. 자음미단의 1,000 mg/kg/d의 농도에서는 에스트로디올과 유사한 정도의 내벽 두께를 관찰할 수 있었다. 따라서 본 실험을 통해 자음미단이 폐경기 여성의 증상인 비만, 대사질환 및 질 건조증의 증상을 완화해줄 수 있으며, 이를 활용한 기능성 식품의 개발이 기대된다. During menopause, elderly woman experience physical and emotional symptoms, including hot flashes, night sweats, weight gain, vaginal dryness, and mood swings. In this study, we investigated the effects of mixed herbal extracts containing Angelica gigas Nakai and Cuscuta chinensis Lam. on menopause symptoms in ovariectomized (OVX) rats. Daily oral treatments of 500 mg/kg and 1,000 mg/kg in OVX rats for 8 weeks significantly reduced weight gain, inhibited hepatic lipid accumulation, and lowered serum lipid levels comparable to the effects induced by estradiol treatments. Mixed herbal extracts (named Jaeummidan) also prevented vaginal dryness as measured by increased vaginal weight. Increased vaginal weight was associated with enlarged epithelium thickness. These data show the preventive actions of Jaeummidan against menopause symptoms and further suggest the possibility of developing functional ingredients in elderly human populations experiencing menopause symptoms.
