      • Effects of PLCE1 Gene Silencing by RNA Interference on Cell Cycling and Apoptosis in Esophageal Carcinoma Cells

        Esophageal squamous cell carcinoma (ESCC) is one of the most malignancies with a poor prognosis. The phospholipase $C{\varepsilon}$ gene (PLCE1) encodes a novel ras-related protein effector mediating the effects of R-Ras on the actin cytoskeleton and membrane protrusion. However, molecular mechanisms pertinent to ESCC are unclear. We therefore designed PLCE1-special small interfering RNA and transfected to esophageal squamous cell (EC) 9706 cells to investigat the effects of PLCE1 gene silencing on the cell cycle and apoptosis of ESCC and indicate its important role in the development of ESCC. Esophageal cancer tissue specimens and normal esophageal mucosa were obtained and assayed by immunohistochemical staining to confirm overexpression of PLCE1 in neoplasias. Fluorescence microscopy was used to examine transfection efficiency, while the result of PLCE1 silencing was examined by reverse transcription (RT-PCR). Flow cytometry and annexin V apoptosis assays were used to assess the cell cycle and apoptosis, respectively. Expression of cyclin D1 and caspase-3 was detected by Western-blotting. The level of PLCE1 protein in esophageal cancer tissue was significantly higher than that in normal tissue. After transfection, the expression of PLCE1 mRNA in EC 9706 was significantly reduced, compared with the control group. Furthermore, flow cytometry results suggested that the PLCE1 gene silencing arrested the cell cycle in the G0/G1 phase; apoptosis was significantly higher than in the negative control group and mock group. PLCE1 gene silencing by RNAi resulted in decreased expression of cyclin D1 and increased expression of caspase-3. Our study suggests that PLCE1 may be an oncogene and play an important role in esophageal carcinogenesis through regulating proteins which control cell cycling and apoptosis.

        Effects of Aqueous Extract of Schizandrae Fructus on Lead-Induced Change of Monoamine Neurotransmitters in Hippocampus

        The effects of aqueous extract of Schizandrae Fructus (AESC) on lead (Pb)-induced changes of monoamine neurotransmitters in the hippocampus (HIP) of adult rats were investigated. Male Sprague-Dawley rats were received intraperitoneal (i.p.) administration of Pb acetate (5 mg/kg/d) for 28 days and sacrificed 7 days after the last administration. Concentrations of norepinephrine (NE), dopamine (DA), serotonin (5-HT), 5-hydroxyindole acetic acid (5-HIAA) in HIP were measured by HPLC. There were significant decreases of NE, DA, 5-HT and 5-HIAA in Pb treated rats (P < 0.05), while pretreatment with AESC (100 mg/kg/d or 300 mg/kg/d, p.o., 2 h before Pb) greatly inhibited the decrease of monoamine transmitters, respectively (P < 0.05). Also, AESC (300 mg/kg/d) significantly increased the reduction of glutathione contents and superoxide dismutase activities in HIP induced by chronic Pb. These results suggest that AESC ameliorates Pb-induced depletion of monoamine neurotransmitters in HIP through its antioxidant activity.

      • Diagnostic Accuracy of 18F-FDG-PET in Patients with Testicular Cancer: a Meta-analysis

        Objective: Fluorine-18-fluorodeoxyglucose positron emission tomography (18F-FDG-PET) is a new technique for identifying different malignant tumors using different uptake values between tumor cells and normal tissues. Here we assessed the diagnostic accuracy of 18F-FDG-PET in patients with testicular cancer by pooling data of existing trials in a meta-analysis. Methods: PubMed/MEDLINE, Embase and Cochrane Central Trials databases were searched and studies published in English relating to the diagnostic value of FDG-PET for testicular cancer were collected. The summary receiver operating characteristic (SROC) curve was used to examine the FDG-PET accuracy. Results: A total of 16 studies which included 957 examinations in 807 patients (median age, 31.1 years) were analyzed. A meta-analysis was performed to combine the sensitivity and specificity and their 95% confidence intervals (CIs), from diagnostic odds ratio (DOR), positive likelihood ratios (PLR), negative likelihood ratio (NLR). SROC were derived to demonstrate the diagnostic accuracy of FDG-PET for testicular cancer. The pooled sensitivity and specificity were 0.75 (95% confidence interval (CI), 0.70-0.80) and 0.87 (95% CI, 0.84-0.89), respectively. The pooled DOR was 35.6 (95% CI, 12.9-98.3). The area under the curve (AUC) was 0.88. The pooled PLR and pooled NLR were 7.80 (95% CI, 3.73-16.3) and 0.31 (95% CI, 0.23-0.43), respectively. Conclusion: In patients with testicular cancer, 18F-FDG-PET demonstrated a high SROC area, and could be a potentially useful tool if combined with other imaging methods such as MRI and CT. Nevertheless, the literature focusing on the use of 18F-FDG-PET in this setting still remains limited.

