        Qualitative and Quantitative Magnetic Resonance Imaging Phenotypes May Predict CDKN2A/B Homozygous Deletion Status in Isocitrate Dehydrogenase-Mutant Astrocytomas: A Multicenter Study

        Objective: Cyclin-dependent kinase inhibitor (CDKN)2A/B homozygous deletion is a key molecular marker of isocitrate dehydrogenase (IDH)-mutant astrocytomas in the 2021 World Health Organization. We aimed to investigate whether qualitative and quantitative MRI parameters can predict CDKN2A/B homozygous deletion status in IDH-mutant astrocytomas. Materials and Methods: Preoperative MRI data of 88 patients (mean age ± standard deviation, 42.0 ± 11.9 years; 40 females and 48 males) with IDH-mutant astrocytomas (76 without and 12 with CDKN2A/B homozygous deletion) from two institutions were included. A qualitative imaging assessment was performed. Mean apparent diffusion coefficient (ADC), 5th percentile of ADC, mean normalized cerebral blood volume (nCBV), and 95th percentile of nCBV were assessed via automatic tumor segmentation. Logistic regression was performed to determine the factors associated with CDKN2A/B homozygous deletion in all 88 patients and a subgroup of 47 patients with histological grades 3 and 4. The discrimination performance of the logistic regression models was evaluated using the area under the receiver operating characteristic curve (AUC). Results: In multivariable analysis of all patients, infiltrative pattern (odds ratio [OR] = 4.25, p = 0.034), maximal diameter (OR = 1.07, p = 0.013), and 95th percentile of nCBV (OR = 1.34, p = 0.049) were independent predictors of CDKN2A/B homozygous deletion. The AUC, accuracy, sensitivity, and specificity of the corresponding model were 0.83 (95% confidence interval [CI], 0.72–0.91), 90.4%, 83.3%, and 75.0%, respectively. On multivariable analysis of the subgroup with histological grades 3 and 4, infiltrative pattern (OR = 10.39, p = 0.012) and 95th percentile of nCBV (OR = 1.24, p = 0.047) were independent predictors of CDKN2A/B homozygous deletion, with an AUC accuracy, sensitivity, and specificity of the corresponding model of 0.76 (95% CI, 0.60–0.88), 87.8%, 80.0%, and 58.1%, respectively. Conclusion: The presence of an infiltrative pattern, larger maximal diameter, and higher 95th percentile of the nCBV may be useful MRI biomarkers for CDKN2A/B homozygous deletion in IDH-mutant astrocytomas.

        Measuring economic sentiment using ordinary response options

        Economic sentiment is typically measured using ordinary response options. The University of Michigan and the United States Conference Board are two widely used major indexes that have separately established independent consumer sentiment indexes based on three-level ordinary response options: positive, neutral, and negative. Notwithstanding, limited attention has been paid to the structural differences in their built-in formulas, which are referred to the disparate micro scoring schemes applied to an individual question. This paper examines the structural difference of the two indexes and then addresses situations where one is more reliable than the other. Real data from business tendency surveys of the Organization for Economic Cooperation and Development are used to illustrate our points empirically. As a conclusion, it is stressed that the two indexes should be handled with care when applied to economic sentiment comparison studies.

        Background: Chemotherapy-refractory acute leukemia is only salvaged by allogeneic stem cell transplantation(ASCT). We intended to analyze clinical outcome of patients with acute leukemia who received ASCT in chemo-refractory state. With this analysis, we tried to find subset who could be salvaged by ASCT. Methods: We retrospectively analyzed patients who received ASCT for chemo-refractory acute leukemia. We included patients who received ASCT while the disease was not in complete remission. Results: A total of 226 patients were analyzed. Disease included acute myeloid leukemia (AML, N=156), acute lymphocytic leukemia (ALL, N=52), and acute biphenotypic leukemia (ABL, N=18). Proportion of secondary leukemia was 15.9%. 41.6% of patients were primary chemo-refractory, while 58.4% of patients developed chemo-refractoriness after achievement of CR at least once. CR was achieved in 79.6% of patients by ASCT. CR rate seemed to be higher in AML (84%) compared to ALL (71%) or ABL (67%) (p=0.058). OS was significantly superior in patients with AML or ABL compared to ALL (p=0.020). Five-years OS rate was more than 20% in patients with AML or ABL, while 5-years OS rate was less than 5% in patients with ALL. Patients who received sibling ASCT had better survival outcome compared to unrelated ASCT or haploidentical ASCT (p<0.001). Primary chemo-refractoriness (p=0.284) and conditioning intensity (p=0.404) did not affect survival outcome. Karyotypic risk grouping had prognostic value in these patients with shorter OS observed in patients with poor risk group compared to standard or good risk group (p=0.006). Conclusions: Among acute leukemias, only AML and ABL are salvaged by ASCT when they are chemo-refractory. However, outcome of primary chemo-refractory disease is not inferior, hence ASCT should be considered in this disease. For ASCT in chemo-refractory acute leukemia, sibling ASCT has better outcome than unrelated ASCT.

