<P><B>ABSTRACT</B></P><P>Correct temporal and spatial control of actin dynamics is essential for the cytotoxic T cell effector function against tumor cells. However, little is known whether actin engineering in tumor-targeted T cells can enhance their antitumor responses, thereby potentiating the adoptive T cell therapy. Here, we report that TAGLN2, a 22-KDa actin-stabilizing protein which is physically associated with lymphocyte function-associated antigen-1 (LFA-1), potentiates the <I>OTI TCR</I> CD8<SUP>+</SUP> T cells to kill the intercellular adhesion molecule-1 (ICAM-1)-positive/OVA-presenting E0771 cells, but not ICAM-1-negative OVA-B16F10 cells, suggesting an ‘inside-out’ activation of LFA-1, which causes more efficient immunological synapse formation between T cells and tumor cells. Notably, recombinant TAGLN2 fused with the protein transduction domain (TG2P) overcame the disadvantages of viral gene delivery, leading to a significant reduction in tumor growth in mice. TG2P also potentiated the CD19-targeted, chimeric antigen receptor (CAR)-modified T cells to kill Raji B-lymphoma cells. Our findings indicate that activating the TAGLN2–actin–LFA-1 axis is an effective strategy to potentiate the adoptive T-cell immunotherapy.</P>
ZBTB2, a novel master regulator of the p53 pathway.

<P>We found that ZBTB2, a POK family transcription factor, is a potent repressor of the ARF-HDM2-p53-p21 pathway important in cell cycle regulation. ZBTB2 repressed transcription of the ARF, p53, and p21 genes, but activated the HDM2 gene. In particular, ZBTB2 repressed transcription of the p21 gene by acting on the two distal p53 binding elements and the proximal Sp1 binding GC-box 5/6 elements. ZBTB2 directly interacted with Sp1 via its POZ domain and zinc fingers, which was important in the repression of transcription activation by Sp1. ZBTB2 and Sp1 competed with each other in binding to the GC-box 5/6 elements and the two p53 binding elements. ZBTB2 directly interacted with p53 via its zinc fingers, inhibiting p53 binding and repressing transcription activation by p53. The POZ domain, required for transcription repression, interacted with corepressors such as BCoR, NCoR, and SMRT. The interactions deacetylated histones Ac-H3 and -H4 at the proximal promoter. Although ectopic ZBTB2 stimulated cell proliferation, knock-down of ZBTB2 expression decreased cell proliferation and DNA synthesis. Overall, our data suggest that ZBTB2 is a potential proto-oncogenic master control gene of the p53 pathway and, in particular, is a potent transcription repressor of the cell cycle arrest gene p21 by inhibiting p53 and Sp1.</P>

<P>ZNF509 is unique among POK family proteins in that four isoforms are generated by alternative splicing. Short ZNF509 (ZNF509S1, -S2 and -S3) isoforms contain one or two out of the seven zinc-fingers contained in long ZNF509 (ZNF509L). Here, we investigated the functions of ZNF509 isoforms in response to DNA damage, showing isoforms to be induced by p53. Intriguingly, to inhibit proliferation of HCT116 and HEK293 cells, we found that ZNF509L activates <I>p21/CDKN1A</I> transcription, while ZNF509S1 induces <I>RB</I>. ZNF509L binds to the <I>p21/CDKN1A</I> promoter either alone or by interacting with MIZ-1 to recruit the co-activator p300 to activate <I>p21/CDKN1A</I> transcription. In contrast, ZNF509S1 binds to the distal <I>RB</I> promoter to interact and interfere with the MIZF repressor, resulting in derepression and transcription of <I>RB</I>. Immunohistochemical analysis revealed that ZNF509 is highly expressed in normal epithelial cells, but was completely repressed in tumor tissues of the colon, lung and skin, indicating a possible role as a tumor suppressor.</P>

