        원격장 와전류 배관 탐상 시스템 개발

        배관의 관벽투과 특성을 지닌 원격장 와전류 비파괴 검사법은 열교환기 세관 검사에서부터 천연가스 수송관에 이르기까지 다양한 형태의 배관과 결함을 대상으로 적용 연구가 진행되고 있다. 공칭 외경 100㎜의 주철관은 주로 상수도 관로의 배수관으로 활용되고 있는데, 차량 진동과 부식에 의한 결합의 생성으로 정수의 누수 문제를 일으킨다. 그러나, 탐상 경제성 및 효용성 문제로 관내 검사장치 투입과 같은 직접적인 검사 방법의 수행이 제한되어 왔다. 원격장 와전류 방법은 시스템 구성이 용이하고, 내·외부 부식 결함에 대한 정량적인 평가가 가능하기 때문에 경제성 있는 장치 개발이 가능하다. 본 연구에서는 원격장 와전류 기술을 이용하여 상수도 관망의 특성을 고려한 매설 배관 검사시 스템 개발을 수행하였다. 세부적으로는 관내 투입 원격장 와전류 검사의 기구부 설계 및 제작, 원격장 와전류 신호의 획득 및 처리 프로그램 개발, 그리고 개발 장치의 운용 방법 및 절차 검토 등으로 되어 있다. Remote field eddy current testing (RFECT) with through-wall transmission characteristic is being applied to pipes ranging from small tubes of heat exchanger to natural gas supply pipelines. Cast iron popes with nominal diameter of 100㎜ are used primarily as the waterline pipes. The leakage of water occurs due to defects in the pipes caused by vibration of automobiles and corrosion. But, the use of direct inspection methods such as insertion of inspection equipment inside the pipelines has been limited due to its lack of economical efficiency. Economical development of inspection equipments is possible since RFECT method can be easily employed for system integration and quantitative evaluation of both inside and outside defects. In this study, the development of underground pipeline inspection system was carried out by using RFECT method in consideration of the characteristics of waterline network. This paper specifically describes the design and production of RFECT pipeline inspection pig using centralizer mechanism, development of remote field eddy current signal acquisition and processing software, and review of RFECT system operation procedures.

        배관 결함부 원거리장 와전류 신호 정량화 연구

        다양한 기계 가공 결함을 유기한 공칭 외경 100mm의 덕타일 주철관을 대상으로 원거리장 와전류 검사를 수행하였다. 상수도 배수관으로 사용되는 주철관은 제조 단계의 큰 허용 오차 때문에 두께가 일정하지 않고 단면이 비대칭적인 특징을 지니므로, 원거리장 와전류 탐상 신호에는 배관 축방향에 걸친 장범위 잡음이 존재하게 된다. 본 논문에서는 장범위에 걸친 배경잡음을 효과적으로 제거하기 위하여 이동구간 평균법을 응용하였고, 결함의 깊이와 원주방향 정도를 정량적으로 평가하기 위하여 voltage plane 극좌표(VPPP) 방법을 활용하였다. 이동구간 평균처리로서 신호비를 일차적으로 향상시킨 다음 VPPP 기법을 이용하여 결함신호를 얻은 결과, 결함신호가 VPPP상의 기준점에서 x 축과 이루는 각이 결함 깊이와 직선적인 상관 관계가 있음을 확인하였다. 따라서 배관과 동축으로 놓인 탐촉코일을 이용한 원거리장 와전류 검사로서 매설 상수도 배관 내ㆍ외부에 존재하는 부식결함을 비파괴 정량평가할 수 있는 기반을 구축하였다. The remote field eddy current (RFEC) inspection was performed on the ductile cast iron pipes with nominal outer diameter of 100mm, which were machined with various shapes and sizes of defects. Ductile cast iron pipes which are used as water supply pipe have the non-uniform thickness and asymmetric cross section due to relatively high degree of allowable errors during the manufacturing processes. These characteristics of ductile cast iron pipes cause the long range background noises in RFEC signals along the pipe. In this study, the machined defects in pipes were effedctively classified by the moving window average (MWA) method which eliminated the long -range noise. The voltage plane polar plots (VPPP) signatures showed method was used to quantitatively evaluate the depth and circumferential degree of defects. The VPPP signatures showed that the angle between defect signature and the naormalized in-phase component on the VPPP is linear to the depth of defects. The nondestructive RFEC technique proved to be capable of quantitatively evaluating the machined defects of underground water supply pipe.

