        Characteristics of Cholesterol-lowering Lactobacillus casei subsp. casei strain GL-03 Isolated from Cheese

        Cholesterol-lowering effect of lactic acid bacteria iswell-known. In the present study, nine cholesterol-loweringLactobacillus strains from Chinese traditional cheese, pickle, andyoghurt were screened and characterized for their potential use. The microbial contents of all strains significantly decreased at pH1.5; however, the residual counts of Lactobacillus casei subsp. casei GL-03, L. plantarum ZP-Z, L. plantarum ZP-05, and L. brevis ZP-04 were more than 107 CFU/mL after incubation for 6h. All nine strains of Lactobacillus indicated good tolerance to bileat concentration less than 0.2% after incubation for 2 to 6 h. L. plantarum ZP-W had maximum hydrophobicity towards xylene,whereas GL-03 strain possessed maximum hydrophobicity forboth hexadecane and octane. ZP-05 strain had more effectiveinhibitory activity against both Staphylococcus aureus and Bacillussubtilis than other eight strains. Furthermore, GL-03 strainsignificantly reduced cholesterol TC and TG levels in hyperlipidemiamice fed high-cholesterol diet. The growth of GL-03 strain waspromoted by five kinds of Chinese herbal medicines, and theChinese hawthorn at concentration of 0.0125% showed thehighest promoting effect. These results suggest that L. casei subsp. casei GL-03 may be effective as a probiotic with cholesterollowingactivities.

        Astilbin alleviates sepsis-induced acute lung injury by inhibiting the expression of macrophage inhibitory factor in rats

        Sepsis is a systemic inflammatory responsesyndrome caused by severe infections. Astilbin is a dihydroflavonolderivative found in many medicinal and foodplants with multiple pharmacological functions. To investigatethe effects of astilbin on sepsis-induced acute lunginjury (ALI), cecal ligation and puncture was performed onrats to establish a sepsis-induced ALI model; these ratswere then treated with astilbin at different concentrations. Lung injury scores, including lung wet/dry ratio, proteinleakage, myeloperoxidase activity, and inflammatory cellinfiltration were determined to evaluate the effects ofastilbin on sepsis-induced ALI. We found that astilbintreatment significantly attenuates sepsis-induced lunginjury and improves survival rate, lung injury scores, lungwet/dry ratio, protein leakage, myeloperoxidase activity,and inflammatory cell infiltration. Astilbin treatment alsodramatically decreased the production of inflammatorycytokines and chemokines in bronchoalveolar lavage fluid. Further, astilbin treatment inhibited the expression andproduction of macrophage inhibitory factor (MIF), whichinhibits the inflammatory response. Collectively, these datasuggest that astilbin has a protective effect against sepsisinducedALI by inhibiting MIF-mediated inflammatoryresponses. This study provides a molecular basis for astilbinas a new medical treatment for sepsis-induced ALI.

      • Protein-based soft micro-optics fabricated by femtosecond laser direct writing

        <P>In this work, we report a novel soft diffractive micro-optics, called 'microscale kinoform phase-type lens (micro-KPL)', which is fabricated by femtosecond laser direct writing (FsLDW) using bovine serum albumin (BSA) as building blocks and flexible polydimethylsiloxane (PDMS) slices as substrates. By carefully optimizing various process parameters of FsLDW (e. g., average laser power density, scanning step, exposure time on a single point and protein concentration), the as-formed protein micro-KPLs exhibit excellent surface quality, well-defined three-dimensional (3D) geometry and distinctive optical properties, even in relatively harsh operation environments (for instance, in strong acid or base). Laser shaping, imaging and other optical performances can be easily achieved. More importantly, micro-KPLs also have unique flexible and stretchable properties as well as good biocompatibility and biodegradability. Therefore, such protein hydrogel-based micro-optics may have great potential applications, such as in flexible and stretchable photonics and optics, soft integrated optical microsystems and bioimplantable devices.</P>

        Effectiveness and safety of acupuncture for angina pectoris: An overview of systematic reviews

