        Metabolism Characteristics of Mycoplasma pneumoniae Infection in Asthmatic Children

        Purpose: Studies have shown that Mycoplasma pneumoniae (Mp) infection can aggravate symptoms in asthmatics. However, the mechanism by which Mp infection exacerbates asthma remains unclear. Metabolomics can help identify the mechanism of Mp aggravating asthma in children, thereby providing more a potential target for improving clinical treatment programs. In this article, we analyzed the metabolic level of patients to explain how Mp aggravates asthma in children. Methods: We divided the subjects into the asthma, Mp infection, asthma combined with Mp infection and healthy groups. Patients’ peripheral blood was collected for metabolic and interaction analysis. Cytokine levels were measured via serum and exhaled breath condensate (EBC). Results: A total of 150 participating subjects were divided into four groups after exclusion. We found out that there were different metabolic pathways between the healthy and disease groups. The major pathways of both asthma and asthma combined with Mp infection were valine, leucine and isoleucine biosynthesis; malate-aspartate shuttle was the main differential pathway for Mp infection. Moreover, even though three disease groups involved 81 metabolites at the same time, compared with asthma combined with Mp infection, 2 single disease groups still involved different amino acid pathways (phenylalanine, tyrosine and tryptophan biosynthesis; valine, leucine and isoleucine biosynthesis). Interaction analysis showed that Mp infection in asthmatic patients not only activated cytokines, but also activated Toll-like receptors (TLRs) 2 and 6. Finally, the levels of interleukin (IL)-4, IL-8, IL-13 and tumor necrosis factor-α in EBC with asthma combined with Mp infection were significantly higher than the 2 single disease groups. Conclusions: Mp infection in asthmatic children can cause changes in the levels of various amino acids in the body, which were enriched in the pathways such as valine, leucine and isoleucine biosynthesis. Palmitic acid can activate TLR2, and iloprost reduces IL-10 levels, ultimately leading to the increased airway inflammation.

        Methanobactin-Mediated One-Step Synthesis of Palladium Nanoparticles

        The development of palladium nanoparticles (PdNPs) with a narrow size distribution is an important aspect of nanotechnology. Methanobactin (Mb) is a copper-binding small peptide that appears to function as an agent for copper sequestration and uptake in methanotrophs. Here, Mb was shown to bind and catalytically reduce Pd (II) to Pd (0). The one-step synthesis of monodisperse PdNPs using Mb as both coordination agent and reduction agent is reported. Fluorescence spectra, UV-visible spectra, X-ray photoelectron spectroscopy (XPS) and Fourier transform-infrared spectroscopy (FT-IR) suggested that the Mb molecules catalytically reduce Pd (II) to Pd (0) with the concomitant production of PdNPs. The Mb is then adsorbed onto the surface of the PdNPs to form an Mb–PdNPs coordination compound. This avoids secondary nucleation. The PdNPs are small with high monodispersity and are easily synthesized in Mb solution. The PdNPs were extremely stable and resisted aggregation even after several months.

        Melanin in Auricularia auricula: biosynthesis, production, physicochemical characterization, biological functions, and applications

        Auricularia auricular (A. auricula), a nutritious fungus and traditional medicinal resource, is known for melanin. This review aims to summarize the research progress on melanin in A. auricula, specifically focusing on biosynthesis, fermentation production, extraction processes, physicochemical characterization, biological functions, and applications. The biosynthesis of melanin in A. auricula primarily involves the oxidative polymerization reaction of phenolic compounds. To enhance melanin production, strategies such as deep fermentation culture, selection of optimal fermentation materials, and optimization of the culture medium have been employed. Various extraction processes have been compared to determine their impact on the physicochemical properties and stability of melanin. Moreover, the antioxidant and antibiofilm activities of A. auricula melanin, as well as its potential beneficial effects on the human body through in vivo experiments, have been investigated. These findings provide valuable insights into the application of A. auricula melanin and serve as a reference for future research in this field.

        Splat‑boundary reinforcement induced by graphene in plasma‑sprayed hydroxyapatite composite coating

        Plasma-sprayed HA coatings on metallic implants are widely used for clinical applications. However, typical lamellar structure along with plasma-sprayed coatings usually leads to weak inter-splat adhesion and impair their mechanical properties. In this research, graphene nanosheet (GNS) reinforced HA coatings were fabricated using plasma spray; these GNSs retained their original structure and distributed homogeneously in the as-sprayed coatings. On the basis of instrumented microindentation tests with and without multiple partial unloading, as compared with the monolithic HA coating, the inter-splat friction force increased by ~ 8.7% for the 1.0 wt% GNS/HA coating, and it slightly decreased to ~ 6.5% for the 2.0 wt% GNS/ HA coating due to GNS agglomeration. Meanwhile, the added GNSs contributed greatly to the indentation yield strength of the HA coatings. These results illustrated that these embedded GNSs at splat boundaries are potential in splat-boundary strengthening and resisting splat sliding.