        Pharmacogenetic Impact on Korean Patients Receiving Antiepileptic Drugs

        간질은 가장 흔한 만성 신경 장애로 70% 이상의 환자에서 항전간제로 증상의 조절이 가능하다. 약물의 치료효과를 예측하는데 있어 개인이 가진 유전적 다형성이 부분적인 영향을 주는 것으로 알려지면서 항전간제의 효과에 영향을 주는 유전자 연구가 빠르게 진행되고 있다. 본 연구에서는 83명의 간질 환자를 대상으로 간질 환자의 약물대사와 관련된 것으로 알려진 유전자(CYP2C9, CYP2C19, ABCB1, SCN1A)의 다형성과 약물부작용의 관계에 대해 연구하였다. 연구 결과, 약물 이상 반응과 약물 용량의 상관관계는 통계적으로 유의한 수준은 관찰되지 않았다. 한편, carbamazepine 계열에서의 환자군에서는 SCN1A 유전자의 인트론 유전자 유전형 CC와 CT 유전형에 비해 TT 유전형에서 약물용량과 연관을 보였으며, 500 mg 이상의 용량을 투약한 환자에서는 TT 유전형에 비해 CC와 CT를 가진 유전형에서 통계적으로 유의한 상관성을 보였다. Epilepsy is the most prevalent chronic neurological disorder and can be controlled by antiepileptic drugs (AEDs) in up to 70% of patients. We performed an association study between adverse drug reactions and the genetic polymorphisms of CYP2C9, CYP2C19, ABCB1, and SCN1A. The clinical data of 83 epilepsy patients who had received AEDs containing carbamazepine (CBZ) were collected. We extracted genomic DNA from peripheral blood and then genotyped CYP2C9 (CYP2C9*2, CYP2C9*3), CYP2C19 (CYP2C19*2, CYP2C19*3), ABCB1 (C3435T), and SCN1A (IVS5N+5 G>A) using direct sequencing. The allele frequencies of CYP2C9*3, CYP2C19*2, CYP2C19*3, ABCB1 (3435C>T), and SCN1A (IVS5N+5 G>A) were 0.93, 0.72, 0.91, 0.61, and 0.55, respectively. Statistically significant differences were indicated from the data obtained. Patients with SCN1A genotype CC or CT were compared with patients with SCN1A genotype TT while using more than 500mg of carbamazepine. We have associated functional polymorphisms with the dose used in regular clinical practice for Korean epilepsy patients who had received antiepileptic drugs (AEDs) containing carbamazepine. For AEDs, we found that one of the SCN1A genotypes is associated with a 500 mg dose. There was no association found with CNS ADR caused by AEDs.

        Wafer-scale colloidal lithography based on self-assembly of polystyrene nanospheres and atomic layer deposition

        <P>This paper reports a facile and robust wafer-scale colloidal lithography using polystyrene nanospheres. This technique is based on a combination of two simple and scalable processes: the self-assembly of polystyrene nanospheres by spin coating and the atomic layer deposition. Patterns, such as two dimensional hole-arrays, can be routinely fabricated over 4 inch diameter wafers owing to high reproducibility of this novel method. The hole size and pitch can be controlled by heat treatment and the use of polystyrene nanospheres with different diameters, respectively. We demonstrate five- to ten-fold enhancement in the photoluminescence of phosphor films by constructing two dimensional SiN<SUB><I>x</I></SUB> photonic crystals on the films using this technique.         Clusterin protects H9c2 cardiomyocytes from oxidative stress-induced apoptosis $via$ Akt/GSK-$3{\beta}$ signaling pathway

