        Identification of polymorphic loci in the deiodinase 2 gene and their associations with head dimensions in geese

        Objective: This study was conducted to clone and compare the molecular characteristics of the deiodinase 2 (DIO2) gene between Sichuan White geese and Landes geese, and to analyze the association between polymorphisms of the DIO2 gene and head dimensions in Tianfu meat geese. Methods: The coding sequence of the DIO2 gene was cloned by polymerase chain reaction and vector ligation and aligned by DNAMAN software. A total of 350 Tianfu meat geese were used to genotype the polymorphisms of the DIO2 gene and measure the head dimensions. Association analysis between the polymorphisms of the DIO2 gene and head dimensions was carried out. Results: An 840-bp coding sequence of the DIO2 gene was obtained and comparison analysis identified four polymorphic loci between Sichuan White geese and Landes geese. Further analysis showed that the dominant alleles for the four polymorphic loci were G, G, A, and T and the frequency of the heterozygous genotype was higher than that of the homozygous genotype in Tianfu meat geese. Compared to that in the population of non-knob geese of Tianfu meat geese, the head dimensions in the population of knob geese were significantly higher except for nostril height. However, in the non-knob geese, beak width 1, beak width 2, nostril length, cranial width 1, and maxillary length had significant differences among different genotypes or haplotypes/diplotypes. Conclusion: These results suggested that polymorphisms of the DIO2 gene could be considered molecular markers to select larger heads of geese in the population of non-knob geese.

      • HPV16 CTL Epitope Peptide-activated Dendritic Cell and Natural Killer Co-culture for Therapy of Cervical Cancer in an Animal Model

        There is increasing evidence that natural killer (NK) cells play an important role in antitumor immunity following dendritic cell (DC) vaccination. Little is known, however, about the optimal stimulation of DCs by epitopes and NK interactions for cytotoxicity in tumors. In this study, DC cells activated by the HPV16E7.49-57 epitope and LPS were co-cultured with NK cells in vitro, and then used ot immunize mice to study CTL activity of TC-1, which constitutively expresses HPV16E6E7, with an LDH release assay. Cytotoxicity in mice immunized with DC loaded with epitope HPVE7.49-57 vaccine co-cultured with NK was enhanced significantly (p<0.01). In conclusion, talk-across between DC and NK cells enhances their functions, also improving cytotoxicity againsttumor cells, suggesting that activated DC-NK by epitopes has potential application for cancer-specific immuno-cellular therapy.

      • Calpain-10 SNP43 and SNP19 Polymorphisms and Colorectal Cancer: a Matched Case-control Study

        Objective: Insulin resistance (IR) is an established risk factor for colorectal cancer (CRC). Given that CRC and IR physiologically overlap and the calpain-10 gene (CAPN10) is a candidate for IR, we explored the association between CAPN10 and CRC risk. Methods: Blood samples of 400 case-control pairs were genotyped, and the lifestyle and dietary habits of these pairs were recorded and collected. Unconditional logistic regression (LR) was used to assess the effects of CAPN10 SNP43 and SNP19, and environmental factors. Both generalized multifactor dimensionality reduction (GMDR) and the classification and regression tree (CART) were used to test gene-environment interactions for CRC risk. Results: The GA+AA genotype of SNP43 and the Del/Ins+Ins/Ins genotype of SNP19 were marginally related to CRC risk (GA+AA: OR = 1.35, 95% CI = 0.92-1.99; Del/Ins+Ins/Ins: OR = 1.31, 95% CI = 0.84-2.04). Notably, a high-order interaction was consistently identified by GMDR and CART analyses. In GMDR, the four-factor interaction model of SNP43, SNP19, red meat consumption, and smoked meat consumption was the best model, with a maximum cross-validation consistency of 10/10 and testing balance accuracy of 0.61 (P < 0.01). In LR, subjects with high red and smoked meat consumption and two risk genotypes had a 6.17-fold CRC risk (95% CI = 2.44-15.6) relative to that of subjects with low red and smoked meat consumption and null risk genotypes. In CART, individuals with high smoked and red meat consumption, SNP19 Del/Ins+Ins/Ins, and SNP43 GA+AA had higher CRC risk (OR = 4.56, 95%CI = 1.94-10.75) than those with low smoked and red meat consumption. Conclusions: Though the single loci of CAPN10 SNP43 and SNP19 are not enough to significantly increase the CRC susceptibility, the combination of SNP43, SNP19, red meat consumption, and smoked meat consumption is associated with elevated risk.

      • Functional regression with dependent error and missing observation in reproducing kernel Hilbert spaces

        In this paper, we focus on the partial functional linear model with missing observation, which allows the responses or part of the covariates or responses and part of the covariates simultaneously missing at random, where the regression error is a linear process deduced by not necessarily independent random variables. Under the reproducing kernel Hilbert space setting, we construct the estimators of the slope parameter and coefficient function in the model based on the inverse probability weighting methods, and establish their asymptotic normality and weak convergence with rates, respectively. Meanwhile, the penalized estimator of the parameter is defined by the SCAD penalty and its oracle property is investigated. In addition, we construct a test statistic to check a linear hypothesis of the nonzero parameters and discuss its asymptotic distribution. Simulation study and real data analysis are conducted to investigate the finite sample performance of the proposed methods.

