The ethanol extracts from bark of red pine, Pinus densiflora Siebold et Zuccarini, was examined for single-dose and seven-day repeated-dose toxicity with Sprague-Dawley rats. The single oral dose of the extracts at 5000 ㎎/㎏ did not produce mortality or abnormal gross appearance of the internal organs of rats. In seven-day repeated-dose toxicity study, the extracts administered at doses of 100, 1000, or 5000 ㎎/㎏ for 7 days. There were no significant changes of body weights and organ weights during the experimental period. Hematological analysis and clinical blood chemistry have no revealed toxicity effect of the ethanol extracts from bark of Pinus densiflora. Pathologically, neither gross abnormalities nor histopathological changes observed between controls and treated rats both sexes. Collectively, these data suggest that the ethanol extracts from bark of Pinus densiflora have a high margin of safety.
흰쥐에서 땃두릅 에탄올 추출물의 14일 반복경구투여에 의한 독성시험

본 연구는 시험물질 국내산 땃두릅나무 추출물의 반복 경구투여에 의한 독성을 질적ㆍ양적으로 평가하기 위하여 식품의약안전청의 의약품 등의 독성시험기준에 따라 Sprague-Dawley 계열의 암ㆍ수 흰쥐에 시험 물질을 0, 500, 1,000 및 2,000 ㎎/㎏의 용량으로 14일간 반복경구 투여한 후 사망률, 일반증상, 체중변화, 혈액 및 혈액생화학적 변화, 부검소견, 조직학적인 변화를 관찰하였다. 시험기간 중 암ㆍ수 모든 군에서 시험물질 투여에 기인한 일반적인 증상변화는 관찰되지 않았고, 시험물질의 반복 투여로 인한 사망례 역시 관찰되지 않았다. 시험 물질의 투여에 기인한 유의적인 체중감소 또한 나타나지 않았으며, 상기 이외의 육안적인 부검소견에서도 시험물질 투여에 기인한 어떠한 이상소견도 발견되지 않았다. 혈액학적 분석 결과 일부 시험물질 투여군에서 WBC, MCHC, PLT의 수치가 유의적인 변화를 보였으나, 정상범위 내에서의 변화로 시험물질에 의한 독성이라고 판단하기는 어렵다. 혈청중의 ALP 및 뇨의 BUN의 유의적인 감소가 시험물질 투여군에서 관찰되었으나 정상 생리 범위 내에서의 변화로 시험물질에 의한 독성이라 판단 하기는 어렵다. 간장과 신장의 조직학적인 관찰에서 시험물질 투여에 의한 변화는 관찰되지 않았다. 본 시험결과는 흰쥐에서 땃두릅나무 에탄올추출물의 14일 반복경구 투여는 2,000 ㎎/㎏ 이하의 용량에서 어떠한 독성영향도 유발하지 않았으며, 본 시험 조건하에서 최소치사량은 2,000 ㎎/㎏ 이상이라는 것으로 나타났다. 따라서 본 시험을 통하여 땃두릅나무 추출물은 투여가능 최대 용량에서도 독성이 없는 안전한 천연물이라는 것을 확인하였고, 의약품이나 기능성식품으로서의 개발 가능성을 확인하였다. Oplopanax elatus (O. elatus) is a tall deciduous shrub that has traditionally been used for treating a variety of ailments such as diabetes, coughing, rheumatism, gastro-intestinal disorders, and wounds. In order to examine the safety of the ethanol extracts of O. elatus, we performed a 14-day repeated-dose toxicity study with Sprague-Dawley rats. The rats were treated with daily doses of the O. elatus ethanol extracts by gavage at 0, 500, 1000, and 2000 ㎎/㎏/day for 14 consecutive days. We recorded clinical signs of toxicity, body weight, hematology, organ weights, gross and histological changes in target organs, and clinical chemistry analysis data for all rats. There were no significant changes in body and organ weights during the experimental period. The hematological analysis and clinical blood chemistry data revealed no toxic effects from the O. elatus ethanol extracts. Pathologically, neither gross abnormalities nor histopathological changes were observed between the control and treated rats of both sexes. Collectively, these data suggest that the ethanol extracts of O. elatus have a high margin of safety.