        Clusterin is a secretory glycoprotein, which is highly up-regulated in a variety of normal and injury tissues undergoing apoptosis including infarct region of the myocardium. Here, we report that clusterin protects H9c2 cardiomyocytes from $H_2O_2$-induced apoptosis by triggering the activation of Akt and GSK-$3{\beta}$. Treatment with $H_2O_2$ induces apoptosis of H9c2 cells by promoting caspase cleavage and cytochrome c release from mitochondria. However, co-treatment with clusterin reverses the induction of apoptotic signaling by $H_2O_2$, thereby recovers cell viability. The protective effect of clusterin on $H_2O_2$-induced apoptosis is impaired by PI3K inhibitor LY294002, which effectively suppresses clusterin-induced activation of Akt and GSK-$3{\beta}$. In addition, the protective effect of clusterin is independednt on its receptor megalin, because inhibition of megalin has no effect on clusturin-mediated Akt/GSK-$3{\beta}$ phosphoylation and H9c2 cell viability. Collectively, these results suggest that clusterin has a role protecting cardiomyocytes from oxidative stress and the Akt/GSK-$3{\beta}$ signaling mediates anti-apoptotic effect of clusterin.

        Baicalin (baicalein-7-glucuronide) is a flavonoid purified from Scutellaria baicalensis Georgi that has traditionally been used for treatment of hypertension, cardiovascular diseases, and viral hepatitis. In this study, the effects of intestinal microbiota on the pharmacokinetics of baicalin were investigated in normal and antibiotic-pretreated rats following p.o. administration of 100 mg/kg baicalin by using liquid chromatography/ion trap mass spectrometry. When rats were pretreated orally with cefadroxil, oxytetracycline and erythromycin for 3 days to control the number of intestinal bacteria, the pharmacokinetic parameters of oral baicalin were significantly affected by antibiotics: Cmax, T1/2(β), Kel and AUC values were significantly changed compared to those in normal rats. These results indicate that intestinal microbiota might play a key role in the oral pharmacokinetics of baicalin.

        Genetic Polymorphisms of UGT1A and their Association with Clinical Factors in Healthy Koreans

        Glucuronidation by the uridine diphosphateglucuronosy-ltransferase 1A enzymes (UGT1As) is a major pathway for elimination of particular drugs and endogenous substances, such as bilirubin. We examined the relation of eight single nucleotide polymorphisms (SNPs) and haplotypes of the UGT1A gene with their clinical factors. For association analysis, we genotyped the variants by direct sequencing analysis and polymerase chain reaction (PCR) in 218 healthy Koreans. The frequency of UGT1A1 polymorphisms, -3279T>G, -3156G>A, -53 $(TA)_{6>7}$, 211G>A, and 686C>A, was 0.26, 0.12, 0.08, 0.15, and 0.01, respectively. The frequency of -118 $(T)_{9>10}$ of UGT1A9 was 0.62, which was significantly higher than that in Caucasians (0.39). Neither the -2152C>T nor the -275T>A polymorphism was observed in Koreans or other Asians in comparison with Caucasians. The -3156G>A and -53 $(TA)_{6>7}$ polymorphisms of UGT1A were significantly associated with platelet count and total bilirubin level (p=0.01, p=0.01, respectively). Additionally, total bilirubin level was positively correlated with occurrence of the UGT1A9-118 $(T)_{9>10}$ rare variant. Common haplotypes encompassing six UGT1A polymorphisms were significantly associated with total bilirubin level (p=0.01). Taken together, we suggest that determination of the UGT1A1 and UGT1A9 genotypes is clinically useful for predicting the efficacy and serious toxicities of particular drugs requiring glucuronidation.