어린이집 교사의 다문화가정 유아 경험을 통한 교육적 노력과 실천

본 연구는 어린이집 교사의 다문화가정 유아 경험을 통하여 교사들이 어떠한 노력과 실천들을 수행하였는가를 탐색해 보고 이 유아들에 대한 이해를 높이며 교사들에게 필요한 정보와 자료를 제공하고자함에 목적을 두었다. 이에 현상학적 질적 연구방법을 활용하여 연구를 수행하였으며, 다문화가정 유아를 담당하는 보육교사 6명을 대상으로 심층면접을 실시한 결과, 총 18개의 소주제들이 발굴되어 2개의 범주에서 8개의 주제들로 정리되었다. 첫째, 다문화가정 유아에 대한 경험의 범주에서 유아들의 발음 부정확, 어휘력 부족, 주의력 결핍, 혼자놀이 즐김, 소극적이며 모방행동을 하는 모습들이 포착되었다. 식습관은 편식을 하며, 의생활에서는 계절에 맞지 않는 옷을 착용하는 경향이 높았다고 한다. 반면, 이들은 신체활동에는 적극적이고, 그림/숫자를 활용한 학습에서 몰두하는 모습들이 나타났다. 둘째, 교사의 노력과 실천 범주에서, 교사가 유아의 개별적 수준과 특성을 고려한 언어 및 맞춤형 학습지도, 또래관계 개입, 의생활과 식습관 지도 등에서 적극적이었음이 발견되었다. 그 결과, 다문화가정 유아들은 전반적으로 행동의 변화와 함께 언어적 발달, 학습력 향상, 식습관 변화를 보였다고 한다. 하지만, 참여교사들의 이러한 교육적 노력과 실천은 소수자인 다문화가정 유아가 어린이집, 학교, 사회 등에 적응하기 위한 기본적인 소양을 가르치고자 한 것으로 해석된다. 이러한 맥락에서 어린이집 교사 대상 다문화교육 및 연수가 체계적으로 제공될 필요가 있으며 이 유아를 지도하기 위한 교사 지침서와 가이드라인 등을 준비하여 배포 할 것을 제안한다. 또한, 교사들이 현장에서 다양한 유아들을 지도하기 위한 다문화적 교수역량을 강화해야 함을 시사하는 바이다. This aim of this study was to discover the educational efforts and practices of daycare teachers with children from multicultural families. Six teachers of daycare centers participated in this study by sharing their hands-on experiences with the children. From their experiences studied in depth, eight main themes were discovered under two categories. The first category is about teachers’ daily experiences and the second category is about teachers’ teaching practices and efforts. In the first category, main findings were about the teachers’ daily experiences with the children regarding the differences in their language development, peer relations, behavior, eating habits, and ways of being dressed compared to their classmates. Under the second category, the main findings were on teachers’ practices and efforts with the children in regards to their Korean language skills, readiness for school, peer relations, eating habits, and ways of being dressed. From the teachers’ viewpoints, all children changed and developed greatly in all areas. Under the perspective of multicultural education, however, teachers’ efforts and practices with the children were mostly based on the assimilation perspective to help them become members of the Korean society regardless of their multicultural backgrounds. This study suggests that daycare center teachers need to be retrained or educated to better understand multiculturalism in order to teach all children under the multicultural perspective. Also, daycare teacher’s guidebooks or guidelines under the multicultural perspective should be prepared and provided to them.
Inkjet Printing of Polymeric Resistance Humidity Sensor Using UV-Curable Electrolyte Inks

UV-curable electrolytes inks were prepared by dissolving [2-[(methacryloyloxy)ethyl]dimethyl]propylaminium bromide (MEPAB), 1,5-[bis(2-methacryloxyethyl)dimethyl ammono]penthane dibromide (MDAPD), hexafunctional reactive oligomer (SP1013), trimethylolpropane triacrylate (TMPTA) and a photoinitiator in methanol/2-methoxyethanol. The compositions of the electrolyte inks were MEPAB/MDAPD/SP1013/TMPTA=6/24/17.2/12.8, 15/15/17.2/12.8 and 24/6/17.2/12.8. Humid-membranes were deposited on a gold electrode/alumina electrode using an ink-jet printer equipped with a UV lamp followed by immediate UV curing. The humidity-sensing characteristics including hysteresis, frequency and temperature dependence, response time and water durability were estimated.
First Report of Gray Mold Disease on Endangered Species Cypripedium japonicum

Cypripedium japonicum is known to be the indigenous plant to Korea, Japan, and China. However, C. japonicum represents the most critically endangered plant species in South Korea. The plant is esthetically pleasing due to its flower, which is larger than any other orchidaceous species. Disease symptoms relating to gray mold were observed on C. japonicum in May 2019. The suspected pathogen was successfully isolated from the symptomatic leaf tissue and conducted a pure culture of the fungi. The conidia formed consisted of a colorless or light brown single cell, which was either egg or oval-shaped with a size of 7.1 to 13.4×5.2 to 8.6 lm. Molecular phylogenetic relationship analysis was also confirmed that the pathogen concerned belonging to the family of Botrytis cinerea. Therefore, the findings confirmed that the pathogen isolated from C. japonicum was consistent with the unique properties of B. cinerea.