        Background: The number of systematic reviews or meta-analyses (SRs/MAs) on the effectiveness of acupuncture for angina pectoris (AP) is increasing. Due to the inconsistent conclusions and unknown quality of these SRs/MAs, this overview aimed to systematically evaluate and synthesize the existing SRs/MAs, attempting to provide more reliable evidence for the effectiveness and safety of acupuncture in the treatment of AP. Methods: SRs/MAs were searched via eight databases from inception to March 14, 2022. The risk of bias was evaluated using the Risk of Bias in Systematic reviews (ROBIS) tool. The quality of the methodology, reporting, and evidence were assessed by the Assess Systematic Reviews 2 (AMSTAR-2), the Preferred Reporting Item for Systematic Review and Meta-analysis for Acupuncture (PRISMA-A), and the Grading of Recommendations Assessment, Development, and Evaluation (GRADE) system, respectively. Results: Sixteen SRs/MAs were included and fifteen SRs/MAs were considered being of critically low quality according to AMSTAR-2. Only three SRs/MAs were rated at low risk of bias. No study reported all the items listed in the PRISMA-A checklist. No high-quality evidence with GRADE assessment was found. With the low-quality evidence, acupuncture combined with other interventions was superior to monotherapy (medications or Chinese medicine) in the angina symptom and electrocardiogram recovery. No adverse effects owing to acupuncture were reported. Conclusions: Owing to the lack of high-quality evidence provided by the current SRs/MAs, the effective-ness of acupuncture for AP still warrants further proof. Further researches with more critical design and methodology are needed for providing more convincing evidence.       • ANXA2 Regulates the Behavior of SGC-7901 Cells

        ANXA2, a member of the annexin family, is overexpressed and plays important roles in tumor development. However, the significance of ANXA2 expression in gastric carcinoma has not been clarified.To elucidate its roles in growth of gastric cancer, ANXA2 expression in SGC-7901 cells was inhibited with a designated siRNA, then cell proliferation, cell cycling, apoptosis and motility were determined by MTT assay, flow cytometry, Hoechst 33342 staining and wound healing assay, respectively. To further assess the behavior of ANXA2 deleted SGC-7901 cells, changes of microstructures were observed under fluorescence microscopy, laser scanning confocal microscopy and electron microscopy. We found that inhibition of ANXA2 expression caused cell proliferation to decrease significantly with G1 arrest, motility to be reduced with changes in pseudopodia/filopodia structure and F-actin and ${\beta}$-tubulin expression, and apoptosis to be enhanced albeit without significance. At the same time, ANXA2 deletion resulted in fewer pseudopodia/filopodia, non-stained areas were increased, contact inhibition among cells reappeared, and expression of F-actin and ${\beta}$-tubulin was decreased, with induction of polymerized disassembled forms. Taken together, these data suggest that ANXA2 overexpression is important to maintain the malignancy of cancer cells, and this member of the annexin family has potential to be considered as a target for the gene therapy of gastric carcinoma.

        Tetrabromobisphenol A (TBBPA) generation and removal paths analysis in printed circuit board (PCB) industrial wastewater: Lab-scale investigations

        With the rapid development of printed circuit board (PCB) industry, the problem of the release of tetrabromobisphenol A (TBBPA) during PCB manufacturing processes was recognized and harmful to the receiving water body. In this study, TBBPA generation paths, fates in industrial wastewater in the PCB manufacturing processes were analyzed. Gas chromatograph-mass spectrometer (GC-MS) scanning results showed that the raw materials including solder mask ink, character ink and dry film mainly contained TBBPA. The PCB fabrication procedures, mainly including development, etching, stripping of inner layer manufacturing process, and pattern plating, tin-plating of outer layer manufacturing process, and development during the forming and surface treatment process would introduce TBBPA into wastewater. Flocculation and activated sludge process (ASP) that were commonly used for industrial wastewater treatment were selected to assess their removal capability for TBBPA in the wastewater, both which could remove more than 80% TBBPA. Low pH below 7, increased PAC and PAM dosages, and proper dosing during the slow-stirring stage could enhance the TBBPA removal of flocculation-sedimentation process. The ASP removes TBBPA mainly by the path of adsorption. High activated sludge concentration, low pH, medium-temperature and limited competitive adsorbates (NH4+, Cu2+) were beneficial to remove TBBPA by the ASP.