        OsSPL13 controls grain size in cultivated rice

        <P>Although genetic diversity has a cardinal role in domestication, abundant natural allelic variations across the rice genome that cause agronomically important differences between diverse varieties have not been fully explored. Here we implement an approach integrating genome-wide association testing with functional analysis on grain size in a diverse rice population. We report that a major quantitative trait locus, GLW7, encoding the plant-specific transcription factor OsSPL13, positively regulates cell size in the grain hull, resulting in enhanced rice grain length and yield. We determine that a tandem-repeat sequence in the 5'UTR of OsSPL13 alters its expression by affecting transcription and translation and that high expression of OsSPL13 is associated with large grains in tropical japonica rice. Further analysis indicates that the large-grain allele of GLW7 in tropical japonica rice was introgressed from indica varieties under artificial selection. Our study demonstrates that new genes can be effectively identified on the basis of genome-wide association data.</P>

        Experimental investigation and stability analysis on dense-phase pneumatic conveying of coal and biomass at high pressure

        Conveying characteristics and flow stability are very important for design and control of a conveying system at high pressure. The influences of operating parameters and material properties on conveying characteristics were investigated in an experimental test facility with a conveying pressure up to 4MPa. Wavelet transform and Shannon entropy analysis were applied to analyzing pressure drops through horizontal pipe in order to obtain the stability criterion. Results indicated that the mass flow rate of biomass decreased, while the mass flow rate of pulverized coal increased at first and then decreased with the increase in fluidization velocity. Solid loading ratios for four kinds of powders decreased with the increase in fluidization velocity. Conveying phase diagrams and pressure drops through different test sections of pulverized coal and biomass at high pressure were obtained and analyzed. The influences of coal category,fracture characteristics and particle size on conveying characteristics were determined.

        Atractylenolides (I, II, and III): a review of their pharmacology and pharmacokinetics

        Atractylodes macrocephala Koidz is a widelyused as a traditional Chinese medicine. Atractylenolides(-I, -II, and -III) are a class of lactone compounds derivedfrom Atractylodes macrocephala Koidz . Research intoatractylenolides over the past two decades has shown thatatractylenolides have anti-cancer, anti-infl ammatory, antiplatelet,anti-osteoporosis, and antibacterial activity; protectthe nervous system; and regulate blood glucose and lipids. Because of structural diff erences, both atractylenolide-I andatractylenolide-II have remarkable anti-cancer activities,and atractylenolide-I and atractylenolide-III have remarkableanti-infl ammatory and neuroprotective activities. Wetherefore recommend further clinical research on the anticancer,anti-infl ammatory and neuroprotective eff ects ofatractylenolides, determine their therapeutic eff ects, aloneor in combination. To investigate their ability to regulateblood glucose and lipid, as well as their anti-platelet, antiosteoporosis,and antibacterial activities, both in vitro andin vivo studies are necessary. Atractylenolides are rapidlyabsorbed but slowly metabolized; thus, solubilization studiesmay not be necessary. However, due to the inhibitory eff ectsof atractylenolides on metabolic enzymes, it is necessaryto pay attention to the possible side eff ects of combiningatractylenolides with other drugs, in clinical application. Inshort, atractylenolides have considerable medicinal valueand warrant further study.

        Protective Effect of Schisandra chinensis Polysaccharides Against the Immunological Liver Injury in Mice Based on Nrf2/ARE and TLR4/NF-κB Signaling Pathway

        We have previously demonstrated the hepatoprotective effect of Schisandra chinensis polysaccharides (SCP) against the liver injury induced by alcohol, high-fat diet, and carbon tetrachloride in mice. In this study, we investigated the effect of SCP against the immunological liver injury induced by concanavalin A (Con A) in mice. The results showed that SCP could significantly reduce the level of alanine aminotransferase (ALT), aspartate aminotransferase (AST), tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), interleukin-1β (IL-1β), and interleukin-6 (IL-6) in the serum of mice with immunological liver injury. SCP could significantly decrease the content of malondialdehyde (MDA) and nitric oxide (NO) and increase the activity of superoxide dismutase (SOD) and glutathione (GSH) in the liver tissue. SCP could significantly increase the number of CD4+ and decrease the number of CD8+ in the peripheral blood, and elevate the ratio of CD4+/CD8+. SCP could significantly downregulate the expression of Kelch-like ECH-associated protein 1 (Keap1) and upregulate the expression of nuclear factor-erythroid 2-related factor2 (Nrf2) and downstream gene heme oxygenase-1 (HO-1), and downregulate the expression of toll-like receptor 4 (TLR4), myeloid differentiation primary response gene 88 (MyD88), and nuclear factor-kappa B (NF-κB) proteins. This study indicates that SCP can reduce the release of a large number of inflammatory factors to inhibit the oxidative stress in mice with the immunological liver injury induced by Con A, and its mechanism is closely related to the regulation of Nrf2/antioxidant response element and TLR4/NF-κB signaling pathways.
