      • Collapse-resistant performance of a single-story frame assembly and multi-story sub-frame under an internal column-removal scenario

        To elucidate the differences in the collapse behavior between a single-story beam-column assembly and multi-story frame, two 1/3-scale two-bay composite frames, including a single-story composite beam–column assembly and a three-story composite sub-frame, were designed and quasi-statically tested. The load–displacement responses, failure modes, and internal force development of the two frames were analyzed and compared in detail. Furthermore, the resistance mechanisms of the two specimens were explored, and the respective contributions of different load-resisting mechanisms to the total resistances were quantitatively separated to gain deeper insights. The experimental tests indicated that Vierendeel action was present in the two-dimensional multi-story frames, which led to an uneven internal force distribution among the three stories. The collapse resistance of TSDWA-3S in the flexural stage was not significantly increased by the structural redundancy provided by the additional story, as compared to that of TSDWA-1S. Although the development of the load response was similar in the two specimens at flexural stage, the collapse mechanisms of the multi-story composite frame were much more complicated than those of the single-story beam–column assembly, and the combined action between stories was critical in determining the internal force redistribution and rebalancing of the remaining structure.

        Antioxidant and Immunomodulatory Activities of Polysaccharides from Moxa (Artemisia argyi) Leaf

        The purpose of the current study was to characterize the polysaccharides isolated from the moxa (Artemisia argyi) leaf and investigate their bioactivities. The molecular weights of the purified polysaccharides were at a range from 6,000 to 10,000 Da and consisted mostly of carbohydrates (>90%), mainly the monosaccharide of xylose, arabinose, mannose, and glucose. The isolated polysaccharides were further fractionated to form 4subfractions and all of the subfractions exhibited high antioxidant capability on both hydroxide (IC50 ranging from 8 to 18 μg/mL) and superoxide anion radicals (IC50ranging from 10 to 30 μg/mL). The moxa leaf polysaccharides were also capable of enhancing the Concanavallin A (ConA)-induced T cell proliferation, but not the lipopolysaccharides (LPS)-induced B cell proliferation in the in vitro immunological tests. Furthermore, the polysaccharides also strongly facilitate ConA-induced secretion of interferon-γ (IFN-γ) and interleukin-2 (IL-2) in a dose-dependent manner. The results suggested that the moxa leaf polysaccharides may have potential applications as antioxidants and immune enhancers.

        Functional Definition of LuxS, an Autoinducer-2 (AI-2) Synthase and Its Role in Full Virulence of Streptococcus suis Serotype 2

        Quorum sensing is a widespread chemical communication in response to fluctuation of bacterial population density, and has been implicated into bacterial biofilm formation and regulation of expression of virulence factors. The luxS gene product, S-ribosylhomocysteinase, catalizes the last committed step in biosynthetic pathway of autoinducer 2 (AI-2), a signaling molecule for inter-species quorum sensing. We found a luxS homologue in 05ZYH33, an epidemic strain of Streptococcus suis serotype 2 (SS2) in China. A luxS null mutant (ΔluxS) of 05ZYH33 strain was obtained using an approach of homologous recombination. LuxS was determined to be required for AI-2 production in 05ZYH33 strain of S. suis 2. Inactivation of luxS gene led to a wide range of phenotypic changes including thinner capsular walls, increased tolerance to H_2O_2, reduced adherence capacity to epithelial cells, etc. In particular, loss of LuxS impaired dramatically its full virulence of SS2 in experimental model of piglets, and functional complementation restored it nearly to the level of parent strain. Genome-wide transcriptome analyses suggested that some known virulence factors such as CPS are down-regulated in the ΔluxS mutant, which might in part explain virulence attenuation by luxS deletion. Similarly, 29 of 71 genes with different expression level were proposed to be targets candidate regulated by LuxS/AI-2-dependent quorum sensing.