        Measurement of undesirable neutron spectrum in a 120 MeV linac

        Photoneutron background spectroscopy observations at linac are essential for directing accelerator shielding and subtracting background signals. Therefore, we constructed a Bonner Sphere Spectrometer (BSS) system based on an array of BF3 gas proportional counter tubes. Initially, the response of the BSS system was simulated using the MCNP5 code. Next, the response of the system was calibrated by using neutrons with energies of 2.86 MeV and 14.84 MeV. Then, the system was employed to measure the spectrum of the 241AmeBe neutron source, and the results were unfolded by using the Gravel and EM algorithms. Using the validated system, the undesirable neutron spectrum of the 120 MeV electron linac was finally measured and acquired. In addition, it is demonstrated that the equivalent undesirable neutron dose at a distance of 3.2 m from the linac is 19.7 mSv/h. The results measured by the above methods could provide guidance for linac-related research.

        PL Property of Al-N Codoped p-type ZnO Thin Films Fabricated by DC Magnetron Sputtering

        High-quality Al-N doped p-type ZnO thin films were deposited on Si and buffer layer/Si by DC magnetron sputtering in a mixture of N2 and O2 gas. The target was ceramic ZnO mixed with Al2O3 (2 wt%). The p-type ZnO thin films showed a carrier concentration in the range of 1.5x1015~2.93x1017 cm-3, resistivity in the range of 131.2~2.864 Ωcm, mobility in the range of 3.99~31.6 cm2V-1s-1, respectively. It was easier to dope p-type ZnO films on Si substrates than on buffer layer/Si. The film grown on Si showed the highest quality of photoluminescence (PL) characteristics. The Al donor energy level depth (Ed) of Al-N codoped ZnO films was reduced to about 50 meV, and the N acceptor energy level depth (Ea) was reduced to 63 meV.

        Transcriptional Analysis of 10 Selected Genes in a Model of Penicillin G Induced Persistence of Chlamydophila psittaci in HeLa Cells

        Chlamydophila psittaci is an important intracellular pathogen. Persistent infection is an important state of the host-parasite interaction in this chlamydial infection, which plays a significant role in spreading the organism within animal populations and in causing chronic chlamydiosis and serious sequelae. In this study, a C. psittaci persistent infection cell model was induced by penicillin G, and real-time quantitative PCR was used to study the transcriptional levels of 10 C. psittaci genes (dnaA, dnaK, ftsW, ftsY, grpE, rpsD, incC, omcB, CPSIT_0846, and CPSIT_0042) in acute and penicillin-G-induced persistent infection cultures. Compared with the acute cultures, the penicillin-G-treated cultures showed a reduced chlamydial inclusion size and a significantly decreased number of elementary body particles. Additionally, some enlarged aberrant reticulate body particles were present in the penicillin- G-treated cultures but not the acute ones. The expression levels of genes encoding products for cell division (FtsW, FtsY) and outer membrane protein E encoding gene (CPSIT_0042) were downregulated (p < 0.05) from 6 h post-infection onward in the persistent infection cultures. Also from 6 h post-infection, the expression levels of DnaA, DnaK, IncC, RpsD, GrpE, and CPSIT_0846 were upregulated (p < 0.05); however, the expression level of OmcB in the persistent infection was almost the same as that in the acute infection (p > 0.05). These results provide new insight regarding molecular activities that accompany persistence of C. psittaci, which may play important roles in the pathogenesis of C. psittaci infection.

        Analysis of Photoluminescence for N-doped and undoped p-type ZnO Thin Films Fabricated by RF Magnetron Sputtering Method

        N-doped ZnO thin films were deposited on n-type Si(100) and homo-buffer layer, and undoped ZnO thin film was also deposited on homo-buffer layer by RF magnetron sputtering method. After deposition, all films were in-situ annealed at 800 oC for 5 minutes in ambient of O2 with pressure of 10 Torr. X-ray diffraction shows that the homo-buffer layer is beneficial to the crystalline of N-doped ZnO thin films and all films have preferable c-axis orientation. Atomic force microscopy shows that undoped ZnO thin film grown on homo-buffer layer has an evident improvement of smoothness compared with N-dope ZnO thin films. Hall-effect measurements show that all ZnO films annealed at 800 oC possess p-type conductivities. The undoped ZnO film has the highest carrier concentration of 1.145x1017 cm-3. The photoluminescence spectra show the emissions related to FE, DAP and many defects such as VZn, ZnO, Oi and OZn. The p-type defects (Oi, VZn, and OZn) are dominant. The undoped ZnO thin film has a better p-type conductivity compared with N-doped ZnO thin film.

        Fabrication of Stiffness Gradient Nanocomposite Hydrogels for Mimicking Cell Microenvironment

        It has attracted much attention to develop a gradient hydrogel for real mimic of the extracellular matrix. Despite simplicity and ease of electrophoresis preparation method, some drawbacks limit the application of the method including toxic organic crosslinker residual, specific ionic monomer types, as well as narrow gradient gaps. In this work, we successfully prepared gradiently crosslinked nonionic PNIPAm/Laponite nanocomposite (NC) hydrogels with controllable gradient structures by adjusting the intensity of the direct current electric field. The results showed that the stiffness, surface property and thermosensitivity of the hydrogel changed with gradient distributions of Laponite. The average stiffness gradients varied from 0.18 kPa/mm, 0.41 kPa/mm to 1.28 kPa/mm by simply adjusting the intensity of the direct current electric field from 0.13 V/mm, 0.20 V/mm to 0.27 V/mm accordingly. The stiffness reached in a range of 13.8~33.0 kPa. The growth behaviors of human cervical cancer cells (HeLa cells) on the gradient NC hydrogel surface were discussed. Spreading and proliferation, as well as morphology of HeLa cells were closely related to the stiffness of the gradient NC hydrogels. This work develops a stiffness-controllable gradient NC hydrogel and opens up a new insight into the better understanding of cells behaviors in different tissues and organs in vivo.