      • Awareness of putative risk factors for Alzheimer’s disease among elderly Koreans

        <P>Objective</P><P>Although there are rapidly growing concerns about the high rates of cognitive dysfunction in Korea, the knowledge of risk factors for Alzheimer’s disease (AD) among the general public in Korea remains to be elucidated.</P><P>Methods</P><P>A total of 2767 randomly selected subjects from the Ansan Geriatric Study were questioned on their knowledge of putative risk factors for AD. Their answers were compared with their sociodemographic data and other variables.</P><P>Results</P><P>The most common stated risk factor was being older (59.6%), followed by head trauma (33.6%) and cerebrovascular disease (30.4%). However, a substandard education, which is a known risk factor, was considered significant by only 9.5% of the subjects. Predictors for a worse knowledge of the risk factors for AD were being older, a lower level of education, lower economic status and the attitude that dementia is not curable.</P><P>Conclusion</P><P>This study revealed that misunderstanding about AD is more prevalent in older subjects and those with a lower level of education, and so public health education on the basic concepts of AD should be targeted at this population.</P>

        Sexual Maturation May Affect the Levels of n-6 PUFA in Muscle Tissues of Male Mice

        Lipid metabolism in mature male mice may be different from immature male mice, but the relationship of lipid metabolism, especially n-6 fatty acid metabolism, and sexual maturation is not clearly established. This study was carried out to elucidate whether sexual maturation may affect the metabolism of functional n-6 fatty acids of lipid components by investigating the composition of fatty acids in the longissimus muscle tissues of mature and immature male mice with GC and analyzing the expression of genes and proteins for synthesis of n-6 fatty acids with real-time PCR and western blotting, respectively. Mature male mice showed significantly higher testosterone level in the sera. Similarly, n-6 fatty acids, levels of linoleic acid (LA 18:2n-6) and total n-6 PUFA (Polyunsaturated fatty acids) were increased, but the levels of ${\gamma}$-linolenic acid (GLA; 18:3n-6), dihomo-${\gamma}$-linolenic acid (DGLA; 20:3n-6) and arachidonic acid (AA; 20:4 n-6) were decreased in the mature male mice. mRNA levels of ${\Delta}5$-desaturase (FASD1) and elongase (ELOVL5) genes related to n-6 fatty acid metabolism increased. However, the level of FADS1 protein only increased in mature male mice. In conclusion, this study suggested that sexual maturation of male mice affected n-6 fatty acid metabolism by stimulating the expression of enzyme FADS1 of n-6 PUFA metabolism.

      • Two Distinct Isozymes of Repair Protein Carboxyl O-Methyltransferase from Porcine Brain

        Protein carboxyl 0-methyltransferase (PCMT) catalyzes the transfer of a methyl group from S-adenosyl-L-methionine to free carboxyl groups of methyl-accepting substrate proteins. Two isozymes were separated by DEAE-Sephacel chromatography from porcine brain cytosol and designated PCMT I and II. Isozymes I and II were further purified by adenosy1homocysteine-Sepharose 4B and Superose HR 12 chromatography. The molecular weights of the purified PCMT I and II were determined by mass spectrometry to be 20,138 Da and 25,574 Da, respectively. The two enzymes displayed different isoelectric points; 7.9 for PCMT I and 5.3 for PCMT II. Isozymes I and II exhibited similar substrate specificities when tested with various methyl-accepting proteins. Myelin basic protein, a component of myelinated neurons, was found to be an excellent methyl-accepting substrate for both PCMT isozymes with different K_m values, 21.1 μM for PCMT I and 10.6 μM for PCMT II. The PCMT activity and methyl-accepting capacity displayed similar distribution in the various brain regions with an exception of the lower values in the cerebellum. The overall distribution may relate to a general function of protein repair by PCMT in the brain.

      • Amplified circularly polarized phosphorescence from co-assemblies of platinum( <small>II</small> ) complexes

        <▼1><P>Co-assemblies of platinum(<SMALL>II</SMALL>) complexes produce amplified circularly polarized phosphorescence emission.</P></▼1><▼2><P>Molecules capable of producing zero-field circularly polarized phosphorescence (CPP) are highly valuable for chiroptoelectronic applications that rely on triplet exciton. However, the paucity of tractable molecular design rules for obtaining CPP emission has inhibited full utilization. We report amplification of CPP by the formation of helical co-assemblies consisting of achiral square planar cycloplatinated complexes and small fractions of homochiral cycloplatinated complexes. The latter has a unique Pfeiffer effect during the formation of superhelical co-assemblies, enabling versatile chiroptical control. Large dissymmetry factors in electronic absorption (<I>g</I><SUB>abs</SUB>, 0.020) and phosphorescence emission (<I>g</I><SUB>lum</SUB>, 0.064) are observed from the co-assemblies. These values are two orders of magnitude improved relative to those of individual molecules. In addition, photoluminescence quantum yields (PLQY) also increase by a factor of ten. Our structural, photophysical, and quantum chemical investigations reveal that the chiroptical amplification is attributable to utilization of both the magnetically allowed electronic transition and asymmetric coupling of excitons. The strategy overcomes the trade-off between <I>g</I><SUB>lum</SUB> and PLQY which has frequently been found for previous molecular emitters of circularly polarized luminescence. It is anticipated that our study will provide new insight into the future research for the exploitation of the full potential of CPP.</P></▼2>

        In recent image processing, beyond the object recognition problem, deep learning has been used in various aspects such as object detection, semantic segmentation. In addition, classic technique-based detection has been performed variously. These technologies are applied in various systems such as factory automation systems, automatic target recognition (ATR) systems, autonomous driving systems, etc. Object detection is performed in various categories such as people, vehicles and animals, etc. And it is operated for various situations which contain different object size, image size, distance range from near to remote, changeable environment, etc. For the situation analysis, indicators need to be used appropriately. And when researchers make some algorithm for object detection, if there are no any evaluation indicators, the algorithm can’t be demonstrated. So, it is important to know about performance indicators of object detection. Various indicators are used in object detection. As a result, this paper introduces performance indicators of object detection. The main purpose of the survey is that researchers find the proper performance indicator for object detection. And It can help to compare the detection result with a different algorithm result, exactly and effectively.