      • Integrated Biosensor for Rapid and Point-of-Care Sepsis Diagnosis

        <P>Sepsis is an often fatal condition that arises when the immune response to an infection causes widespread systemic organ injury. A critical unmet need in combating sepsis is the lack of accurate early biomarkers that produce actionable results in busy clinical settings. Here, we report the development of a point-of-care platform for rapid sepsis detection. Termed IBS (integrated biosensor for sepsis), our approach leverages (i) the pathophysiological role of cytokine interleukin-3 (IL-3) in early sepsis and (ii) a hybrid magneto-electrochemical sensor for IL-3 detection. The developed platform produces test results within 1 h from native blood samples and detects IL-3 at a sensitivity of <10 pg/mL; this performance is >5-times faster and >10-times more sensitive than conventional enzyme-linked immunoadsorbent assays, the current gold standard. Using clinical samples, we show that elevated plasma IL-3 levels are associated with high organ failure rate and thus greater risk of mortality, confirming the potential of IL-3 as a sepsis diagnostic biomarker. With further system development (<I>e</I>.<I>g</I>., full automation, data security measures) and rigorous validation studies, the compact and fast IBS could be a practical clinical tool for timely diagnosis and proactive treatment of sepsis.</P> [FIG OMISSION]</BR>

      • Multivariate Analysis of the Prognosis of 37 Chondrosarcoma Patients

        Objective: The current study aimedto screen for possible factors which affect prognosis of chondrosarcoma. Methods: Thirty seven cases were selected and analyzed statistically. The patients received surgical treatment at our hospital between December 2005 and March 2008. All of them had complete follow-up data. The survival rates were calculated by univariate analysis using the Kaplan-Meier method and tested by Log-rank. ${\chi}^2$ or Fisher exact tests were carried out for the numeration data. The significant indexes after univariate analysis were then analyzed by multivariate analysis using COX regression model. Based on the literature, factors of gender, age, disease course, tumor location, Enneking grades, surgical approaches, distant metastasis and local recurrence were examined. Results: Univariate analysis showed that there were significant differences in Enneking grades, surgical approaches and distant metastasis related to the patients' 3-year survival rate after surgery (P<0.001). No significant difference was not found in gender, age, disease course, tumor location or local recurrence (P>0.05). Multivariate analysis showed that Enneking grade (P=0.007) and surgical approaches (P=0.010) were independent factors affecting the prognosis of chondrosarcoma, but distant metastasis was not (P=0.942). Conclusion: Enneking grades, surgical approaches and distant metastasis are risk factors for prognosis of chondrosarcoma, among which the former two are independent factors.

      • Ubiquitination of p53 is Involved in Troglitazone Induced Apoptosis in Cervical Cancer Cells

        Peroxisome proliferator-activated receptor gamma (PPAR-${\gamma}$), a ligand-dependent nuclear transcription factor, has been found to widely exist in tumor tissues and plays an important role in affecting tumor cell growth. In this study, we investigated the effect of PPAR-${\gamma}$ on aspects of the cervical cancer malignant phenotype, such as cell proliferation and apoptosis. Cell growth assay, Western blotting, Annexin V and flow cytometry analysis consistently showed that treatment with troglitazone (TGZ, a PPAR-${\gamma}$ agonist) led to dose-dependent inhibition of cervical cancer cell growth through apoptosis, whereas T0070907 (another PPAR-${\gamma}$ antagonist) had no effect on Hela cell proliferation and apoptosis. Furthermore, we also detected the protein expression of p53, p21 and Mdm2 to explain the underlying mechanism of PPAR-${\gamma}$ on cellular apoptosis. Our work, finally, demonstrated the existence of the TGZ-PPAR-${\gamma}$-p53 signaling pathway to be a critical regulator of cell apoptosis. These results suggested that PPAR-${\gamma}$ may be a potential therapeutic target for cervical cancer.

        Influence factors of non-uniform phase transformation in hot stamping process of ultra-high-strength steel sheet

        In the hot stamping process of ultra-high-strength steel sheets, it is a significant issue to reveal the nonuniformity of cooling rate in space–time domain by finite element method and even uncover the inner cause, which contributes to the further adjustment of the phase transformation. In this work, a series of heat transfer experiments between dies and sheets were conducted on the self-developed experimental apparatus. The temperature evolution curves of die and BR1500HS ultra-high-strength steel sheets under different pressures and holding time were obtained. Moreover, the transient heat transfer coefficients (HTC) under different mean interface temperatures and pressures were calculated by the inverse heat transfer algorithm. Subsequently, based on the HTC curves, a thermal–mechanical-phase dynamic coupling finite element model was developed for modeling the hot stamping process, and a series of simulations for analyzing the non-uniform microstructures distribution in hot stamping parts were implemented. Finally, the simulation results were validated by actual hot stamping experiments. Two significant influence factors on the nonuniform distribution of microstructures were summarized as follows: the existence of incomplete contact between steel sheets and dies due to the sheet thickness reduction in sidewall and circular bead regions, and the temperature differences between dies and steel sheets.