        A new iterative near-field coherent subspace method for rub-impact fault localization using AE technique

        Acoustic emission (AE) localization is an important method to detect defects in bearing of rotatory machine for faults maintenance. However, only the faults near the sensor array can be detected due to severe attenuation in the recorded AE signals. Therefore, we propose a highly reliable new Iterative near-field Coherent subspace method (IN-CSM) for multiple rub-impact faults localization. The proposed approach contains four improved processes: Modal plate wave theory (MPWT) analysis for the multi-modes decomposition and group velocity revision; Discrete wavelet transform (DWT) for the useful narrow band extraction; Near field Multiple signal classification (N-MUSIC) method for the preliminary position estimations; the IN-CSM algorithm for the multiple coherent sources separation and the precise localizations. The simulations based on N-MUSIC and IN-CSM methods were compared by rubbing teston the test rig of rotation machinery. The results indicate that the proposed method can effectively locate multiple coherent rubbing faults at once. Thus, it is an effective analysis tool for rub-impact fault detection.

      • A Multi-center Survey of HPV Knowledge and Attitudes Toward HPV Vaccination among Women, Government Officials, and Medical Personnel in China

        Objectives: To assess knowledge of HPV and attitudes towards HPV vaccination among the general female population, government officials, and healthcare providers in China to assist the development of an effective national HPV vaccination program. Methods: A cross-sectional epidemiologic survey was conducted across 21 urban and rural sites in China using a short questionnaire. 763 government officials, 760 healthcare providers, and 11,681 women aged 15-59 years were included in the final analysis. Data were analyzed using standard descriptive statistics and logistic regression. Results: Knowledge of HPV among the general female population was low; only 24% had heard of HPV. Less than 20% of healthcare providers recognized sexually na$\ddot{i}$ve women as the most appropriate population for HPV vaccination. There was high acceptance of the HPV vaccine for all categories of respondents. Only 6% of women were willing to pay more than US $300 for the vaccine. Conclusions: Aggressive education is necessary to increase knowledge of HPV and its vaccine. Further proof of vaccine safety and efficacy and government subsidies combined with increased awareness could facilitate development and implementation of HPV vaccination in China.

      • AE Sound Source Localization Using Nearfield MUSIC Algorithm Based on Fourth-Order Cumulants

        Acoustic Emission (AE) source localization is a useful implement to diagnose the incipient faults in rotating machine. This paper proposes an improved Near-field Multiple Signal Classification method using four-order cumulants (NFC-MUSIC) to accurately locate the faults position. In order to overcome dispersion effect and revised velocity, the feature sub narrow band is extracted by Wavelet Packet reconstitution (WPR) and Modal Plate Wave Theory (MPWT). For multi-source decorrelation and increasing localized resolution, the four-order cumulants of observed signal can be selected for localization. The experiment results indicate that the improved method can accurately locate multi-rubbing faults. It is an efficient way to assist incipient rubbing faults diagnosis.

        A ‘‘micro-explosion” strategy for preparing membranes with high porosity, permeability, and dye/salt separation efficiency

        In this study, inspired by ‘‘micro-explosion” strategies, a separation membrane with high porosity andpermeability, and highly efficient separation performance was prepared. With the use of polyvinylidenefluoride (Solvay6015) as membrane material, azodicarbonamide (AC) as an ‘‘explosion center point” tothe casting membrane solution, and NaOH as an ‘‘external stimulus” in the coagulation bath, the twochemicals undergo in-situ foaming reaction to form a loose nanofiltration membrane. FTIR, XPS, andTGA results demonstrated that the decomposition of AC was complete, which produced gases thatincreased the porosity of the membrane. The optimized membrane has a higher flux(101.72 L m2 h1 at 0.3 Mpa), higher negative surface charge, and better mechanical properties underthe premise of separating CR/NaCl. In the separation of pollutants with different molecular weights,the permeation flux of the optimized membrane increased by more than double. This foaming technologywas also applied to another membrane material, ethylene vinyl alcohol, from which we found that themembrane also had higher porosity and better permeability. Together, this paper presents an in-situfoaming method for preparing separation membranes and lays the foundation for solving the trade-offbetween membrane permeability and rejection in dye/salt separation.

