Design of Distributed Data Handling Architecture for Large-scale Science Data

One of key components for data centric researches on cyber-infrastructures is data handling. Recent researches, like high-energy physics, uses huge-number-of and huge-size-of data. Because the size of data that researchers treat is so huge, they usually use grids and distribute data into several distributed sites. To handle those distribute data efficiently, data handling system should have a robust function for the distribution, management, and service of data. In this paper, we propose a data handling architecture on distributed environment, like grids. The proposed architecture can distribute loads, adapt to user's request, scale in proportion to the number of distributed sites, and provide automatic replication of data.

Preparation of recipient stallions is critical step to produce donor spermatogonial stem cell (SSC) derived sperm using transplantation technique. This study was conducted to evaluate the effects of intravenous busulfan infusion on germ cell depletion, semen production, and libido in stallions. Six Thoroughbred stallions were separated into two treatment groups: 1) a multiple low-dose (2.5 mg/kg bw for the first 4 weeks and 5 mg/kg bw for the 5th week); and 2) control group treated with PBS. Testicular samples were obtained at 11 weeks and classified into three different patterns of spermatogenesis, such as normal, Sertoli cell only, and destroyed. Semen collection and libido experiments were performed 1 week before treatment, and 4 and 8 weeks after treatment. For the sperm analysis, total spermatozoa and motility were measured using a light microscope with a motility analyzing system. In the multiple low-dose group, the numbers of tubules categorized as Sertoli cell only were significantly higher than those in the control as well as the total population and total/progressive motility of sperm were significantly decreased 8 weeks after the start of the treatment. The sperm production and motility in the multiple low-dose group appears to be reduced, while libido was maintained. In conclusion, multiple administration of 2.5 mg/kg bw busulfan depletes endogenous germ cells in the stallion recipients for SSC transplantation.

The spermatogonial stem cell (SSC) transplantation technique has been utilized for the production of donor-derived sperms in the recipient animals. Depletion of the endogenous germ cells in the seminiferous tubules is the key technique for successful SSCs transplantation because it provides a place for transplanted donor germ cells settle in the seminiferous tubules. We previously reported that intra-testicular injection of 70% glycerin partially removed endogenous germ cells, but the effect varied among stallions. The main objective of this study was to find out an optimal dose and injection method by which endogenous germ cells of stallions are successfully depleted without disturbing functions of sperm production and libido. Each group of stallions (n=3) were treated with 1) 15 mg of busulfan/kg of BW, all at once (group 1), 2) 15 mg of busulfan/kg of BW, at 5 different times (group 2), or 3) none (control, group 3). The percentages of survival rate in group 1, 2, and 3 were 33 (1/3), 100 (3/3), and 100% (3/3), respectively. The percentage of Sertoli cell only cross section of seminiferous tubules were significantly higher (p<0.05) in group 2 (78.2±7%) compared to group 3 (0%). The averages of testicular weight (g) between group 2 (143.5±8.5 g) and 3 (179±1.0 g) were not significantly different (p<0.05). In group 2, the number of sperm production and motility (total and progressive) decreased after busulfan treatment. However, libido of stallions was not altered after treatment. In conclusion, weekly split IV injection of busulfan can be utilized to deplete endogenous germ cells for the purpose of preparing recipient stallions for spermatogonial stem cell transplantation.

Depletion of endogenous germ cells in the recipient model is the key process for successful SSC transplantation. The main objectives of this study are 1) to evaluate the effect of local irradiation or testicular infusion of 70% glycerin on depletion of germ cells, 2) to evaluate germ cell transplantation in the goat testes using ultrasound guided rete-testis transplantation technique. The testes of two recipient goats were treated with irradiation with does of 10 or 15 Gy, respectively. Testicular tissue biopsy was performed to analyze patterns of spermatogenesis at 6 weeks after treatment. Fifteen milliters of 70% glycerin was infused into the core of each testes at 1 cc/min. After 4 weeks, germ cells (500×106) stained with PKH-26 dye were transplanted using ultrasound guided rete testis injection technique. Testes were castrated at 2 weeks after transplantation and seminiferous tubules were isolated to monitor the presence of PKH-26 positive germ cells. Spermatogenesis patterns in cross section of seminiferous tubules at each dose of 10 and 15 Gy were categorized and quantified as following; normal (5.1 and 25.3%), Sertoli cell only (35.3 and 11.3%), and destroyed (59.4 and 63.3%). However, abnomal testicular tissue condition was noticed; tissue was not as firm as normal testes as well as effusion was evidenced. The testes treated with 70% glycerin also showed 3 different patterns of normal (19.6%), destroyed (72.8%), and Sertoli cell only (7.6%). In this testis, the severe inflammation area was evidenced. Germ cells stained with PKH-26 were successfully identified within seminiferous tubules. In conclusion, the local radiation and testicular infusion of 70% glycerin are not ideal method for the depletion of endogenous germ cells in the goat testes. The ultrasound guided rete-testis transplantation technique can be utilized.
