        Mechanical behavior of composite gel periodic structures with the pattern transformation

        When the periodic cellular structure is loaded or swelling beyond the critical value, the structure may undergo a pattern transformation owing to the local elastic instabilities, thus leading to structural collapse and the structure changing to a new configuration. Based on this deformation-triggered pattern, we have proposed the novel composite gel materials. This designed material is a type of architectural material possessing special mechanical properties. In this study, the mechanical behavior of the composite gel periodic structure with various gel inclusions is studied further through numerical simulations. When pattern transformation occurs, it results in a different elastic relationship compared with the material at untransformed state. Based on the obtained nominal stress versus nominal strain behavior, the Poisson‟s ratio and corresponding deformed structure patterns, we investigate the performance of designed composite materials and the effects of the uniformly distributed gel inclusions on composite materials. A better understanding of the characteristics of these composite gel materials is a key to develop its potential applications on new soft machines.

        Melting Cell Based Compensated Design Method for Improving Dimensional Accuracy of Additively Manufactured Thin Channels

        Powder-bed fusion additive manufacturing technology makes it possible to produce parts with complicated geometry and high accuracy. However, dimensional deviation caused by powder overmelting and dross formation is still a challenge for manufacturing thin channels. In this study, the origins of the overmelting of printed thin channels were analyzed and a concept called “melting cell” is proposed to describe and quantify the geometric error. Based on the geometrical relationship between the melting cell and target channel, a method for predicting and optimizing the final geometry of thin channels is outlined. In order to verify the method, geometries of thin horizontal circular channels in various sizes are studied as examples. The predicted results by the proposed method show a remarkable agreement with available experimental results. Moreover, a new egg-shaped compensated design, which is able to improve the dimensional accuracy of thin horizontal circular channels, is presented. The proposed method is simple yet very effective. It can be easily extended to the manufacturing of thin channels with various shapes, materials, and different powder bed fusion processes.

        Upregulation of miR-27b Facilitates Apoptosis of TNF-α-Stimulated Fibroblast-Like Synoviocytes

        Purpose: The aim of this study was to explore the function of microRNA-27b (miR-27b) in fibroblast-like synoviocytes (FLSs)stimulated by tumor necrosis factor α (TNF-α). Materials and Methods: mRNA expression of miR-27b in FLS cells (MH7A) treated with or without TNF-α was determined by q-PCR. MiR-27b mimics was transfected into MH7A cells to upregulate miR-27b expression. MTT assay and flow cytometry analysiswere performed to investigate the effect of miR-27b on MH7A cell viability and apoptosis. The targets of miR-27b were predictedby TargetScan. The direct regulation of miR-27b on IL-1β expression was verified by luciferase assay. The protein expressionlevels of apoptosis-related proteins, IL-1β, and NF-κB signaling-related proteins were detected by Western blot. Results: We discovered that miR-27b expression was decreased in MH7A cells stimulated by TNF-α. Upregulation of miR-27b bymiR-27b mimics significantly inhibited the proliferation and promoted the apoptosis of TNF-α-stimulated MH7A cells. Consistently,upregulation of miR-27 decreased the level of Bcl-2 and increased Bax and caspase-3 expression in MH7A cells stimulated byTNF-α. Luciferase assay revealed that IL-1β was indeed a target of miR-27b. By quantitative real-time PCR and Western blot, wefound that the expression of IL-1β is negatively regulated by miR-27b. Moreover, the NF-κB signaling pathway was significantlyinhibited by miR-27b. Conclusion: Taken together, our results illustrated that enhanced miR-27b expression results in the suppression of proliferationand the promotion of apoptosis in FLSs stimulated by TNF-α, partially by regulating IL-1β expression and NF-κB signaling.

        The Correlation between Thyrotropin and Dyslipidemia in a Population-based Study

        This study investigated the relationship between serum thyrotrophin levels and dyslipidemia in subclinical hypothyroid and euthyroid subjects. A total of 110 subjects with subclinical hypothyroidism and 1,240 euthyroid subjects enrolled in this study. Patients with subclinical hypothyroidism had significantly lower high density lipoprotein cholesterol (HDL-C) levels than those who were euthyroid. The lipid profiles were each categorized and mean thyrotrophin levels were higher in subjects in the dyslipidemia subclasses than subjects in the normal subclasses. Thyrotrophin was positively associated with serum triglyceride and negatively associated with serum HDL-C in women. Thyrotrophin was also positively associated with total cholesterol (TC) in the overweight population along with TC and LDL-C in overweight women. In the euthyroid population, thyrotrophin was positively associated with TC in the overweight population. In conclusion, serum thyrotrophin was correlated with dyslipidemia in subclinical hypothyroid and euthyroid subjects; the correlation was independent of insulin sensitivity.