        Construction and effect of intramolecular hydrogen bond on solvent resistance of polymeric membranes and their application in impermeable membranes

        Hydrogen bonds, which can be divided into many types along with the deepening of research, exist inmany compounds and have certain impact on properties of substances. In this work, effect of intramolecularhydrogen bond on solvent resistance of polymeric membranes was studied from two aspects. On theone hand, the existence of intramolecular hydrogen bond on solvent resistance of polymeric membraneswas studied by constructing and shielding special hydroxyl groups in typical polymer polyurethane. Onthe other hand, content of intramolecular hydrogen bond on solvent resistance was investigated by buildingdifferent ratios of inter- and intra-molecular hydrogen bonds in polyvinyl alcohols. The structures,solvent resistance and hydrogen bonding interactions were systematically investigated by FT-IR, XRD,Ds and performances before and after immersed in solvents. Permeabilities of the membranes for practicalapplication were also characterized. The results showed that distinct type and ratio of hydrogenbonds played different role on solvent resistance. A good agreement between the swelling performancesand content of intramolecular hydrogen bond proved the positive effect of intramolecular hydrogen bondon solvent durability of polymeric membranes. In addition, polyurethane membranes exhibited goodimpermeability to water vapor, oxygen and chloride ions which showed potential application in manyfields.

        Anatomical Study of the Accessory Tendon of the Extensor Hallucis Longus Muscle and Its Clinical Application

        Background: The accessory tendon of the extensor hallucis longus (ATEHL) muscle is a common abnormal structure, and its clinical significance remains debatable. In this study, we provide the incidence of the ATEHL and characterize its morphological types in Asian cadavers and investigate its clinical applications. Methods: The tendons from 50 adult cadaveric feet, fixed in 10% formalin, were analyzed. We measured the length and width of both the ATEHL and the extensor hallucis brevis (EHB). Results: All dissected specimens had an ATEHL. The first metatarsophalangeal joint was surrounded by an accessory tendon that inserted onto the joint capsule and the dorsal base of the proximal phalanx. We classified the ATEHL into 3 types based on their directions. Differences in ATEHL type based on sex were not statistically significant. Conclusions: We found an ATEHL in all cadaveric specimens in this study. We surmise that the ATEHL acts as an antagonist with the EHB when the toe is extending, which might help prevent the occurrence of hallux valgus deformity.

        G Protein–Coupled Receptor 30 Mediates the Anticancer Effects Induced by Eicosapentaenoic Acid in Ovarian Cancer Cells

        Purpose: While numerous epidemiological studies have indicated that omega-3 polyunsaturated fatty acids have anticancer properties in various cancers, the effects and mechanisms of eicosapentaenoic acid (EPA) in ovarian cancer cell growth are poorly understood. Materials and Methods: ES2 ovarian clear cell carcinoma cells and SKOV3 adenocarcinoma cells were treated with palmitic acid or EPA, followed by flow cytometry and cell counting to measure apoptosis and proliferation, respectively. A modified protein lipid overlay assay was used to further verify whether EPA was a ligand of G protein–coupled receptor 30 (GPR30) in ES2 cells. The levels of apoptosis-related genes, phosphorylated AKT, and phosphorylated ERK1/2 were detected to explore the underlying mechanism. Finally, inhibitory effect of EPA on tumor growth via GPR30 was determined in vitro and in vivo. Results: EPA suppressed ES2 ovarian clear cell carcinoma cells growth via GPR30, a novel EPA receptor, by inducing apoptosis. As a ligand of GPR30, EPA activated the GPR30-cAMP– protein kinase A signaling pathway. When GPR30 was suppressed by siRNA or its inhibitor G15, the antiproliferative action of EPA was impaired. Furthermore, EPA inhibited tumor growth by blocking the activation of AKT and ERK. In the mouse xenograft model, EPA decreased tumor volume and weight through GPR30 by blocking tumor cell proliferation. Conclusion: These results confirm that EPA is a tumor suppressor in human ovarian clear cell carcinoma cells and functions through a novel fatty acid receptor, GPR30, indicating a mechanistic linkage between omega-3 fatty acids and cancers.