        Risk Factors for Anxiety in Major Depressive Disorder Patients

        Objective: To analyze the sociodemographic and clinical factors related to anxiety in patients with major depressive disorder (MDD). Methods: This study involved a secondary analysis of data obtained from the Diagnostic Assessment Service for People with Bipolar Disorders in China (DASP), which was initiated by the Chinese Society of Psychiatry (CSP) and conducted from September 1, 2010 to February 28, 2011. Based on the presence or absence of anxiety-related characteristics, 1,178 MDD patients were classified as suffering from anxious depression (n=915) or non-anxious depression (n=263), respectively. Results: Compared with the non-anxious group, the anxious-depression group had an older age at onset (t=−4.39, p<0.001), were older (t=−4.69, p<0.001), reported more lifetime depressive episodes (z=−3.24, p=0.001), were more likely to experience seasonal depressive episodes (χ2=6.896, p=0.009) and depressive episodes following stressful life events (χ2=59.350, p <0.001), and were more likely to have a family history of psychiatric disorders (χ2=6.091, p=0.014). Their positive and total scores on the Mood Disorder Questionnaire (MDQ) and the 32-item Hypomania Checklist (HCL-32) (p<0.05) were also lower. The logistic regression analysis indicated that age (odds ratio [OR]=1.03, p<0.001), a lower total MDQ score (OR=0.94, p=0.011), depressive episodes following stressful life events (OR=3.04, p<0.001), and seasonal depressive episodes (OR=1.75, p=0.039) were significantly associated with anxious depression. Conclusion: These findings indicate that older age, fewer subclinical bipolar features, an increased number of depressive episodes following stressful life events, and seasonal depressive episodes may be risk factors for anxiety-related characteristics in patients with MDD.

        Adsorption of Lead(II) from Aqueous Solution by Two Modified Porous Chelating Resins Based on (3-Mercaptopropyl) Trimethoxysilane

        Two novel types of thiol-based chelating resins were fabricated by modifying poly(2-hydroxyethyl methacrylate- co-glycerol dimethacrylate) (P(HEMA-GDMA)) and poly(2-hydroxyethyl methacrylate-co-ethylene glycol dimethacrylate) (P(HEMA-EGDMA)) with (3-mercaptopropyl) trimethoxysilane(MPS), and used to adsorb lead(II) from aqueous solution. The effects of initial lead(II) concentrations, solution pH, coexisting ions on adsorption of lead(II) were investigated. After modifying, the product P(HEMA-GDMA)-SH has excellent adsorption capacity (118.56 mg/g) for lead(II) at initial lead(II) concentration of 200 mg/L. The resins could easily be regenerated and were reused 6 times without significant loss of adsorption capacity. Besides, the presence of coexisting ions did not markedly influence the adsorption capacities of the resins. The results indicate that the functional thiol-based resins are efficient adsorbents for the removal of toxic lead(II) ions in water treatment.

      • Imatinib Mesylate Versus Allogeneic Hematopoietic Stem Cell Transplantation for Patients with Chronic Myelogenous Leukemia

        Purpose: To compare the relative merits of imatinib and allogeneic hematopoietic stem cell transplantation (allo-HSCT) for chronic myelogenous leukemia (CML). Materials and Methods: This cohort study was designed to compare the outcomes of imatinib (n=292) versus allo-HSCT (n=141) for CML, the clinical data of these patients being retrospectively analyzed so as to compare the event free survival (EFS) and overall survival (OS) between these two groups with patients in the chronic phase (CP) and advanced phases, including accelerate (AP) and blast phases (BP). Results: (1) Patients treated with imatinib (278 in the CP) demonstrated superior EFS, OS, 5-year EFS and 5-year OS rates of 88.5% versus 70.0% (P<0.05), 93.2% versus 80.0% (P<0.05), 84% versus 75.0% (P<0.05) and 92% versus 79.0% (P<0.05), respectively, to those treated with allo-HSCT (120 patients in the CP). (2) Both treatments resulted in similar survival, with EFS and OS rates of 42.9% versus 47.6% (P>0.05), 42.9% versus 57.1% (P> 0.05), respectively, for imatinib (14 patients in the AP and BP) and allo-HSCT (21 patients in the AP and BP). Conclusions: Imatinib confers significant survival advantage (EFS and OS) for CML patients with CP compared with allo-HSCT treatment. However, the outcomes are equally good with both treatments in AP and BP patients.