      • Optimal Tracking Control of MISO Nonlinear Systems Based on Multi-dimensional Taylor Network Only by Output Feedback

        The control input item is added to constitute the nonlinear dynamic model, on the basis of the original multi-dimensional Taylor network in this paper. And this nonlinear dynamic model is used to optimally control MISO nonlinear system only by output feedback without the disturbance estimation of the system or needing the state observer. The back-propagation algorithm is used to train the parameters of the multi-dimensional network with the control input item. Through the simulation, it is demonstrated the multi-dimensional Taylor network used as the optimal MISO nonlinear system tracking controller is effective.

        Clinical Profles and Short-Term Outcomes of Acute Disseminated Encephalomyelitis in Adult Chinese Patients

        Background and PurposezzAcute disseminated encephalomyelitis (ADEM) is an infammatory demyelinating disorder that predominantly afects children. Previous studies have mostly involved children in Western developed countries. MethodszzTis study retrospectively reviewed the clinical profles of ADEM in adult Chinese patients. ResultszzADEM occurred during summer and autumn in about two-thirds of the 42 included patients. Prior infection was found in fve patients and no preimmunization was recorded. Te most frequent clinical presentations were alterations in consciousness (79%) and behavior changes (69%), followed by motor defcits (64%) and fever (50%). About one-quarter (26%) of the patients showed positive results for oligoclonal bands, and about half of them exhibited increases in the IgG index and 24-hour IgG synthesis rate. Magnetic resonance imaging showed white- and gray-matter lesions in 83% and 23% of the patients, respectively. Steroids were the main treatment, and full recovery occurred in 62% of the patients, with residual focal neurological defcits recorded in a few patients. Afer a mean follow-up period of 3.4 years, two patients exhibited recurrence and one patient exhibited a multiphasic course. One patient was diagnosed with multiple sclerosis (MS). ConclusionszzWith the exception of the seasonal distribution pattern and prior vaccine rate, the clinical profles of ADEM in adult Chinese patients are similar to those in pediatric populations. No specifc markers are available for distinguishing ADEM from MS at the initial presentation. Careful clinical evaluations, cerebrospinal fuid measurements, and neuroradiological examinations with long-term follow-up will aid the correct diagnosis of ADEM.

      • Inhibition by Imatinib of Expression of O-glycan-related Glycosyltransferases and Tumor-associated Carbohydrate Antigens in the K562 Human Leukemia Cell Line

        Objective: To study changes of tumor associated carbohydrate antigen (TACAs) expression and mRNA levels for tumor associated glycosyltransferases, and assess subcellular localizations of N-acetyl galactosyltransferases (GalNAc-Ts) in the K562 leukemia cell line after imatinib treatment. Methods: RT-PCR was performed to analyze the expression of glycosyltransferases which synthesize O-glycan in tumor-associated carbohydrate antigens (TCTAs). The expression of Tn antigen, T antigen and sialyl T antigen on K562 cell membranes was measured by flow cytometry after treatment with different concentrations of imatinib. Co-localization of GalNAc-Ts and ER (endoplasmic reticulum) was determined by confocal laser scanning microcopy. Results: Transcript expression levels of several glycosyltransferases related to TCTAs were decreased after imatinib ($0-0.3{\mu}M$) treatment. Expression of Tn antigen and T antigen was increased while that of sialyl T antigen was decreased. Co-localization of GalNAc-Ts and ER was reduced by $0.2{\mu}M$ of imatinib. Conclusion: Imatinib inhibited the expression of O-glycan related TACAs and several related glycosyltransferases, while decreasing the co-localization of GalNAc-Ts and ER and normalizing O-glycosylation in the K562 human leukemia cell.