본 연구의 목적은 향신료로서 한국인의 식생활에 중요한 위치를 차지하고 있는 홍고추가루 수용성 추출물의 항염증 활성을 검토해보고자 수행되었다. 그 결과 홍고추가루의 수용성 추출물은 LPS 처리에 의한 NO 및 PGE₂의 생성을 현저히 억제시키는 것을 관찰할 수 있었으며, NO의 생합성효소인 iNOS 단백질의 발현 또한 억제시킴을 확인할 수 있었다. 홍고추가루의 수용성 추출물은 염증성 사이토카인인 IL-6의 생성억제 효과도 탁월하였다. 이러한 연구결과로 볼 때 고추의 수용성 추출물이 대식세포에 의해 생성되는 염증반응의 매개물질인 NO, PGE₂, IL-6의 생성을 억제함으로 염증반응을 완화시켜 주는 것으로 판단된다. 차후 항염증 활성을 갖는 홍고추가루의 수용성 추출물의 성분을 동정하는 연구가 필요할 것으로 사료되며, 고추가 식품유래의 항염증 활성을 갖는 제품 개발에 있어 유용한 식품자원 원료로 사용될 수 있을 것으로 생각된다. Inflammation is a pivotal component of a variety of diseases, such as atherosclerosis and tumour progression. Various naturally occurring phytochemicals exhibit antiinflammatory activity and are considered to be potential drug candidates against inflammation-related pathological processes. Red pepper is the most consumed species in Korea. However, the antiinflammatory effects of red pepper have not been characterized. Thus, the present study was designed to evaluate the effects of the aqueous extract from red pepper (RPAE) on lipopolysaccharide (LPS)-induced inflammatory responses in murine macrophages. RPAE demonstrated strong antiinflammatory activity through its ability to reduce nitric oxide and prostaglandin E2 production in the LPS-stimulated mouse macrophage cell, RAW264.7. It also inhibited the production of interleukin-6 (IL-6) on the LPS-stimulated RAW264.7 cells. Further study indicated that LPS-stimulated induction of inducible nitric oxide synthase and cyclooxygenase-2 was significantly inhibited by RPAE exposure (1,000 ㎎/mL) in RAW264.7 cells. Collectively, these data suggest that the use of RPAE may be a useful therapeutic approach to various inflammatory diseases.

The primary aim of this study is to examine if the awareness of one`s body attractiveness affects her sexual satisfaction and behavioral intention. The objects of the study were taken from 5 of local fitness center and culture center within “A” district; a group of participants consist of 262 married wives who were participating in dance-for-all activities. Exploratory factor analysis and credibility test were followed by simple regression analysis. The significance level was set at .05. The results of the tests were as follows. First, one`s body attractiveness shows beneficial effect on her sexual satisfaction, and it turns out that there is a static causality in the process. Second, awareness of body attractiveness also brings about fruitful influence on one`s behavioral intention. There is a static causality in the process. Last but not least, one`s sexual satisfaction evidently has an invaluable impact on her behavioral intension. There is a static causality in the process as well. From the results mentioned above, some proposals can be drawn. It is necessary to bring Partner`s perception of participant`s body attractiveness and sexual appeal and sexual satisfaction to light. Furthermore, there need to be more profound and insightful understanding about bodily discourse such as body image satisfaction and eroticism.
Antigenicity of HM10760 in Guinea Pigs

HM10760 is a recombinant human erythropoietin that has been developed as a drug for anemia. In this study, antigenic potential of HM10760 was examined by active systemic anaphylaxis in guinea pigs and passive cutaneous anaphylaxis in a guinea pig-guinea pig system. HM10760 was subcutaneously administered at 0, 2, and 20 ㎍/㎏ and also as a suspension with adjuvant (20 ㎍/㎏ + FCA). Ovalbumin as a suspension with adjuvant was administered to induce positive control responses. In the active systemic anaphylaxis test, no symptoms except rubbing or licking nose and urination that were considered as physiological phenomena were observed at 0 ㎍/㎏. Four of 5 animals at 2 ㎍/㎏ and all the 5 animals at 20 ㎍/㎏ showed cyanosis and lying on side. All animals in the adjuvant mixture group showed relatively mild symptoms such as rubbing or licking nose, urination, and evacuation. In the passive cutaneous anaphylaxis test, 0/5, 3/5, and 5/5 serum samples from the animals immunized with 0, 2, and 20 ㎍/㎏, respectively, showed positive reactions against HM10760. All 5 sera collected from the animals immunized with an adjuvant mixture contained HM10760-specific antibodies. These results suggest HM10760 have antigenicity in guinea pigs.
Poly-adenine-Coupled LAMP Barcoding to Detect <i>Apple Scar Skin Viroid</i>

<P><I>Apple Scar Skin Viroid</I> (ASSVd), a nonprotein coding, circular RNA pathogen is relatively difficult to detect by immunoassay. We report here a one-step reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay to improve selectivity for diagnostic use in detecting ASSVd in plants. ASSVd RT-LAMP was accelerated using loop primers and was found to be highly sensitive with a detection limit of 10<SUP>4</SUP> copies of cDNA-ASSVd within 30 min. Real-time LAMP and melting curve analysis could differentiate between the true-positive LAMP amplicons and false-positive nonspecific primer amplification products. The optimized RT-LAMP was then followed by the addition of nonthiolated AuNP:poly-adenine (A10)-ASSVd LAMP barcodes, showing a high authentication capacity with colorimetric changes. This type of barcoding assay is a potential alternative for rapid and multiple viroid diagnosis, providing for visible sensing in the field that can be applied to viroid-free planting.</P> [FIG OMISSION]</BR>
